miR-106b promotes cancer progression in hepatitis B virusassociated hepatocellular carcinoma

AIM: To investigate the effect of mi R-106 b on tumor progression in hepatitis B virus(HBV)-associated hepatocellular carcinoma(HCC).METHODS: A total of 120 patients who underwent liver resection for HCC at National Cheng Kung University Hospital were enrolled in the present study. Micro RNA(mi RNA) array was first used to screen the mi RNA expression profiles in HCC patients. The clinical records were retrospectively analyzed, and correlations with the mi RNA expression profiles were evaluated. The m RNA expression levels of the mi R-106b-25 cluster(mi R-106 b, mi R-93 and mi R-25), and MCM7 in tumor and non-tumor samples were quantitated using quantitative real-time reverse transcription-polymerase chain reaction(q-RT-PCR) analysis, and correlations in the levels of mi R-106 b, mi R-93 and mi R-25 expression were calculated. Kaplan-Meier overall and diseasefree survival rates of HBV-associated HCC patients were analyzed using the log-rank test based on mi R-106 b expression. The comparison of the mi R-106 b expression levels in patients with different clinical outcomes was analyzed using Mann-Whitney U tests. Furthermore, a hepatitis B virus X protein(HBx) expression plasmid was transfected into Huh7 and Hep 3B cells. The expression levels of the mi R-106b-25 cluster and MCM7 in HBx-expressing Huh7 and Hep 3B cells were detected using q-RT-PCR. RESULTS: mi RNA array screening showed that mi R-106 b and its cluster, mi R-93 and mi R-25 were upregulated in HCC patients(P < 0.01). The value of mi R-106 b expression in HBV-associated HCC patients was significantly higher than that in HCV-(P < 0.05) or non-B/non-C-(P < 0.001) associated HCC patients. The expression of the mi R-106b-25 cluster was significantly higher in tumor tissue(P < 0.001) and associated with the host gene, MCM7, in clinical specimens from HBVassociated HCC patients. Furthermore, the expression levels of mi R-106 b, mi R-93 and mi R-25 were positively correlated in HBV-associated HCC tissues(mi R-106 vs mi R-93, r = 0.75; mi R-93 vs mi R-25, r =

AIM To investigate the clinicopathological significance of progesterone receptor membrane component 1(PGRMC1) and PGRMC2 in hepatocellular carcinoma(HCC). METHODS We performed immunohistochemical staining to evaluate the estrogen receptor(ER), progesterone receptor(PR), PGRMC1, and PGRMC2 in a clinical cohort consisting of 89 paired HCC and non-tumor liver samples. We also analyzed HCC data(n = 373) from The Cancer Genome Atlas(TCGA). We correlated the expression status of PGRMC1 and PGRMC2 with clinicopathological indicators and the clinical outcomes of the HCC patients. We knocked down or overexpressed PGRMC1 in HCC cell lines to evaluate its biological significance in HCC cell proliferation, differentiation, migration, and invasion. RESULTS We found that few HCC cases expressed ER(5.6%) and PR(4.5%). In contrast, most HCC cases expressed PGRMC1(89.9%) and PGRMC2(100%). PGRMC1 and PGRMC2 exhibited significantly lower expression in tumor tissue than in non-tumor tissue(P < 0.001). Lower PGRMC1 expression in HCC was significantly associated with higher serum alpha-fetoprotein expression(P = 0.004), poorer tumor differentiation(P = 0.045) and liver capsule penetration(P = 0.038). Low PGRMC1 expression was an independent predictor for worse disease-free survival(P = 0.002, HR = 2.384,CI: 1.377-4.128) in our cases, as well as in the TCGA cohort(P < 0.001, HR = 2.857, CI: 1.781-4.584). The expression of PGRMC2 did not relate to patient outcome. PGRMC1 knockdown promoted a poorly differentiated phenotype and proliferation of HCC cells in vitro, while PGRMC1 overexpression caused the opposite effects.CONCLUSION PGRMC1 is a non-classical hormonal receptor that negatively regulates hepatocarcinogenesis. PGRMC1 down-regulation is associated with progression of HCC and is a poor prognostic indicator.

We investigated the structural behavior and bearing capacity of system scaffolds.The research showed that the critical load of a system scaffold structure without diagonal braces is similar to that of a door-shaped steel scaffold structure.Joint stiffness between vertical props in system scaffolds can be defined based on a comparison between analytical and experimental results.When the number of scaffold stories increases,the critical loads of system scaffolds decrease.Diagonal braces markedly enhance the critical load of system scaffolds.The coupling joint position between vertical props should be kept away from story-to-story joints to prevent a reduction in critical loads.The critical load of a system scaffold decreases as the quantity of extended vertical props at the bottom of the structure increases.A large Christmas tree set up by system scaffolds under various loads was used as an example for analysis and to check the design of system scaffolds.

Applications of FBG-based sensors to ground stability monitoring

The four diversion tunnels at Jinping Ⅱ hydropower station represent the deepest underground project yet conducted in China, with an overburden depth of 1500-2000 m and a maximum depth of 2525 m.The tunnel structure was subjected to a maximum external water pressure of 10.22 MPa and the maximum single-point groundwater inflow of 7.3 m^3/s. The success of the project construction was related to numerous challenging issues such as the stability of the rock mass surrounding the deep tunnels, strong rockburst prevention and control, and the treatment of high-pressure, large-volume groundwater infiltration. During the construction period, a series of new technologies was developed for the purpose of risk control in the deep tunnel project. Nondestructive sampling and in-situ measurement technologies were employed to fully characterize the formation and development of excavation damaged zones（EDZs）, and to evaluate the mechanical behaviors of deep rocks. The time effect of marble fracture propagation, the brittleeductileeplastic transition of marble, and the temporal development of rock mass fracture and damage induced by high geostress were characterized. The safe construction of deep tunnels was achieved under a high risk of strong rockburst using active measures, a support system comprised of lining, grouting, and external water pressure reduction techniques that addressed the coupled effect of high geostress, high external water pressure, and a comprehensive early-warning system. A complete set of technologies for the treatment of high-pressure and large-volume groundwater infiltration was developed. Monitoring results indicated that the Jinping II hydropower station has been generally stable since it was put into operation in 2014.

The current report focuses on two patients of the same age who presented similar appearances on initial anteroposterior chest images. Follow-up images showed superoanterior and superoposterior mediastinal lesions. The first patient with noninvasive cystic thymoma was suspected before surgery, while the pathologic diagnosis was intrathoracic phrenic nerve schwannoma. The second patient was with an asymmetric, dumbbell-shaped paravertebral tumor over T3 and T4 on the left side. The preoperative diagnostic images were interpreted as showing a neurogenic tumor. However, the pathologic report was cell chondrosarcoma.

Selection of the Optimal Cooling Parameters to the Multi-Cavity Die

This study is subject to the finite element and abd uc tive network method application in the multi-cavity die. In order to select the optimal cooling system parameters to minimize the warp of a die-casting die, t he Taguchi’s method and the abductive network are used. These methods are appli ed to create an efficient model with functional nodes for the considered problem . Once the cooling system parameters are developed, this network can be used to predict the warp for the die-casting die accurately. A simulated annealing (SA) optimization algorithm with a performance index is then applied to the neur al network for searching the optimal cooling system parameters, and obtain rathe r satisfactory result as compared with the corresponding finite element veri fication.

This system was composed of independently developed ECG Signal Acquisition Model, DAQ Signal Acquisition Card, and the Lab VIEW Graphics System. Regarding the ECG Signal Acquisition Model, this system uses the TL082 Operational Amplifier to realize the amplification, and uses the Filter Circuits along with the ISO122 to isolate IC. Regarding the ECG oscillogram display and ECG Signal Acquisition, the Lab VIEW Graphics System developed by the American company "National Instruments"（NI） was adopted. Also, the Lab VIEW system and the DAQ signal acquisition card of the Data Acquisition Package developed by the NI Company was adopted as well.The DAQ signal acquisition card can engage in A/D, DI/O, number counting and time counting applications. At last, it can use the TCP/IP Protocol to send the ECG waveform data by the internet to the computers with remote surveillance monitors. The remote surveillance computers can be installed in the hospital. Professional doctors in the hospital can observe the patient’s ECG waveform data and statistics, and engage in the job of remote care.

Endoscopic diagnosis of cervical esophageal heterotopic gastric mucosa with conventional and narrow-band images

AIM:To compare the diagnostic yield of heterotopic gastric mucosa(HGM)in the cervical esophagus with conventional imaging(CI)and narrow-band imaging(NBI).METHODS:A prospective study with a total of 760patients receiving a CI examination(mean age 51.6years;47.8%male)and 760 patients undergoing NBI examination(mean age 51.2 years;45.9%male).The size of HGM was classified as small(1-5 mm),medium(6-10 mm),or large(>1 cm).A standardized questionnaire was used to obtain demographic characteristics,social habits,and symptoms likely to be related to cervical esophageal HGM,including throat symptoms(globus sensation,hoarseness,sore throat,and cough)and upper esophageal symptoms(dysphagia and odynophagia)at least 3 mo in duration.The clinicopathological classification of cervical esophageal HGM was performed using the proposal by von Rahden et al.RESULTS:Cervical esophageal HGM was found in 36of 760(4.7%)and 63 of 760(8.3%)patients in the CI and NBI groups,respectively(P=0.007).The NBI mode discovered significantly more small-sized HGM than CI(55%vs 17%;P<0.0001).For the 99 patients with cervical esophageal HGM,biopsies were performed in 56 patients;37(66%)had fundic-type gastric mucosa,and 19 had antral-type mucosa.For the clinicopathological classification,77 patients(78%)were classified as HGMⅠ(asymptomatic carriers);21 as HGMⅡ(symptomatic without morphologic changes);and one as HGMⅢ(symptomatic with morphologic change).No intraepithelial neoplasia or adenocarcinoma was found.CONCLUSION:NBI endoscopy detects more cervical esophageal HGM than CI does.Fundic-type gastric mucosa constitutes the most common histology.One-fifth of patients have throat or dysphagic symptoms.
