Naked-eye visualization of lymph nodes using fluorescence nanoprobes in non-human primate-animal models

Despite sufficient studies performed in non-primate animal models,there exists scanty information obtained from pilot trials in non-human primate animal models,severely hindering nanomaterials moving from basic research into clinical *** herein present a pioneering demonstration of nanomaterials based optical imaging-guided surgical operation by using macaques as a typical kind of non-human primate-animal ***,taking advantages of strong and stable fluorescence of the small-sized(diameter:~5 nm)silicon-based nanoparticles(SiNPs),lymphatic drainage patterns can be vividly visualized in a real-time manner,and lymph nodes(LN)are able to be sensitively detected and precisely excised from small animal models(e.g.,rats and rabbits)to non-human primate animal models(e.g.,cynomolgus macaque(Macaca fascicularis)and rhesus macaque(Macaca mulatta)).Compared to clinically used invisible near-infrared(NIR)lymphatic tracers(i.e.,indocyanine green(ICG);etc.),we fully indicate that the SiNPs feature unique advantages for naked-eye visible fluorescence-guided surgical operation in long-term *** toxicological analysis in macaque models further provides confirming evidence of favorable biocompatibility of the SiNPs *** expect that our findings would facilitate the translation of nanomaterials from the laboratory to the clinic,especially in the field of cancer treatment.

Chilling-induced accumulation of reactive oxygen species(ROS) is harmful to plants,which usually produce anthocyanins to scavenge ROS as protection from chilling *** a tropical crop,cassava is hypersensitive to chilling,but the biochemical basis of this hypersensitivity remains *** previously generated Me MYB2-RNAi transgenic cassava with increased chilling *** we report that Me MYB2-RNAi transgenic cassava accumulated less ROS but more cyanidin-3-O-glucoside than the wild type under early chilling *** this stress,the anthocyanin biosynthesis pathway was more active in Me MYB2-RNAi lines than in the wild type,and several genes involved in the pathway,including Me TT8,were up-regulated by Me MYB2-RNAi in the transgenic *** MYB2 bound to the Me TT8 promoter and blocked its expression under both normal and chilling conditions,thereby inhibiting anthocyanin *** TT8 was shown to bind to the promoter of Dihydroflavonol 4-reductase(Me DFR-2)and increased Me DFR-2 *** MYB2 appears to act as an inhibitor of chilling-induced anthocyanin accumulation during the rapid response of cassava to chilling stress.

In eukaryotes, the Mre11-Rad50-Nbs1 (MRN) complex, which resides at the crossroads of DNA repair and checkpoint signaling, rapidly forms prominent foci at damage sites following double-strand break (DSB) induction. This complex carries out the initial processing of the DSB ends. Mutations in the genes that encode components of this complex result in DNA-damage hypersensitivity, genomic in- stability, telomere shortening, and aberrant meiosis. Therefore, the MR proteins are highly conserved during evolution. The bacterial orthologs of Mre11 and Rad50 are the SbcD and SbcC proteins, respec- tively. Deinococcus radiodurans, an extremely radioresistant bacterium, is able to mend hundreds of radiation-induced DSBs. The SbcD and SbcC proteins were identified as the products of the Dr1921 and Dr1922 genes. Disruption of the sbcD gene, by direct reverse-orientation insertional mutagenesis tech- nology, remarkably increases the cells’ sensitivity to various types of DNA damaging agents, such as ionizing radiation, ultraviolet irradiation, hydrogen peroxide, and mitomycin C. We also provide evidence that the drSbcD protein plays an important role in both growth and DNA repair in this organ- ism, especially in repair of DSBs generated after cellular exposure to 6000 Gy of IR. These results demonstrate that the drSbcD protein plays an important role in DSBs repair in D. radiodurans.

Cardiovascular diseases such as myocardial infarction(MI)are among the major causes of death *** intramyocardial injection of hydrogels can effectively enhance the ventricular wall,this approach is limited because of its restriction to the poor vascularization in the infarcted ***,we reported a new type of hydrogel composed of alginate(ALG)and hyaluronic acid(HA)with lyophilized platelet-rich fibrin(Ly-PRF)for releasing abundant growth factors to realize their respective *** results of in vitro studies demonstrated favorable mechanical property and release ability of ALG-HA with *** injected into the infarcted myocardium,this composite hydrogel preserved heart function and the Ly-PRF within the hydrogel promoted angiogenesis and increased vascular density in both infarcted and border zone,which rescued the ischemic *** beneficial effects were also accompanied by macrophage polarization and regulation of myocardial ***,the autologous origin of Ly-PRF with ALG-HA hydrogel offers myriad advantages including safety profile,easiness to obtain and ***,this study demonstrated the versatile therapeutic effects of a novel composite hydrogel ALG-HA with Ly-PRF,which optimizes a promising vascularized substitution strategy for improving cardiac function after MI.

Oligo probe staining is a low-cost and efficient chromosome identification *** this study,oligo genomic in situ hybridization(Oligo-GISH)technology was established in *** A and B subgenome-specific interspersed repeat(IR)oligo probe sets were developed based on clustering and electronic localization of tandem repeat sequences in the reference genome of *** OligoGISH kit was then used to perform staining of 15 Arachis *** A-subgenome probe set stained the chromosomes of A-and E-genome Arachis species,the B-subgenome probe set stained those of B-,F-,K-,and E-genome species,and neither set stained those of H-genome *** results indicate the relationships among the genomes of these Arachis *** Oligo-GISH kit was also used for batch staining of the chromosomes of 389 seedlings from the irradiated M1generation,allowing 67 translocation and deletion lines to be *** Oligo-FISH karyotyping,FISH using single-copy probe libraries,and trait investigation identified seven homozygous chromosomal variants from the M3generation and suggested that there may be genes on chromosome 4B controlling seed number per *** findings demonstrate that the IR probe sets and method developed in this study can facilitate research on distant hybridization and genetic improvement in peanut.

The construction of advanced electrode materials is key to the field of energy ***,a free-standing anatase titania(TiO_(2))nanocrystal/carbon nanotube(CNT)film is reported using a simple and scalable sol-gel method,followed by *** unique free-standing film comprises ultra-small TiO_(2) nanocrystals(~5.9 nm)and super-aligned CNTs,with ultra-dispersed TiO_(2) nanocrystals on the surfaces of the *** the one hand,these TiO_(2) nanocrystals can significantly decrease the diffusion distance of the charges and on the other hand,the cross-linked CNTs can act as a three-dimensional(3D)conductive network,allowing the fast transport of *** addition,the film is free-standing,without requiring electrode fabrication and additional conductive agents and *** to these above synergistic effects,the film is directly used as an anode in Li-ion batteries,and delivers a high discharge capacity of~105 mAh·g^(−1) at high rate of 60 C(1 C=170 mA·g^(−1))and excellent cycling performance over 2,500 cycles at 30 *** results indicate that the free-standing anatase TiO_(2) nanocrystal/CNT film affords a superior performance among the various TiO_(2) materials and can be a promising anode material for fast-charging Li-ion ***,the TiO_(2)/CNT film exhibits an areal capacity of up to 2.4 mAh·cm^(−2),confirming the possibility of its practical use.
