In this paper, the issues of stochastic stability analysis and fault estimation are investigated for a class of continuoustime Markov jump piecewise-affine(PWA) systems against actuator and sensor faults. Firstly, a novel mode-dependent PWA iterative learning observer with current feedback is designed to estimate the system states and faults, simultaneously, which contains both the previous iteration information and the current feedback mechanism. The auxiliary feedback channel optimizes the response speed of the observer, therefore the estimation error would converge to zero rapidly. Then, sufficient conditions for stochastic stability with guaranteed performance are demonstrated for the estimation error system, and the equivalence relations between the system information and the estimated information can be established via iterative accumulating representation.Finally, two illustrative examples containing a class of tunnel diode circuit systems are presented to fully demonstrate the effectiveness and superiority of the proposed iterative learning observer with current feedback.

To enhance the catalytic activity of copper ferrite(CuFe_(2)O_(4))nanoparticle and promote its application as combustion catalyst,a low-cost silicon dioxide(SiO_(2))carrier was employed to construct a novel CuFe_(2)O_(4)/SiO_(2)binary composites via solvothermal method.The phase structure,morphology and catalytic activity of CuFe_(2)O_(4)/SiO_(2)composites were studied firstly,and thermal decomposition,combustion and safety performance of ammonium perchlorate(AP)and 1,3,5-trinitroperhydro-1,3,5-triazine(RDX)with it affecting were then systematically analyzed.The results show that CuFe_(2)O_(4)/SiO_(2)composite can remarkably either advance the decomposition peak temperature of AP and RDX,or reduce the apparent activation energy at their main decomposition zone.Moreover,the flame propagation rate of RDX was promoted by about 2.73 times with SiO_(2)content of 3 wt%,and safety property of energetic component was also improved greatly,in which depressing the electrostatic discharge sensitivity of pure RDX by about 1.89 times.In addition,the effective range of SiO_(2)carrier content in the binary catalyst is found to be 3 to 5 wt%.Therefore,SiO_(2)opens a new insight on the design of combustion catalyst carrier and will promote the application of CuFe_(2)O_(4)catalyst in solid propellant.

A double low-density parity-check (D-LDPC) coding system was proposed in[1]as a typical joint source-channel coding(JSCC) scheme[1–5].In this system,two LDPC coding matrices perform source and channel coding,respectively.An important concept in a D-LDPC coding system is the introduction of a linking matrix between the check nodes (CNs)of the source coding matrix and the variable nodes (VNs) of the channel LDPC coding matrix.

BACKGROUND Pulmonary lymphoepithelioma-like carcinoma(PLELC)is a rare type of nonsmall-cell lung cancer.Stomach lymphoepithelioma-like carcinoma(LELC)metastasis secondary to PLELC has not been reported recently.CASE SUMMARY A 64-year-old female was admitted to our hospital for a regular gastroscopy examination with a 6-year history of surgical resection for left PLELC.Positron emission tomography/computed tomography suggested high accumulation of 18F-fludeoxyglucose in the gastric cardia region.Upper gastrointestinal endoscopy confirmed a large mass at the stomach fundus.Immunohistochemistry(IHC)of the biopsy suggested metastatic stomach LELC.Proximal gastrectomy showed that this 6.5 cm×5.0 cm mass was located in the stomach fundus near the cardia.Histopathological examination showed a poorly differentiated carcinoma with prominent lymphoplasmacytic infiltration.IHC demonstrated that the tumor was positive for CK(AE1/AE3),p63,p40,p53,Ki-67(70%),and EGFR(3+)and negative for CK7,CK20,Her2,and CD10.In situ hybridization analysis showed positive staining Epstein-Barr virus-encoded RNA.Tumor programmed cell death ligand 1(PD-L1)expression score was 98%,and the combined positive score was 100,with no evidence of microsatellite instability.Thus,the patient was unequivocally diagnosed with metastatic stomach LELC secondary to pulmonary LELC.After discharge,this patient underwent PD-1 inhibitor treatment(toripalimab,240 mg)every 3 wk for ten cycles,and she has had no tumor recurrence.CONCLUSION For gastric LELC metastasis,PD-1 inhibitor therapy could become a new therapeutic approach,though there is still no evidence from large data sets to support this.

Multi-omics analysis of adamantinomatous craniopharyngiomas reveals distinct molecular subgroups with prognostic and treatment response significance

Background: Adamantinomatous craniopharyngioma (ACP) is the commonest pediatric sellar tumor. No effective drug is available and interpatient heterogeneity is prominent. This study aimed to identify distinct molecular subgroups of ACP based on the multi-omics profiles, imaging findings, and histological features, in order to predict the response to anti-inflammatory treatment and immunotherapies.Methods: Totally 142 Chinese cases diagnosed with craniopharyngiomas were profiled, including 119 ACPs and 23 papillary craniopharyngiomas. Whole-exome sequencing (151 tumors, including recurrent ones), RNA sequencing (84 tumors), and DNA methylome profiling (95 tumors) were performed. Consensus clustering and non-negative matrix factorization were used for subgrouping, and Cox regression were utilized for prognostic evaluation, respectively.Results: Three distinct molecular subgroups were identified: WNT, ImA, and ImB. The WNT subgroup showed higher Wnt/β-catenin pathway activity, with a greater number of epithelial cells and more predominantly solid tumors. The ImA and ImB subgroups had activated inflammatory and interferon response pathways, with enhanced immune cell infiltration and more predominantly cystic tumors. Mitogen-activated protein kinases (MEK/MAPK) signaling was activated only in ImA samples, while IL-6 and epithelial-mesenchymal transition biomarkers were highly expressed in the ImB group, mostly consisting of children. The degree of astrogliosis was significantly elevated in the ImA group, with severe finger-like protrusions at the invasive front of the tumor. The molecular subgrouping was an independent prognostic factor, with the WNT group having longer event-free survival than ImB (Cox,P = 0.04). ImA/ImB cases were more likely to respond to immune checkpoint blockade (ICB) therapy than the WNT group (P <0.01). A decline in mucosal vascularity is a histological hallmark of oral submucous fibrosis (OSF), a premalignant disease that is largely induced by betel quid chewing. However, the lack of available models has challenged studies of angiogenesis in OSF. Here, we found that the expression of thrombospondin 1 (THBS1), an endogenous angiostatic protein, was elevated in the stroma of tissues with OSF. Using a fibroblast-attached organoid (FAO) model, the overexpression of THBS1 in OSF was stably recapitulated in vitro. In the FAO model,treatment with arecoline, a major pathogenic component in areca nuts, enhanced the secretion of transforming growth factor (TGF)-β1 by epithelial cells, which then promoted the expression of THBS1 in fibroblasts. Furthermore, human umbilical vein endothelial cells (HUVECs)were incorporated into the FAO to mimic the vascularized component. Overexpression of THBS1 in fibroblasts drastically suppressed the sprouting ability of endothelial cells in vascularized FAOs (v FAOs). Consistently, treatment with arecoline reduced the expression of CD31in v FAOs, and this effect was attenuated when the endothelial cells were preincubated with neutralizing antibody of CD36, a receptor of THBS1. Finally, in an arecoline-induced rat OSF model, THBS1 inhibition alleviated collagen deposition and the decline in vascularity in vivo. Overall, we exploited an assembled organoid model to study OSF pathogenesis and provide a rationale for targeting THBS1.

Immune checkpoint inhibitors(ICIs) are used to relieve and refuel anti-tumor immunity by blocking the interaction, transcription, and translation of co-inhibitory immune checkpoints or degrading co-inhibitory immune checkpoints. Thousands of small molecule drugs or biological materials, especially antibody-based ICIs, are actively being studied and antibodies are currently widely used. Limitations, such as anti-tumor efficacy, poor membrane permeability, and unneglected tolerance issues of antibody-based ICIs, remain evident but are thought to be overcome by small molecule drugs. Recent structural studies have broadened the scope of candidate immune checkpoint molecules, as well as innovative chemical inhibitors. By way of comparison, small molecule drug-based ICIs represent superior oral bioavailability and favorable pharmacokinetic features. Several ongoing clinical trials are exploring the synergetic effect of ICIs and other therapeutic strategies based on multiple ICI functions, including immune regulation, anti-angiogenesis, and cell cycle regulation. In this review we summarized the current progression of small molecule ICIs and the mechanism underlying immune checkpoint proteins, which will lay the foundation for further exploration.

Satellite glial cells in sensory ganglia play a wider role in chronic pain via multiple mechanisms

Satellite glial cells are unique glial cells that surround the cell body of primary sensory neurons.An increasing body of evidence suggests that in the presence of inflammation and nerve damage,a significant number of satellite glial cells become activated,thus triggering a series of functional changes.This suggests that satellite glial cells are closely related to the occurrence of chronic pain.In this review,we first summarize the morphological structure,molecular markers,and physiological functions of satellite glial cells.Then,we clarify the multiple key roles of satellite glial cells in chronic pain,including gap junction hemichannel Cx43,membrane channel Pannexin1,K channel subunit 4.1,ATP,purinergic P2 receptors,and a series of additional factors and their receptors,including tumor necrosis factor,glutamate,endothelin,and bradykinin.Finally,we propose that future research should focus on the specific sorting of satellite glial cells,and identify genomic differences between physiological and pathological conditions.This review provides an important perspective for clarifying mechanisms underlying the peripheral regulation of chronic pain and will facilitate the formulation of new treatment plans for chronic pain.

Paradigm for Determining the Optimal Ultradeep and Superthick Saline Aquifer for High-TDS Mine Water Geological Storage

Saline aquifers are the most popular waste and CO_(2)injection and storage reservoirs worldwide.This project proposes that several optimal injection positions should be investigated as hydraulic pressure-focused positions,in order to relieve the high demands of pump performance.The comprehensive indices(F_(i))representing the injectivity of different burial depths were obtained by using information entropy,based on the mercury injection experimental data of 13 rock samples.The results demonstrated that the burial depths of No.4,No.1 and No.2 in the Liujiagou Formation were the most suitable positions for hydraulic focused injection,which means the upper 30 m thickness could be regarded as the hydraulic focused range in the saline aquifer with an average thickness of 400 m.In addition,some laboratory experiments and in situ tests were carried out for the purpose of certifying and analyzing results,including SEM,XRD,brittleness index and logging.The results suggested that the rock samples at the No.4,No.1 and No.2 burial depth ranges have loose microstructure,weak cementation,as well as dual pores and fractures.The lithology is mainly quartz and feldspar,but the clay mineral content is high(10%-25%),which is positive for dissolution.The lithology is suitable for hydraulic fracturing to form extended cracks and micro-fissures during high-TDS(total dissolved solids)mine water injection,because of the high brittleness index.Finally,a theoretical and technical framework for high-TDS mine water injection was established,based on operating pilot engineering.Some theoretical defects and drawbacks learned from the field practices were summarized and solutions proposed.The research in this study could provide guidance and a paradigm for the inexpensive treatment of high-TDS mine water by injection and storage.

BACKGROUND Venous thromboembolism significantly contributes to patient deterioration and mortality.Management of its etiology and anticoagulation treatment is intricate,necessitating a comprehensive consideration of various factors,including the bleeding risk,dosage,specific anticoagulant medications,and duration of therapy.Herein,a case of lower extremity thrombosis with multiple primary malignant tumors and high risk of bleeding was reviewed to summarize the shortcomings of treatment and prudent anticoagulation experience.CASE SUMMARY An 83-year-old female patient was admitted to the hospital due to a 2-wk history of left lower extremity edema that had worsened over 2 d.Considering her medical history and relevant post-admission investigations,it was determined that the development of left lower extremity venous thrombosis and pulmonary embolism in this case could be attributed to a combination of factors,including multiple primary malignant tumors,iliac venous compression syndrome,previous novel coronavirus infection,and inadequate treatment for prior thrombotic events.However,the selection of appropriate anticoagulant medications,determination of optimal drug dosages,and establishment of an appropriate duration of anticoagulation therapy were important because of concurrent thrombocytopenia,decreased quantitative fibrinogen levels,and renal insufficiency.CONCLUSION Anticoagulant prophylaxis should be promptly initiated in cases of high-risk thrombosis.Individualized anticoagulation therapy is required for complex thrombosis.
