3D assessment of intervertebral disc degeneration in zebrafish identifies changes in bone density that prime disc disease

Back pain is a common condition with a high social impact and represents a global health *** disc disease(IVDD)is one of the major causes of back pain;no therapeutics are currently available to reverse this *** impact of bone mineral density(BMD)on IVDD has been controversial,with some studies suggesting osteoporosis as causative for IVDD and others suggesting it as protective for *** studies to evaluate the influence of genetic components of BMD in IVDD could highlight opportunities for drug development and *** taking a holistic 3D approach,we established an aging zebrafish model for spontaneous *** BMD in aging,detected by automated computational analysis,is caused by bone deformities at the ***,aged zebrafish spines showed changes in bone morphology,microstructure,mineral heterogeneity,and increased fragility that resembled *** of the discs recapitulated IVDD symptoms found in humans:the intervertebral ligament(equivalent to the annulus fibrosus)showed disorganized collagen fibers and herniation,while the disc center(nucleus pulposus equivalent)showed dehydration and cellular *** manipulated BMD in young zebrafish by mutating sp7 and cathepsin K,leading to low and high BMD,***,we detected IVDD in both groups,demonstrating that low BMD does not protect against IVDD,and we found a strong correlation between high BMD and *** learning was applied to high-resolution synchrotron\iCJ image data to analyze osteocyte 3D lacunar distribution and morphology,revealing a role of sp7 in controlling the osteocyte lacunar 3D *** findings suggest potential avenues through which bone quality can be targeted to identify beneficial therapeutics for IVDD.

Abnormal subchondral bone remodeling leading to sclerosis is a main feature of osteoarthritis(OA), and osteomodulin(OMD), a proteoglycan involved in extracellular matrix mineralization, is associated with the sclerotic phenotype. However, the functions of OMD remain poorly understood, specifically in vivo. We used Omd knockout and overexpressing male mice and mutant zebrafish to study its roles in bone and cartilage metabolism and in the development of OA. The expression of Omd is deeply correlated with bone and cartilage microarchitectures affecting the bone volume and the onset of subchondral bone sclerosis and spontaneous cartilage lesions. Mechanistically, OMD binds to RANKl and inhibits osteoclastogenesis, thus controlling the balance of bone remodeling. In conclusion, OMD is a key factor in subchondral bone sclerosis associated with OA. It participates in bone and cartilage homeostasis by acting on the regulation of osteoclastogenesis. Targeting OMD may be a promising new and personalized approach for OA.

The age and stroke-associated decline in musculoskeletal strength degrades the ability to perform daily human tasks using the upper ***,we introduce an intelligent upper-limb exoskeleton system that utilizes deep learning to predict human intention for strength *** embedded soft wearable sensors provide sensory feedback by collecting real-time muscle activities,which are simultaneously computed to determine the user’s intended *** deep learning predicts four upper-limb joint motions with an average accuracy of 96.2%at a 500–550 ms response rate,suggesting that the exoskeleton operates just by human *** addition,an array of soft pneumatics assists the intended movements by providing 897 newtons of force while generating a displacement of 87mm at *** intent-driven exoskeleton can reduce human muscle activities by 3.7 times on average compared to the unassisted exoskeleton.

Understanding how plants respond to nitrogen in their environment is crucial for determining how they use it and how the nitrogen use affects other processes related to plant growth and development. Under nitrogen limitation the activity and affinity of uptake systems is increased in roots, and lateral root formation is regulated in order to adapt to low nitrogen levels and scavenge from the soil. Plants in the legume family can form associations with rhizobial nitrogen-fixing bacteria, and this association is tightly regulated by nitrogen levels. The effect of nitrogen on nodulation has been extensively investigated, but the effects of nodulation on plant nitrogen responses remain largely unclear. In this study, we integrated molecular and phenotypic data in the legume Medicago truncatula and determined that genes controlling nitrogen influx are differently expressed depending on whether plants are mock or rhizobia inoculated. We found that a functional autoregulation of nodulation pathway is required for roots to perceive, take up, and mobilize nitrogen as well as for normal root development. Our results together revealed that autoregulation of nodulation, root development, and the location of nitrogen are processes balanced by the whole plant system as part of a resource-partitioning mechanism.

Is Business School Performance Impacted by Market Orientation Toward Students, Employers of Students, and Parents of Students？ Views from Accounting Department Chairpersons, Business School Deans, and Academic Vice-Presidents of AACSB Business Schools

This paper is part of a continuing research stream dealing with organizational behavior and performance in higher education, specifically within AACSB-Intemational business schools. Using responses to a national survey sent to AACSB-International members schools located in the United States, we report market orientation levels toward students, parents of students, and employers of students as well as levels of organizational performance reported by accounting department chairpersons, business school deans, and academic vice-presidents. Theory and empirical research suggest that higher levels of market orientation result in higher levels of organizational performance. Comparisons of the various input scores for each customer group （students, parents of students, and employers of students） submitted by the survey respondents are made against a benchmark established for businesses in the marketing literature and then scores are compared by administrative groups against one another. Finally, regression analysis is used to determine if the reported levels of organizational performance are impacted by the levels of reported market orientation. Altogether, 101 accounting department chairpersons, 130 business school deans, and 110 academic vice-presidents responded. The paper presents details of the research process, findings, statistical inferences, and discusses the implications of the research for schools of business and academic accounting departments.

Heat shock protein 90 inhibition by 17-DMAG lessens disease in the MRl/Ipr mouse model of systemic lupus erythematosus

Elevated expression of heat shock protein 90 （HSP90） has been found in kidneys and serum of systemic lupus erythematosus （SlE） patients and MRlIMp-FasIprIFasJpr（MRlIIpr） autoimmune mice. We investigated if inhibition of HSP90 would reduce disease in MRl/ Ipr mice. In vitro, pretreatment of mesangial cells with HSP90 inhibitor Geldanamycin prior to immune-stimulation showed reduced expression of Il-6, Il-12 and NO. In vivo, we found HSP90 expression was elevated in MRl/Ipr kidneys when compared to C57Bl/6 mice and MRIJIpr mice treated with HSP90 inhibitor 17-DMAG. MRIJIpr mice treated with 17-DMAG showed decreased proteinuria and reduced serum anti-dsDNA antibody production. Glomerulonephritis and glomerular IgG and C3 were not significantly affected by administration of 17-DMAG in MRIJIpr. 17-DMAG increased CD8＋ T cells, reduced double-negative T cells, decreased the CD4/CD8 ratio and reduced follicular B cells. These studies suggest that HSP90 may play a role in regulating T-cell differentiation and activation and that HSP90 inhibition may reduce inflammation in lupus.

Nitrogen molecules Nx have been the subject of much recent research because of their potential as high-energy materials. Many nitrogen molecules dissociate with very low barriers, including molecules such as acyclic N4 that are essentially unbound. A number of studies have reported the ability of heteroatoms to stabilize complex nitrogen molecules. In the present study, the energetic and electronic properties of scandium（Ⅰ） and titanium（Ⅱ） complexes with N2 and N4 are calculated and discussed. Dissociation energies and singlet-triplet energies are determined by theoretical calculations using second-order perturbation theory （MP2） in conjunction with the Dunning basis sets.

Improved Interpretive Accuracy of Sugar Content on Beverage labels Following an Educational Intervention for Children

To address childhood obesity, it is important to help children make better choices with regards to intake of sugary drinks and snacks. The purpose of this study was to evaluate the validity of a novel educational intervention designed to convert grams of sugar into teaspoons on labeled beverages. A convenience sample of children （n = 21） at a charter elementary school in Washington, D. C was studied. A pre-test required an estimation of teaspoons of sugar on labels, followed by a practice demonstration and song. A post-test was given requiring estimation of teaspoons of sugar in on the same labels. Two weeks later, a retention test as well as qualitative questions assessed participant learning and information usage. Test scores revealed increased accuracy from pre-test （26.2%） to post-test （79.8%）. Estimations were more accurate after the demonstration, as evidenced by the decreased range in average variation of responses （＋25.6 tsp to ＋1.8 tsp）. Two-week recall maintained accuracy （66.7%）, with an average variance of ＋3.8 teaspoons. Qualitative responses revealed participants had learned and used the conversion concepts. These results suggest that the educational intervention was effective. Qualitative data suggest behavior change, increased awareness, and the potential for change extending to caregivers.
