Chronic myelogenous leukemia(CML)is a malignancy from bone marrow myeloid stem cells mainly driven by the fusion gene BCR-ABL.In addition to BCR-ABL,other genes including RNF6 are also dysregulated in CML cells.1 RNF6,a ubiquitin ligase of the RING family,promotes various cancer cell proliferation,chemoresistance,and tumor growth in vivo by targeting various proteins for ubiquitination and degradation,including SHP1,Tle3,FOXA1,and MAD1.^(2) However,its specific mechanism in CML is not known.

Recently,Peter et al reported a novel form of cell death(cuproptosis)that was different from other known death mechanisms.1 They found that accumulation of copper ions could induce destabilization of Fe-S cluster proteins and aggregation of mitochondrial lipoylated proteins,ultimately resulting in cuproptosis.1 of note,ten genes were identified as cuproptosis-related genes(CRGs)in copper ion-induced cell death,including PDHB,PDHA1,DLAT,DLD,LIPT1,LIAS,FDX1,CDKN2A,GLS,and MTF1.1 Since dysregulation of copper metabolism is involved in many cancers,cuproptosis and CRGs may play vital roles in cancer development and treatment.1.2 Herein,our pancancer analysis revealed the coordinated upregulation of 9 of 10 CRGs in human liver hepatocellular carcinoma(LIHC)across 33 solid tumors(Fig.S1A;Fig.1A),suggesting the distinct role of CRGs in LIHC.We then explored the genetic landscape,biological function,and clinical significance of CRGs in LIHC,and validated our bioinformatic findings by in vitro experiments and clinical cohorts.The detailed methods were described in the supplementary material.

Optimization of Gas-Flooding Fracturing Development in Ultra-Low Permeability Reservoirs

Ultra-low permeability reservoirs are characterized by small pore throats and poor physical properties, which areat the root of well-known problems related to injection and production. In this study, a gas injection floodingapproach is analyzed in the framework of numerical simulations. In particular, the sequence and timing of fracturechanneling and the related impact on production are considered for horizontal wells with different fracturemorphologies. Useful data and information are provided about the regulation of gas channeling and possible strategiesto delay gas channeling and optimize the gas injection volume and fracture parameters. It is shown that inorder to mitigate gas channeling and ensure high production, fracture length on the sides can be controlled andlonger fractures can be created in the middle by which full gas flooding is obtained at the fracture location in themiddle of the horizontal well. A Differential Evolution (DE) algorithm is provided by which the gas injectionvolume and the fracture parameters of gas injection flooding can be optimized. It is shown that an improvedoil recovery factor as high as 6% can be obtained.

The influences of perforating phase and bedding planes on the fracture deflection in laminated shale

The perforating phase leads to complex and diverse hydraulic fracture propagation behaviors in laminated shale formations. In this paper, a 2D high-speed imaging scheme which can capture the interaction between perforating phase and natural shale bedding planes was proposed. The phase field method was used to simulate the same conditions as in the experiment for verification and hydraulic fracture propagation mechanism under the competition of perforating phase and bedding planes was discussed.The results indicate that the bedding planes appear to be no influence on fracture propagation while the perforating phase is perpendicular to the bedding planes, and the fracture propagates along the perforating phase without deflection. When the perforating phase algins with the bedding planes, the fracture initiation pressure reserves the lowest value, and no deflection occurs during fracture propagation. When the perforating phase is the angle 45°, 60°and 75°of bedding planes, the bedding planes begin to play a key role on the fracture deflection. The maximum deflection degree is reached at the perforating phase of75°. Numerical simulation provides evidence that the existence of shale bedding planes is not exactly equivalent to anisotropy for fracture propagation and the difference of mechanical properties between different shale layers is the fundamental reason for fracture deflection. The findings help to understand the intrinsic characteristics of shale and provide a theoretical basis for the optimization design of field perforation parameters.

An Improved YOLOv7 tiny Algorithm for Vehicle and Pedestrian Detection with Occlusion in Autonomous Driving

Future transportation is advancing in the direction of intelligent transportation systems, where an essential part is vehicle and pedestrian detection. Due to the complex urban traffic environment, vehicles and pedestrians in road monitoring have different forms of occlusion problems, resulting in the missed detection of objects. We design an improved you only look once version 7(YOLOv7) tiny algorithm for vehicle and pedestrian detection under occlusion, with the following four main improvements. In order to locate the object more accurately, 1 × 1 convolution and identity connection are added to the 3 × 3 convolution, and convolution reparameterization is used to enhance the inference speed of the network model. In view of the complex road background and more interference, the coordinate attention was added to the connection part of backbone and neck to enhance the network's capacity to detect the object and lessen interference from other targets. At the same time, before being sent to the detection head, global attention mechanism is added to improve the accuracy of model detection by capturing three-dimensional features. Considering the issue of imbalanced training samples, we propose focal complete intersection over union(CIOU) loss instead of CIOU loss to become the bounding box regression loss, so that the regression process attention to high-quality anchor boxes. Experiments show that the improved YOLOv7 tiny algorithm achieves 82.2%map@0.5 in Pattern Analysis, Statistical Modelling and Computational learning Visual Object Classes(PASCAL VOC) dataset, which is 2.8% higher than before the improvement. The performance of map@0.5:0.95 is 5.2% better than the previous improvement. The proposed improved algorithm can availably to detect partial occlusion objects.

Background:Oleanolic acid(OA),a pentacyclic triterpenoid exhibiting speciﬁc anti-cancer properties and highly effective antioxidant activity,was isolated from traditional Chinese medicinal herbs.Conversely,the OA that impacts colon cancer(CC)cells and its underlying mechanisms remain poorly understood.Methods:The cytotoxic effect of OA alone or OA-5-Fluorouracil(5-FU)combination on normal and CC cells was analyzed by methyl thiazolyl diphenyl-tetrazolium bromide(MTT).Then,the impact of OA on CC cell lines(LoVo and HT-29)proliferation and stemness were measured using colon formation and tumorsphere formation assays.Octamer-binding transcription factor 4(Oct4),Prominin-1(CD133),Nanog,and transcription factor SOX-2(SOX2)are cell stemness-related indicators whose expression was assessed usingﬂuorescence qPCR assay,Western blotting,and immunohistochemistry.The effect of OA on the proliferative potency of CC cells was evaluated using an in vivo model.Results:The stem-like characteristics and clone production of colon cancer cells were markedly reduced by OA alone or in combination with OA-5-FU.Moreover,OA increases the susceptibility of CC cells to 5-FU by blocking the cell stemness-related markers(CD133,Nanog,SOX2,and Oct4)expression levels both in vitro and in vivo,as well as by inactivating the activator of transcription 3(STAT3 signaling)and Janus kinase 2/signal transducer(JAK2).Conclusion:Theseﬁndings imply that oleanolic acid,both in vitro and in vivo,suppresses the JAK2/STAT3 pathway,which in turn reverses chemoresistance and decreases colon cancer cell stemness.Therefore,by reducing the recommended amount of 5-FU,this strategy may improve chemotherapeutic effectiveness and minimize undesired side effects.

Existence of multi-hump generalized homoclinic solutions for a class of reversible systems

In this paper, we investigate a class of reversible dynamical systems in four dimensions. The spectrums of their linear operators at the equilibria are assumed to have a pair of positive and negative real eigenvalues and a pair of purely imaginary eigenvalues for the small parameterμ> 0, where these two real eigenvalues bifurcate from a double eigenvalue 0 forμ=0. It has been shown that this class of systems owns a generalized homoclinic solution with one hump at the center （a homoclinic solution exponentially approaching a periodic solution with a small amplitude）. This paper gives a rigorous existence proof of two-hump solutions.These two humps are far away and are glued by the small oscillations in the middle if some appropriate free constants are activated. The obtained results are also applied to some classical systems. The ideas here may be used to study the existence of 2k-hump solutions.

The spatial arrangement of activating ligands is known to have great influence on T cell activation.However,independently studying each ligand’s spatial organization parameter that affects T cell activation remains a great challenge.Here,with DNA origami,we precisely organized the CD3ɛantibodies simulating T cell receptor(TCR)ligands and CD28 antibodies simulating co-stimulatory ligands to interrogate the independent role of TCR-ligand spacing and local copy numbers as well as the spacing between TCR ligands and co-stimulatory ligands on T cell activation.We found that T cell activation benefited fromlocally concentrated TCR ligands with a shorter spacing and was maximized by an∼38 nm spacing between TCR ligands and co-stimulatory ligands.The T cell expander constructed based on our findings could efficiently expand CD8+T cells for tumor immunotherapy.Thus,the DNA nanostructurebased ligands’precise arrangement can be a unique tool in studying immune cell activations and cellbased immunotherapies.

AOYOLO Algorithm Oriented Vehicle and Pedestrian Detection in Foggy Weather

In the context of complex foggy environments, the acquired images often suffer from low visibility,high noise, and loss of detailed information. The direct application of general object detection methods fails to achieve satisfactory results. To address these issues, this paper proposes a foggy object detection method based on YOLOv8n,named AOYOLO. The all-in-one dehazing network(AOD-Net), a lightweight defogging network, is employed for data augmentation. Additionally, the ResCNet module is introduced in the backbone to better extract features from low-illumination images. The GACSP module is proposed in the neck to capture multi-scale features and effectively utilize them, thereby generating discriminative features with different scales. The detection head is improved using WiseI oU, which enhances the accuracy of object localization. Experimental evaluations are conducted on the publicly available datasets annotated realworld task-driven testing set(RTTS) and synthetic foggy KITTI dataset. The results demonstrate that the proposed AOYOLO algorithm outperforms the original YOLOv8n algorithm with an average mean average precision improvement of 3.3% and 4.6% on the RTTS and KITTI datasets, respectively. The AOYOLO method effectively enhances the performance of object detection in foggy scenes. Due to its improved performance and stronger robustness, this experimental model provides a new perspective for foggy object detection.

Targeted delivery of anti-tumor drugs and overcoming drug resistance in malig-nant tumor cells remain significant clinical challenges.However,there are only few effective methods to address these issues.Extracellular vesicles(EVs),actively secreted by cells,play a crucial role in intercellular information transmis-sion and cargo transportation.Recent studies have demonstrated that engineered EVs can serve as drug delivery carriers and showed promising application prospects.Nevertheless,there is an urgent need for further improvements in the isolation and purification of EVs,surface modification techniques,drug assem-bly processes,and precise recognition of tumor cells for targeted drug delivery purposes.In this review,we summarize the applications of engineered EVs in cancer treatment and overcoming drug resistance,and current challenges asso-ciated with engineered EVs are also discussed.This review aims to provide new insights and potential directions for utilizing engineered EVs as targeted delivery systems for anti-tumor drugs and overcoming drug resistance in the near future.
