A few of snake venom phospholipases A have been demonstrated to inhibit the binding of the antagonists to muscarinic acetylcholine receptor(mAChR).However,their pharmacological effect on mAChR has been unknown so *** this study,the effect of a phospholipase A from the venom of Naja atra(Chinese cobra)with high affinity for mAChR on guinea-pig ileum was *** active principle,called muscarinic protein(MP),was isolated by column chromatography on Sephadex G-50,Sephadex G-150, CM-Sepharose Fast Flow,and PorosCM 4.6/100 Perfusion Chromatography *** final preparation was homogeneous as determined by SDS-PAGE and *** analysis showed one peak of major protein with the molecular weight of 13.3 *** N-terminal amino acid sequence of MP was shown to be highly homologous with that of phospholipase A from the venom of genus *** MP hydrolyzed phosphatidylcholine in a dose-and a time-dependent manner,and was determined as a snake venom phospholipase *** MP inhibited the muscarinic antagonist,[H]quinuclidinyl benzilate([H]QNb),binding to rat cortex synaptosomes in a dose-dependent *** IC and K value of its antagonist-displacing activity is about 28.0 nM and 10.1 nM,*** the longitudinal muscle preparation from guinea-pig ileum,the MP produced a rapid and dose-dependent contraction,which persisted for more than 120 *** EC value of MP was determined as 30 riM,which is similar to that of carbachol(48 nM).However,the maximum force of contraction caused by MP was only approximately 43% of that obtained from carbachol. The action of MP was reversed by 1 μM atropine indicating the involvement of *** findings suggest for the first time that the phospholipase A(MP)with high affinity for mAChR from Chinese cobra venom may be a muscarinic agonist.

In this study,a high performance liquid chromatography(HPLC)coupled with a variable wavelength detector(VWD)has been developed for the simultaneous determination of three anthraquinones(including emodin,physcion,emodine-8-O-β-D-glucoside)and one stilbene((E)-2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside)in prepared roots of Polygonum multiflorum Thunb.(***).It is widely used in the prescription of traditional Chinese medicine(TCM),and also edible as an *** method was simple,reliable and reproducible,and was successfully applied to determine the four target compounds in the prepared *** for the quality *** addition,it was the first time reported to identify another one naphthalene derivation and three stilbenes by this ESI/MS method.

A new,rapid and sensitive method was developed for the quantifying of rhodamine 123(R123) in rat plasma using liquid chromatography tandem mass spectrometry(LC-MS/MS).Rhodamine 6G(R6G) was used as the internal standard(IS).R123 and IS were extracted from aliquots of plasma with ethyl acetate and dichloromethane (4:1) as the *** separation was performed using a Zorbax Eclipse Plus C18 *** mobile phase was composed of A:ammonium formate-formic acid buffer containing 5 mM ammonium formate and 0.1%formic acid and b:methanol(A:b,5:95,v/v).To quantify R123 and IS respectively,multiple reaction monitoring(MRM) transition of m/z 345.2→285.2 and m/z 443.3→415.2 were *** analysis lasted for 4 min,in the positive mode,the results of which were presented as a linear calibration curve over the concentration range of 1-200 ng/ml and sampling volume of 2μ*** lowest limit of quantification(LLOQ) reached 1 ng/*** intra and inter-day precision were 6.7%and 9.2%for the low quality control(QC) sample (2 ng/ml),*** and inter-day precision of the medium QC sample(100 ng/ml) were both less than 3.4%.Intra and inter-day precision of high QC sample(150 ng/ml) were both less than 2.1%,while the intra and inter-day relative errors ranged between -7.4%and 9.1%for the above three QC concentration *** LC-MS/MS method proved to be simple,accurate,reliable and with a shorter running time and has been successfully applied to an absorption experiment in the rat.

bruton’s tyrosine kinase inhibitors(bTKis)have revolutionized the treatment of b-cell ***,safety issues related to the use of bTKis may hinder treatment continuity and further affect clinical efficacy.A comprehensive and systematic expert consensus from a pharmacological perspective is lacking for safety issues associated with bTKi treatment.A multidisciplinary consensus working group was established,comprising 35 members from the fields of hematology,cardiovascular disease,cardio-oncology,clinical pharmacy,and evidencebased *** evidence-based expert consensus was formulated using an evidence-based approach and the Delphi *** Joanna briggs Institute Critical Appraisal(JbI)tool and Grading of Recommendations Assessment,Development,and Evaluation(GRADE)approach were used to rate the quality of evidence and grade the strength of recommendations,*** consensus provides practical recommendations for bTKis medication based on nine aspects within three domains,including the management of common adverse drug events such as bleeding,cardiovascular events,and hematological toxicity,as well as the management of drug-drug interactions and guidance for special *** multidisciplinary expert consensus could contribute to promoting a multi-dimensional,comprehensive and standardized management of bTKis.

Role of Apolipoprotein A-I in Protecting against Endotoxin Toxicity

High density lipoprotein (HDL) binds lipopolysaccharide (LPS or endotoxin) and neutralizes its toxicity. We investigated the function of Apolipoprotein A-I (ApoA-I), a major apolipoprotein in HDL, in this process. Mouse macrophages were incubated with LPS, LPS+ApoA-I, LPS+ApoA-I+LFF (lipoprotein-free plasma fraction d>1.210 g/ml), LPS+HDL, LPS+HDL+LFF, respectively. MTT method was used to detect the mortality of L-929 cells which were attacked by the release-out cytokines in LPS-activated macrophages. It was found that ApoA-I significantly decreased L-929 cells mortality caused by LPS treatment (LPS vs. LPS+ApoA-I, Pbecame even more significant when LFF was utilized (LPS vs. LPS+ApoA-I+LFF, Pbetween LPS+ApoA-I+LFF and LPS+HDL+LFF treatment, indicating that ApoA-I was the main factor. We also investigated in vivo effects ofApoA-I on mouse mortality rate and survival time after LPS administration. We found that the mortality in LPS+ApoA-I group (20%) and in LPS+ApoA-I+LFF group (10%) was signifi- cantly lower than that in LPS group (80%) (Pbinding study to investigate the binding capacity of ApoA-I and ApoA-I+LFF to fluorescence labeled LPS (FITC-LPS). It was shown that both ApoA-I and ApoA-I+LFF could bind with FITC-LPS, however, the binding capacity of ApoA-I+LFF to FITC-LPS (64.47 ± 8.06) was significantly higher than that of ApoA-I alone (24.35 ± 3.70) (Pbility to bind with and protect against LPS; (2) LFF enhances the effect of ApoA-I; (3) ApoA-I is the major contributor for HDL anti-endotoxin function.

Under physiological conditions, γ-aminobutyric acid poorly crosses the blood-brain barrier. It is likely that a non-toxic derivative of γ-aminobutyric acid which enters the brain easily will have useful anticonvulsant activity. 16 derivatives of γ-aminobutyric acid with an imine link to a lipophilic carrier were prepared and tested for anticonvulsant activity; six compounds show anticonvulsant activity.
