Spinal cord injury(SCI)is a devastating traumatic disease seriously impairing the quality of life in patients.Expectations to allow the hopeless central nervous system to repair itself after injury are unfeasible.Developing new approaches to regenerate the central nervous system is still the priority.Exosomes derived from mesenchymal stem cells(MSC-Exo)have been proven to robustly quench the inflammatory response or oxidative stress and curb neuronal apoptosis and autophagy following SCI,which are the key processes to rescue damaged spinal cord neurons and restore their functions.Nonetheless,MSC-Exo in SCI received scant attention.In this review,we reviewed our previous work and other studies to summarize the roles of MSC-Exo in SCI and its underlying mechanisms.Furthermore,we also focus on the application of exosomes as drug carrier in SCI.In particular,it combs the advantages of exosomes as a drug carrier for SCI,imaging advantages,drug types,loading methods,etc.,which provides the latest progress for exosomes in the treatment of SCI,especially drug carrier.

Dormancy of Amaranthus retroflexus L. Seeds and Physiological Response Seedlings to Acifluorfen Sodium

Amaranthus retroflexus L. is a serious and widespread malignant weed in soybean fields in Heilongjiang Province. Exploring the dormancy characteristics of A. retroflexus L. seeds and the physiological response of its seedlings to acifluorfen sodium can provide a basis for further researches on its resistance mechanism. Using newly harvested and stored A. retroflexus L. seeds for one year as experimental materials, the effects of different concentrations of HCl, NaOH, water temperature, gibberellic acid(GA) and polyethylene glycol(PEG) on the dormancy and germination of A. retroflexus L. seeds were studied. The sensitivity of A. retroflexus L.to acifluorfen sodium was determined using bioassay. The effects on leaf chlorophyll content and target enzyme activity were studied at a normal dosage of 360 g a.i. hm^(-2) and a doubling dosage of 720 g a.i. hm^(-2) of acifluorfen sodium. Newly harvested seeds exhibiting dormancy were soaked in water of various temperatures and in different concentrations of NaOH and HCl, which were ineffective in breaking the seed dormancy. GA could break seed dormancy, and the highest seed germination rate reached 93.33% when they were soaked at 3 000 mg·L^(-1) for 72 h and 4 000 mg·L^(-1) for 48 h. The drought stress was simulated with a 15%-25% polyethylene glycol solution, which had no significant effect on the seed germination rate. The GR_(50) value of acifluorfen sodium for A. retroflexus L. was 705.7 g a.i. hm^(-2), which was 1.96 times the recommended dose in the field. After the application of different doses of acifluorfen sodium, the chlorophyll content of A. retroflexus L. reached its minimum value 3 days after treatment(DAT), and then gradually increased. The activity of the target enzyme protoporphyrinogen oxidase(PPO) reached the highest value at 7 DAT under different dosages, and gradually returned to normal levels thereafter. Chlorogenic acid (CA) is a natural plant-derived polyphenol compound that is widely present in beverages (coffee and tea)， fruits (blueberry， apple， and mulberry)， and medicinal plants (Lonicera japonica， Eucommia ulmoides， and Cichorium intybus). In recent years， CA has attracted extensive attention due to its various health benefits， such as anti-inflammatory， immunomodulatory， anti-obesity， anti-diabetes mellitus， and neuroprotection activities. Interestingly， the low bioavailability of CA corresponds to high bioactivity， which raises a key scientific question of how low bioavailability CA can achieve high biological activity. In this review， we highlight the gut microbiota as a key target for connecting the two by summarizing a large amount of evidence. Specifically， the composition and abundance of gut microbiota， its metabolites， intestinal immunity， barrier function and so on were all changed under different pathological conditions such as inflammatory bowel disease， liver disease， kidney disease， cardiovascular disease， diabetes mellitus， obesity， etc. Conversely， these changes could be reversed by CA. In a word， CA could achieve pharmacological activity as well as health-protective effects by modulating intestinal immune and barrier function， gut microbiota composition， gut microbiota metabolites， and signaling pathways. This review provides new targets for the prevention and treatment of diseases by CA. Meanwhile， regulating the composition of the gut microbiota through natural products may be a potential strategy to achieve health protection.

The scope of stereochemistry recognition usually occurs near the chiral scaffold of a ligand or catalyst.Remote stereocontrol,which can surpass the limits of stereorecognition of remote prochiral centers,has long been a challenging object of great interest in asymmetric catalysis.The current work realized the remote stereocontrol of 1,7-zwitterion intermediates formed from Huang's o-amino aryl MBH carbonates.With simple and easily accessibleβ-ICD as the bifunctional catalyst,multifunctionalized tetrahydroquinoline derivatives could be synthesized via(4+2)cycloadditions with excellent enantioselectivity and diastereoselectivity under mild conditions.The strategy possesses broad substrate scope,and three types of electron-deficient enones are successfully applied.Mechanistic studies disclosed the Lewis base-catalyzed reaction pathway,and H-bonding between the catalyst and enones is crucial for long-range stereocontrol.Scale-up reaction and transformations of the tetrahydroquinoline products demonstrated the potential of this strategy.

Clinical analysis of multiple primary gastrointestinal malignant tumors:A 10-year case review of a single-center

BACKGROUND Multiple primary malignant tumors(MPMTs)was first described by Billroth as early as 1889,with the first report published by Warren and Gates in 1932.Since then,numerous cases have been reported.A literature review of 1104269 patients with cancer revealed that the incidence of MPMTs ranged from 0.73 to 11.7%.In recent years,however,there has been a significant upward trend in the incidence of this phenomenon,which may be associated with many different factors,including the advancement of modern diagnostic procedures facilitating the examination and diagnosis of more MPMTs,increased exposure to chemotherapy and radiotherapy that exacerbate the risk of new malignant tumors in patients with cancer,and prolonged survival of patients with cancer allowing sufficient time for the development of new primary cancers.AIM To analyze the incidence,clinical features,treatment factors,prevalence,and prognosis of patients with MPMTs in the gastrointestinal tract treated in a single center.Additionally,we analyzed the different tumor combinations,time interval between the occurrence of tumors,and staging.METHODS This retrospective cohort study analyzed 8059 patients with pathologically confirmed gastrointestinal malignant tumors treated at the Gansu Province Hospital in Lanzhou,Gansu,China between June 2011 and June 2020.Of these,85 patients had MPMTs.The clinical features,treatment factors,prevalence,and prognosis of this latter cohort were analyzed.RESULTS The incidence of MPMTs in patients with gastrointestinal malignant tumors was 1.05%(85/8059),including 83 double primary malignant tumors and two triple primary malignant tumors of which 57(67.06%)were synchronous MPMTs(SMPMTs)and 28(32.94%)were metachronous MPMTs(MMPMTs).The most frequent associations were found between the rectum colon cancers within the SMPMT category and the gastric-colon cancers within the MMPMT category.For the MMPMTs,the median interval was 53 months.The overall 1-,3-and 5-year survival rates from diagnosis of the first primary cancer were 9

Health indicator(HI)construction is a crucial task in degradation evaluation and facilitates the prognostic and health management(PHM)of rotating machinery.Excluding interference from artificial labeling,the HI construction approaches in an unsupervised manner have attracted substantial attention.Nevertheless,current unsupervised methods generally struggle with two problems:(1)ignorance of both redundancy between features and global variability of features during the feature selection process;(2)inadequate utilization of information from different sampling moments.To tackle these problems,this work develops a novel unsupervised approach for HI construction that integrates multi-criterion feature selection and the Attentive Variational Autoencoder(Attentive VAE).Explicitly,a multi-criterion feature selection(Mc FS)algorithm together with an elaborately designed metric is proposed to determine a superior feature subset,considering the relevance,the redundancy,and the global variability of features simultaneously.Then,for the adequate utilization of the information from distinct sampling moments,a deep learning model named Attentive VAE is established.The Attentive VAE is solely fed with the selected features in the health state for model training and the HI is derived through the reconstruction error to reveal the degradation degree of machinery.Two case studies based on genuine experimental datasets are involved to quantitatively evaluate the superiority of the developed approach,demonstrating its superiority over other unsupervised methods for characterizing degradation processes.The effectiveness of both the Mc FS algorithm and the Attentive VAE is verified by ablation experiments,respectively.

The hypoxic microenvironment is an essential characteristic of most malignant tu-mors.Notably,hypoxia-inducible factor-1 alpha(HIF-1a)is a key regulatory factor of cellular adaptation to hypoxia,and many critical pathways are correlated with the biological activity of organisms via HIF-1a.In the intra-tumoral hypoxic environment,HIF-1αis highly expressed and contributes to the malignant progression of tumors,which in turn results in a poor prog-nosis in patients.Recently,it has been indicated that HiF-1αinvolves in various critical pro-cesses of life events and tumor development via regulating the expression of HiF-1a target genes,such as cell proliferation and apoptosis,angiogenesis,glucose metabolism,immune response,therapeutic resistance,etc.Apart from solid tumors,accumulating evidence has re-vealed that HiF-1αis also closely associated with the development and progression of hemato-logical malignancies,such as leukemia,lymphoma,and multiple myeloma.Targeted inhibition of HiF-1a can facilitate an increased sensitivity of patients with malignancies to relevant ther-apeutic agents.In the review,we elaborated on the basic structure and biological functions of HIF-1a and summarized their current role in various malignancies.It is expected that they will have future potential fortargeted therapy.

The eye,as a specialized visual organ,is directly exposed to the external environment,and,therefore,it faces constant chal-lenges from external pathogenic organisms and toxins.In the ocular mucosa(OM)of mammals,mucosal-associated lymphoid tissues(MALTs)constitute the primary line of defense.However,the immune defense role of the OM remains unknown in aquatic vertebrates.To gain insights into the immune processes within the OM of teleost fish,we developed an infection model of rainbow trout(Oncorhynchus mykiss)OM using a parasite,Ichthyophthirius multifiliis(Ich).Immunofluorescence,qPCR,and H&E staining revealed that Ich successfully infiltrates the OM of rainbow trout,leading to pathological struc-tural changes,as evidenced by A&B staining.Importantly,the qPCR results indicate an up-regulation of immune-related genes following Ich infection in the OM.Moreover,transcriptome analyses were conducted to detect immune responses and impairments in eye function within the OM of rainbow trout with Ich infection.The results of the transcriptome analysis that Ich infection can cause an extensive immune response in the OM,ultimately affecting ocular function.To the best of our knowledge,our findings represent for the first time that the teleost OM could act as an invasion site for parasites and trigger a strong mucosal immune response to parasitic infection.

Spacelike Framed Curves with Lightlike Components and Singularities of Their Evolutes and Focal Surfaces in Minkowski 3-space

In this paper,we define the evolute and focal surface of a spacelike framed curve with lightlike components in Minkowski 3-space.It is a generalization of the previous results of regular spacelike curves,since singularities are allowed in the original spacelike curves studied by spacelike framed curves with lightlike components.Meanwhile,we show a new geometric invariant to characterise singularities of the focal surface.Then,the classification theorem and recognition theorem for the singularities of the focal surface in generic are also given.
