Psoriasis is a chronic autoimmune disease featured by patches on the skin.It is caused by malfunction of immune cells and keratinocytes with inflammation as one of its key features.Apigenin(API)is a natural flavonoid with anti-inflammatory and immunoregulatory properties.Therefore,we speculated that API can ameliorate psoriasis,and determined its effect on the development of psoriasis by using imiquimod(IMQ)-induced psoriasis mouse model.Our results showed that API attenuated IMQ-induced phenotypic changes,such as erythema,scaling and epidermal thickening,and improved splenic hyperplasia.Abnormal differentiation of immune cells was restored in API-treated mice.Mechanistically,we revealed that API is a key regulator of signal transducer activator of transcription 3(STAT3).API regulated immune responses by reducing interleukin-23(IL-23)/STAT3/IL-17A axis.Moreover,it suppressed IMQ-caused cell hyperproliferation by inactivating STAT3 through regulation of extracellular signal-regulated kinase 1/2 and nuclear factor-κB(NF-κB)pathway.Furthermore,API reduced expression of inflammatory cytokines through inactivation of NF-κB.Taken together,our study demonstrates that API can ameliorate psoriasis and may be considered as a strategy for psoriasis treatment.

Effects of Abscisic Acid on Cold Resistance in Digitaria sanguinalis(L.)Scop.

[Objectives]This study was conducted to detect the protective effect of abscisic acid on chilling injury of Digitaria sanguinalis(L.)Scop,and whether this effect is related to antioxidant enzymes and osmotic adjustment.[Methods]D.sanguinalis plants were sprayed with abscisic acid solution,and exposed to cold stress at 15℃for 3 d after one day and then at 5℃for 25 to 30 d in a growth chamber.The changes of plant osmotic potential under this treatment were detected.[Results]Under low temperature stress,the osmotic potential of plants in the abscisic acid treatment and the control increased,but the osmotic potential level of the abscisic acid treatment plants was lower.The SOD activity of plants in the ABA treatment and the control decreased under low temperature stress.Under low temperature stress,the activity of catalase and peroxidase in ABA-treated plants was higher than that in control plants.[Conclusions]This study provides a theoretical basis for the impact of abscisic acid on the physiological response of D.sanguinalis to cold injury.

L-Ergothioneine ameliorates MPTP-induced Parkinson's disease in C57BL/6J mice by activating DJ-1

Parkinson’s disease (PD) is one of the most common neurodegenerative diseases. The loss of dopaminergic (DAergic) neurons in the substantia nigra and the decrease of dopamine (DA) levels accelerate the process of PD. L-Ergothioneine (EGT) is a natural antioxidant derived from microorganisms， especially in edible mushrooms. EGT can penetrate blood-brain barrier and its levels are significantly decreased in the plasma of PD patients. Therefore， we speculated that EGT could ameliorate PD， and determined its effect on PD development by using 1-methyl-4-phenyl-1，2，3，6-tetrahydropyridine (MPTP)-induced PD mouse models and neurotoxin 1-methyl-4-phenylpyridinium (MPP+)-induced cell models. Our results show that EGT alleviated MPTP-induced behavioral dysfunction in mice. Mechanistically， we innovatively revealed that EGT is a key regulator of DJ-1. EGT restored DA levels by activating the DJ-1-Nuclear receptor-related factor 1(Nurr1) axis. Furthermore， it reduced reactive oxygen species (ROS) levels by regulating the DJ-1-Nuclear factor erythroid 2-related factor 2 (Nrf2) pathway， which inhibited oxidative stress-induced DAergic neuronal apoptosis. Combined treatment with DJ-1-siRNA transfection revealed that blocking DJ-1 reversed EGT upregulated Nurr1 and Nrf2 expression in the nucleus， which significantly decreased the benefits of EGT. Taken together， our study suggests that EGT can ameliorate PD and be considered as a strategy for PD treatment.

高速铁路动检数据对轨道养护维修具有重要意义,其中轴箱加速度和轮轨力是两种能够反映轨道短波不平顺信息且相互关联的高频采样数据。然而,这两种数据之间存在里程误差,以至于在评价轨道平顺性时无法建立参考基准。现有的动检数据里程修正方法大多需要结合线路台账信息,主要针对轨道几何不平顺数据的里程对齐,鲜有适用于无线路超高信息且高频采样下的轴箱加速度和轮轨力里程对齐方法。提出一种二阶段窗长收敛的里程误差修正模型,结合线性插值方法调整全局波形。根据某高速铁路实测轴箱加速度、轮轨力数据修正结果表明:该线路区间的里程误差在第一阶段修正后从1 km左右减小至42 m以内,经第二阶段精确修正后误差在99.7%的置信度下可控制在1.55 m以内。数据能量趋势结果表明,修正后轴箱加速度、轮轨力数据趋势线性相关性在全区段显著提升。

Identification of epitopes targeted following virus infection or vaccination can guide vaccine design and development of therapeutic interventions targeting functional sites,but can be laborious.Herein,we employed peptide microarrays to map linear peptide epitopes(LPEs)recognized following SARS-CoV-2 infetion and vaccination.LPEs detected by nonhuman primate(NHP)and patient IgMs after SARS-CoV-2 infection extensively overlapped,localized to functionally important virus regions,and aligned with reported neutralizing antibody binding sies.Similar LPE overlap occurred atfter infection and vaccination,with LPE clusters specifc to each stimulus,where strong and conserved LPEs mapping to sites known or likely to inhibit spike protein function.Vaccine-specifc LPEs tended to map to sites known or likely to be afected by structural changes induced by the proline substitutions in the mRNA vaccine's S protein.Mapping LPEs to regions of known functional importance in this manner may acelerate vaccine evaluation and discovery of targets for sile secific therapeutic interventions.

β-Cyclodextrin and Its Derivatives Functionalized Magnetic Nanoparticles for Targeting Delivery of Curcumin and Cell Imaging

β-Cyclodextrin （β-CD） and its derivatives functionalized magnetic nanoparticles （MNPs） with high saturated magnetism were fabricated successfully by an effective grafting method. The resultant carboxymethyl/hydroxy- propyl/sulfobutyl ether-β-CD-MNPs （CM/HP/SBE-β-CD-MNPs） nanocomposites were characterized by the TEM, FTIR, DLS, Zeta potential, XRD and VSM. In addition, the loading and release performance of the as-prepared nanocarriers for the hydrophobic anti-cancer drug curcumin was also investigated. The results revealed that the SBE-fl-CD-MNPs possessed the highest loading and release capacity in comparison with other two nanosystems. Cellular uptake and imaging suggested that the SBE-β-CD-MNPs entered into the cell, and curcumin could be suc-cessfully delivered into the cell by SBE-β-CD-MNPs nanocarrier. Moreover, cell toxicity experiments demonstrated the SBE-β-CD-MNPs were non-toxic, while curcumin loaded SBE-β-CD-MNPs showed high potential to kill the HepG2 cells. The as-prepared magnetic composites were expected to expand their potential applications in bio- medical field.

Theoretical investigation of electron-impact ionization of W8+ion

The electron-impact single ionization cross section for W8+ion has been calculated using flexible atomic code,employing the level-to-level distorted-wave approximation.This calculations takes into account contributions form both direct ionization(DI)and excitation autoionization(EA).However,the theoretical predictions,based solely on the ground state,tends to underestimate the experimental values.This discrepancy can be mitigated by incorporation contributions from excited states.We extended the theoretical analysis,including the contributions from the long-lived metastable states with lifetimes exceeding 1.5×10-5 s.We employed two statistical models to predict the fraction of ground state ions in the parent ion beam.Assuming a 79%fraction of parent ions in ground configuration,the experiment measurements align with the predictions.Furthermore we derived the theoretical cross-section for the ground state as correlated plasma rate coefficients,and compared it with existing data.Despite the uncertainty in our calculations,our results are still acceptable.

轨道几何动、静检测数据间的精确匹配对探明高速铁路线路服役状态和制定准确可靠的养护维修策略具有关键作用。针对动静里程匹配算法研究较少的现状,提出利用动、静态实测数据波形匹配,建立基于互相关函数与动态时间规划相结合的两阶段修正算法,并以距离误差作为评价指标。结合某高铁线路轨道几何检测数据的案例分析,以静态检测数据作为参考基准,对动态检测数据进行里程误差评估与修正。结果表明,两阶段算法修正效果显著,累积距离误差降幅超过93%,修正后的动、静态检测数据严格对齐保证了动态检测数据的可靠性与有效性。

[目的]明确砜吡草唑·噻吩磺隆复配剂对大豆-玉米间作田杂草的防除效果及对大豆、玉米的安全性。[方法]采用Gowing法评价砜吡草唑和噻吩磺隆复配后对稗草的联合作用,并提出最佳配比。采用室内生物测定法和田间小区试验评价其除草活性及安全性。[结果]联合作用评价试验表明砜吡草唑+噻吩磺隆的最佳复配有效成分用量为150+22.5 g a.i./hm^(2)。室内生物测定试验表明:150+22.5 g a.i./hm^(2)时砜吡草唑·噻吩磺隆复配剂药后15 d对稗草及反枝苋的鲜质量抑制率分别为97.56%、100%,且对大豆、玉米株高和鲜质量无明显抑制。田间试验结果表明:在有效成分用量为150+22.5 g a.i./hm^(2)施药后60 d,对田间禾本科杂草和阔叶类杂草的鲜质量防效均在90%以上,通过目测观察对大豆、玉米幼苗生长发育无不良影响。[结论]砜吡草唑·噻吩磺隆复配剂能够有效防除大豆-玉米间作田一年生杂草,并对大豆、玉米生长无影响。筛选出一种一次性封闭控草方案,以期为有效解决大豆-玉米间作田草害问题。
