Early evaluation of growth traits of Larix kaempferi clones

Early selection is an important method to shorten the breeding cycle for tree species, which may differ in the time for early selection. To evaluate the early selected time for Larix kaempferi, tree height and diameter at breast height of 57 L. kaempferi clones were measured over many different growth years. The results indicated that, except for age × clone interaction for diameter at breast height （P = 0.741）, there were significant differences among all variation sources （P 〈 0.01）. The coefficient of phenotypic variation ranged from 14.89 to 35.65% for height and from 19.17 to 23.86% for diameter at breast height in different growth years. The repeatability of height and of diameter at breast height among clones was high, ranging from 0.6181 to 0.8531 （height） and from 0.8443 to 0.8497 （diameter at breast height）, in different growth years. There were significant positive correlations between all pairs of growth traits except between height in the 2nd growth year and height in the 30th growth year; and between height in the 2nd growth year × diameter at breast height in the 30th growth year. With a comprehen- sive evaluation method and a selection ratio of 10%, L65, L1, L62, L9, L15, and L78 were selected as excellent clones in the 30th growth year. Their average values of height and diameter at breast height were 9.81 and 16.57% higher than the overall average, representing genetic gains of 6.46 and 13.99%, respectively. This study provides a theoretical foundation for the genetic improvement of L. kaempferi.

Study on the low mechanical anisotropy of extruded Mg-Zn-Mn-Ce-Ca alloy tube in the compression process

In this study,the extruded Mg-Zn-Mn-Ce-Ca alloy tube with a low compression anisotropy along the ED,45ED and TD was prepared.The effect of the second phases,initial texture and deformation behavior on this low mechanical anisotropy was investigated.The results revealed that the alloy tube contains the high content(Mg1-xZnx)11Ce phase and the low content of Mg12Ce phase.These second phases are respectively incoherent and coherent with the Mg matrix,and their influence can be ignored.Additionally,the alloy tube exhibited a weak basal fiber texture,where the c-axis was aligned along the 0°∼30°tilt from TD to ED.Such a texture made the initial deformation(at 1.0%∼1.6%strain)of the three samples controlled by comparable basalslip.As deformation progressed(1.6∼9.0%strain),larger amounts of ETWs nucleated and gradually approached saturation in the three samples,re-orienting the c-axis to a 0°∼±30°deviation with respect to the loading directions.Meanwhile,the prismatic and pyramidalslips replaced the dominant deformation progressively until fracture.Eventually,the similar deformation mechanisms determined by the weak initial texture in the three samples contribute to the comparable strain hardening rates,resulting in the low compressive anisotropy of the alloy tube.

Parkinson's disease(PD)is a neurodegeneration disease withα-synuclein accumulated in the substantia nigra pars compacta(SNpc)and most of the dopaminergic neurons are lost in SNpc while patients are diagnosed with PD.Exploring the pathology at an early stage contributes to the development of the disease-modifying strategy.Although the“gut–brain”hypothesis is proposed to explain the underlying mechanism,where the earlier lesioned site in the brain of gastricα-synuclein and howα-synuclein further spreads are not fully understood.Here we report that caudal raphe nuclei(CRN)are the early lesion site of gastricα-synuclein propagating through the spinal cord,while locus coeruleus(LC)and substantia nigra pars compacta(SNpc)were further affected over a time frame of 7 months.Pathologicalα-synuclein propagation via CRN leads to neuron loss and disordered neuron activity,accompanied by abnormal motor and non-motor behavior.Potential neuron circuits are observed among CRN,LC,and SNpc,which contribute to the venerability of dopaminergic neurons in SNpc.These results show that CRN is the key region for the gastricα-synuclein spread to the midbrain.Our study provides valuable details for the“gut–brain”hypothesis and proposes a valuable PD model for future research on early PD intervention.

Moyamoya disease(MMD)is a chronic occlusive cerebrovascular disease with the development of a network of abnormal vessels.Immune inflammation is associated with the occurrence and development of MMD.However,the mechanisms underlying the formation of the abnormal vascular network remain unclear.Twenty-eight patients with MMD,26 ischemic stroke patients,and 26 unrelated healthy volunteers were enrolled in this study The data showed that the levels of granulocyte-macrophage colony-stimulating factor(GM-CSF)were higher in MMD patients than in healthy controls(P<0.01),and GM-CSF was mainly from Th1 and Th17 cells in MMD.We found that increased GM-CSF drove monocytes to secrete a series of cytokines associated with angiogenesis,inflammation,and chemotaxis.In summary,our findings demonstrate for the first time the important involvement of GM-CSF in MMD and that GM-CSF is an important factor in the formation of abnormal vascular networks in MMD.

Photo-induced Ag modulating carbon dots:Greatly improved fluorescent properties and derived sensing application

Carbon dots(CDs)have been attracted much attention and widely studied due to their excellent fluorescence(FL)properties,better biocompatibility and outstanding photo/chemical stability.However,the disadvantage of lower quantum yield(QY)still limits its wide application.Herein,we reported a novel and convenient strategy to prepare photo-induced Ag/CDs(p-Ag/CDs)by irradiating the mixed Ag+and hydrophobic CDs(h-CDs)acetone solution with ultraviolet(UV)light.The obtained p-Ag/CDs exhibit a greatly enhanced FL emission together with a blue shift(460 nm)than h-CDs(520 nm).The QY of p-Ag/CDs is measured to be 51.1%,which is 10.4 times higher than that of h-CDs(4.9%),indicating that photo-induced Ag modulation can effectively improve the optical properties of CDs.The mechanisms for the FL enhancement and blue shift of h-CDs are studied in detail.The results prove that the greatly enhanced FL emission is from the generated Ag nanoparticles(AgNPs)by UV light irradiation based on metal-enhanced fluorescence(MEF),and the increased oxygen-contained groups in this process lead to the blue shift in CDs fluorescence.Interestingly,the p-Ag/CDs exhibit higher sensitivity and selectivity for sulfide ions(S2-)detection than that of h-CDs,which have a lower response to S2-.This work not only offers a novel strategy to improve the FL properties of materials but also endows them with new functions and broadens their application fields.

ARL14 在结直肠癌中的表达及其与肿瘤相关成纤维细胞浸润的关系

目的:探究二磷酸腺苷核糖基化因子样蛋白14(adenosine diphosphate-ribosylation factor-like protein 14,ARL14)在结直肠癌(colorectal cancer,CRC)中的表达水平及其与肿瘤微环境中浸润细胞的关系。方法:分析TCGA-CRC数据集中607例CRC组织与51例正常组织中ARL14的表达差异及其与预后之间的关系,并预测参与上游表达调控的转录因子和miRNA。应用生物信息学方法分析ARL14表达与肿瘤微环境中各细胞浸润的关系。收集2020年1月至2021年1月于天津医科大学肿瘤医院院行手术治疗的45例CRC患者的肿瘤及配对正常组织,使用免疫组织化学染色检测ARL14、平滑肌肌动蛋白-α(smooth muscle actin-α,α-SMA)和成纤维细胞活化蛋白(fibroblast activation protein,FAP)的表达情况,验证ARL14表达与成纤维细胞活化的关系。结果:差异分析显示CRC中ARL14的表达量显著低于正常组织(P<0.001)。与ARL14高表达患者相比,低表达患者的预后更差(P=0.025)。MCPcounter分析显示ARL14表达与成纤维细胞的浸润呈负相关(P<0.0001)。免疫组织化学染色结果显示ARL14在肿瘤中的表达显著低于正常组织(P<0.01),ARL14与间质中α-SMA、FAP的表达均为负相关。结论:ARL14可能在CRC中发挥肿瘤抑制基因的作用,并可能抑制成纤维细胞的活化。

Assessment of tree-ring mercury radial translocation and age effect in Masson pine:Implications for historical atmospheric mercury reconstruction

The tree ring has been regarded as an emerging archive to reconstruct historical atmospheric mercury(Hg)trends,but with the large knowledge gaps in the reliability.In this study,we comprehensively evaluated the Hg source,radial translocation and age effect of Masson pine(Pinus massoniana)tree ring at Mt.Jinyun in Chongqing,to assess the suitability of such tree ring as the archive of atmospheric Hg.Results showed that distinct variabilities among Masson pine tree-ring Hg concentration profiles.The Hg concentration significantly increased along with stem height(P<0.05),indicating the Hg in tree rings mainly derived from foliage uptake atmospheric Hg.We found a distinct age effect that the tree ring of young trees had the higher Hg concentration.Besides,we used the advection-diffusion model to demonstrate how Hg concentration shifted by the advection or/and diffusion in tree rings.The modeling results showed that the advection induced radial translocation during the young growth period of tree was a plausible mechanism to result in the tree-ring Hg record largely different from the trend of anthropogenic Hg emissions in Chongqing.We finally suggest that in further Hg dendrochemistry,better discarding the tree-ring Hg profile of the young growth period to reduce impacts of the radial translocation and age effect.

Enrichment of anammox biomass during mainstream wastewater treatment driven by achievement of partial denitrification through the addition of bio-carriers

Anammox is widely considered as the most cost-effective and sustainable process for nitrogen removal.However,how to achieve the enrichment of anammox biomass remains a challenge for its large-scale application,especially in mainstream wastewater treatment.In this study,the feasibility of enrichment of anammox biomass was explored through the realization of partial denitrification and the addition of bio-carriers.By using ordinary activated sludge,a sequencing batch reactor(SBR)followed by an up-fow anaerobic sludge bed(UASB)was operated at 25±2℃ for 214 days.The long-term operation was divided into five phases,in which SBR and UASB were started-up in Phases I and II,respectively.By eliminating oxygen and adjusting the infow ratios in Phases III-V,advanced nitrogen removal was achieved with the effuent total nitrogen being 4.7 mg/L and the nitrogen removal efficiency being 90.5%in Phase V.Both in-situ and ex-situ activity tests demonstrated the occurrence of partial denitrification and anammox.Moreover,16S rRNA high-throughput sequencing analysis revealed that Candidatus Brocadia was enriched from below the detection limit to in biofilms(0.4%in SBR,2.2%in UASB)and the foc sludge(0.2%in SBR,1.3%in UASB),while Thauera was mainly detected in the foc sludge(8.1%in SBR,8.8%in UASB),which might play a key role in partial denitrification.Overall,this study provides a novel strategy to enrich anammox biomass driven by rapid achievement of partial denitrification through the addition of bio-carriers,which will improve large-scale application of anammox processes in mainstream wastewater treatment.

Performance and mechanism of La-Fe metal-organic framework as a highly efficient adsorbent for fluoride removal from mine water

Water fluoride pollution has caused non-negligible harm to the environment and humans,and thus it is crucial to find a suitable treatment technology.In this study,La-Fe@PTA adsorbent was synthesized for the defluoridation of mine water.The results showed that the optimum conditions for defluoridation by La-Fe@PTA were p H close to 7.0,the initial F-concentration of 10 mg/L,the dosage of 0.5 g/L and the adsorption time of 240 min.Compared with SO_4^(2-),Cl^(-),NO_(3)^(-),Ca^(2+)and Mg^(2+),CO_(3)^(2-)and HCO_(3)^(-)presented severer inhibition on fluoride uptake by La-Fe@PTA.The adsorption process fits well with the pseudo-second-order kinetic model and Freundlich model,and the maximum adsorption capacity of Langmuir model was 95 mg/g.Fixed-bed adsorption results indicated that fluoride in practical fluorinated mine water could be effectively removed from 3.6 mg/L to less than 1.5 mg/L within130 bed volume(BV)by using 1.5 g La-Fe@PTA.Furthermore,the adsorbent still had good adsorption capacity after regeneration,which confirms the great application potential of La-Fe@PTA as a fluoride ion adsorbent.The mechanism analysis showed that La-Fe@PTA adsorption of fluorine ions is a physicochemical reaction driven by electrostatic attraction and ion exchange.
