Prediction and Regulation of Delamination at Flexible Film/Finite-Thickness-Substrate Structure Interfaces

Regulating the surface instability of thin film/substrate structures has been successfully applied to prepare new ductile electronic devices.However,such electronic devices need to be subjected to external loads during operation,which can easily induce delamination of the thin-film electronic device from the substrate.This study aims to investigate the instability characteristics of hard films on flexible substrate surfaces from theoretical analysis and numerical simulation perspectives.Considering finite-thickness substrates,this paper establishes theoretical models for pure bending,bent wrinkle,partial delamination,and total delamination buckling of film/substrate structures based on the nonlinear Euler–Bernoulli beam theory and the principle of minimum energy;then the effects of material and geometric parameters of the structure,interfacial adhesion strength,and pre-strain on the evolutionary path of the four patterns are discussed.The study results show that:the greater Young’s modulus of the substrate is,the larger the parameter region where partial delamination of the film/substrate structure occurs,and the smaller the parameter region where bent wrinkle occurs.By varying Young’s modulus,thickness of the film and substrate,interfacial adhesion coefficient,and pre-strain,the buckling pattern of the structure can be predicted and regulated.The parametric design intervals for each pattern are summarized in the phase diagram.The results of this paper provide theoretical support for the design and reliability evaluation of flexible electronic devices.

Proper regulation of synapse formation and elimination is critical for establishing mature neuronal circuits and maintaining brain function.Synaptic abnormalities,such as defects in the density and morphology of postsynaptic dendritic spines,underlie the pathology of various neuropsychiatric disorders.Protocadherin 17(PCDH17)is associated with major mood disorders,including bipolar disorder and depression.However,the molecular mechanisms by which PCDH17 regulates spine number,morphology,and behavior remain elusive.In this study,we found that PCDH17 functions at postsynaptic sites,restricting the number and size of dendritic spines in excitatory neurons.Selective overexpression of PCDH17 in the ventral hippocampal CA1 results in spine loss and anxiety-and depression-like behaviors in mice.Mechanistically,PCDH17 interacts with actin-relevant proteins and regulates actin filament(F-actin)organization.Specifically,PCDH17 binds to ROCK2,increasing its expression and subsequently enhancing the activity of downstream targets such as LIMK1 and the phosphorylation of cofilin serine-3(Ser3).Inhibition of ROCK2 activity with belumosudil(KD025)ameliorates the defective F-actin organization and spine structure induced by PCDH17 overexpression,suggesting that ROCK2 mediates the effects of PCDH17 on F-actin content and spine development.Hence,these findings reveal a novel mechanism by which PCDH17 regulates synapse development and behavior,providing pathological insights into the neurobiological basis of mood disorders.

Brain-derived extracellular vesicles participate in interorgan communication after traumatic brain injury by transporting pathogens to initiate secondary injury.Inflammasome-related proteins encapsulated in brain-derived extracellular vesicles can cross the blood‒brain barrier to reach distal tissues.These proteins initiate inflammatory dysfunction,such as neurogenic heterotopic ossification.This recurrent condition is highly debilitating to patients because of its relatively unknown pathogenesis and the lack of effective prophylactic intervention strategies.Accordingly,a rat model of neurogenic heterotopic ossification induced by combined traumatic brain injury and achillotenotomy was developed to address these two issues.Histological examination of the injured tendon revealed the coexistence of ectopic calcification and fibroblast pyroptosis.The relationships among brain-derived extracellular vesicles,fibroblast pyroptosis and ectopic calcification were further investigated in vitro and in vivo.Intravenous injection of the pyroptosis inhibitor Ac-YVAD-cmk reversed the development of neurogenic heterotopic ossification in vivo.The present work highlights the role of brain-derived extracellular vesicles in the pathogenesis of neurogenic heterotopic ossification and offers a potential strategy for preventing neurogenic heterotopic ossification after traumatic brain injury.

Hot-exciton effects on exciton diffusion and circular polarization dynamics in a single PbI_(2)nanoflake

As one of the emerging two-dimensional lead halide materials,lead iodide(PbI_(2))nanosheets have proven to possess strong application potential in the fields of high-energy radiation detection and highly efficient perovskite solar cells.However,the underlying photophysical properties such as hot-exciton-related carrier dynamics remain unclear for PbI_(2)nanosheets.Here,we report the exciton dynamics of a single PbI_(2)nanoflake prepared by an aqueous solution method.Through a three-dimensional(3D)diffusion model,we obtain the exciton annihilation radius and diffusion coefficient of a single PbI_(2)nanoflake under nonresonant and resonant excitation conditions of band-edge exciton state.As initial exciton densities increase,we find the carrier recombination mechanism for a single PbI_(2)nanoflake gradually changes from exciton-exciton annihilation to free-carrier recombination.Finally,we reveal the room-temperature circular polarization of a single PbI_(2)nanoflake is due to free-carrier recombination with a band-edge exciton dissociation time of~120 fs under the resonant excitation condition.

Gabapentinoids for the treatment of stroke

Gabapentinoid drugs(pregabalin and gabapentin) have been successfully used in the treatment of neuro pathic pain and in focal seizure prevention.Recent research has demonstrated their potent activities in modulating neurotransmitter release in neuronal tissue,oxidative stress,and inflammation,which matches the mechanism of action via voltage-gated calcium channels.In this review,we briefly elaborate on the medicinal history and ligand-binding sites of gabapentinoids.We systematically summarize the preclinical and clinical research on gabapentinoids in stroke,including ischemic stro ke,intracerebral hemorrhage,subarachnoid hemorrhage,seizures after stro ke,cortical spreading depolarization after stroke,pain after stroke,and nerve regeneration after stro ke.This review also discusses the potential to rgets of gabapentinoids in stroke;however,the existing results are still unce rtain regarding the effect of gabapentinoids on stroke and related diseases.Further preclinical and clinical trials are needed to test the therapeutic potential of gabapentinoids in stroke.Therefore,gabapentinoids have both opportunities and challenges in the treatment of stroke.

Knockdown of RCN1 contributes to the apoptosis of colorectal cancer via regulating IP3R1

Background:The incidence of colorectal cancer(CRC)has been increasing in recent years.Thus,the discovery of factors that can assist in alleviating CRC is urgently warranted.Methods:To identify a potential factor involved in the development of CRC,we screened the upregulated genes in tumor tissues through four datasets from an online database.The expression of reticulocalbin 1(RCN1),a Ca2+-binding protein,was upregulated in the four datasets.Based on loss-offunction experiments,the effect of RCN1 on cell viability was assessed by Cell Counting Kit-8(CCK-8)assay.The regulatory effect of RCN1 on apoptosis was evaluated through Annexin V-fluorescein 5-isothiocyanate(FITC)/propidium iodide(PI)staining assay and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL)assay in RKO and SW480 cells.Activation of endoplasmic reticulum(ER)stress signaling pathways was confirmed by estimating the phosphorylation and expression of PRKR-like ER kinase(PERK),inositol-requiring kinase-1(IRE1),transcription factor 6(ACT6),and CCAAT/enhancer-binding protein-homologous protein(CHOP).The intracellular Ca2+homeostasis regulated by RCN1 was determined through the detection of Ca2+concentration and mitochondrial membrane potential(MMP)measurement.Moreover,whether inositol 1,4,5-trisphosphate receptor type 1(IP3R1)was involved in the regulation of RCN1 in CRC was verified through the depletion of IP3R1 in RKO cells.Results:Knockdown of RCN1 reduced cell viability and facilitated apoptosis in RKO and SW480 cells.Phosphorylation of PERK and IRE1,activation of ATF6,and upregulation of CHOP were induced by the absence of RCN1,suggesting that the unfolded protein response(UPR)was activated in CRC cells.The concentration of Ca2+in mitochondria was increased after RCN1 depletion,followed by reduction in the MMP and release of cytochrome c from mitochondria to the cytoplasm in RKO and SW480 cells.Moreover,it was demonstrated that IP3R1 mediates the effect of RCN1 on apoptosis induced by ER stress in CRC cells.The downregulat

Efficient privacy-preserving user authentication scheme with forward secrecy for industry 4.0

Industry 4.0, which combines information technology, network and industrial production, is expected to have a tremendous impact on our daily lives. In such a complex and security-critical system with resource-constrained sensor nodes, the design of a secure user authentication scheme for preventing real-time data from unauthorized access is full of challenges, and the main crux lies in how to realize the important property of forward secrecy. Existing schemes either fail to achieve forward secrecy or achieve forward secrecy with high computation cost on sensor nodes. Besides, they often fail to conform to the development trend of industry 4.0 systems where a cloud center is necessary to help intelligent decision-making and alleviate computation and storage pressure. Therefore, in this paper, we propose an efficient privacy-preserving user authentication scheme with forward secrecy for industry 4.0, and formally prove its security in the random oracle model. Compared with previous schemes, it has three advantages:(1) all eleven state-of-the-art criteria are achieved;(2) its computation cost on sensor nodes is comparable to those insecure schemes that employ only symmetric cryptographic algorithms, and is superior to those that also use asymmetric cryptographic algorithms;(3) it takes the advantage of the computation and storage capabilities of the cloud center to achieve user anonymity and the resistance to offline dictionary attack without performing any asymmetric cryptographic algorithms on gateways. Our computation cost on gateways is the smallest among all state-of-the-art relevant schemes for comparison.

Carbon dots(CDs)have wide application potentials in optoelectronic devices,biology,medicine,chemical sensors,and quantum techniques due to their excellent fluorescent properties.However,synthesis of CDs with controllable spectrum is challenging because of the diversity of the CD components and structures.In this report,machine learning(ML)algorithms were applied to help the synthesis of CDs with predictable photoluminescence(PL)under the excitation wavelengths of 365 and 532 nm.The combination of precursors was used as the variable.The PL peaks of the strongest intensity(λ_(s))and the longest wavelength(λ_(l))were used as target functions.Among six investigated ML models,the random forest(RF)model showed outstanding)performance in the prediction of the PL peaks.

Artificial intelligence(AI)is developing rapidly and is being used in several medical capacities,including assisting in diagnosis and treatment decisions.As a result,this raises the conceptual and practical problem of how to distribute responsibility when AI-assisted diagnosis and treatment have been used and patients are harmed in the process.Regulations on this issue have not yet been established.It would be beneficial to tackle responsibility attribution prior to the development of biomedical AI technologies and ethical guidelines.In general,human doctors acting as superiors need to bear responsibility for their clinical decisions.However,human doctors should not bear responsibility for the behavior of an AI doctor that is practicing medicine inde-pendently.According to the degree of fault-which includes internal institutional ethics,the AI bidding process in procurement,and the medical process-clinical institutions are required to bear corresponding responsibility.AI manufacturers are responsible for creating accurate algorithms,network security,and insuring patient privacy protection.However,the AI itself should not be subjected to legal evaluation since there is no need for it to bear responsibility.Corresponding responsibility should be borne by the employer,in this case the medical institution.
