Characterization of the osteogenic potential of mesenchymal stem cells from human periodontal ligament based on cell surface markers

Mesenchymal stem cell （MSC）-mediated therapy has been shown to be clinically effective in regenerating tissue defects. For improved regenerative therapy, it is critical to isolate homogenous populations of MSCs with high capacity to differentiate into appropriate tissues. The utilization of stem cell surface antigens provides a means to identify MSCs from various tissues. However, few surface markers that consistently isolate highly regenerative MSCs have been validated, making it challenging for routine clinical applications and making it all the more imperative to identify reliable surface markers. In this study, we used three surface marker combinations： CD51/CD140a, CD271, and STRO-1/CD146 for the isolation of homogenous populations of dental mesenchymal stem cells （DMSCs） from heterogeneous periodontal ligament cells （PDLCs）. Fluorescence-activated cell sorting analysis revealed that 24% of PDLCs were CD51＋/CD140a＋, 0.8% were CD271＋, and 2.4% were STRO-1＋/CD146＋. Sorted cell populations were further assessed for their multipotent properties by inducing osteogenic and chondrogenic differentiation. All three subsets of isolated DMSCs exhibited differentiation capacity into osteogenic and chondrogenic lineages but with varying degrees. CD271＋ DMSCs demonstrated the greatest osteogenic potential with strong induction of osteogenic markers such as DLX5, RUNX2, and BGLAP. Our study provides evidence that surface marker combinations used in this study are sufficient markers for the isolation of DMSCs from PDLCs. These results provide important insight into using specific surface markers for identifying homogenous populations of DMSCs for their improved utilization in regenerative medicine.

Clay minerals in the late Quaternary sediment of Tulare Lake,California: Implications for climate change, weathering, and erosion processes

The clay mineralogy of Tulare Lake sediment was examined to investigate hydroclimatic and environmental changes in the southern Sierra Nevada Mountains(SNM) since the most recent glacial *** of clay mineral assemblages elucidates significant changes in weathering, erosion, and hydroclimatic condition in the catchment. During the last glacial period(24.4–15.1 cal ka BP), low illite content implies less physical erosion of the granitic batholith rocks and a cold and arid environment in the southern SNM. Abrupt increases of illite content at 21.8–20.8 and 17.6 cal ka BP resulted from the glacier advances to the ablation zone and illite-rich glacier flour was transported down to the lake. The gradual increase of smectite induced by progressive depletion of illite-rich glacier flour from 17.6 cal ka BP toward the end of this period indicates climate was beginning to get warm and wet. From 11.9 to5.3 cal ka BP, two warm and wet periods(10.7–9.4 and 8.2–5.2 cal ka BP) were characterized by high smectite/illite content ratios and low illite crystallinity values, suggesting intensive rainfall precipitation and more physical erosion in the highland and lowland catchment as well as more smectite formation in the terrace soils. Since the last glacial period, physical erosion, in comparison to the chemical weathering,was the dominant process responding to the hydroclimatic change in the Tulare Lake *** to weak chemical weathering was signified by the mostly low illite chemical weathering index of the core sediments. Such results suggest that vegetation cover in the southern SNM was low and limited.

A time fractional model to represent rainfall process

This paper deals with a stochastic representation of the rainfall process. The analysis of a rainfall time series shows that cumulative representation of a rainfall time series can be modeled as a non-Gaussian random walk with a log-normal jump distribution and a time-waiting distribution following a tempered a-stable probability law. Based on the random walk model, a fractional Fokker-Planck equation （FFPE） with tempered a-stable waiting times was obtained. Through the comparison of observed data and simulated results from the random walk model and FFPE model with tempered a-stable waiting times, it can be concluded that the behavior of the rainfall process is globally reproduced, and the FFPE model with tempered a-stable waiting times is more efficient in reproducing the observed behavior.

Tropical Arenosols may be challenging for agricultural use,particularly in semi-arid *** aim of this study was to evaluate the impact of the addition of increasing shares of biochar and clay on the nutrient sorption capacity of a tropical *** batch equilibrium experiments,the sorption of ammonium-N(NH_(4)^(+)-N),nitrate-N(NO_(3)^(−)-N),potassium(K^(+)),and phosphate-P(PO_(4)^(3−)-P)was quantified for mixtures of an Arenosol with increasing shares of biochar and clay(1%,2.5%,5%,10%,100%)and the unmixed Arenosol,biochar,and *** mid-temperature biochar was produced from Prosopis juliflora feedstock;the clayey material was taken from the sedimentary parent material of a temporarily dry *** the Arenosol-biochar mixture with 10%biochar addition and the biochar increased the NH_(4)^(+)-N maximum sorption capacity(q_(max))of the Arenosol,by 34%and 130%,*** q_(max)of PO_(4)^(3−)-P slightly increased with ascending biochar shares(1-10%)by 14%,30%,26%,and 42%,whereas the undiluted biochar released PO_(4)^(3−)-*** addition slightly reduced NO_(3)^(−)-N release from the Arenosol but strongly induced K^(+)*** the other hand,clay addition of 10%and clay itself augmented q_(max)of NH_(4)^(+)-N by 30%and 162%;ascending clay rates(1-100%)increased q_(max)for PO_(4)^(3−)-P by 78%,130%,180%,268%,and 712%.Clay rates above 5%improved K^(+)sorption;however,no q_(max)values could be *** of NO_(3)^(−)-N remained unaffected by clay ***,clay addition proved to enhance the nutrient sorption capacity of the Arenosol more effectively than biochar;nonetheless,both materials may be promising amendments to meliorate sandy soils for agricultural use in the semi-arid tropics.

Deprescribing antihypertensive treatment in nursing home patients and the effect on blood pressure

Background It is debatable whether treating multimorbid nursing home patients with antihypertensive drugs produces beneficial effects. Most cardiovascular guidelines promote treatment; few have advice on how to deprescribe when treatment may no longer be necessary. We investigated the effect of medication review on antihypertensive drug use and the association between cognition, blood pressure, and prescribing. Methods From August 2014 to December 2015, 765 patients from 72 units （clusters） in 32 Norwegian nursing homes were included in a 4-month, multicentre, cluster-randomized, controlled trial, with 9-month follow-up. Patients ≥ 65 years old with antihypertensive treatment （n = 295, 39%） were randomized to systematic medication review where the physician received support from peers （collegial mentoring） or were given care as usual （control condition）. Outcome measures were the number of antihypertensive drugs, systolic blood pressure, and pulse. We used hospitalizations and deaths as criteria to assess harm. Results At baseline, each patient used 9.2 ± 3.5 regular drugs, and 1.6 ± 0.7 antihypertensives. Mean blood pressure was 128/71 mmHg and 9% had a systolic pressure ≥ 160 mmHg. Between baseline and month four, antihypertensives were deprescribed to a significantly higher extent in the intervention group （n = 43, 32%） compared to control （n = 11, 10%）; Incidence Rate Ratio = 0.8, 95% CI = 0.7？0.9. In the intervention group, there was an immediate increase in systolic blood pressure when antihypertensives were reduced, from baseline 128 ± 19.5 mmHg to 143 ± 25.5 mmHg at month four. However, at month nine, the blood pressure had reverted to baseline values （mean 134 mmHg）. Deprescription did not affect pulse and systolic pressure. The number of hospitalizations was higher in control patients at month four （P = 0.031） and nine （P = 0.041）. Conclusion A systematic medication review supported by collegial mentoring significantly decreased the use of antihypertensive

Mesenchymal stem cells （MSCs） are characterized by their self-renewing capacity and differentiation potential into multiple tissues. Thus, management of the differentiation capacities of MSCs is important for MSC-based regenerative medicine, such as craniofacial bone regeneration, and in new treatments for metabolic bone diseases, such as osteoporosis. In recent years, histone modification has been a growing topic in the field of MSC lineage specification, in which the Su（var）3-9, enhancer-of-zeste, trithorax （SET） domain-containing family and the Jumonji C （JmjC） domain-containing family represent the major histone lysine methyltransferases （KMTs） and histone lysine demethylases （KDMs）, respectively. In this review, we summarize the current understanding of the epigenetic mechanisms by which SET domain-containine KMTs and JmiC domain-containinlz KDMs balance the osteogenic and adipogenic differentiation of MSCs.

Using Statistical Downscaling to Quantify the GCM-Related Uncertainty in Regional Climate Change Scenarios: A Case Study of Swedish Precipitation

There are a number of sources of uncertainty in regional climate change scenarios. When statistical downscaling is used to obtain regional climate change scenarios, the uncertainty may originate from the uncertainties in the global climate models used, the skill of the statistical model, and the forcing scenarios applied to the global climate model. The uncertainty associated with global climate models can be evaluated by examining the differences in the predictors and in the downscaled climate change scenarios based on a set of different global climate models. When standardized global climate model simulations such as the second phase of the Coupled Model Intercomparison Project （CMIP2） are used, the difference in the downscaled variables mainly reflects differences in the climate models and the natural variability in the simulated climates. It is proposed that the spread of the estimates can be taken as a measure of the uncertainty associated with global climate models. The proposed method is applied to the estimation of global-climate-model-related uncertainty in regional precipitation change scenarios in Sweden. Results from statistical downscaling based on 17 global climate models show that there is an overall increase in annual precipitation all over Sweden although a considerable spread of the changes in the precipitation exists. The general increase can be attributed to the increased large-scale precipitation and the enhanced westerly wind. The estimated uncertainty is nearly independent of region. However, there is a seasonal dependence. The estimates for winter show the highest level of confidence, while the estimates for summer show the least.

On Northern Hemisphere Wave Patterns Associated with Winter Rainfall Events in China

During extended winter （November-April）, 43% of the intraseasonal rainfall variability in China is explained by three spatial patterns of temporally coherent rainfall, These patterns were identified with empirical orthogonal teleconnection （EOT） analysis of observed 1982-2007 pentad rainfall anomalies and connected to midlatitude disturbances. However, ex- amination of individual strong EOT events shows that there is substantial inter-event variability in their dynamical evolution, which implies that precursor patterns found in regressions cannot serve as useful predictors. To understand the physical nature and origins of the extratropical precursors, the EOT technique is applied to six simulations of the Met Office Unified Model at horizontal resolutions of 200-40 km, with and without air-sea coupling. All simulations reproduce the observed precursor patterns in regressions, indicating robust underlying dynamical processes. Further investigation into the dynamics associated with observed patterns shows that Rossby wave dynamics can explain the large inter-event variability. The results suggest that the appaxently slowly evolving or quasi-stationaxy waves in regression analysis are a statistical amalgamation of more rapidly propagating waves with a variety of origins and properties.

An Innovative Method to Accommodate Chinese Medicine Pattern Diagnosis within the Framework of Evidence-Based Medical Research

Pattern diagnosis is an integral aspect of Chinese medicine （CM）. CM differentiates biomedical diseases into patterns, based upon the patient＇s symptoms and signs. Pattern identification （PI） is used to diagnose, direct the treatment principle and determine the treatment protocol. Most CM research has used fixed formula treatments for Western-defined diseases with outcomes measured using objective biomedical markers. This article presents an innovative method used in a randomised controlled pilot study using acupuncture for participants with hepatitis C virus. Each participant＇s CM patterns were identified and quantified at baseline which directed the treatment protocol for the treatment group. Data identified that while each participant expressed different patterns at baseline all participants displayed multiple patterns. Six patterns showed some expression by all 16 participants; Liver （Gan） yin vacuity expressing a group aggregate mean percentage of 47.2, binding depression of Liver qi 46.9, and Liver Kidney （Shen） yin vacuity 45.1. Further sub category gender grouping revealed that pattern ranking changed with gender; Liver yin vacuity （male 53.4%, female 51.93%）, binding depression of Liver qi （male 50.0%, female 42.86%） and Liver Kidney yin vacuity （male 42.9%, female 47.96%）. The quantification of CM patterns described in this article permitted statistical evaluation of presenting CM patterns. Although this methodology is in its infancy it may have potential use in the integration of PI with rigorous evidence based clinical research. Biomedical markers often do not relate to symptom/signs and therefore this innovative measure may offer an additional CM evaluation methodology and further CM PI understanding.

Iterative Factors Favoring Collaboration for Interorganizational Resilience:The Case of the Greater Montréal Transportation Infrastructure

Between members of a network, interorganizational resilience is favored by effective collaboration and coordination during a crisis. The quality of that collaboration depends on various iterative factors present between these organizations before the occurrence of a *** find that these factors are iterative since collaboration factors follow a mutually reinforcing cycle: collaboration within a crisis management network is conditioned by a general agreement, which is in turn conditioned by the extent to which the institutions coordinate themselves prior to crisis. We evaluated the factors that promote collaboration between public and private organizations that manage the Greater Montréal transportation *** factors are based on adaptive management processes such as mutual agreements, common organizational culture, knowledge and financial resources, levers of power,regulations, and pressure. Crisis management coordination represents the ability to build and assess the effectiveness of common response plans to risks to which they are exposed. We show how these processes vary depending on the links between private and public organizations.
