Lipidomics coverage improvement is essential for functional lipid and pathway construction.A powerful approach to discovering organism lipidome is to combine various data acquisitions,such as full scan mass spectrometry(full MS),data-dependent acquisition(DDA),and data-independent acquisition(DIA).Caenorhabditis elegans(***)is a useful model for discovering toxic-induced metabolism,highthroughput drug screening,and a variety of human disease *** determine the lipidome of *** and investigate lipid disruption from the molecular level to the system biology level,we used integrative data *** methyl-tert-butyl ether method was used to extract L4 stage *** after exposure to triclosan(TCS),perfluorooctanoic acid,and nanopolystyrene(nPS).Full MS,DDA,and DIA integrations were performed to comprehensively profile the *** lipidome by Q-Exactive Plus *** annotated lipids were then analyzed using lipid ontology and pathway *** annotated up to 940 lipids from 20 lipid classes involved in various functions and *** biological investigations revealed that when *** were exposed to nPS,lipid droplets were disrupted,whereas plasma membrane-functionalized lipids were likely to be changed in the TCS treatment *** nPS treatment caused a significant disruption in lipid ***,glycerophospholipid,and ether class lipids were those primarily hindered by ***,toxicant exposure frequently involved numerous lipid-related pathways,including the phosphoinositide 3-kinase/protein kinase B *** conclusion,an integrative data acquisition strategy was used to characterize the *** lipidome,providing valuable biological insights into hypothesis generation and validation.

日本俯冲带与IBM俯冲带位于太平洋板块、菲律宾海板块和欧亚板块三者的交汇地带,是典型的"俯冲工厂"地区,具有重要的研究意义.本文利用震源分布资料与卫星重力数据对日本俯冲带与IBM俯冲带进行了研究.通过空间重力异常反映了俯冲带地区的区域构造形态,在此基础上基于艾利模式计算了均衡异常以反映地壳均衡特征.利用震源分布资料,分别从垂直俯冲带走向与沿俯冲带走向划定了横截剖面(cross-sections)进行了地震提取,讨论了俯冲带地区的Wadati-Benioff带形态特征,并借助于俯冲带地震等深线图直观描述了俯冲带的俯冲形态.在日本俯冲带与伊豆—小笠原俯冲带各选取了一条典型剖面进行了重力2.5D反演,研究了俯冲带地区的壳幔结构特征.研究结果表明,九州—帕劳海脊与IBM岛弧在均衡异常上存在差异,前者已逐渐趋向于地壳均衡.IBM的Wadati-Benioff带存在明显的南北差异,反映出伊豆—小笠原俯冲板片停留在了660km转换带中,而马里亚纳俯冲板片很可能垂直穿过了这一转换带,造成这种南北差异的原因与板块相对运动、岩石圈黏性和年龄差异以及俯冲板片的重力效应等因素有关.在IBM的中部和南部存在板片撕裂现象.日本俯冲带的俯冲洋壳密度随俯冲深度变化较小,洋幔存在一定程度的蛇纹岩化,地幔楔蛇纹岩化作用不典型,海沟处有一范围较小的含水畸变带;伊豆—小笠原俯冲带俯冲洋壳密度随深度增大而明显增大,洋幔蛇纹岩化程度较日本俯冲带低,地幔楔蛇纹岩化作用强烈,板块交汇处存在明显的蛇纹岩底辟.日本俯冲带与IBM俯冲带一线自北向南板片俯冲变陡,两侧板块耦合度降低,与俯冲带两侧的板块运动速率差异有关.

The effect of diet-induced insulin resistance on DNA methylation of the androgen receptor promoter in the penile cavernosal smooth muscle of mice

Population studies have suggested an association between diabetes and the symptoms of testosterone deficiency. Recently, the expression of the androgen receptor （AR） has been shown to be decreased in diabetic patients. Furthermore, diabetes has been shown to induce global methylation. In this study, we used an animal model to investigate whether diabetes results in increased methylation of the AR promoter and whether these changes are associated with the decreased expression of AR in penile cavernosal smooth muscle tissue. Twenty C57BL/6J mice were divided into two groups, receiving either high- （mature diabetic） or low- （mature control） caloric meals for 14 weeks. Another 10 mice were killed at 1 week （young control）. Animals in the mature diabetic group showed decreased testosterone levels, although this was not statistically significant. In both control groups, no significant methylation was observed in the AR promoter region CpG island （-85 to ＋339）. In the mature diabetic group, significant methylation was observed at ＋ 185 and ＋200 of the AR promoter. These changes were associated with increased homeostatic model assessment for insulin resistance （HOMA-IR） and decreased corpus cavernosal tissue mass and expression of AR mRNA and protein. We conclude that in these animals, insulin resistance increased the methylation of the GC-rich regions of the AR promoter, leading to decreased AR expression.

Removal of primary iron rich phase from aluminum-silicon melt by centrifugal separation

Recycling is a major consideration in continued aluminum use due to the enormous demand for high quality products. Some impurity elements gradually accumulate through the repetitive reuse of aluminum alloy scrap. Of them, the iron content should be suppressed under the allowed limit. In the present research, a novel separation method was introduced to remove primary iron-rich intermetallic compounds by centrifugation during solidification of AI-Si-Fe alloys. This method does not use the density difference between two phases as in other centrifugal methods, but uses the order of solidification in AI-Si-Fe alloys, because iron promotes the formation of intermetallic compounds with other alloying elements as a primary phase. Two AI-Si-Fe alloys which have different iron contents were chosen as the starting materials. The iron-rich phase could be efficiently removed by centrifuging under a centrifugal force of 40 g. Coarse intermetallic compounds were found in the sample inside the crucible, while rather fine intermetallic compounds were found in the sample outside the crucible. Primary intermetallic compounds were linked to each other via aluminum-rich matrix, and formed like a network. The highest iron removal fraction is 67% and the lowest one is 7% for AI-12Si-1.7Fe alloy. And they are 82% and 18% for AI-12Si-3.4Fe alloy, respectively.

Enhanced Stability of Anisotropic Gold Nanoparticles by Poly(N-isopropylacrylamide)

A simple synthetic method has been described to prepare anisotropic gold nanoparticles （AuNPs） possessing unique optical and structural properties at room temperature, and subsequently the nanoparticles have been stabilized by temperature-sensitive poly（N-isopropylacrylamide）, or poly（NIPAM）. Although poly（NIPAM） does not exhibit a strong binding affinity to gold, simply introducing poly（NIPAM） to these unstable anisotropic AuNPs can maintain the original structures and absorption properties for several weeks. This increased stability is presumably caused by the adsorbed polymer layer around the anisotropic AuNPs. The existence of adsorbed linear poly（NIPAM） around the AuNPs is confirmed through the reversible absorption properties of the nanoparticles upon heating and cooling. To verify the presence of weak attractive forces （e.g., van der Waals, dipole-dipole, and possible hydrogen bonding） between the polymer and the AuNPs, various concentrations of linear poly（NIPAM） are introduced during the formation of the AuNPs resulting in the systematical control of the size and roughness of the nanoparticles. In addition, the preferential attachment of pre-formed anisotropic AuNPs on cross-linked poly（NIPAM） nanoparticles indicates the presence of weak attractive forces between AuNPs and poly（NIPAM）. As such, poly（NIPAM） and its derivatives can serve as a useful stabilizing and capping agent to preserve the properties of the anisotropic AuNPs.

Epigenetic mechanisms involved in the neuroprotective effect of scorpion extract in a Parkinson's disease murine model based on multi-omics approach

OBJECTIVE: To investigate whether scorpion extract elicits a neuroprotective effect in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-treated mice models, and the genes associated with the therapeutic effects using RNA sequencing(seq)***: This study investigated the changes in interaction between messenger ribonucleic acid(m RNA) expression and deoxyribonucleic acid(DNA) methylation related to the protective effects of scorpion extracts, in the substantia nigra(SN)region of a MPTP-induced Parkinson's disease(PD)***: In this model, scorpion extracts attenuated the motor impairment as demonstrated by the rotarod and open field tests. Scorpion extracts consistently attenuated the decrease of tyrosine hydroxylase(TH) positive neural cells in the SN and striatum of mice. We profiled genomewide DNA methylation using Methyl-Seq and measured the transcriptome using RNA-Seq in murine SN in the following groups: vehicle-treated MPTP-induced PD mice and scorpion extracttreated MPTP-induced PD mice. In total, 13 479 differentially expressed genes were identified in association with the anti-PD effect of the scorpion extract, mainly in the promoter and coding *** them, 47 were negatively correlated downregulated genes. Nineteen genes out of 47 downregulated genes were negatively correlated with the expression of the other 28 genes. Among these genes, SGSM1 was related to dopaminergic neurons including dopamine transporters, TH, dihydroxyphenylalanine decarboxylase, and dopamine D2 ***: This study provides insights into the anti-parkinsonian effects of scorpion extract and reveals the epigenetic targets in its therapeutic mechanism.

Identification of Aristolactam Derivatives That Act as Inhibitors of human Immunodeficiency Virus Type 1 Infection and Replication by Targeting Tat-Mediated Viral Transcription

Despite the success of antiretroviral therapy(ART),efforts to develop new classes of antiviral agents have been hampered by the emergence of drug ***-indole-bearing aristolactams are compounds that have been isolated from various plants species and which show several clinically relevant effects,including anti-inflammatory,antiplatelet,and antimycobacterial ***,the effect of these compounds on human immunodeficiency virus type 1(HIV-1)infection has not yet been *** this study,we discovered an aristolactam derivative bearing dibenzo[cd,f]indol-4(5 H)-one that had a potent anti-HIV-1 effect.A structure-activity relationship(SAR)study using nine synthetic derivatives of aristolactam identified the differing effects of residue substitutions on the inhibition of HIV-1 infection and cell *** the compounds tested,1,2,8,9-tetramethoxy-5-(2-(piperidin-1-yl)ethyl)-dibenzo[cd,f]indol-4(5 H)-one(Compound 2)exhibited the most potent activity by inhibiting HIV-1 infection with a half-maximal inhibitory concentration(IC50)of 1.03 lmol/L and a half-maximal cytotoxic concentration(CC50)of 16.91 lmol/L(selectivity index,16.45).The inhibitory effect of the compounds on HIV-1 infection was linked to inhibition of the viral replication ***-of-action studies showed that the aristolactam derivatives did not affect reverse transcription or integration;instead,they specifically inhibited Tat-mediated viral *** together,these findings show that several aristolactam derivatives impaired HIV-1 infection by inhibiting the activity of Tat-mediated viral transcription,and suggest that these derivatives could be antiviral drug candidates.

Associated factors for a hyperechogenic pancreas on endoscopic ultrasound

AIM: To identify the associated risk factors for hyperechogenic pancreas (HP) which may be observed on endoscopic ultrasound (EUS) and to assess the relationship between HP and obesity. METHODS: From January 2007 to December 2007, we prospectively enrolled 524 consecutive adults who were scheduled to undergo EUS. Patients with a history of pancreatic disease or with hepatobiliary or advanced gastrointestinal cancer were excluded. Finally,284 patients were included in the analyses. We further analyzed the risk of HP according to the categories of visceral adipose tissue (VAT) and subcutaneous adipose tissue in 132 patients who underwent abdominal computed tomography scans. RESULTS: On univariate analysis, age older than 60 years, obesity (body mass index > 25 kg/m 2 ), fatty liver, diabetes mellitus, hypertension and hypercholesterolemia were identified as risk factors associated with HP (P < 0.05). On multivariate analysis, fatty liver [P = 0.008, odds ratio (OR) = 2.219], male gender (P = 0.013, OR = 2.636), age older than 60 years (P = 0.001, OR = 2.874) and hypertension (P = 0.044, OR = 2.037) were significantly associated with HP. In the subgroup analysis, VAT was a statistically significant risk factor for HP (P = 0.010, OR = 5.665, lowest quartile vs highest quartile). CONCLUSION: HP observed on EUS was associated with fatty liver, male gender, age older than 60 years, hypertension and VAT.

Effects of Cr addition(0,3,and 6 wt%) on Charpy impact properties of Fe-C-Mn-Cr-based steels were studied by conducting dynamic compression tests at room and cryogenic *** room temperature,deformation mechanisms of Charpy impacted specimens were observed as twinning induced plasticity(TWIP) without any transfo rmation induced plasticity(TRIP) in all the *** cryogenic temperature,many twins were populated in the Cr-added steels,but,interestingly,fine ε-martensite was found in the OCr steel,satisfying the Shoji-Nishiyama(S-N) orientation relationship,{111}γ//{0002}ε and γ//ε.Even though the cryogenic-temperature staking fault energies(SFEs) of the three steel were situated in the TWIP regime,the martensitic transformation was induced by Mn-and Cr-segregated *** the OCr steel,SFEs of low-(Mn,Cr) bands lay between the TWIP and TRIP regimes which were sensitively affected by a small change of *** dynamic compressive test results well showed the relation between segregation bands and the *** of Cr were known as not only increasing the SFE but also promoting the carbide *** order to identify the possibility of carbide formation,a precipitation kinetics simulation was conducted,and the predicted fractions of precipitated M23C6 were negligible,0.4-1.1×10-5,even at the low cooling rate of 10℃/s.
