目的：通过体内实验和体外实验的方法研究aquaporin5的表达及其与不同分化程度肺癌的关系。方法：对于肺癌组织，采用免疫组织化学的方法检测高、低分化程度的肺鳞癌和肺腺癌细胞AQP5的表达，采用TUNEL的方法检测高、低分化程度的肺鳞癌和肺腺癌细胞的凋亡指数。对于体外实验，采用全反式维甲酸诱导A549细胞形成高、低分化的肺腺癌细胞模型来进行AQP5的表达和凋亡关系的研究。采用MTT法测试A549的细胞增殖;采用Periodic Acid Schiff染色和CEA的免疫组织化学的方法来鉴定细胞模型。采用免疫荧光的方法检测细胞模型的AQP5的表达，采用流式细胞术、Caspase3和Hoechst33342荧光染色来测试细胞模型的细胞凋亡指数。结果：对于肺鳞癌组织，高分化鳞癌的AQP5光密度(0,283±0,02)显著高于低分化鳞癌的AQP5光密度(0,219±0,05)(p<0.05);高分化肺鳞癌细胞的直径(27,02±0,83μm)显著高于低分化肺鳞癌细胞的直径(17,85±0,82μm)(p<0.05)。对于肺腺癌组织，高分化肺腺癌的AQP5光密度(0,281±0,03)显著高于低分化肺腺癌的AQP5光密度(0,238±0,02)(p<0.05).;高分化肺腺癌细胞的直径(13,60±0,82μm)显著低于低分化肺腺癌细胞的直径(28,01±1,01μm)(p<0.05)。高分化肺腺癌细胞的凋亡指数(20,20±2,40)显著高于低分化肺腺癌细胞的凋亡指数(18,53±1,73)(p<0.05)。相关分析检测结果显示：高、低分化肺腺癌细胞的AQP5表达与细胞凋亡指数之间呈显著正相关(高分化肺腺癌r=0.524，p=0.045;低分化肺腺癌r=0.661， p=0.007);高、低分化的肺鳞癌细胞的AQP5表达与细胞凋亡指数之间无显著相关关系(高分化鳞癌r=-0.069，p=0.807;低分化鳞癌， r=-0.299;p=0.279)。对于体外实验，全反式维甲酸以剂量依赖方式降低细胞的增殖，同时，AQP5的表达也依赖细胞的分化。Caspase3和流式细胞仪测试测试结果也呈现维甲酸的剂量依赖。AQP5表达的光密度和细胞凋亡指数（AI）的测试结果如下：对照组（(MOD=1,29±0,14;AI=1,01%)，5μg/ml组(MOD=1,31±0,15;AI=5,67%)，10μg/ml组(MOD=1,32±0,10;AI=6,13%) and15μg/ml组(MOD=1,33±0,12;AI=7,15%)。A549的AQP5的表达与其凋亡指数呈显著正相关(r=0.970;p=0.0304)。结论：AQP5的表达无论是在体外还是体内均呈肿瘤类型和肿瘤分化程度的依赖。在高、低分化肺腺癌组织，随着AQP5表达的增加，其细胞凋亡也在增加;而在高、低分化鳞癌组织，AQP5的表达与其细胞凋亡之间无显著的相关性。腺癌的凋亡指数较鳞癌的高。在体外实验，全反式维甲酸能够促进A549细胞的分化、抑制其增殖和促进AQP5的表达，同时，促进A549的细胞凋亡。上述研究表明，AQP5的高表达能够促进肺腺癌的凋亡。

An Analysis of Pain Following Traumatic Spinal Cord Injury among Adults in Zimbabwe

The purpose of the study was to analyse on pain following traumatic spinal cord injury, its prevalence, the types of pain present, the common treatments used and their perceived effectiveness in the management of these reported pain types. A cross sectional study was carried out at St Giles Rehabilitation Centre and from members of the Spinal Injuries Association of Zimbabwe (SIAZ). A researcher-administered questionnaire was used to collect data from 24 participants with traumatic spinal cord injury. The questionnaire elicited information on demographic data, pain characteristics and the perceived effectiveness of the common treatments used. Among the 24 participants in the study, 17 were males (70.8%) and 7 were females (29.2%). Pain prevalence was 79.2% among the study participants and approximately a fifth (21.03%) of all participants rated their pain as severe. Eight (33.3%) of the participants had neuropathic pain while 11 (45.8%) had both nociceptive and neuropathic pain types. However, no association was found between sex, age, time post injury when tested against the presence of pain (p > 0.05). Weather changes aggravated almost every type of pain reported. Both pharmacological and non-pharmacological methods were used to manage the pain but their perceived effectiveness was rated as low. The majority of the traumatic spinal cord injured people experienced some pain and this pain was severe in a fifth of all participants. Pain significantly affected their quality of life. Physiotherapists and other medical professionals need to be aware of this and should employ pain reducing modalities and empathy when dealing with these patients.

Aquaporin 5 Expression and Its Relationship to Apoptosis in Different Grades of Differentiated Non-Small Cell Lung Carcinoma

Aquaporin 5 has been recently found as an important oncogenic marker whose expression levels seem to be determined by the level of cellular differentiation. Despite aquaporin volume decrease (AVD) being the most conserved earliest event in apoptosis, there is still a paucity of studies exploring on aquaporin expression and its relationship with apoptosis in cancer. The aim of this study was to investigate the expression of aquaporin 5 channel protein and to explore on its relationship with apoptosis in well and poorly differentiated non-small cell lung carcinoma both in-vivo and in-vitro. Findings from the study showed that the expression of AQP5 both in-vivo and in-vitro was dependent on the type and degree of tumour differentiation. In-vivo, an increase in aquaporin 5 expression was associated with an increased apoptosis in both poorly and highly differentiated adenocarcinoma (AC) while there was no association between aquaporin 5 expression and apoptosis in both poorly and highly differentiated squamous cell carcinoma (SCC). In vitro, differentiation therapy in the form of ATRA decreased both cell proliferation and increased the expression of AQP5 in A549 cells. The cytomorphological changes, expression of differentiation markers and flow cytometry apoptotic results were dependent on the dose of ATRA treatment. In conclusion, a higher expression of aquaporin 5 was found to promote the rate of the apoptotic process in lung adenocarcinoma (AC).
