目的　评价菊花倍半萜烯内酯类化合物 (SL s)对鼻咽癌的抗肿瘤作用。方法　采用 SL s的活性成分parthenolide (PN)为受试物作用于人鼻咽癌高分化细胞株 CNE1,检测细胞毒性和诱导性凋亡指标 ,进行时间 -效应和剂量 -效应作用观察。结果　 10～ 10 0 μm ol/ L 的 PN作用 6～ 36 h后与阴性对照组比较 ,CNE1细胞乳酸脱氢酶 (L DH)漏出率明显增加、细胞计数减少、细胞增殖抑制率升高 (P<0 .0 5 ) ;细胞出现凋亡特征形态学改变 ,TU NEL阳性细胞、Sub G1亚群细胞百分数增加 ,细胞失贴壁率明显增高 (P<0 .0 5 ) ;上述指标与 PN剂量存在明显相关 (P<0 .0 5 )和时间循序变化。结论　 PN可通过对人鼻咽癌细胞毒性作用和诱导细胞凋亡发挥其抗肿瘤作用 ,提示

背景:鼻咽癌是我国南方重点防治的恶性肿瘤之一,探讨天然草本菊花活性成分在NPC化学预防的作用,对建立现代康复体系具有重要意义。目的:研究菊花活性成分倍半萜烯内酯(SLs)对人鼻咽癌细胞中线粒体功能和半胱天冬酶活化途径的影响。设计:完全随机对照分组设计。地点和对象:由中山大学公共卫生学院预防医学系完成,对象为低分化CNE2细胞株。干预:对parthenolide(PN)进行剂量-反应和时间效应作用观察,采用噻唑蓝(MTT)颜色反应检测细胞线粒体功能,底物荧光光谱法测定细胞内caspase-9和-3活性,蛋白免疫印迹法检测线粒体内细胞色素C释放和caspase-3酶原降解片段;并应用特异性抑制剂进行细胞内cas-pase途径阻断实验。主要观察指标:细胞线粒体功能、细胞内caspase-9和caspase-3活性、线粒体细胞色素C的释放和caspase-3酶原的降解。结果:PN(1～100μmol/L)作用12h和24h后,MTT颜色反应抑制率随剂量显著增高,呈明显剂量依赖性(Pearson'sr=0.7322,0.7703,P<0.05),IC50分别为252.94μmol/L和49.63μmol/L;但caspase-9和-3活性未见升高,未见细胞色素C释放和caspase-3酶原降解片段形成。PN与caspase抑制剂联合作用后,caspase-9和-3活性明显低于对照和单独PN作用(t=9.146,8.280,27.325,27.450,P<0.05)。

Heading date and photoperiod sensitivity are fundamental traits that determine rice adaptation to a wide range of geographic environments. By quantitative trait locus （QTL） mapping and candidate gene analysis using whole- genome re-sequencing, we found that Oryza sativa Pseudo-Response Regulator37 （OsPRR37; hereafter PRR37） is respon- sible for the Early heading7-2 （EH7-2）/Heading date2 （Hd2） QTL which was identified from a cross of late-heading rice ＇Milyang23 （M23）＇ and early-heading rice ＇H143＇. H143 contains a missense mutation of an invariantly conserved amino acid in the CCT （CONSTANS, CO-like, and TOC1） domain of PRR37 protein. In the world rice collection, different types of nonfunctional PRR37 alleles were found in many European and Asian rice cultivars. Notably, the japonica varieties harboring nonfunctional alleles of both Ghd7/Hd4 and PRR37/Hd2 flower extremely early under natural long-day condi- tions, and are adapted to the northernmost regions of rice cultivation, up to 53~ N latitude. Genetic analysis revealed that the effects of PRR37 and Ghd7 alleles on heading date are additive, and PRR37 down-regulates Hd3a expression to suppress flowering under long-day conditions. Our results demonstrate that natural variations in PRR37/Hd2 and GhdT/ Hd4 have contributed to the expansion of rice cultivation to temperate and cooler regions.

目的对比研究气相色谱-四极杆飞行时间串联质谱(GC/QTOF-MS)法与气相色谱-四极杆质谱(GC/Q-MS)法在人体尿液代谢组学研究中的应用。方法将采集到的94份环境镉暴露人群与对照人群尿液样品采用相同的前处理,通过两种气质联用技术采集质谱数据,比较两组人群尿液代谢组谱图差异,并利用质谱数据建立模式识别模型,考察建模效果。结果 GC/QTOF-MS技术采集到的尿液代谢组信息较GC/Q-MS技术更为丰富、全面;基于GC/QTOF-MS的数据建立的偏最小二乘模型(R2Y=0.757,Q2=0.5)效果不及基于GC/Q-MS的数据建立的模型(R2Y=0.875,Q2=0.558)。结论 GC/QTOF-MS法具有超高的分辨率与质量准确度,决定该技术在代谢组学研究中的代谢产物识别与结构鉴定方面有着传统GC/Q-MS法无法比拟的优势;但该技术由于较传统GC/Q-MS技术能够采集更多的碎片离子,而显著加强了碎片子离子之间存在的共线性关系,导致模式识别建模效果有所下降,但在代谢组学研究中仍然充满应用前景。

Assessment of left ventricular ejection fraction: comparison of two dimensional echocardiography, cardiac magnetic resonance imaging and 64-row multi-detector computed tomography

Objectives To compare left ventricular ejection fraction (LVEF) determined from 64-row multi-detector computed tomography (64-row MDCT) with those determined from two dimensional echocardiography (2D echo) and cardiac magnetic resonance imaging (CMR). Methods Thirty-two patients with coronary artery disease underwent trans-thoracic 2D echo, CMR and contrast-enhanced 64-row MDCT for assessment of LVEF within 48 hours of each other. 64-row MDCT LVEF was derived using the Syngo Circulation software; CMR LVEF was by Area Length Ejection Fraction (ALEF) and Simpson method and 2D echo LVEF by Simpson *** The LVEF was 49.13 ± 15.91% by 2D echo, 50.72 ± 16.55% (ALEF method) and 47.65 ± 16.58%(Simpson method) by CMR and 50.00 ± 15.93% by 64-row MDCT. LVEF measurements by 64-row MDCT correlated well with LVEF measured with CMR using either the ALEF method (Pearson correlation r = 0.94, P ＜0.01) or Simpson method (r = 0.92, P＜0.01). It also correlated well with LVEF measured using 2D echo (r = 0.80, P ＜ 0.01). Conclusion LVEF measurements by 64-row MDCT correlated well with LVEF measured by CMR and 2D echo. The correlation between 64-row MDCT and CMR was better than the correlation between 2D echo with CMR. Standard data set from a 64-row MDCT coronary study can be reliably used to calculate the LVEF.

Feasibility and accuracy of coronary imaging in elderly patients using the 64-row multi-detector computed tomography: a correlation study with conventional coronary angiography

Background Elderly patients generally have higher occurrence of coronary calcification, increased heart rate and difficulty with prolonged breath-holding. The aim of our study was to investigate the feasibility and accuracy of using 64-row multi-detector computed tomography (MDCT) in the assessment of coronary artery stenoses in elderly patients. Methods One hundred and fifty two patients with suspected or known coronary artery disease were divided into 4 groups according to their age (Group A: 40-49 years,n=34; Group B: 50-59 years, n=57; Group C: 60-69 years, n=48; Group D: 70 years and above; n=13). Coronary CT angiography (CTA) using a 64-row MDCT was performed and the findings were compared with that of conventional coronary angiography (CCA).Using axial images, multi-planar reconstructions (MPR) and maximum intensity projections (MIP), coronary segments of lumen diameter = 1.5mm were analyzed for the presence of significant stenosis (= 50% ). Results Percentages of poor image quality from coronary CTA preventing reliable correlations with CCA were 21%, 14%, 19% and 62% in Groups A to D respectively. Patients in Group D had significantly higher calcium scores compared with the other groups (P＜0.001). In patients where CTA images were of acceptable quality, percentages of accurate correlations with CCA were 89.8%, 93.4%, 86.6% and 78.0% for Groups A to D *** were no significant difference in serum creatinine, heart rate and contrast volume between the 4 groups. Conclusions The 64-row MDCT coronary angiography was less accurate and feasible for patients aged 70 years or above due to heavy coronary calcification and inability to perform a satisfactory breath-hold. However, a high diagnostic accuracy with the MDCT is possible in patients aged less than 70 years.

AIM: Hydrogen sulfide (H2S) is a prominent gaseous constituent of the gastrointestinal (GI) tract with known cytotoxic properties. Endogenous concentrations of H2S are reported to range between 0.2-3.4 mmol/L in the GI tract of mice and humans. Considering such high levels we speculate that, at non-toxic concentrations, H2S may interact with chemical agents and alter the response of colonic epithelium cells to such compounds. The GI tract is a major site for the absorption of phytochemical constituents such as isothiocyanates, flavonoids, and carotenoids, with each group having a role in the prevention of human diseases such as colon cancer. The chemopreventative properties of the phytochemical agent β-phenyethyl isothiocyanate (PEITC) are well recognized. However, little is currently known about the physiological or biochemical factors present in the GI tract that may influence the biological properties of ITCs. The current study was undertaken to determine the effects of H2S on PEITC mediated apoptosis in colon cancer ***: Induction of apoptosis by PEITC in human colon cancer HCT116 cells was assessed using classic apoptotic markers namely SubG1 population analysis,caspase-3 like activity and nuclear fragmentation and condensation coupled with the MTT (3-(4,5-dimethylthiazol2-yl)-2,5-diphenyl tetrasodium bromide) viability assay and LDH ***: PEITC significantly induced apoptosis in HGT116cells as assessed by SubG1 population formation, nuclear condensation, LDH leakage and caspase-3 activity after 24 h, these data being significant from control groups (P＜0.01). In contrast, co-treatment of cells with physiological concentrations of H2S (0.1-1 mmol/L) prevented PEITC mediated apoptosis as assessed using the parameters ***: PEITC effectively induced cell death in the human adenocarcinoma cell line HCT116 in vitro through classic apoptotic mechanisms.
