RAD-seq data reveals robust phylogeny and morphological evolutionary history of Rhododendron

Rhododendron is famous for its high ornamental value.However,the genus is taxonomically difficult and the relationships within Rhododendron remain unresolved.In addition,the origin of key morphological characters with high horticulture value need to be explored.Both problems largely hinder utilization of germplasm resources.Most studies attempted to disentangle the phylogeny of Rhododendron,but only used a few genomic markers and lacked large-scale sampling,resulting in low clade support and contradictory phylogenetic signals.Here,we used restriction-site associated DNA sequencing(RAD-seq)data and morphological traits for 144 species of Rhododendron,representing all subgenera and most sections and subsections of this species-rich genus,to decipher its intricate evolutionary history and reconstruct ancestral state.Our results revealed high resolutions at subgenera and section levels of Rhododendron based on RAD-seq data.Both optimal phylogenetic tree and split tree recovered five lineages among Rhododendron.Subg.Therorhodion(cladeⅠ)formed the basal lineage.Subg.Tsutsusi and Azaleastrum formed cladeⅡand had sister relationships.CladeⅢincluded all scaly rhododendron species.Subg.Pentanthera(cladeⅣ)formed a sister group to Subg.Hymenanthes(cladeⅤ).The results of ancestral state reconstruction showed that Rhododendron ancestor was a deciduous woody plant with terminal inflorescence,ten stamens,leaf blade without scales and broadly funnelform corolla with pink or purple color.This study shows significant distinguishability to resolve the evolutionary history of Rhododendron based on high clade support of phylogenetic tree constructed by RAD-seq data.It also provides an example to resolve discordant signals in phylogenetic trees and demonstrates the application feasibility of RAD-seq with large amounts of missing data in deciphering intricate evolutionary relationships.Additionally,the reconstructed ancestral state of six important characters provides insights into the innovation of key characters in Rhododendron.

M6PR interacts with the HA2 subunit of influenza A virus to facilitate the fusion of viral and endosomal membranes

Influenza A virus(IAV) commandeers numerous host cellular factors for successful replication. However, very few host factors have been revealed to be involved in the fusion of viral envelope and late endosomal membranes. In this study, we identified cation-dependent mannose-6-phosphate receptor(M6PR) as a crucial host factor for the replication of IAV. We found that siRNA knockdown of M6PR expression significantly reduced the growth titers of different subtypes of IAV, and that the inhibitory effect of M6PR siRNA treatment on IAV growth was overcome by the complement of exogenously expressed M6PR. When A549 cells were treated with siRNA targeting M6PR,the nuclear accumulation of viral nucleoprotein(NP) was dramatically inhibited at early timepoints post-infection, indicating that M6PR engages in the early stage of the IAV replication cycle. By investigating the role of M6PR in the individual entry and post-entry steps of IAV replication, we found that the downregulation of M6PR expression had no effect on attachment, internalization, early endosome trafficking,or late endosome acidification. However, we found that M6PR expression was critical for the fusion of viral envelope and late endosomal membranes. Of note, M6PR interacted with the hemagglutinin(HA) protein of IAV, and further studies showed that the lumenal domain of M6PR and the ectodomain of HA2 mediated the interaction and directly promoted the fusion of the viral and late endosomal membranes,thereby facilitating IAV replication. Together, our findings highlight the importance of the M6PR–HA interaction in the fusion of viral and late endosomal membranes during IAV replication.

Acral melanoma(AM)is the most common histologic subtype of melanoma in dark-skinned patients and is associated with a worse prognosis and a high mortality rate,largely due to the inconspicuous nature of early-stage lesions,which can lead to late diagnosis.Because of the overlapping clinical and histopathological features of AM with other forms of cutaneous melanomas,early detection of AM requires a multidisciplinary approach that integrates various diagnostic modalities,including clinical examination,dermoscopy,histopathology,molecular testing,radiological imaging,and blood tests.While surgery is the preferred method of treatment for AM,other therapeutic options may be employed based on the stage and underlying etiology of the disease.Immune checkpoint inhibitors,molecular targeted therapy,radiotherapy,chemotherapy,and oncolytic virotherapy represent promising advanced treatment options for AM.In this review,we provide an overview of the latest advancements in diagnostic and therapeutic methods for AM,highlighting the importance of early detection and the prompt,individualized management of this challenging disease.

Construction of a bivalent vaccine candidate against HAdV4/HAdV7 based on capsid-display strategy via Red-homologous recombination and counter-selection methodology

Human adenoviruses(HAdvs)are major respiratory pathogens.Specifically,human adenovirus type 4(HAdV4)and human adenovirus type 7(HAdV7)are known for causing fever and pneumonia,with docu-mented cases of fatalities among the population.In recent years,HAdV4/HAdv7 has been implicated in caus-ing substantial outbreaks,leading to increased morbidity in multiple countries.Most HAdV4 and HAdV7 infections have been reported in North America,Asia,Europe,Africa,South America,Oceania,and the Middle East.Most fatalities occurred in North America(the United States)and Asia(China and Singapore).Engineered recombinant adenoviruses have played a crucial role as vaccine vectors.In this study,we con-structed a recombinant adenovirus,Ad4ITRmut-Ad7E3,and evaluated it in vitro and in vivo.We observed that the replication rate of Ad4ITRmut-Ad7E3 was lower than that of the RI-67 strain,indicating that the mutation of inverted terminal repeats(ITRs)weakened the replication ability of HAdV4.Immunization of BALB/c mice with the bivalent Ad4ITRmut-Ad7E3 vaccine strain,administered by intraperitoneal injection and oral gavage,resulted in the elicitation of neutralizing antibodies targeting HAdV4 and HAdv7.This finding not only pro-vides a novel method and technique for the efficient construction of a polyvalent recombinant adenovirus vac-cine candidate against HAdV4 and HAdv7 but also against other prevalent adenovirus serotypes such as HAdV3,HAdV11,HAdV14,and HAdv55,from various regions.

Microbial growth causes lamb spoilage. This study explored the spoilage ability of Latilactobacillus sakei (L. sakei)， Serratia proteamaculans (S. proteamaculans) and Hafnia proteus (H. proteus) in vacuum-packed raw lamb， including growth ability， degradation of protein and lipid， and change of volatile organic compounds (VOCs) profile， meanwhile screened the key VOCs produced by the targeted strains with meat background excluding， finally confirmed the volatile spoilage marker of vacuum-packaged lamb by comparing with our previous work. The results showed that L. sakei， S. proteamaculans and H. proteus had excellent growth ability. L. sakei inoculated group significantly reduced the pH value， showed higher TCA-soluble peptides content， and excellently degraded sarcoplasmic and myofibrillar proteins. About free amino acids， L. sakei significantly degraded serine， arginine and aspartic acid， while S. proteamaculans and H. proteus significantly degraded serine and lysine. In addition， L. sakei had a strongest effect on promoting free fatty acids production， followed by S. proteamaculans and finally H. proteus. Evaluating from various indicators， the co-culture of the three strains did not have any effect. The key volatiles produced by L. sakei were 1-hexanol， acetic acid and hexanoic aicd， S. proteamaculans were 1-hexanol and acetoin， and H. proteus was 1-hexanol， acetic acid and acetoin. In the end， 1-hexanol， hexanoic acid and acetoin were proven to be spoilage markers for vacuum-packaged and chilled lamb. This study can provide fundamental information for inhibiting and rapid identification of spoilage in vacuum-packaged lamb.

Surface electromyography(sEMG)is widely used for analyzing and controlling lower limb assisted exoskeleton robots.Behavior intention recognition based on sEMG is of great significance for achieving intelligent prosthetic and exoskeleton control.Achieving highly efficient recognition while improving performance has always been a significant challenge.To address this,we propose an sEMG-based method called Enhanced Residual Gate Network(ERGN)for lower-limb behavioral intention recognition.The proposed network combines an attention mechanism and a hard threshold function,while combining the advantages of residual structure,which maps sEMG of multiple acquisition channels to the lower limb motion states.Firstly,continuous wavelet transform(CWT)is used to extract signals features from the collected sEMG data.Then,a hard threshold function serves as the gate function to enhance signals quality,with an attention mechanism incorporated to improve the ERGN’s performance further.Experimental results demonstrate that the proposed ERGN achieves extremely high accuracy and efficiency,with an average recognition accuracy of 98.41%and an average recognition time of only 20 ms-outperforming the state-of-the-art research significantly.Our research provides support for the application of lower limb assisted exoskeleton robots.

Transfer printing, as an important assembly technique, has attracted much attention due to its valuable merits to develop novel forms of electronics such as stretchable inorganic electronics requiring the heterogeneous integration of inorganic materials with soft elastomers. Here, we report on a laser-driven programmable non-contact transfer printing technique via a simple yet robust design of active elastomeric microstructured stamp that features cavities filled with air and embedded under the contacting surface, a micro-patterned surface membrane that encapsulates the air cavities and a metal layer on the inner-cavity surfaces serving as the laser-absorbing layer. The micro-patterned surface membrane can be inflated dynamically to control the interfacial adhesion, which can be switched from strong state to weak state by more than three orders of magnitude by local laser heating of the air in the cavity with a temperature increase below 100℃. Theoretical and experimental studies reveal the fundamental aspects of the design and fabrication of the active elastomeric microstructured stamp and the operation of non-contact transfer printing. Demonstrations in the programmable transfer printing of micro-scale silicon platelets and micro-scale LED chips onto various challenging receivers illustrate the extraordinary capabilities for deterministic assembly that are difficult to address by existing printing schemes, thereby creating engineering opportunities in areas requiring the heterogeneous integration of diverse materials such as curvilinear electronics and Micro LED displays.

新零售模式下,高时效性的物流末端配送对提高顾客满意度具有重要作用,而对末端配送中心位置和数量的确定,是优化末端配送网络的关键。首先,综合考虑影响配送中心位置与数量的模糊因素,构建以配送时间最少、配送中心覆盖范围最大以及系统成本最低为目标的模糊多目标规划模型;其次,采用结构元加权序准则法求解模型,生成三角模糊数简化求解过程;最后,以算例检验方法的有效性。结果表明,所提出的模型能够将模糊因素融入模糊决策过程中,且结构元加权序准则法有助于决策者灵活调整方案和决策,从而对新零售末端配送网络布局进行优化。

针对采茧机采茧质量差、效率低等问题,基于TRIZ理论设计一种采茧机器人。采用九屏幕分析法、组件分析法和因果轴分析法,得出采茧机效率低、质量差等问题的根本原因:存在下茧、适应性不足、顶杆维修效率低等;运用技术矛盾法、物理矛盾法和物—场模型法对采茧机存在的问题进行求解,创新设计执行器、姿态转换器、仿形压头及剔茧装置等关键部件,并结合实际工况完成采茧机器人的整机设计。利用有限元仿真软件,进行顶杆与仿形压头采茧试验,分析蚕茧的变形和动力学响应,结果表明:采用仿形压头采茧,蚕茧总变形最大值减少96%,等效应力最大值减少91.7%,等效弹性应变最大值减少88.2%。
