目的探讨成人心脏直视术后手术部位感染（SSI）发生的危险因素,为控制SSI的发生提供依据.方法对本院心脏外科2001年1月至2009年12月间体外循环心脏直视术后发生SSI 54例成年患者临床资料进行分析,按1：3比例配对选取对照组.对2组患者的临床资料中SSI的潜在危险因素进行单因素和多因素条件Logistic回归分析.结果单因素条件Logistic分析：左心室射血分数（LVEF）〈50%（OR=2.134,95%CI：1.095～4.159,P=0.026）,心功能NYHA≥Ⅲ级（OR=2.390,95%CI：1.218～4.690,P=0.011）,糖尿病（OR=3.275,95%CI：1.391-7.708,P=0.007）,慢性阻塞性肺疾病（COPD）（OR=5.408,95%CI：1.248～23.445,P=0.024）,体外循环时间〉90 min（OR=3.045,95%CI：1.540～6.024,P=0.001）,手术时间〉4 h（OR=3.281,95%CI：1.610～6.685,P=0.0131）,血液制品用量〉2 U（OR=1.929,95%CI：1.018～3.675,P=0.044）,切口连续缝合（OR=2.344,95%CI：1.221～4.498,P=0.010）,二次开胸止血（OR=6.625,95%CI：1.597～27.491,P=0.009）,术后高血糖（OR=3.510,95%CI：1.596～7.718,P=0.002）,重症监护病房入住〉72 h（OR=3.281,95%CI：1.505～7.150,P=0.003）与SSI发生相关.多因素条件Lgistic回归分析显示：手术时间〉4 h（OR=3.100,95%CI：1.470～6.537,P=0.003）、切口皮下层连续缝合（OR=2.340,95%CI：1.183～4.692,P=0.015）、术后高血糖（OR=3.272,95%CI：1.427～7.505,P=0.005）是SSI的独立危险因素.结论手术时间〉4 h、切口皮下连续缝合及术后高血糖是心脏直视术后SSI发生的危险因素.

Understanding hydrological responses to rising levels of greenhouse gases are essential for climate and impact *** is,however,often limited by a lack of long record of observational data to provide a basis for understanding the longterm behavior of the climate *** reconstructed data and(global climate and hydrological)model simulations will help us to better understand the variability of climate and hydrology over timescales ranging from decades to *** this study,we proposed an integrated approach to study flood variability in the upper reach of the Yangtze River over the last millennium to the end of the 21st *** accomplish this,we first drove hydrological models using the precipitation and temperature from four Global Climate Models(GCM),BCC-CSM1.1,MIROC,MRI-CGCM3,and CCSM4,to simulate daily discharge for the upper reach of the Yangtze River during the period of the last millennium(850–1849),historical period(1850–2005),and a future period(2006–2099).Then,we evaluated whether the modeled precipitation,temperature,and extreme discharge had statistical properties similar to those shown in the documented dry-wet periods,temperature anomalies,and paleoflood ***,we explored the extreme discharge variability using model *** results indicate that:(1)The MIROC-ESM model,differing from the other three GCM models,revealed positive temperature changes from the warm period(Medieval Climate Anomaly;MCA)to the cold period(Little Ice Age;lia),while the temperature variability of the other models was similar to the records.(2)The BCC-CSM1.1 model performed better than the others regarding correlations between modeled precipitation and documented dry-wet periods.(3)Over most of the subbasins in the upper Yangtze River,the magnitude of extreme discharge in the BCC-CSM1.1 model results showed that there was a decrease from the MCA to the lia period and an increase in the historical period relative to the cold period,whil

Two new platinum（II） complexes 5 a and 5 b with methyl hydrazinecarbodithioate derivatives of indolin-2-one have been synthesized and characterized by single-crystal X-ray diffraction,NMR and *** activity of the two complexes and their ligands 4 a and 4 b against HCT-116,MCF-7 and MDA-MB-231 cell lines was determined by the MTS *** 5 a and 5 b were more cytotoxic against three cell lines than compounds 4 a and 4 b（IC50,1.89-5.60 versus 6.52-35.13 μM）.Moreover,treatment of HCT-116 cells with the complexes resulted in an obvious sub-G1 peak by cell cycle profile analysis,and an increase of cleaved PARP1 levels by immunoblotting *** cell imaging showed that nucleus shrinkage and condensation started to appear when MCF-7 cells were treated with 5 a for 8 *** spectrophotometric analysis revealed that the complexes physically associated with calf thymus *** together,our results indicated that the two complexes 5 a and 5 b bind to DNA with high affinity and exhibit cytotoxicity against cancer cells by inducing apoptosis.

Anthocyanins are essential for the quality of perennial horticultural crops,such as *** grapes,ELONGATED HYPOCOTYL 5(HY5)and MYBA1 are two critical transcription factors that regulate anthocyanin *** previous work has shown that Vitis vinifera B-box protein 44(VvBBX44)inhibits anthocyanin synthesis and represses VvHY5 expression in grape ***,the regulatory mechanism underlying this regulation was *** this study,we found that loss of VvBBX44 function resulted in increased anthocyanin accumulation in grapevine ***44 directly represses VvMYBA1,which activates ***1,but not VvBBX44,directly modulates the expression of grape UDP flavonoid 3-O-glucosyltransferase(VvUFGT).We demonstrated that VvBBX44 represses the transcriptional activation of VvUFGT and VvBBX44 induced by ***,VvBBX44 and VvMYBA1 did not physically interact in *** application of exogenous anthocyanin stimulated VvBBX44 expression in grapevine suspension cells and tobacco *** findings suggest that VvBBX44 and VvMYBA1 form a transcriptional feedback loop to prevent overaccumulation of anthocyanin and reduce metabolic *** work sheds light on the complex regulatory network that controls anthocyanin biosynthesis in grapevine.

为了准确而又经济地现场测试研究高速运输过程中由风作用而在墙面产生的非均衡载荷,利用空气动力学的三维模型研究了预制木结构房在高速运输过程中的各个部分的风压力分布,优化了风压力测试传感器的数量和安装位置.主要结论为:构建的三维动力学模型能够很好地模拟运输过程中由于风作用而在墙面和屋顶上产生的压力分布;模型分析结果表明在迎风和背风墙面上均出现了显著的风压力梯度,迎风墙面处压力值最大可达645Pa;在侧面和屋顶处压力分布比较均衡,没有出现明显的压力分布梯度;为了精确和经济地在运输现场实施实地风压力测试,在迎风墙面的单位面积应该多布置传感器.

目的制备自身失活型慢病毒载体为基础的鼠基因工程调节性T细胞（Treg细胞），检测其表型变化，并观察其增殖能力及免疫抑制能力。方法构建含鼠Foxp3基因及内部核糖体进入位点序列（IRES）和绿色荧光蛋白基因（GFP）的双顺反子自身失活型慢病毒载体。采用脂质体转染法将慢病毒载体三质粒转染293T细胞，经共培养法感染免疫磁珠分离的小鼠CD4^＋CD25-T细胞，流式细胞术（FCM）检测基因转染效率及细胞Foxp3、CD25、GITR、CTLA-4的表达，CCK-8法、ELISA法检测其增殖、免疫抑制能力及细胞因子分泌的变化。结果成功构建了慢病毒表达质粒pXZ208-IRES—GFP、pXZ208·Foxp3-IRES—GFP，包装的重组慢病毒滴度达10^6IU／ml。以pXZ208-IRES—GFP为阴性对照组，共培养法感染CD4^＋CD25-γ细胞，实验组细胞Foxp3、CD25、CTLA-4、GITR表达升高。在抗CD3e单抗、抗原呈递细胞存在条件下，实验组细胞增殖能力及IL-2、IL-4、IL-10和IFN-γ的分泌均低于阴性对照组（P〈0．01）；且该细胞可显著抑制CI）4^＋CD25-T细胞的增殖。结论成功构建了自身失活型慢病毒载体介导的鼠基因工程Treg细胞；基因工程Treg细胞具有与CD4^＋ CD25^＋ Treg细胞相似的表型及低增殖性和免疫抑制性。

新生隐球菌是临床重要致病真菌之一,主要导致获得性免疫缺陷综合征(AIDS)、器官移植等免疫抑制病人产生隐球菌病.它的主要致病途径为鸽粪、桉树等中的隐球菌播散入空气,经呼吸道吸入后,多数产生一过性肺炎,被肺泡巨噬细胞吞噬,进入血液,形成隐球菌菌血症.可能由于颅内的高浓度的儿茶酚胺、菌与胶质细胞的配受体作用及其他可能的机制,新生隐球菌可表现为嗜神经性,临床上表现为隐球菌性脑膜炎或脑膜脑炎,症状重、病死率高.据统计,非洲地区AIDS病人的隐球菌发病率高达30%,是AIDS的三大致病死因之一.临床上最重要的隐球菌性脑膜炎,病人症状重、治疗困难、病死率高[1,2].目前认为巨噬细胞是抗新生隐球菌的第一线作用细胞,并且新生隐球菌可以在巨噬细胞内长期生存[3].因此,新生隐球菌如何在巨噬细胞长期生存是其致病的关键环节,对此进行深入的研究具有重要意义.

目的探讨吻合血管的足部跖趾及指间关节移植修复手部掌指关节及指间关节缺损的临床疗效。方法自1993年4月至2010年11月，分别对4例掌指关节及5例近位指间关节和3例掌指关节连同近位指间关节一并缺损的患者，应用吻合血管的第二跖趾关节、近位趾间关节或同一足趾的跖趾关节连同近位指间关节一并移植进行修复。结果12例关节移植均全部成活，术后创口均一期愈合，移植关节的骨愈合时间为5-10周，术后随访时间为4-28个月。移植关节均无明显骨及关节的退变。其修复后的掌指关节伸屈活动为0°-70°，近位指间关节的伸屈活动10°-90。，参照关节活动度TAM／TAF评定标准评定：优5例，良4例，可2例，差1例，优良率为75％。结论应用吻合血管的跖趾关节及趾问关节移植修复掌指关节及指间关节缺损，其愈后功能恢复比较满意，关节活动能达到基本正常的工作和生活需求。可获得较好的关节功能。
