Transcriptome profiling reveals regulatory mechanisms underlying corolla senescence in petunia

The genetic regulatory mechanisms that govern natural corolla senescence in petunia are not well *** identify key genes and pathways that regulate the process,we performed a transcriptome analysis in petunia corolla at four developmental stages,including corolla fully opening without anther dehiscence(D0),corolla expansion,2 days after anthesis(D2),corolla with initial signs of senescence(D4),and wilting corolla(D7).We identified large numbers of differentially expressed genes(DEGs),ranging from 4626 between the transition from D0 and D2,1116 between D2 and D4,a transition to the onset of flower senescence,and 327 between D4 and D7,a developmental stage representing flower *** analysis showed that the auxin-and ethylene-related hormone biosynthesis and signaling transduction pathways were significantly activated during the flower development and highly upregulated at onset of flower *** emission was detected at the D2 to D4 transition,followed by a large eruption at the D4 to D7 ***,large numbers of transcription factors(TFs)were activated over the course of *** analysis by virus-induced gene silencing(VIGS)experiments demonstrated that inhibition of the expression of TFs,such as ethylene-related ERF,auxin-related ARF,bHLH,HB,and MADS-box,significantly extended or shortened flower *** data suggest that hormonal interaction between auxin and ethylene may play critical regulatory roles in the onset of natural corolla senescence in petunia.

Gastric polyposis is a rare disease. Not all polyps progress to cancer. Monoallelic mutation in Fanconi anemia(FA) genes, unlike biallelic gene mutations that causes typical FA phenotype, can increase risks of cancers in a sporadic manner. Aberrations in the FA pathway were reported in all molecular subtypes of gastric cancer. We studied a patient with synchronous gastric cancer from gastric polyposis by conducting a 13-year long-term follow up. Via pathway-driven massive parallel genomic sequencing, a germline mutation at FANCA D1359Y was identified. We identified several recurrent mutations in DNA methylation(TET1, V873I), the β-catenin pathway(CTNNB1, S45F) and RHO signaling pathway(PLEKHG5, R203C) by comparing the genetic events between benign and malignant gastric polyps. Furthermore, we revealed gastric polyposis susceptible genes and genetic events promoting malignant transformation using pathway-driven targeted gene sequencing.

Training gastroenterology fellows to perform gastric polypectomy using a novel ex vivo model

AIM:To evaluate the effect of hands-on training of gastroenterology fellows in gastric polypectomy using an ex vivo ***:Eight gastroenterology fellows at Mackay Memorial Hospital,Taipei were evaluated in gastricpolypectomy techniques using a pig stomach with artificial polyps created by a rubber band ligation *** performance of four second year(year-2)fellows who had undergone one year of clinical training was compared with that of four f irst year(year-1)fellows both before and after a 4-h workshop using the ex vivo *** workshop allowed for hands-on train-ing in the removal of multiple artif icial polyps and the placement of hemoclips at the excision *** included observation of technical skills,procedure time,and the fellows' conf idence ***:One week after the workshop,the year-1 fellows were re-evaluated and had significantly im-proved mean performance scores(from 17.9 ± 1.8 to 22.5 ± 0.7),conf idence scale(from 4.5 ± 1.0 to 7.8 ± 0.5)and procedure time(from 615.0 ± 57.4 s to 357.5 ± 85.0 s)compared with their baseline *** 4 h of training using the ex vivo simulator,the skills of the year-1 fellows were statistically similar to those of the year-2 ***:Use of this ex vivo simulator significantly improved the endoscopic gastric polypectomy skills of gastroenterology fellows who had not had previous clinical training in gastric polypectomy.

Enhancement of Swimming Endurance by Herbal Supplement M3P

Objective:To investigate the effect of M3P(containing Deer antler,Cordyceps sinensis,Rhodiola rosea,and Panax ginseng);an herbal remedy with the function of tonifying Kidney(Shen)and invigorating Spleen(Pi),replenishing qi and nourishing blood;on fatigue alleviation,endurance capacity and ***:Swimming with weight-loading of 24 male ICR mice was used to evaluate the endurance capacity,and fatiguerelated plasma biomarkers were *** were randomly assigned to control or M3P treatment groups with 6 mice for each group and were orally administered with M3P everyday for 8 weeks at doses 0,10,33 or 100 mg/*** time to exhaustion was measured in a specialized water *** and kidney functions,body weight,and hematological profile were determined to evaluate the safety and toxicity after long-term M3P ***:M3P supplementation 100 mg/kg significantly increased swimming endurance time up to approximate 2.4 folds of controls(P0.05).The liver and kidney functions muscle damage biomarker creatine,body weight,and hemograms were not altered in M3P supplementation(P>0.05).Conclusion:M3P supplementation may improve swimming endurance accompanied by increasing hepatic glycogen content and serum cortisol level without major toxicity.

Cell polarization in ischemic stroke: molecular mechanisms and advances

Ischemic stroke is a cerebrovascular disease associated with high mortality and disability rates. Since the inflammation and immune response play a central role in driving ischemic damage, it becomes essential to modulate excessive inflammatory reactions to promote cell survival and facilitate tissue repair around the injury site. Various cell types are involved in the inflammatory response, including microglia, astrocytes, and neutrophils, each exhibiting distinct phenotypic profiles upon stimulation. They display either proinflammatory or anti-inflammatory states, a phenomenon known as ‘cell polarization.’ There are two cell polarization therapy strategies. The first involves inducing cells into a neuroprotective phenotype in vitro, then reintroducing them autologously. The second approach utilizes small molecular substances to directly affect cells in vivo. In this review, we elucidate the polarization dynamics of the three reactive cell populations(microglia, astrocytes, and neutrophils) in the context of ischemic stroke, and provide a comprehensive summary of the molecular mechanisms involved in their phenotypic switching. By unraveling the complexity of cell polarization, we hope to offer insights for future research on neuroinflammation and novel therapeutic strategies for ischemic stroke.

Pancreatic stent improves the success rate of needle-knife papillotomy in patients with difficult biliary cannulation

BACKGROUND Needle-knife precut papillotomy(NKP)is typically performed ***,it remains unclear whether pancreatic stent(PS)placement can improve the outcomes of *** To explore whether PS placement improves the success rate of NKP in patients with difficult biliary *** This single-center retrospective study included 190 patients who underwent NKP between January 2017 and December 2021 after failed conventional biliary *** cases with incidental pancreatic duct cannulation during conventional biliary cannulation,the decision for pre-NKP PS placement was made at the endoscopist's *** primary outcome was the difference in the NKP success rate between patients with and without PS placement;the secondary outcome was the adverse event *** Among the 190 participants,82 received pre-NKP PS(PS-NKP group)whereas 108 did not[freehand or freehand NKP(FH-NKP)group].Post-NKP selective biliary cannulation was successful in 167(87.9%)patients,and the PS-NKP had a significantly higher success rate than the FH-NKP group(93.9%vs 83.3%,P=0.027).The overall adverse event rates were 7.3%and 11.1%in the PS-NKP and FH-NKP groups,respectively(P=0.493).A periampullary diverticulum(PAD)and significant intraoperative bleeding during NKP were independently associated with NKP failure;however,a pre-NKP PS was the only predictor of NKP *** the 44 participants with PADs,the PS-NKP group had a non-significantly higher NKP success rate than the FH-NKP group(87.5%and 65%,respectively;P=0.076).CONCLUSION PS significantly improved the success rate of NKP in patients with difficult biliary cannulation.

Brain-derived neurotrophic factor is a key factor in stress adaptation and avoidance of a social stress behavioral *** studies have shown that brain-derived neurotrophic factor expression in stressed mice is brain region–specific,particularly involving the corticolimbic system,including the ventral tegmental area,nucleus accumbens,prefrontal cortex,amygdala,and *** how brain-derived neurotrophic factor participates in stress processing in different brain regions will deepen our understanding of social stress *** this review,we discuss the expression and regulation of brain-derived neurotrophic factor in stress-sensitive brain regions closely related to the pathophysiology of *** focused on associated molecular pathways and neural circuits,with special attention to the brain-derived neurotrophic factor–tropomyosin receptor kinase B signaling pathway and the ventral tegmental area–nucleus accumbens dopamine *** determined that stress-induced alterations in brain-derived neurotrophic factor levels are likely related to the nature,severity,and duration of stress,especially in the above-mentioned brain regions of the corticolimbic ***,BDNF might be a biological indicator regulating stress-related processes in various brain regions.

Effect of boundary conditions on downstream vorticity from counter-rotating swirlers

Particle image velocimetry（PIV） is utilized to measure the non-reacting flow field in a reflow combustor with multiple and single swirlers. The velocity field, vortex structure and total vorticity levels are experimentally obtained using two different boundary conditions, representing a single confined swirler and multiple swirlers in an annular combustor. The influence of the boundary conditions on the flow field at several locations downstream of the swirlers is experimentally investigated, showing that the central vortex in the multi-swirler case is more concentrated than in the single-swirler case. The vorticity of the central vortex and average cross-sectional vorticity are relatively low at the swirler outlet in both cases. Both of these statistics gradually increase to the maximum values near 20 mm downstream of the swirler outlet, and subsequently decrease. It is also found that the central vortex in the multi-swirler case is consistently greater than the single-swirler case. These results demonstrate the critical influence of boundary conditions on flow characteristic of swirling flow, providing insight into the difference of the experiments on testbed combustor and the full-scale annular combustors.

Unraveling the therapeutic potential of Calculus Bovis in liver cancer:A novel step for targeted cancer treatment

Hepatocellular carcinoma is one of the leading causes of cancer-related deaths globally,and effective treatments are urgently *** present study aimed to investigate the inhibitory effect of Calculus Bovis(CB)on liver cancer and the underlying *** inhibited M2 tumor-associated macrophage polarization and modulated the Wnt/β-catenin signaling pathway,thereby suppressing the proliferation of liver cancer *** inhibitory effect on liver cancer growth was confirmed by both in vivo and in vitro experiments(detailed by Huang et al).The present study provides a theoretical basis for the application of CB for the treatment of liver cancer,providing new avenues for liver cancer treatment.
