Biomimetic materials that use natural wisdom to solve practical problems are developing rapidly.The trend for systematic biomimicry is towards in-situ characterization of naturalcreatures with high spatial resolutions.Furthermore,rapid reconstruction of digital twin models with the same complex features as the prototype is indispensable.However,it faces bottlenecks and limits in fast characterization and fabrication,precise parameter optimization,geometricdeviations control,and quality prediction.To solve these challenges,here,we demonstrate astate-of-the-art method taking advantage of micro-computed tomography and three-dimensional printing for the fast characterization of the pitcher plant Nepenthes x ventrata and fabrication of its biomimetic model to obtain a superior drainage controller with multiscale structures withprecise surface morphology optimization and geometric deviation control.Thefilm-rupture-based drainage dynamic and mechanisms are characterized by x-ray and high-speed videography,which determines the crucial structures for unique directionaldrainage.Then the optimized artificial pitchers are further developed into sustained drainage devices with novel applications,such as detection,reaction,and smoke control.

Early Holocene environment at a key location of the northwest boundary of the Asian summer monsoon: a synthesis on chronologies of Zhuye Lake, Northwest China

The intensified monsoon increases summer rainfall and creates wet conditions in the Asian summer monsoon region during the early Holocene. Along with millennial-scale changes of the monsoon intensity, it is still unclear whether the boundary of the monsoon region changes according to monsoon variability. Investigations into the early Holocene environment in monsoon marginal zones are crucial for understanding the monsoon boundary changes. Zhuye Lake is located at the northwest edge of the Asian summer monsoon, the northern Qilian Mountains, which are less affected by modern summer monsoon water vapor. Previous studies have reached different conclusions regarding the early Holocene climatic and environmental changes based on different dating methods（^14C and OSL（optically stimulated luminescence）） and materials（shells, carbonate, pollen concentrates and bulk organic carbon）. In this study, we synthesized 102 ^14C dates and 35 OSL dates from ten Holocene sedimentary sections and ten paleo-shorelines in the lake basin. A comparison between ages from different dating methods and materials generally shows that carbon reservoir effects are relatively slight in Zhuye Lake while the disordered chronologies are mainly related to the erosion processes and reworking effects. In addition, proxy data, including lithology, pollen, total organic carbon and carbonate, were collected from different sites of Zhuye Lake. According to the new synthesis, the early Holocene environment was relatively humid, associated with high runoff and lake water levels. The result indicates that the monsoon boundary moves to the north during the period of the intensified monsoon. A typical arid-area lake was formed during the mid-Holocene when carbonate accumulation and high organic matter contents were the main features of this period. The lake retreated strongly during the late Holocene, showing a drought trend. A paleo-hydrological simulation experiment and its verification in an inland basin

Hydrological circulation,as the most basic material cycle and active natural phenomenon on earth,exerts a significant in fluence on climate change.The mid-Holocene is an important period to better understand modern environmental change;however,little research has focused on its quantitative simulation of paleo-hydrological process.In this research,we first collected chronological evidence and sediment records from six Holocene sedimentary sections in the Shiyang River Ba sin to reconstruct the mid-Holocene environment and terminal paleo-lake area.Secondly,we comprehensively analyzed modern pollen combinations and their propagation characteristics in surface soil,air,river and lacustrine sediments in the Shiyang River Basin,and combined the pollen records,as well as quantitatively reconstructed the millennial-scale vegeta tion zones.Finally,based on the land-cover adjustment results during the mid-Holocene,we successfully used the Soil and Water Assessment Tool(SWAT)model,a modern distributed hydrological watershed model,to simulate mid-Holocene runoff in the basin.Results show that the reconstructed climate in the basin was warmer and moister than that in recent times.Vegetation types in the mid-Holocene mainly consisted of sub-alpine shrub distributed between 2,550 m and 2,750 m,forest at an elevation of 2,550 2,750 m,steppe at an elevation of 1,550 2,150 m and desert steppe below 1,550 m.The up stream,midstream,downstream and average annual runoff of the mid-Holocene in the basin were 16.76×10^8 m^3,22.86×10^8 m^3,9.00×10^8 m^3 and 16.20×10^8 m^3 respectively,compared to 15.61×10^8 m^3 of modern annual runoff.Also,the area of terminal paleo-lake in the mid-Holocene was 628 km^2.Thus,this study provides a new quantitative method for paleo-hy drological simulation.

半胱天冬氨酸酶(caspase)在哺乳动物中参与调控细胞凋亡过程,控制着多种疾病的发生发展。本研究利用基因敲除和回补的方法,获得了禾谷镰孢中半胱天冬氨酸酶基因FgCas4的敲除突变体ΔFgCas4和回补体ΔFgCas4-C。通过观察分析发现,基因FgCas4的缺失并不影响禾谷镰孢(Fusarium graminearum)的生长速率、菌落形态、分生孢子产量、致病和产毒。但是,敲除突变体ΔFgCas4的菌丝分支变多且具有3个隔膜的分生孢子比例较野生型和回补体增加了8.7%。对外界环境胁迫因子耐受性测定显示,ΔFgCas4对杀菌剂和金属离子的敏感性显著增加,同时其细胞内脂滴含量增多,对渗透胁迫因子抗性变强。亚细胞定位发现,半胱天冬氨酸酶FgCas4定位于液泡中。此外,基因FgCas4的缺失导致F.graminearum细胞自噬过程明显提前。上述研究结果表明,F.graminearum中半胱天冬氨酸酶FgCas4主要在菌体无性繁殖、对不同环境胁迫的应答以及细胞自噬过程中发挥重要作用。

Relationship between pollen assemblages and organic geochemical proxies and the response to climate change in the Zhuye Lake sediments

This paper examines the relationship among organic geochemical proxies （TOC, C/N ratio and ~13C） and pollen assemblages in Zhuye Lake sediments since the Late Glacial. Results show that the reaction extent of organic geochemical proxies and pollen as- semblages to environment changes are different. Organic geochemical proxies are sensitive to overall environmental change, while pollen assemblages indicate detailed information of environmental change. For the entire sedimentary section （except the sand layer fi＇om the bottom of the section）, when values of TOC, C/N ratio and total pollen concentrations are high, 813C values are low, and vice versa. The different responses of organic geochemical proxies and pollen records in Zhuye Lake are mainly due to their different sensitivity and diverse influencing factors in different environmental conditions.

干旱区盐碱土是中国重要的土地资源,对其进行改良利用对于推进中国绿色可持续发展具有重要意义,关系着国家粮食安全与生态安全。微生物作为土壤生态系统的重要组成部分,在改良治理盐碱土、提高植物耐盐性等方面发挥着重要作用。开展干旱区盐碱土微生物多样性、群落结构、功能特征及其影响因素的深入研究,可以为干旱区盐碱土的修复和生态重建提供微生物方面的重要参考。本文整理了干旱区盐碱土的概况,分析盐碱土中的微生物多样性、群落结构和生态功能特征,梳理盐碱环境对土壤微生物群落的影响因素,提出干旱区盐碱土微生物研究中存在的问题及今后的发展方向,以期为中国干旱区盐碱土开发利用和微生物资源管理提供参考。

为研发高免疫原性的新型狂犬病颗粒样疫苗,利用PCR方法扩增狂犬病病毒HEP-Flury株的G蛋白和M蛋白基因序列,将其依次克隆至Bac-to-Bac杆状病毒表达系统穿梭载体pFastBac dual上,构建重组质粒pFD-GM。制备基于pFD-GM的重组杆粒,转染至Sf9细胞,得到重组杆状病毒rb-GM,在Sf-9细胞中表达获得病毒样颗粒VLP-GM。经SDS-PAGE、Western blot和间接免疫荧光鉴定,重组杆状病毒成功表达了G蛋白(约56 kDa)和M蛋白(约23 kDa);经电镜观察,VLP-GM大小约为100 nm×50 nm,呈表面带有纤突的子弹形状,与典型的狂犬病病毒粒子类似。将VLP-GM免疫犬,经测定犬在二次免疫后产生较灭活疫苗更高的中和抗体水平,达到7.81 U/mL,并且与单独免疫G蛋白相比可刺激更多细胞因子如白细胞介素4(IL-4)和γ-干扰素(IFN-γ)生成。本研究成功表达了狂犬病病毒的病毒样颗粒VLP-GM,证明了其免疫原性,为后续的疫苗研制和抗体制备奠定了基础。

Aiming at the indoor positioning by using SIFT algorithm, because the structure information is more or similar, and there is no obvious identification, which will lead to low accuracy and poor applicability, this paper proposes an improved SIFT algorithm for image matching. Choosing Junction feature which is more suitable for indoor structure, and combining with the improved CPW energy function method to eliminate mismatching points. By testing the actual image to compare and analyze get the feasibility and validity of the new algorithm.
