Anti-aging Effects of Alu Antisense RNA on Human Fibroblast Senescence Through the MEK-ERK Pathway Mediated by KIF15

Objective:To investigate whether human short interspersed nuclear element antisense RNA(Alu antisense RNA;Alu asRNA)could delay human fibroblast senescence and explore the underlying mechanisms.Methods:We transfected Alu asRNA into senescent human fibroblasts and used cell counting kit-8(CCK-8),reactive oxygen species(ROS),and senescence-associated beta-galactosidase(SA-β-gal)staining methods to analyze the anti-aging effects of Alu asRNA on the fibroblasts.We also used an RNA-sequencing(RNA-seq)method to investigate the Alu asRNA-specific mechanisms of anti-aging.We examined the effects of KIF15 on the anti-aging role induced by Alu asRNA.We also investigated the mechanisms underlying a KIF15-induced proliferation of senescent human fibroblasts.Results:The CCK-8,ROS and SA-β-gal results showed that Alu asRNA could delay fibroblast aging.RNA-seq showed 183 differentially expressed genes(DEGs)in Alu asRNA transfected fibroblasts compared with fibroblasts transfected with the calcium phosphate transfection(CPT)reagent.The KEGG analysis showed that the cell cycle pathway was significantly enriched in the DEGs in fibroblasts transfected with Alu asRNA compared with fibroblasts transfected with the CPT reagent.Notably,Alu asRNA promoted the KIF15 expression and activated the MEK-ERK signaling pathway.Conclusion:Our results suggest that Alu asRNA could promote senescent fibroblast proliferation via activation of the KIF15-mediated MEK-ERK signaling pathway.

Alu antisense RNA ameliorates methylglyoxal-induced human lens epithelial cell apoptosis by enhancing antioxidant defense

AIM:To determine whether an antisense RNA corresponding to the human Alu transposable element(Aluas RNA)can protect human lens epithelial cells(HLECs)from methylglyoxal-induced apoptosis.METHODS:Cell counting kit-8(CCK-8)and 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assays were used to assess HLEC viability.HLEC viability/death was detected using a Calcein-AM/PI double staining kit;the annexin V-FITC method was used to detect HLEC apoptosis.The cytosolic reactive oxygen species(ROS)levels in HLECs were determined using a reactive species assay kit.The levels of malondialdehyde(MDA)and the antioxidant activities of total-superoxide dismutase(T-SOD)and glutathione peroxidase(GSH-Px)were assessed in HLECs using their respective kits.RT-q PCR and Western blotting were used to measure m RNA and protein expression levels of the genes.RESULTS:Aluas RNA rescued methylglyoxal-induced apoptosis in HLECs and ameliorated both the methylglyoxalinduced decrease in Bcl-2 m RNA and the methylglyoxalinduced increase in Bax m RNA.In addition,Aluas RNA inhibited the methylglyoxal-induced increase in Alu sense RNA expression.Aluas RNA inhibited the production of ROS induced by methylglyoxal,restored T-SOD and GSHPx activity,and moderated the increase in MDA content after treatment with methylglyoxal.Aluas RNA significantly restored the methylglyoxal-induced down-regulation of Nrf2 gene and antioxidant defense genes,including glutathione peroxidase,heme oxygenase 1,γ-glutamylcysteine synthetase and quinone oxidoreductase 1.Aluas RNA ameliorated methylglyoxal-induced increases of the m RNA and protein expression of Keap1 that is the negative regulator of Nrf2.CONCLUSION:Aluas RNA reduces apoptosis induced by methylglyoxal by enhancing antioxidant defense.

Evaluating the performance and influencing factors of three portable black carbon monitors for field measurement

Black carbon(BC)is associated with adverse human health and climate change.Mapping BC spatial distribution imperatively requires low-cost and portable devices.Several portable BC monitors are commercially available,but their accuracy and reliability are not always satisfactory during continuous field observation.This study evaluated three models of portable black carbon monitors,C12,MA350 and DST,and investigates the factors that affect their performance.The monitors were tested in urban Beijing,where portable devices running for one month alongside a regular-size reference aethalometer AE33.The study considers several factors that could influence the monitors'performance,including ambient weather,aerosol composition,loading artifacts,and built-in algorithms.The results show that MA350and DST present considerable discrepancies to the reference instrument,mainly occurring at lower concentrations(0-500 ng/m^(3))and higher concentrations(2500-8000 ng/m^(3)),respectively.These discrepancies were likely caused by the anomalous noise of MA350 and the loading artifacts of DST.The study also suggests that the ambient environment has limited influence on the monitors'performance,but loading artifacts and accompanying compensation algorithms can result in unrealistic data.Based on the evaluation,the study suggests that C12 is the best choice for unsupervised field measurement,DST should be used in scenarios where frequent maintenance is available,and MA350 is suitable for research purposes with post-processing applicable.The study highlights the importance of assigning portable BC monitors to appropriate applications and the need for optimized real-time compensation algorithms.

Three-dimensional morphological and fluorescent imaging of zebrafish with a continuous-rotational light-sheet microscope

Light-sheet fluorescence microscopy(LSFM)has been widely used to image the three-dimensional(3D)structures and functions of various millimeter-size bio-specimen such as zebrafish.However,the sample adsorption and scattering cause shading of the light-sheet illumination,preventing the even 3D image of thick samples.Herein,we report a continuous-rotational light-sheet microscope(CR-LSM)that enables simultaneous 3D bright-field and fluorescence imaging.With a high-accuracy rotational stage,CR-LSM records the outline projections and the fluorescent images of the sample at multiple rotation angles.Then,3D morphology and fluorescent structure were reconstructed with a developed algorithm.Using CR-LSM,zebrafish’s whole-fish contour and blood vessel structures were obtained simultaneously.

Conduction-convection coupled heat transfer around a hollow cylinder under different buoyancy forces

In order to clarify the effect of a buoyancy force on conduction–convection coupled heat transfer in a hollow cylinder, the flow and thermal characteristics were analyzed using an RNG k-ε turbulence model. The Reynolds number was fixed at 1.014 × 10^(6), and the Rayleigh number varied from 1.122 × 10^(10)to 1.088 × 10^(11). Results have shown that, when considering the effect of an opposed buoyancy force, increasing the Rayleigh number has a positive impact on the rate of change and uniformity of the cylinder temperature. The temperature distributions along the axial and circumferential directions are similar for different Rayleigh numbers, but extreme values differ.Along the axial direction, the maximum temperature is obtained at the interface between the variable-diameter part and the constant-diameter part. The maximum dimensionless temperature value decreases to 0.12 when the Rayleigh number increases to 1.088 × 10^(11). Along the circumferential direction, the temperature distribution is affected by the buoyancy force, which results in the temperature of the upper part being higher than that of the lower part. After nondimensionalization of the temperature and time, a correlation was proposed to illustrate the transient heat transfer process quantitatively. The standard deviation of the maximum relative temperature, representing the temperature uniformity, was also calculated. It was found that the difference in the direction of the buoyancy force made a huge difference. Compared with the opposed buoyancy force, the maximum dimensionless temperature is almost two times higher with an assisted buoyancy force. Similarly, the heat transfer coefficient with an assisted buoyancy force is half of that with an opposed buoyancy force. Overall, an assisted buoyancy force plays a negative role in terms of thermal characteristics. The flow field around the hollow cylinder was also illustrated to reveal the mechanism of the buoyancy force on magnitude and direction aspects.

Laboratory and field evaluation of a low-cost optical particle sizer

Low-cost sensors are widely used to collect high-spatial-resolution particulate matter data that traditional reference monitoring devices cannot.In addition to the mass concentration,the number concentration and size distribution are also fundamental in determining the origin and hazard level of particulate pollution.Therefore,low-cost optical sensors have been improved to establish optical particle sizers(OPSs).In this study,a low-cost OPS,the Nova SDS029,is introduced,and it is evaluated in comparison to two reference instruments—the GRIMM 11-D and the TSI 3330.We first tested the sizing accuracy using polystyrene latex spheres.Then,we assessed the mass and number size distribution accuracy in three application scenarios:indoor smoking,ambient air quality,and mobile monitoring.The evaluations suggest that the low-cost SDS029 rivals research-grade optical sizers in many aspects.For example,(1)the particle diameters obtained with the SDS029 are close to the reference instruments(usually<10%)in the 0.3-5μm range;(2)the number of particles and mass concentration are highly correlated(r≥0.99)with the values obtained with the reference instruments;and(3)the SDS029 slightly underestimates the number concentration,but the derived PM_(2.5)values are closer to monitoring station than the reference instruments.The successful application of the SDS029 in multiple scenarios suggests that a plausible particle size distribution can be obtained in an easy and cost-efficient way.We believe that low-cost OPSs will increasingly be used to map the sources and risk levels of particles at the city scale.

AIM:To address the challenges of data labeling difficulties,data privacy,and necessary large amount of labeled data for deep learning methods in diabetic retinopathy(DR)identification,the aim of this study is to develop a source-free domain adaptation(SFDA)method for efficient and effective DR identification from unlabeled data.METHODS:A multi-SFDA method was proposed for DR identification.This method integrates multiple source models,which are trained from the same source domain,to generate synthetic pseudo labels for the unlabeled target domain.Besides,a softmax-consistence minimization term is utilized to minimize the intra-class distances between the source and target domains and maximize the inter-class distances.Validation is performed using three color fundus photograph datasets(APTOS2019,DDR,and EyePACS).RESULTS:The proposed model was evaluated and provided promising results with respectively 0.8917 and 0.9795 F1-scores on referable and normal/abnormal DR identification tasks.It demonstrated effective DR identification through minimizing intra-class distances and maximizing inter-class distances between source and target domains.CONCLUSION:The multi-SFDA method provides an effective approach to overcome the challenges in DR identification.The method not only addresses difficulties in data labeling and privacy issues,but also reduces the need for large amounts of labeled data required by deep learning methods,making it a practical tool for early detection and preservation of vision in diabetic patients.

A model reduction approach for discrete-time linear time-variant systems with delayed inputs

A model reduction approach is presented for discrete-time linear time-variant input-delayed systems. According to this proposed approach, a dynamical variable is constructed by taking advantage of the current state and historical information of input. It is revealed that the behavior of this dynamical variable is governed by a discrete-time linear delay-free system. It is worth noting that the presented variable transformation does not require the system matrix to be invertible. Based on the reduced delay-free models,stabilizing control laws can be easily obtained for the original delayed system. For the case with a single input delay, the constructed variable is an exact prediction for the future state, and thus the stabilizing control law could be designed by replacing the future state with its prediction. Finally, three discrete-time periodic systems with delayed input are employed to illustrate how to utilize the presented model reduction approaches.

A Novel Disturbance Observer Based Fixed-Time Sliding Mode Control for Robotic Manipulators With Global Fast Convergence

This paper proposes a new global fixed-time sliding mode control strategy for the trajectory tracking control of uncertain robotic manipulators.First,a fixed-time disturbance observer(FTDO) is designed to deal with the adverse effects of model uncertainties and external disturbances in the manipulator systems.Then an adaptive scheme is used and the adaptive FTDO(AFTDO) is developed,so that the priori knowledge of the lumped disturbance is not required.Further,a new non-singular fast terminal sliding mode(NFTSM) surface is designed by using an arctan function,which helps to overcome the singularity problem and enhance the robustness of the system.Based on the estimation of the lumped disturbance by the AFTDO,a fixed-time non-singular fast terminal sliding mode controller(FTNFTSMC)is developed to guarantee the trajectory tracking errors converge to zero within a fixed time.The settling time is independent of the initial state of the system.In addition,the stability of the AFTDO and FTNFTSMC is strictly proved by using Lyapunov method.Finally,the fixed-time NFESM(FTNFTSM) algorithm is validated on a 2-link manipulator and comparisons with other existing sliding mode controllers(SMCs) are performed.The comparative results confirm that the FTNFTSMC has superior control performance.

Effect of Mn addition on microstructure and mechanical properties of GX40CrNiSi25-12 austenitic heat resistant steel

Three types of steels were designed on the basis of GX40CrNiSi25-12 austenitic heat resistant steel by adding different Mn contents(2wt.%,6wt.%,and 12wt.%).Thermodynamic calculation,microstructure characterization and mechanical property tests were conducted to investigate the effect of Mn addition on the microstructure and mechanical properties of the austenitic heat resistant steel.Results show that the matrix structure in all the three types of steels at room temperature is completely austenite.Carbides NbC and M_(23)C_(6)precipitate at grain boundaries of austenite matrix.With the increase of Mn content,the number of carbides increases and their distribution becomes more uniform.With the Mn content increases from 1.99%to 12.06%,the ultimate tensile strength,yield strength and elongation increase by 14.6%,8.0%and 46.3%,respectively.The improvement of the mechanical properties of austenitic steels can be explained by utilizing classic theories of alloy strengthening,including solid solution strengthening,precipitation strengthening,and grain refinement.The increase in alloy strength can be attributed to solid solution strengthening and precipitation strengthening caused by the addition of Mn.The improvement of the plasticity of austenitic steels can be explained from two aspects:grain refinement and homogenization of precipitated phases.
