Association between human glutathione S-transferase omega rs4925 polymorphism and bladder cancer

Glutathione S-transferases (GSTs) play an important role in the detoxification of polycyclic aromatic hydrocarbons and aromatic amines, the toxic substances contained in cigarettes. GST Omega 1 (GSTO1) not only utilizes glutathione in conjugation reaction but also contributes to the biotransformation of several xenobiotics. A single nucleotide polymorphism (Ala-140Asp) of GSTO1 gene causing variations in enzyme activity may influence individual susceptibility to bladder cancer (BC). It is hypothesized that genetic polymorphism of GSTO1 gene has an effect on BC risk in particular by interacting with cigarette smoking. A total of histopathologically confirmed 300 BC patients and 300 cancer-free controls were recruited from February 2002 to February 2009. Genotyping of the GSTO1 Ala140Asp polymorphism was determined using a polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP) method. The odds ratio (OR) and 95% confidence interval (CI) were calculated as a measure of the combined effect of cigarette smoking and the GSTO1 Ala140Asp polymorphism on BC risk. We found that study subjects with the GSTO1 Ala/Ala genotype have a significantly increased BC risk (OR = 1.5;95% CI = 1.1 - 2.7). A statistically significant increased BC risk was also found in ever smokers with the GSTO1 Ala/Ala genotype (OR = 4.9;95%CI = 2.8 - 9.7). In conclusion, this study provides an epidemiologic evidence of a significantly increased BC risk among ever smokers with the GSTO1 Ala/Ala genotype.

Background:The immediate challenges during microvascular reconstruction of hepatic artery(HAR)during liver transplantation(LT)can be ***,in order to give a cross sectional view of these problems this study over a period of 1 year,showing our routine practice,was taken ***:From January 2015 to December 2015,a total of 133 LTs were performed in Kaohsiung Chang Gung Memorial Hospital,*** hepatic artery(HA)reconstructions were performed by a microvascular surgeon under an operating ***:In the 133 patients,one artery was anastomosed in 123(92.5%)patients,two in 9(6.8%)patients and three in 1(0.7%)of the ***(8.3%)arteries were less than 2 mm in size(1–1.9 mm).There were intimal dissections(IDs)involving either the donor or the recipient arteries of mild to severe nature in 9(6.8%)*** following graft arterial anastomosis,either there was no flow or an intraoperative hepatic artery thrombosis(HAT)was found in nine(7.1%—8 LDLT,4.8%—1 DDLT)*** re-do anastomosis was done in all of these patients who did well in the *** overall post-operative success rate was 99.2%.One patient(0.8%)developed postoperative HAT due to infection during follow up and died due to ***:Small vessels or HA injury are the frequently encountered problems by a micro vascular *** other problems could be ID,need to do multiple reconstructions,immediate HAT and ability to re-do the HAR immediately.

Epigenetic changes in virus-associated human cancers

Epigenetics of human cancer becomes an area of emerging research direction due to a growing understanding ofspecific epigenetic pathways and rapid development of detection technologies. Aberrant promoter hypermethylation is aprevalent phenonmena in human cancers. Tumor suppressor genes are often hypermethylated due to the increasedactivity or deregulation of DNMTs. Increasing evidence also reveals that viral genes are one of the key players inregulating DNA methylation. In this review, we will focus on hypermethylation and tumor suppressor gene silencing andthe signal pathways that are involved, particularly in cancers closely associated with the hepatitis B virus, simian virus40 (SV40), and Epstein-Barr virus. In addition, we will discuss current technologies for genome-wide detection ofepigenetically regulated targets, which allow for systematic DNA hypermethylation analysis. The study of epigeneticchanges should provide a global view of gene profile in cancer, and epigenetic markers could be used for early detection,prognosis, and therapy of cancer.

DEHP is the most abundant phthalate plasticizer in polyvinyl chloride（PVC）formulations,including vinyl flooring,wall covering,food containers,gloves,and infant *** can be leached into a liquid that comes in contact with plastics,suggesting concerns about the health risk of human *** animal studies have suggested that maternal exposure to phthalates during fetal and/or neonatal periods may cause reproductive and developmental toxicities in offspring according to their actions as endocrine disrupting chemicals（EDCs）.However,there is no detailed mechanism regarding the effect on placentation during *** hypothesize that DEHP have direct impact on the alternations of formation of *** gave DEHP（125 mg/kg）to pregnant Sprague-Dawley rats by daily with oral gavage from second day of gestation（GD 2）to GD18,and sacrifice at GD *** placentas were fixed by formaldehyde,embedded in OCT,and soaked in liquid *** sections are prepared at 8 urn thickness and stained with hematoxylin and *** thickness and the area of the placentas were *** results suggested no difference in the placental shape and weight between control and the DEHP-treated samples,however,decrease of the thickness in labyrinth was ***,the thickness of spongiotrophoblast area *** results showed as well that increase of widths in labyrinthine vessels lead to labyrinthine morphology change irregularly and vasculature disorganization in DEHP-treated *** addition,DEHP might induce ROS generation,which is able to disrupt the placental organization and its ***,our findings showed that maternal exposure to DEHP influence placental formation and development in rat.

On the basis of ZINDO method and according to the sum over states(SOS) expression, the program is devised for the calculation of nonlinear second order optical susceptibilites β ijk and studying how different substituents on the phenyl ring attached to the atom silicon influence on the nonlinear second order optical properties of substituted silane series of molecules. The properties of (CH 3) 3Si? ?has been studied particularly. The effect of the length of silica chains on the calculated values has been studied too. The regularity summarized from the calculated results has been explained micromechanically.

Technical Note: Identification of CT Texture Features Robust to Tumor Size Variations for Normal Lung Texture Analysis

Normal lung CT texture features have been used for the prediction of radiation-induced lung disease (RILD). For these features to be clinically useful, they should be robust to tumor size variations and not correlated with the normal lung volume of interest, i.e., the volume of the peri-tumoral region (PTR). CT images of 14 lung cancer patients were studied. Different sizes of gross tumor volumes (GTVs) were simulated and placed in the lung contralateral to the tumor. 27 texture features [nine from intensity histogram, eight from the gray-level co-occurrence matrix (GLCM) and ten from the gray-level run-length matrix (GLRM)] were extracted from the PTR. The Bland-Altman analysis was applied to measure the normalized range of agreement (nRoA) for each feature when GTV size varied. A feature was considered as robust when its nRoA was less than the threshold (100%). Sixteen texture features were identified as robust. None of the robust features was correlated with the volume of the PTR. No feature showed statistically significant differences (P 0.05) on GTV locations. We identified 16 robust normal lung CT texture features that can be further examined for the prediction of RILD.
