A deformability-based biochip for precise label-free stratification of metastatic subtypes using deep learning

Cellular deformability is a promising biomarker for evaluating the physiological state of cells in medical *** has emerged as a powerful technique for measuring cellular ***,existing microfluidic-based assays for measuring cellular deformability rely heavily on image analysis,which can limit their scalability for high-throughput ***,we develop a parallel constriction-based microfluidic flow cytometry device and an integrated computational framework(ATMQcD).The ATMQcD framework includes automatic training set generation,multiple object tracking,segmentation,and cellular deformability *** system was validated using cancer cell lines of varying metastatic potential,achieving a classification accuracy of 92.4%for invasiveness assessment and stratifying cancer cells before and after hypoxia *** ATMQcD system also demonstrated excellent performance in distinguishing cancer cells from leukocytes(accuracy=89.5%).We developed a mechanical model based on power-law rheology to quantify stiffness,which was fitted with measured data *** model evaluated metastatic potentials for multiple cancer types and mixed cell populations,even under realworld clinical *** study presents a highly robust and transferable computational framework for multiobject tracking and deformation measurement tasks in *** believe that this platform has the potential to pave the way for high-throughput analysis in clinical applications,providing a powerful tool for evaluating cellular deformability and assessing the physiological state of cells.

Tailored Finite Point Method for Numerical Solutions of Singular Perturbed Eigenvalue Problems

We propose two variants of tailored finite point(TFP)methods for discretizing two dimensional singular perturbed eigenvalue(SPE)problems.A continuation method and an iterative method are exploited for solving discretized systems of equations to obtain the eigen-pairs of the *** study the analytical solutions of two special cases of the SPE,and provide an asymptotic analysis for the *** theoretical results are verified in the numerical *** numerical results demonstrate that the proposed schemes effectively resolve the delta function like of the eigenfunctions on relatively coarse grid.

MicroRNA-142-3p and microRNA-142-5p are downregulated in hepatocellular carcinoma and exhibit synergistic effects on cell motility

MicroRNAs （miRNAs）, an important class of small non-coding RNAs, regulate gene expression at the post-transcriptional level, miRNAs are involved in a wide range of biological processes and implicated in different diseases, including cancers. In this study, miRNA profiling and qRT-PCR validation revealed that miR-142-3p and miR-142-5p were significantly downregulated in hepatocellular carcinoma （HCC） and their expression levels decreased as the disease progressed. The ectopic expression of miR-142 significantly reduced HCC cell migration and invasion. Overexpression of either miR-142-3p or miR-142-5p suppressed HCC cell migration, and overexpression of both synergistically inhibited cell migration, which indicated that miR-142-3p and miR-142-5p may cooperatively regulate cell movement, miR-142-3p and miR-142-5p, which are mature miRNAs derived from the 3＇- and 5＇-strands of the precursor miR-142, target distinct pools of genes because of their different seed sequences. Pathway enrichment analysis showed a strong association of the putative gene targets of miR-142-3p and miR-142-5p with several cell motility-associated pathways, including those regulating actin cytoskeleton, adherens junctions, and focal adhesion. Importantly, a number of the putative gene targets were also significantly upregulated in human HCC cells. Moreover, overexpression of miR-142 significantly abrogated stress fiber formation in HCC cells and led to cell shrinkage. This study shows that mature miR-142 pairs collaboratively regulate different components of distinct signaling cascades and therefore affects the motility of HCC cells.

Images are generally corrupted by impulse noise during acquisition and *** deteriorates the quality of *** remove corruption noise,we propose a hybrid approach to restoring a random noisecorrupted image,including a block matching 3D(BM3D)method,an adaptive non-local mean(ANLM)scheme,and the K-singular value decomposition(K-SVD)*** the proposed method,we employ the morphological component analysis(MCA)to decompose an image into the texture,structure,and edge ***,the BM3D method,ANLM scheme,and K-SVD algorithm are utilized to eliminate noise in the texture,structure,and edge parts of the image,*** results show that the proposed approach can effectively remove interference random noise in different parts;meanwhile,the deteriorated image is able to be reconstructed well.

Chronic hepatitis B virus(HBV)infection(CHB)is a public health concern *** therapies utilizing nucleos(t)ide analogs(NA)have not resulted in a complete cure for ***,patients on long-term NA treatment often develop low-level viremia(LLV).Persistent LLV,in addition to causing the progression of liver disease or hepatocellular carcinoma,may shed light on the current plight of NA ***,we review the literature on LLV,NA treatment,and various doses of entecavir to find a strategy for improving the efficacy of this antiviral *** LLV patients,three therapeutic options are available,switching to another antiviral monotherapy,interferon-αswitching therapy,and continuing *** real-world clinical practice,entecavir overdose has been used in antiviral therapy for CHB patients with NA refractory and persistent LLV,which encouraged us to conduct further in-depth literature survey on dosage and duration related entecavir *** studies of pharmacodynamics and pharmacokinetics show that entecavir has the maximal selected index for safety,and has great potential in inhibiting HBV replication,in all of the *** the particular section of the drug approval package published by the United States Food and Drug Administration,entecavir doses 2.5-20 mg/d do not increase adverse events,and entecavir doses higher than 1.0 mg/d might improve the antiviral *** literature survey led us to two suggestions:(1)Increasing entecavir dose to 1.0 mg/d for the treatment of NA naïve patients with HBV DNA>2×106 IU/mL is feasible and would provide better prognosis;and(2)Further research is needed to assess the long-term toxic effects of higher entecavir doses(2.5 and 5.0 mg/d),which may prove beneficial in treating patients with prior NA treatment,partial virological response,or LLV state.

AIM To explore the relationship between colonic secretory function and colonic *** Using a rat model chronically implanted with intracerebroventricular(ICV) and cecal catheters, we validated the correlation between colonic secretion and colonic motor functions, as well as the role of ICV injection *** Compared to saline controls(5 μL/rat), ICV acyl ghrelin at 1 nmol/5 μL enhanced the total fecal weight, accelerated the colonic transit time, and increased the fecal pellet output during the first hour post-injection, while ICV des-acyl ghrelin at 1 nmol/5 μL only accelerated the colonic transit time. These stimulatory effects on colonic motility and/or secretion from acyl ghrelin and des-acyl ghrelin disappeared when the ICV injection volume increased to 10 μL compared with saline controls(10 μL/rat). Additionally, the ICV injection of 10 μL of saline significantly shortened the colonic transit time compared with the ICV injection of 5 μL of saline. The total fecal weight during the first hour post-injection correlated with the colonic transit time and fecal pellet output after the ICV injection of acyl ghrelin(1 nmol/5 μL), whereas the total fecal weight during the first hour post-injection correlated with the fecal pellet output but not the colonic transit time after the ICV injection of des-acyl ghrelin(1 nmol/5 μL).CONCLUSION Colonic secretion does not always correlate with colonic motility in response to different colonic stimulations. Acyl ghrelin stimulates colonic secretion.
