Construction and validation of a novel prediction system for detection of overall survival in lung cancer patients

BACKGROUND Many factors have an aberrant effect on the overall survival of lung cancer(LC)patients.In recent years,remarkable progress has been made in immunotherapy,targeted treatment,and promising biomarkers.However,the available treatments and diagnostic methods are not specific for all patients.AIM To establish a system for predicting poor survival in patients with LC.METHODS The expression matrix and clinical information for this study were obtained from The Cancer Genome Atlas and Gene Expression Omnibus databases.After the differential analysis of all screened genes,weighted gene coexpression network analysis was performed to analyze hub genes related to patient survival.A logistic regression model was used to construct the scoring system.The expression of the hub genes was verified by performing quantitative reverse transcription-polymerase chain reaction.RESULTS A total of 5007 differentially expressed genes were selected for the Weighted Gene Co-expression Network Analysis algorithm.We found that the turquoise module showed the highest correlation with patient prognosis.The gene module with the greatest positive correlation with patient survival was located in the turquoise area.The Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses performed for the genes contained in the turquoise module indicated the potential roles of the selected genes in the regulation of LC development.In addition,protein–protein interaction analysis was performed to screen hub genes,which identified 100 hub genes located in the core area of the network.We then intersected the 100 hub genes with 75 key genes sorted by module members to identify real hub genes associated with prognosis.Forty-one genes were finally selected.We then used a logistic regression model to determine 11 independent risk genes,namely CCNB2,CDC20,CENPO,FOXM1,HJURP,NEK2,OIP5,PLK1,PRC1,SKA1,UBE2 C and SPARC.CONCLUSION We constructed a predictive model based on 11 independent risk genes to establish a system predicting the survival status of pa

Due to narrowband emission and high quantum efficiencies,polycyclic aromatic heterocycles with multi-resonance thermally activated delayed fluorescence(MR-TADF)properties have recently gained considerable attention in the organic optoelectronic field.Albeit their great promise in the full visible region covering from blue to red,MR-TADF emitters with ultraviolet emission have been rarely reported.Through locking the two ortho-positions of a triphenylamine core by sulfone groups,a simple polycyclic aromatic heterocycle,BTPT,was facilely constructed,exhibiting 368 nm ultraviolet emission with a narrow full width at half maximum(FWHM)of 33 nm.Its neat film exhibited distinct TADF property with a main emission peak at 388 nm.Noteworthily,the enantiomeric crystals of BTPT not only demonstrated significant circularly polarized luminescence(CPL)with large luminescence dissymmetry factor in the 10^(-3) order but also displayed obvious room temperature phosphorescence(RTP).The relationship between this innovative helical unit and unique photophysical properties,including ultraviolet MR-TADF,CPL,and RTP,was reasonably revealed.

Room temperature phosphorescence(RTP) has attracted much attention for the applications in information encryption,bioimaging,displaying,and so on.In this work,persistent RTP emission is mainly achieved by the compact face-to-face packing modes with aromatic/perfluoroaromatic interactions.Moreover,it can be further optimized by halogen substitutions with heavy atom effect,electron-withdrawing property,and steric hindrance,resulting in the prolonged RTP lifetime.Furthermore,the multiple fluorine substitutions endowed these luminogens with the water-resistant property,resulting in the maintained RTP emission with water droplets.Therefore,the aromatic/perfluoroaromatic interactions are not only the efficient approach to achieve persistent RTP,but also extend their application to a humid environment or even in water.

Subdural effusion associated with COVID-19 encephalopathy: A case report

BACKGROUND The precise mechanism by which severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)impacts the central nervous system remains unclear,with manifestations spanning from mild symptoms(e.g.,olfactory and gustatory deficits,hallucinations,and headache)to severe complications(e.g.,stroke,seizures,encephalitis,and neurally demyelinating lesions).The occurrence of single-pass subdural effusion,as described below,is extremely rare.CASE SUMMARY A 56-year-old male patient presented with left-sided limb weakness and slurred speech as predominant clinical symptoms.Through comprehensive imaging and diagnostic assessments,he was diagnosed with cerebral infarction complicated by hemorrhagic transformation affecting the right frontal,temporal,and parietal regions.In addition,an intracranial infection with SARS-CoV-2 was identified during the rehabilitation process;consequently,an idiopathic subdural effusion developed.Remarkably,the subdural effusion underwent absorption within 6 d,with no recurrence observed during the 3-month follow-up.CONCLUSION Subdural effusion is a potentially rare intracranial complication associated with SARS-CoV-2 infection.

At present,with the development of technology,the detection of cryptococcal antigen(CRAG)plays an increasingly important role in the diagnosis of cryptococcosis.However,the three major CRAG detection technologies,latex agglutination test(LA),lateral flow assay(LFA)and Enzyme-linked Immunosorbent Assay,have certain limitations.Although these techniques do not often lead to false-positive results,once this result occurs in a particular group of patients(such as human immunodeficiency virus patients),it might lead to severe consequences.

Zinc-based batteries have long occupied the largest share of the primary battery market,but this advantage has not continued in the secondary battery market.This is mainly because the cycling performance of secondary zinc-based batteries is significantly limited by the poor reversibility of zinc electrodes,including the formation of zinc dendrites,electrode deformation,corrosion,and hydrogen evolution.To solve the above problems,researchers have developed many novel strategies,such as surface coating,use of electrode additives,use of electrolyte additives,and electrode structure design.However,the implementation of these strategies inevitably requires consideration of trade-offs because the core factors that limit the reversibility of zinc electrodes are not isolated but intertwined.Therefore,fully understanding the trade-offs in the zinc electrode design process is necessary to fundamentally improve the cycling performance of the zinc electrode and construct a practical secondary zinc-based battery.This perspective gives an introduction to various problems that limit the cycling of zinc electrodes and discusses the theoretical causes of these problems.The trade-offs in various typical strategies are systematically analyzed,and their positive and negative effects on performance are discussed.This work aims to provide insights for the development of highly reversible zinc anodes for practical secondary zinc-based batteries.

Two-dimensional(2D)materials that combine ferromagnetic,semiconductor,and piezoelectric properties hold significant potential for both fundamental research and spin electronic devices.However,the majority of reported 2D ferromagnetic-semiconductor-piezoelectric materials rely on d-electron systems,which limits their practical applications due to a Curie temperature lower than room temperature(RT).Here,we report a high-crystallinity carbon nitride(CCN)material based on sp-electrons using a chemical vapor deposition *** exhibits a band gap of 1.8 eV and has been confirmed to possess substantial in-plane and out-of-plane piezoelectricity.Moreover,we acquired clear evidences of ferromagnetic behavior at room temperature.Extensive structural characterizations combined with theoretical calculations reveal that incorporating structural oxygen into the highly ordered heptazine structure causes partial substitution of nitrogen sites,which is primarily responsible for generating room-temperature ferromagnetism and piezoelectricity.As a result,the strain in wrinkles can effectively modulate the domain behavior and piezoelectric potential at room temperature.The addition of RT ferromagnetic-semiconductor-piezoelectric material based on sp-electrons to the family of two-dimensional materials opens up numerous possibilities for novel applications in fundamental research and spin electronic devices.

Natural variation of an autophagy-family gene among rice subspecies affects grain size and weight

Elucidating the genetic basis of natural variation in grain size and weight among rice varieties can help breeders develop high-yielding varieties.We identified a novel gene,GW3a(Grain Weight 3a)(LOC_Os03g27350),that affects rice grain size and weight.gw3a mutants showed higher total starch content and dry matter accumulation than the wild type(WT),Nipponbare,suggesting that GW3a negatively regulates grain size and weight.Moreover,our study found that GW3a interacted with OsATG8 by cleaving it,suggesting that GW3a may be involved in the assembly of autophagosomes and starch degradation in plants.The haplotype analysis of GW3a showed functional differences between indica and japonica rice.Taken together,we conclude that GW3a is expressed in the autophagosome pathway regulating starch metabolism in rice,affecting yield-related traits,such as grain size,grain weight and thousand grain weight(TGW).Our findings also shed new light on autophagy-mediated yield trait regulation,proposing a possible strategy for the genetic improvement of high-yield germplasm in rice.

The m^(6)A regulator KIAA1429 stabilizes RAB27B mRNA and promotes the progression of chronic myeloid leukemia and resistance to targeted therapy

Chronic myeloid leukemia(CML)is a common adult leukemia.Both the acute phase of the disease and the adverse effects of anti-cancer treatments can lead to a poor prognosis.The N6-methyladenine(m^(6)A)modification plays an important regulatory role in various physiological and pathological processes.KIAA1429 is a known m^(6)A regulator,but the biological role of KIAA1429 in CML is unclear.In this study,we observed that the m^(6)A levels and KIAA1429 expression were significantly up-regulated in patients with blast phase CML.Notably,KIAA1429 regulated the total level of RNA m^(6)A modification in the CML cells and promoted the malignant biological behaviors of CML cells,including proliferation,migration,and imatinib resistance.Inhibiting KIAA1429 in CML cells reduced the stability of RAB27B mRNA through the m^(6)A/YTHDF1 axis,consequently inhibiting CML proliferation and drug efflux,ultimately increasing the sensitivity of CML cells to imatinib.Moreover,the knockdown of RAB27B also inhibited the proliferation and drug resistance of CML cells and promoted their apoptosis.Rucaparib,a recently developed anti-cancer agent,suppressed the expression of KIAA1429 and CML cell proliferation and promoted cell apoptosis.Rucaparib also inhibited the tumorigenesis of CML cells in vivo.The combined use of rucaparib and imatinib enhanced the sensitivity of CML cells to imatinib.Our study provides evidence that elevated KIAA1429 expression in the blast phase of CML enhances the stability of RAB27B mRNA through the m^(6)A/YTHDF1 axis to up-regulate RAB27B expression,thereby promoting CML progression.Rucaparib exerts inhibitory effects on KIAA1429 expression and thus reduces CML progression.
