Impact of chronic exposure to bevacizumab on EpCAM-based detection of circulating tumor cells

Background： Circulating tumor cells （CTCs） are often undetected through the immunomagnetic epithelial cell adhesion molecule （EpCAM）-based CellSearch~ System in breast and colorectal cancer （CRC） patients treated with bevacizumab （BEv）, where low CTC numbers have been reported even in patients with evidence of progression of disease. To date, the reasons for this discrepancy have not been clarified. This study was carried out to investigate the molecular and phenotypic changes in CRC cells after chronic exposure to BEv in vitro. Methods： The human CRC cell line WiDr was exposed to a clinically relevant dose of BEv for 3 months in vitro. The expression of epithelial and mesenchymal markers and EpCAM isoforms was determined by western blotting and immunofluorescence. To evaluate the impact of EpCAM variant isoforms expression on CTC enumeration by CellSearch, untreated and treated colon cancer cells were spiked into 7.5 mL of blood from a healthy donor and enumerated by CellSearch. Results： Chronic exposure of CRC cell line to BEv induced decreased expression of EpCAM 40 kDa isoform and increased expression EpCAM 42 kDa isoform, together with a decreased expression of cytokeratins （CK）, while no evidence of epithelial to mesenchymal transition （EMT） in treated cells was observed. The recovery rate of cells through CellSearch was gradually reduced in course of treatment with BEv, being 84% , 70% and 40% at l, 2 and 3 months, respectively. Conclusions： We hypothesize that BEv may prevent CellSearch from capturing CTCs through altering EpCAM isoforms.

The efficiency of photo-electrocatalytic（PECa） devices for the production of solar fuels depends on several limiting factors such as light harvesting, charge recombination and mass transport diffusion. We analyse here how they influence the performances in PECa cells having a photo-anode based on Au-modified TiOnanotube（TNT） arrays, with the aim of developing design criteria to optimize the photo-anode and the PECa cell configuration for water photo-electrolysis（splitting） and ethanol photo-reforming *** TNT samples were prepared by controlled anodic oxidation of Ti foils and then decorated with gold nanoparticles using different techniques to enhance the visible light response through heterojunction and plasmonic effects. The activity tests were made in a gas-phase reactor, as well as in a PECa cell without applied bias. Results were analysed in terms of photo-generated current, Hproduction rate and photoconversion efficiency. Particularly, a solar-to-hydrogen efficiency of 0.83% and a Faradaic efficiency of 91%were obtained without adding sacrificial reagents.

The venice Lagoon is exposed to atmospheric pollutants from industrial activities, thermoelectric power plants, petrochemical plants, incinerator, domestic heating, ship traffic, glass factories and vehicular emissions on the mainland. In 2005, construction began on the mobile dams （MOSE）, one dam for each channel connecting the lagoon to the Adriatic Sea as a barrier against high tide. These construction works could represent an additional source of pollutants. PM10 samples were taken on random days between 2007 and 2010 at three different sites： Punta Sabbioni, Chioggia and Malamocco, located near the respective dam construction worksites. Chemical analyses of v, Cr, Fe, Co, Ni, Cu, Zn, As, Mo, Cd, Sb, T1 and Pb in PM10 samples were performed by Inductively coupled plasma- quadrupole mass spectrometry （ICP-QMS） and results were used to identify the main aerosol sources. The correlation of measured data with meteorology, and source apportionment, failed to highlight a contribution specifically associated to the emissions of the MOSE construction works. The comparison of the measurements at the three sites showed a substantial homogeneity of metal concentrations in the area. Source apportionment with principal component analysis （PCA） and positive matrix factorization （PMF） showed that a four principal factors model could describe the sources of metals in PM10. Three of them were assigned to specific sources in the area and one was characterised as a source of mixed origin （anthropogenic and crustal）. A specific anthropogenic source of PM10 rich in Ni and Cr, active at the Chioggia site, was also identified.

We report the charge-changing cross sections(σ_(cc))of 24 p-shell nuclides on both hydrogen and carbon at about 900A Mev,of which^(8,9)Li,^(10–12)Be,^(10,14,15)B,^(14,15,17–22)N and^(16)O on hydrogen and^(8,9)Li on carbon are for the first *** from the data set,we found a new and robust relationship between the scaling factor of the Glauber model calculations and the separation energies of the nuclei of interest on both *** allows us to deduce proton radii(R_p)for the first time from the cross sections on *** identical R_p values are deduced from both target data for the neutron-rich carbon isotopes;however,the R_p from the hydrogen target is systematically smaller in the neutron-rich nitrogen *** calls for further experimental and theoretical investigations.

Effective management of hard-to-close skin wounds is a challenging issue due to several co-morbidities in affected ***,infections represent a major obstacle in wound *** design of efficient wound treatments thus represents an urgent *** drug delivery hydrogels with intrinsic antimicrobial and antifungal properties were herein designed for perspective application in the mini-invasive treatment of hard-to-close ***,an amphiphilic polyurethane was synthesized from Poloxamer■407 macrodiol and N-Boc diethanolamine chain extender(DHP407,M_(w)=33 kDa).Chain-extension reaction step was optimized to maximize the formation of-NH groups along the polymer chains(4.5×10^(20)±1.8×10^(19)-NH groups/g polymer),after Boc-caging group removal(D-DHP407).DHP407 and D-DHP407 water-based solutions were thermosensitive with slightly different Critical Micellar Concentration(17.5μg/mL vs.19.7μg/mL)and cluster hydrodynamic diameter(235.6±19.9 nm vs.260.1±20.5 nm),and similar Critical Micellar Temperature(22.5℃ vs.23.1℃).A polyurethane solution concentration(15%w/v)was selected by tube-inverting test and rheological analysis showing injectability,as evidenced by sol-to-gel transition at 27.7±0.6℃ for DHP407 and 29.7±0.6℃ for D-DHP407,within few minutes,at similar gelation ***407 and D-DHP407 hydrogels showed controlled release of Bovine Serum Albumin(BSA)model protein(1 mg/mL),with no burst *** released from DHP407 and D-DHP407 hydrogels at 24 h was 33.7±5.0% and 24.6±1.2%,respectively.D-DHP407 hydrogel was biocompatible and able to support NIH-3T3 cell ***,D-DHP407 hydrogel showed intrinsic antifungal and antibacterial activity against *** and Gram-positive *** and Gram-negative *** bacteria,injectability and capability to retain shape post-injection,making it promising for future use in the management of hard-to-close skin wounds.

Electrocatalytic conversion of CO_2 to liquid fuels using nanocarbon-based electrodes

Recent advances on the use of nanocarbon-based electrodes for the electrocatalytic conversion of gaseous streams of CO2 to liquid fuels are discussed in this perspective paper. A novel gas-phase electrocatalytic cell, different from the typical electrochemical systems working in liquid phase, was developed. There are several advantages to work in gas phase, e.g. no need to recover the products from a liquid phase and no problems of CO2 solubility, etc. Operating under these conditions and using electrodes based on metal nanoparticles supported over carbon nanotube （CNT） type materials, long C-chain products （in particular isopropanol under optimized conditions, but also hydrocarbons up to C8-C9） were obtained from the reduction of CO2. Pt-CNT are more stable and give in some cases a higher productivity, but Fe-CNT, particular using N-doped carbon nanotubes, give excellent properties and are preferable to noble-metal-based electrocatalysts for the lower cost. The control of the localization of metal particles at the inner or outer surface of CNT is an importact factor for the product distribution. The nature of the nanocarbon substrate also plays a relevant role in enhancing the productivity and tuning the selectivity towards long C-chain products. The electrodes for the electrocatalytic conversion of CO2 are part of a photoelectrocatalytic （PEC） solar cell concept, aimed to develop knowledge for the new generation artificial leaf-type solar cells which can use sunlight and water to convert CO2 to fuels and chemicals. The CO2 reduction to liquid fuels by solar energy is a good attempt to introduce renewables into the existing energy and chemical infrastructures, having a higher energy density and easier transport/storage than other competing solutions （i.e. H2）.

The novel coronavirus disease(COvID-19)has hit the healthcare system *** risk of severe infection and mortality increases with advancing age,especially in subjects with comorbidities such as cardiovascular disease,hypertension,diabetes,obesity and ***,cardiovascular complications such as myocardial injury,heart failure and thromboembolism are frequently observed in COvID-19 cases,and several biomarkers(troponin,NTpro BNP and D-Dimer)have been identified as prognostic indicators of disease severity and worst ***,there is no specific therapy against SARS-Co v-2,although many medications are under *** aim of this review will be to explore the intertwined relationship between COvID-19 disease and the cardiovascular system,focusing on elderly *** available supportive treatments along with the related concerns in elderly patients,due to their comorbidities and polypharmacotherapy,will be explored.
