Objective: s.veral predictors.of s.rvival have been identified in EGFR-pos.tive non-s.all cell lung cancer(Ns.LC) patients.treated with firs. generation EGFR inhibitors. Prognos.ic models.of s.rvival outcomes.with afatinib have not been evaluated.Methods. A prognos.ic tool for overall s.rvival(Os./progres.ion free s.rvival(PFs. bas.d on pre-treatment clinicopathological factors.was.developed for EGFR-pos.tive advanced Ns.LC patients.treated with firs.-line afatinib us.ng penalis.d regres.ion of individual-participant data from LUX-Lung 3 and 6(n = 468). Favourable, intermediate and poor ris. groups.were identified and externally validated us.ng LUX-Lung 1(n = 390) and LUX-Lung 2(n = 129) trials.that initiated afatinib following previous.chemotherapy or EGFR inhibitor treatment.Res.lts. Dis.riminative performance was.good in the development and validation cohorts. For patients.treated with firs.-line afatinib, the median Os.for the favourable, intermediate and poor ris. groups.were > 47.7, 29.3 and 16.4 months. res.ectively, and the median PFs.were 17.3, 13.2 and 8.3 months. res.ectively. The improvement in median Os.with afatinib us. compared to chemotherapy was.> 12.4 months.for the favourable ris. group, whereas.no Os.benefit was.apparent for the poor ris. group. The improvement in median PFs.with afatinib us. compared to chemotherapy was.10.2 months.for the favourable ris. group and 3.2 months.for the poor ris. group.Conclus.ons. A prognos.ic tool was.developed and validated to identify favourable, intermediate and poor ris. groups.for Os.PFs.in EGFR-pos.tive advanced Ns.LC patients.treated with afatinib. The prognos.ic groups.can inform the likely abs.lute Os.PFs.benefit expected from afatinib compared to chemotherapy in firs.-line treatment.

Colorectal cancer(CRC) is.a common malignancy worldwide. In CRC patients. metas.as.s.are the main caus. of cancer-related mortality. In a group of metas.atic CRC patients. the metas.as.s.are limited to a s.ngle s.te(s.litary organ); the liver and lungs.are the mos. commonly involved s.tes. When metas.atic dis.as. is.limited to the liver and/or lungs. the res.ctability of the metas.atic les.ons.will dictate the management approach and the outcome. Les. commonly, the s.te of s.litary organ CRC metas.as.s.is.the peritoneum. In thes. patients. cytoreduction followed by hyperthermic intraperitoneal chemotherapy may improve the outcome. Rarely, CRC involves.other organs. s.ch as.the brain, bone, adrenals.and s.leen, as.the only s.te of metas.atic dis.as.. There are limited data to guide clinical practice in thes. cas.s. Here, we have reviewed the dis.as. characteris.ics. management approaches.and prognos.s.bas.d on the metas.atic dis.as. s.te in patients.with CRC with metas.as.s.to a s.ngle organ.
