Variation of a floral polymorphism at different spatial scales in the Mediterranean geophyte Narcissus assoanus

Aims Habitat fragmentation impacts the spatial extent and isolation of local populations and communities.Although the biological consequences of these impacts have been well studied at the site level,effects directly related to changes in the spatial configuration of populations in the landscape remain less clear.The objective of this study is to examine how changes in the spatial-scale configuration of populations are associated with variability in morph rations in the floral polymorphic Mediterranean geophyte Narcissus assoanus.Methods We performed a nested analysis of morph ratio variation at three spatial scales:a 50×50 km regional scale in SE France,in fourteen 1×1 km landscapes located in the same region,and within 12 spatially extensive population patches.We also quantified variation in the behaviour of pollinators in population patches of contrasting spatial configuration.Important FindingsAt a regional scale,morph ratios show a geographic pattern of increasing isoplethy(1:1 morph ratio)away from centres of human population development and in upland pastures.At the landscape scale,isoplethy of local population patches is more correlated with decreased isolation among patches than with patch size.Finally,within local isoplethic populations,small patches can show significantly biased morph ratios.In small isolated patches,pollinators perform shorter flight distances among consecutive flower visits than in spatially extensive patches.This study of variation in a genetic polymorphism at multiple spatial scales provides new insights into the scale-dependent effects of habitat fragmentation and the potential occurrence of metapopulation dynamics in natural plant populations.

AIM:To investigate the clinical course of untreatable hepatocellular carcinoma(HCC) identified at any stage and to identify factors associated with mortality.METHODS:From January 1999 to December 2010,320 out of 825 consecutive patients with a diagnosis of HCC and not appropriate for curative or palliative treatments were followed and managed with supportive therapy.Cirrhosis was diagnosed by histological or clinical features and liver function was evaluated according to Child-Pugh score.The diagnosis of HCC was performed by Ultra-Sound guided biopsy or by multiphasic contrast-enhanced computed tomography or gadolinium-enhanced magnetic resonance imaging.Data were collected for each patient including all clinical,laboratory and imaging variables necessary for the outcome prediction staging systems considered.HCC staging was performed according Barcelona Clinic Liver Cancer(BCLC) and Cancer of the Liver Italian Program scores.Follow-up time was defined as the number of months from the diagnosis of HCC to death.Prognostic baseline variables were analyzed by multivariate Cox analysis to identify the independent predictors of survival.RESULTS:Seventy-five per cent of patients had hepatitis C.Ascites was present in 169 patients(53%),while hepatic encephalopathy was present in 49 patients(15%).The Child-Pugh score was class A in 105 patients(33%),class B in 142 patients(44%),and class C in 73 patients(23%).One hundred patients(31%) had macroscopic vascular invasion and/or extrahepatic spread of the tumor.A single lesion > 10 cm was observed in 34 patients(11%),while multinodular HCC was present in 189 patients(59%).Thirty nine patients(12%) were BCLC early(A) stage,55(17%) were BCLC intermediate(B) stage,124(39%) were BCLC advanced(C) stage,and 102(32%) were endstage BCLC(D).At the time of this analysis(July 2011),28(9%) patients were still alive.Six(2%) patients who were lost during follow-up were censored at the last visit.The overall median survival was 6.8 mo,and the 1-year survival was 32%.The 1-year survival a

Applicability of tooth derived stem cells in neural regeneration

Within the nervous system, regeneration is limited, and this is due to the small amount of neural stem cells, the inhibitory origin of the stem cell niche and often to the development of a scar which constitutes a mechanical barrier for the regeneration. Regarding these aspects, many efforts have been done in the re- search of a cell component that combined with scaffolds and growth factors could be suitable for nervous regeneration in regenerative medicine approaches. Autologous mesenchymal stem cells represent nowa- days the ideal candidate for this aim, thank to their multipotency and to their amount inside adult tissues. However, issues in their harvesting, through the use of invasive techniques, and problems involved in their ageing, require the research of new autologous sources. To this purpose, the recent discovery of a stem cells component in teeth, and which derive from neural crest cells, has came to the light the possibility of using dental stem cells in nervous system regeneration. In this work, in order to give guidelines on the use of dental stem cells for neural regeneration, we briefly introduce the concepts of regeneration and regenerative medicine, we then focus the attention on odontogenesis, which involves the formation and the presence of a stem component in different parts of teeth, and finally we describe some experimental approaches which are exploiting dental stem cells for neural studies.

Neuroinflammation as a target for glaucoma therapy

The pathogenesis of glaucoma is still not fully clarified but a growing body of evidence suggests that neuroinflammation and immune response are part of the sequence of pathological events leading to the optic neuropathy. Indeed, inflammation-involving the activation and proliferation of resident glial cells(astrocytes, Muller cells and microglia) and the release of a plethora of anti-and pro-inflammatory cytokines, chemokines and reactive oxygen species-has been reported as common features in clinical and experimental glaucoma. In the insulted retina, as for other neuronal tissues, pathogenic and reparative aspects coexist in the inflammatory process, with extent and persistency affecting the final outcome. In view of this, therapies aimed at modulating the immune and inflammatory responses may represent a promising approach for limiting the optic nerve damage and the loss of retinal ganglion cells associated with glaucoma.

Monitoring the abundance of saproxylic red-listed species in a managed beech forest by landsat temporal metrics

Background:Rapid climate changes lead to an increase in forest disturbance,which in turn lead to growing concerns for biodiversity.While saproxylic beetles are relevant indicators for studying different aspects of biodiversity,most are smaller than 2 mm and difficult to sample.This,together with a high number of species and trophic roles,make their study remarkably challenging,time-consuming,and expensive.The Landsat mission provides data since 1984 and represents a powerful tool in this scenario.While we believe that remote sensing data cannot replace on-site sampling and analysis,in this study we aim to prove that the Landsat Time Series(TS)may support the identification of insects’hotspots and consequently guide the selection of areas where to concentrate field analysis.Methods:With this aim,we constructed a Landsat-derived NDVI TS(1984–2020)and we summarised the NDVI trend over time by calculating eight Temporal Metrics(TMs)among which four resulted particularly successful in predicting the amount of saproxylic insects:(i)the slope of the regression line obtained by linear interpolating the NDVI values over time;(ii)the Root Mean Square Error(RMSE)between the regression line and the NDVI TS;(iii)the median,and the(iv)minimum values of the NDVI TS.The study area consists of four monitoring sectors in a Mediterranean-managed beech forest located in the Apennines(Molise,Italy),where 60 window flight traps for flying beetles were installed.First,the saproxylic beetle's biodiversities of monitoring sectors were quantified in terms of species richness and alpha-diversity.Second,the capability of TMs in predicting the richness of saproxylic beetles family and trophic categories was assessed in terms of Pearson's product-moment correlation.Results:The alpha diversity and species richness analysis indicate dissimilarities across the four monitored sectors(Shannon and Simpson's index ranging between 0.67 to 2.31 and 0.69 to 0.88,respectively),with Landsat TS resulting in effective predictors for estimating saproxylic b

Stable and Accurate Second-Order Formulation of the Shifted Wave Equation

High order finite difference approximations are derived for a one-dimensional model of the shifted wave equation written in second-order form. Thedomain is discretized using fully compatible summation by parts operators and theboundary conditions are imposed using a penalty method, leading to fully explicittime integration. This discretization yields a strictly stable and efficient scheme. Theanalysis is verified by numerical simulations in one-dimension. The present study isthe first step towards a strictly stable simulation of the second-order formulation ofEinstein’s equations in three spatial dimensions.

Grafting effects on bioactive compounds,chemical and agronomic traits of‘Corbarino’tomato grown under greenhouse healthy conditions

In tomatoes,grafting onto resistant rootstocks represents an effective strategy to control several soil-borne diseases that reduce crop profitability,especially for landraceswith little or no resistance tomultiple pathogens/pests.In this study,the high-value‘Corbarino’tomato landrace was grafted onto seven commercial Solanum interspecific rootstocks to evaluate graft compatibility,yield performance,and qualitative characteristics under greenhouse healthy conditions.Three rootstocks were themost effective in improving fruit yield without negatively affecting the carpometric,technological,and health-related traits of the traditional tomato.‘Dinafort’rootstock increased fresh and dry yield,harvest index,and fruit/clusters,while mean fruit weight,shape index,total and soluble solid content,simple sugars,organic acids,flavonoids,ascorbic acid,and trans-lycopene content did not differ with respect to the ungrafted‘Corbarino’.‘Interpro’rootstock also showed promising results for the overall yield and quality traits,although it reduced the trans-lycopene content in the fruit.‘Silex’rootstock showed the highest harvest index as result of decreased vigour.The fruit quality induced by‘Silex’was satisfactory and high levels of simple sugars,flavonoids,ascorbic acid,and trans-lycopene were also observed.However,graft success,especially for‘Dinafort’and‘Silex’,needs to be improved by investigating more suitable methods for seedling production,grafting,acclimatization,etc.

Management of hepatocellular carcinoma in the elderly

Mean age of hepatocellular carcinoma(HCC) patients has been progressively increasing over the last decades and ageing of these patients is becoming a real challenge in every day clinical practice. Unfortunately, international guidelines on HCC management do not address this problem exhaustively and do not provide any specific recommendation. We carried out a literature search in MEDLINE database for studies reporting on epidemiology, clinical characteristics and treatment outcome of HCC in elderly patients. Available data seem to indicate that in elderly patients the outcome of HCC is mostly influenced by liver function and tumor stage rather than by age and the latter should not influence treatment allocation. Age is not a risk for resection and older patients with resectable HCC and good liver function could gain benefit from surgery. Mild comorbidities do not seem a contraindication for surgery in aged patients. Conversely, major resection in elderly, even when performed in experienced high-volume centres, should be avoided. Both percutaneous ablation and transarterial chemoembolization are not contraindicated in aged patients and safety profile of these procedures is acceptable. Sorafenib is a viable option for advanced HCC in elderly provided that a careful evaluation of concomitant comorbidities, particularly cardiovascular ones, is taken into account. Available data seem to suggest that in either elderly and younger, treatment is a main predictor of outcome. Consequently, a nihilistic attitude of physicians towards under- or no-treatment of aged patients should not be longer justified.

Association of upper gastrointestinal symptoms with functional and clinical characteristics in the elderly

AIM:To evaluate the prevalence of upper gastrointestinal symptoms and their association with clinical and functional characteristics in elderly outpatients.METHODS:The study involved 3238 outpatients ≥ 60 years consecutively enrolled by 107 general practitioners.Information on social,behavioral and demographic characteristics,function in the activities of daily living (ADL),co-morbidities and drug use were collected by a structured interview.Upper gastrointestinal symptom data were collected by the 15-items upper gastro-intestinal symptom questionnaire for the elderly,a validated diagnostic tool which includes the following five symptom clusters:(1) abdominal pain syndrome;(2) reflux syndrome;(3) indigestion syndrome;(4) bleeding;and (5) non-specific symptoms.Presence and severity of gastrointestinal symptoms were analyzed through a logistic regression model.RESULTS:3100 subjects were included in the final analysis.The overall prevalence of upper gastrointestinal symptoms was 43.0%,i.***.cluster (1) 13.9%,(2) 21.9%,(3) 30.2%,(4) 1.2%,and (5) 4.5%.Upper gastrointestinal symptoms were more frequently reported by females (P < 0.0001),with high number of co-morbidities (P < 0.0001),who were taking higher number of drugs (P < 0.0001) and needed assistance in the ADL.Logistic regression analysis demonstrated that female sex (OR=1.39,95% CI:1.17-1.64),disability in the ADL (OR=1.47,95% CI:1.12-1.93),smoking habit (OR=1.29,95% CI:1.00-1.65),and body mass index (OR=1.06,95% CI:1.04-1.08),as well as the presence of upper (OR=3.01,95% CI:2.52-3.60) and lower gastroenterological diseases (OR=2.25,95%CI:1.70-2.97),psychiatric (OR=1.60,95% CI:1.28-2.01) and respiratory diseases (OR=1.25,95% CI:1.01-1.54) were significantly associated with the presence of upper gastrointestinal symptoms.CONCLUSION:Functional and clinical characteristics are associated with upper gastrointestinal symptoms.A multidimensional comprehensive evaluation may be useful when approaching upper gastrointestinal symptoms in older subjects.
