Improve meat production and virus resistance by simultaneously editing multiple genes in livestock using Cas12i^(Max)

The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated gene(Cas) system is continually optimized to achieve the most efficient gene editing effect. The Cas12i^(Max), a Cas12i variant, exhibits powerful DNA editing activity and enriches the gene editing toolbox. However, the application of Cas12i^(Max)in large domestic animals has not yet been reported. To verify the efficiency and feasibility of multiple gene editing in large animals, we generated porcine fibroblasts with simultaneous knockouts of IGF2, ANPEP, CD163,and MSTN via Cas12i^(Max)in one step. Phenotypically stable pigs were created through somatic cell nuclear transfer technology. They exhibited improved growth performance and muscle quality. Furthermore, we simultaneously edited three genes in bovine fibroblasts. A knockout of MSTN and PRNP was created and the amino acid Q-G in CD18 was precisely substituted. Meanwhile, no off-target phenomenon was observed by sum-type analysis or off-target detection. These results verified the effectiveness of Cas12i^(Max)for gene editing in livestock animals and demonstrated the potential application of Cas12i^(Max)in the field of animal trait improvement for agricultural production.

Autophagy receptor ZmNBR1 promotes the autophagic degradation of ZmBRI1a and enhances drought tolerance in maize

Drought stress is a crucial environmental factor that limits plant growth, development, and productivity.Autophagy of misfolded proteins can help alleviate the damage caused in plants experiencing drought.However, the mechanism of autophagy-mediated drought tolerance in plants remains largely unknown.Here, we cloned the gene for a maize(Zea mays)selective autophagy receptor, NEXT TO BRCA1GENE 1(ZmNBR1), and identified its role in the response to drought stress. We observed that drought stress increased the accumulation of autophagosomes. RNA sequencing and reverse transcriptionquantitative polymerase chain reaction showed that ZmNBR1 is markedly induced by drought stress. ZmNBR1 overexpression enhanced drought tolerance, while its knockdown reduced drought tolerance in maize. Our results established that ZmNBR1 mediates the increase in autophagosomes and autophagic activity under drought stress.ZmNBR1 also affects the expression of genes related to autophagy under drought stress. Moreover, we determined that BRASSINOSTEROID INSENSITIVE1A(ZmBRI1a), a brassinosteroid receptor of the BRI1-like family, interacts with ZmNBR1. Phenotype analysis showed that ZmBRI1a negatively regulates drought tolerance in maize, and genetic analysis indicated that ZmNBR1 acts upstream of ZmBRI1a in regulating drought tolerance. Furthermore, ZmNBR1facilitates the autophagic degradation of ZmBRI1a under drought stress. Taken together, our results reveal that ZmNBR1 regulates the expression of autophagy-related genes, thereby increasing autophagic activity and promoting the autophagic degradation of ZmBRI1a under drought stress, thus enhancing drought tolerance in maize. These findings provide new insights into the autophagy degradation of brassinosteroid signaling components by the autophagy receptor NBR1 under drought stress.

Highly elastic energy storage device based on intrinsically super-stretchable polymer lithium-ion conductor with high conductivity

Stretchable power sources,especially stretchable lithium-ion batteries(liBs),have attracted increasing attention due to their enormous prospects for powering flexible/wearable electronics.Despite recent advances,it is still challenging to develop ultra-stretchable liBs that can withstand large deformation.In particular,stretchable liBs require an elastic electrolyte as a basic component,while the conductivity of most elastic electrolytes drops sharply during deformation,especially during large deformations.This is why highly stretchable liBs have not yet been realized until now.As a proof of concept,a super-stretchable liB with strain up to 1200%is created based on an intrinsically super-stretchable polymer electrolyte as the lithium-ion conductor.The super-stretchable conductive system is constructed by an effective diblock copolymerization strategy via photocuring of vinyl functionalized 2-ureido-4-pyrimidone(VFUpy),an acrylic monomer containing succinonitrile and a lithium salt,achieving high ionic conductivity(3.5×10^(-4)mS cm^(-1)at room temperature(RT))and large deformation(the strain can reach 4560%).The acrylic elastomer containing li-ion conductive domains can strongly increase the compatibility between the neighboring elastic networks,resulting in high ionic conductivity under ultra-large deformation,while VFUpy increases elasticity modulus(over three times)and electrochemical stability(voltage window reaches 5.3 V)of the prepared polymer conductor.At a strain of up to 1200%,the resulting stretchable liBs are still sufficient to power LEDs.This study sheds light on the design and development of high-performance intrinsically super-stretchable materials for the advancement of highly elastic energy storage devices for powering flexible/wearable electronics that can endure large deformation.

Polyimide covalent organic frameworks as efficient solid-state li^(+) electrolytes

Covalent organic frameworks(COFs) are attractive porous crystalline materials with extremely high stability, easy functionalization, and open channels, which are expected to be unique ion conductors/transporters in lithium ion batteries(liBs). Despite recent advances, low ion conductivity and low transference number, resulting in low charging/discharging rate, low energy density, and short battery life, are the main issues that limit their direct application as solid electrolytes in liBs. Here, we designed and synthesized a novel polyimide COF, namely, TAPA-PDI-COF, with abundant C=O groups, which has been successfully employed as high-performance solid electrolytes by doping TAPA-PDI-COF and succinonitrile(SN). Both the well-defined nanochannels of COFs and SN confined in the well-aligned channels restricted the free migration of anions, while C=O on COFs and CN groups of SN enhanced li^(+) transport, thus achieving a high ion conductivity of 0.102 m S cm^(-1)at 80 °C and a high lithium-ion transference number of 0.855 at room temperature. According to density functional theory(DFT)calculations, li-ion migration mainly adopted in-plane transport rather than the axial pathway, which may be due to the shorter hopping distances in the planar pathway. The results suggest an effective strategy for the design and development of all-solidstate ionic conductors for achieving high-performance liBs.

Effects of vibration conditions,spatial confinement and friction on mixing and segregation characteristics of mixed pebble beds for CFETR WCCB blanket

This paper presents the study of the dynamic characteristics of confined mixed pebble beds with different friction under different vibration conditions using the discrete element method.Theλsegre-gation index is used to quantify the degree of particle mixing or segregation.The percolation,convection and diffusion mechanisms are responsible for the segregation patterns of the mixed pebble.The results show the degree of segregation can be suppressed by decreasing the vibration acceleration or free space height below a threshold.Further simulation reveals the threshold of vibration acceleration or free space height both are related to the bed height increment which determines the strength of the percolation mechanism.In addition,the strength of percolation and convection becomes weaker by decreasing the friction of particles and walls,which makes the pebbled bed remains in a mixed state under vibration.These findings are significant to clarify the main factors behind the three segregation mechanisms and hence provide solutions to retaining the mixed state of the li2TiO3&Be12Ti mixed pebble bed.

Trajectory planning and base attitude restoration of dual-arm free-floating space robot by enhanced bidirectional approach

When free-floating space robots perform space tasks,the satellite base attitude is disturbed by the dynamic coupling.The disturbance of the base orientation may affect the communication between the space robot and the control center on earth.In this paper,the enhanced bidirectional approach is proposed to plan the manipulator trajectory and eliminate the final base attitude variation.A novel acceleration level state equation for the nonholonomic problem is proposed,and a new intermediate variable-based Lyapunov function is derived and solved for smooth joint trajectory and restorable base trajectories.In the method,the state equation is first proposed for dual-arm robots with and without end constraints,and the system stability is analyzed to obtain the system input.The input modification further increases the system stability and simplifies the calculation complexity.Simulations are carried out in the end,and the proposed method is validated in minimizing final base attitude change and trajectory smoothness.Moreover,the minute internal force during the coordinated operation and the considerable computing efficiency increases the feasibility of the method during space tasks.

Inflammatory bowel disease(IBD)is characterized by recurrent attacks and long courses,and the number of patients has expanded rapidly year by year.Additionally,current conventional strategies exist serious adverse effects.In this case,it is an urgent issue to find out an effective and safe treatment.Functional oligosaccharides possess safe and excellent physiological activities,and have attracted enormous attention due to their great therapeutic potential for IBD.This review emphasizes the attenuating effects of distinct functional oligosaccharides on IBD and their structure,and summarizes the main mechanisms from the aspects of regulating intestinal fl ora structure,repairing intestinal barrier,modulating immune function and mediating related signaling pathways in order to reveal the relationship between functional oligosaccharides,immune regulation,intestinal epithelial cells,gut fl ora and IBD treatment.Oligosaccharides possess excellent protective effects on IBD,and can be considered as safe and functional ingredients in the health food and pharmaceutical industry.

Ultraviolet(UV)-induced photoaging skin has become an urgent issue.The functional foods and cosmetics aiming to improve skin photoaging are developing rapidly,and the demand is gradually increasing year by year.Collagen peptides have been proven to display diverse physiological activities,such as excellent moisture retention activity,hygroscopicity,tyrosinase inhibitory activity and antioxidant activity,which indicates that they have great potential in amelioration of UV-induced photoaging.The main objective of this article is to recap the main mechanisms to improve photoaging skin by collagen peptides and their physiological activities in photo-protection.Furthermore,the extraction and structural characteristics of collagen peptides are overviewed.More importantly,some clinical trials on the beneficial effect on skin of collagen peptides are also discussed.In addition,prospects and challenges of collagen peptides are emphatically elucidated in this review.This article implies that collagen peptides have great potential as an effective ingredient in food and cosmetics industry with a wide application prospect.

The MEKK1-MKK1/2-MPK4 cascade phosphorylates and stabilizes STOP1 to confer aluminum resistance in Arabidopsis

Aluminum(Al)toxicity can seriously restrict crop production on acidic soils,which comprise 40%of the world’s potentially arable land.The zinc finger transcription factor STOP1 has a conserved and essential function in mediating plant Al resistance.Al stress induces STOP1 accumulation via post-transcriptional regulatory mechanisms.However,the upstream signaling pathway involved in Al-triggered STOP1 accumulation remains unclear.Here,we report that the MEKK1-MKK1/2-MPK4 cascade positively regulates STOP1 phosphorylation and stability.Mutations of MEKK1,MKK1/2,or MPK4 lead to decreased STOP1 stability and Al resistance.Al stress induces the kinase activity of MPK4,which interacts with and phosphorylates STOP1.The phosphorylation of STOP1 reduces its interaction with the F-box protein RAE1 that mediates STOP1 degradation,thereby leading to enhanced STOP1 stability and Al resistance.Taken together,our results suggest that the MEKK1-MKK1/2-MPK4 cascade is important for Al signaling and confers Al resistance through phosphorylation-mediated enhancement of STOP1 accumulation in Arabidopsis.

Integrin β2 and β3:Two plasmatocyte markers deepen our understanding of the development of plasmatocytes in the silkworm Bombyx mori

Insect hemocytes play important biological roles at developmental stages,metamorphosis,and innate immunity.As one of the most abundant cell types,plasmatocytes can participate in various innate immune responses,especially in encapsulation and node formation.Here,2 molecular markers of plasmatocytes,consisting of integrinβ2 andβ3,were identified and used to understand the development of plasmatocytes.Plasmatocytes are widely distributed in the hematopoietic system,including circulating hemolymph and hematopoietic organs(HPOs).HPOs constantly release plasmatocytes with high proliferative activity in vitro;removal of HPOs leads to a dramatic reduction in the circulating plasmatocytes,and the remaining plasmatocytes gradually lose their ability to proliferate in vivo.Our results demonstrated that the release of plasmatocytes from HPOs is regulated by insulin-mediated signals and their downstream pathways,including PI3K/Akt and MAPK/Erk signals.The insulin/PI3K/Akt signaling pathway can significantly irritate the hematopoiesis,and its inhibitor LY294002 could inhibit the hemocytes discharged from HPOs.While the insulin/MAPK/Erk signaling pathway plays a negative regulatory role,inhibiting its activity with U0126 can markedly promote the discharge of plasmatocytes from HPOs.Our results indicate that the circulating plasmatocytes are mainly generated and discharged by HPOs.This process is co-regulated by the PI3K/Akt and MAPK/Erk signals in an antagonistic manner to adjust the dynamic balance of the hemocytes.These findings can enhance our understanding of insect hematopoiesis.
