Ethanol extract from Gynostemma pentaphyllum ameliorates dopaminergic neuronal cell death in transgenic mice expressing mutant A53T human alpha-synuclein

Gynostemma(G.) pentaphyllum(Cucurbitaceae) contains various bioactive gypenosides. Ethanol extract from G. pentaphyllum(GP-EX) has been shown to have ameliorative effects on the death of dopaminergic neurons in animal models of Parkinson’s disease(PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-and 6-hydroxydopamine. PD patients exhibit multiple symptoms, so PD-related research should combine neurotoxin models with genetic models. In the present study, we investigated the ameliorative effects of GP-EX, including gypenosides, on the cell death of dopaminergic neurons in the midbrain of A53 T α-synuclein transgenic mouse models of PD(A53 T). Both GP-EX and gypenosides at 50 mg/kg per day were orally administered to the A53 T mice for 20 weeks.α-Synuclein-immunopositive cells and α-synuclein phosphorylation were increased in the midbrain of A53 T mice, which was reduced following treatment with GP-EX. Treatment with GP-EX modulated the reduced phosphorylation of tyrosine hydroxylase, extracellular signal-regulated kinase(ERK1/2), Bcl-2-associated death promoter(Bad) at Ser112, and c-Jun N-terminal kinase(JNK1/2) due to α-synuclein overexpression. In the A53 T group, GP-EX treatment prolonged the latency of the step-through passive avoidance test and shortened the transfer latency of the elevated plus maze test. Gypenosides treatment exhibited the effects and efficacy similar to those of GP-EX. Taken together, GP-EX, including gypenosides, has ameliorative effects on dopaminergic neuronal cell death due to the overexpression of α-synuclein by modulating ERK1/2, Bad at Ser112, and JNK1/2 signaling in the midbrain of A53 T mouse model of PD. Further studies are needed to investigate GP-EX as a treatment for neurodegenerative synucleinopathies, including PD. This study was approved by the Animal Ethics Committee of Chungbuk National University(approval No. CBNUA-956-16-01) on September 21, 2016.

AIM To investigate the association between postoperative pain control and oncologic outcomes in resected pancreatic ductal adenocarcinoma(PDAC).METHODS From January 2009 to December 2014, 221 patients were diagnosed with PDAC and underwent resection with curative intent. Retrospective review of the patients was performed based on electronic medical records system. One patient without records of numerical rating scale(NRS) pain intensity scores was excluded and eight patients who underwent total pancreatectomy were also excluded. NRS scores during 7 postoperative days following resection of PDAC werereviewed along with clinicopathologic characteristics. Patients were stratified into a good pain control group and a poor pain control group according to the difference in average pain intensity between the early(POD 1, 2, 3) and late(POD 5, 7) postoperative periods. Cox-proportional hazards multivariate analysis was performed to determine association between postoperative pain control and oncologic *** A total of 212 patients were dichotomized into good pain control group(n = 162) and poor pain control group(n = 66). Median follow-up period was 17 mo. A negative impact of poor postoperative pain control on overall survival(OS) was observed in the group of patients receiving distal pancreatectomy(DP group; 42.0 mo vs 5.0 mo, P = 0.001). Poor postoperative pain control was also associated with poor disease-free survival(DFS) in the DP group(18.0 mo vs 8.0 mo, P = 0.001). Patients undergoing pancreaticoduodenectomy or pylorus-preserving pancreaticoduodenectomy(PD group) did not show associations between postoperative pain control and oncologic outcomes. 提出一种简便的具有最大功率点跟踪(maximum power point tracking,MPPT)控制功能的热能收集电路,为自供能微型传感器节点供电。由于热电发生器(thermoelectric generator,TEG)的输出电压足够高,可直接驱动负载应用,故该电路免去了复杂的启动电路和直流-直流(DC-DC)升压转换器。为克服TEG设备和负载应用之间的功率失配,该电路在激活/休眠模式下工作。该热能收集电路基于0.35μm互补式金属氧化物半导体(CMOS)工艺研制。实验结果证明该电路能正常工作,展示了MPPT方案性能。该电路实现了95.5%的峰值功效和高于99%的MPPT精度。

The inhomogeneous hardness distribution of high pressure torsion (HPT) processed IF steel disks along different directions is investigated. The results indicated that there exists inhomogeneous distribution in HPT processed IF steel disks, giving lower hardness in the center and higher hardness in the edge regions. However, on the axisymmetrical section testing plane of the disks’ thickness direction, there is a soft zone near the surface of disks. Further results from radius testing plane of different depths from the surface of HPT processed disks show that the inhomogeneity rules of hardness distribution on the radius direction are similar to that on the thickness direction. Compared with the initial state, different stages of HPT (compression and compression + torsion) can both remarkably increase the hardness of IF steel disks. Microstructure investigation results can give a well support to verify the rules of hardness distribution, showing hardly no change of grains in center and sever plastic deformation in edge. The inhomogeneous distribution of stress and strain with the huge friction between anvil and disks in the process of HPT play an important role of hardness and microstructure distribution.

Post-inflammatory hyperpigmentation is common problem, but its treatment still remains challenging. Tranexamic acid has been used to treat or prevent excessive bleeding loss in various medical conditions. There have been some reports of the effect of oral and topical tranexamic acid for treatment of pigmented disorder. Herein we report on a case of female patient who showed improvement of PIH after oral and topical tranexamic acid administration.

AIM:To determine the clinical data that might be useful for differentiating benign from malignant gallbladder (GB) polyps by comparing radiological methods,including abdominal ultrasonography (US) and computed tomography (CT) scanning,with postoperative pathology fi ndings. METHODS:Fifty-nine patients underwent laparoscopic cholecystectomy for a GB polyp of around 10 mm. They were divided into two groups,one with cholesterol polyps and the other with non-cholesterol polyps. Clinical features such as gender,age,symptoms,size and number of polyps,the presence of a GB stone,the radiologically measured maximum diameter of the polyp by US and CT scanning,and the measurements of diameter from postoperative pathology were recorded for comparative analysis. RESULTS:Fifteen of the 41 cases with cholesterol polyps (36.6%) were detected with US but not CT scanning,whereas all 18 non-cholesterol polyps were observed using both methods. In the cholesterol polyp group,the maximum measured diameter of the polyp was smaller by CT scan than by ***,the discrepancy between those two scanning measurements was greater than for the non-cholesterol polyp group. CONCLUSION:The clinical signs indicative of a cholesterol polyp include:(1) a polyp observed by US but not observable by CT scanning,(2) a smaller diameter on the CT scan compared to US,and (3) a discrepancy in its maximum diameter between US and CT measurements. In addition,US and the CT scan had low accuracy in predicting the polyp diameter compared to that determined by postoperative pathology.
