INTRODUCTIONAcute pancreatitis (AP) is a common severe illness of the digestive tract with variable involvement of other regional tissues and / or remote organ sysems[1-3],Mild disease is associated with minimal organ dysfunction and rapid recovery ,while severe disease is associated with multiple organ system failure and local complications such as necrosis , abscess , fistulas and pseudocyst formation [4-6].

Sodium dimercaptopropane sulfonate is used in place of dimercaprol in the treatment of poisoning by heaving metals and metalloids as a well-known specific antidote. It is claimed to be less toxic. In early stage, the pharmacologists and clinical doctors in Wenzhou medical college found this drug as antidote against non-metallic pesticides as nereistoxin insecticides, Chlordimeform, bactercide 402, and tetramine. We did a lot of animal experiments and clinical trials to confirm that this antagonist possesses a strong antidotal effect. Now it is widely used in china. Great break through has been made not only in the theory but also in its practice. Furthermore, the combination use of this antidote and central anticonvulsant as diazepam indicated synergistic antidotal effect, which is probably the best utilization in the treatment of acute poisoning by tetramine.

BACKGROUND: Recent researches demonstrate that onset of cerebral infarction always accompanies with obvious changes of function of thyroid axis; while, high-homocysteic acidemia has been proved as one of risk factors of vascular dementia and Alzheimer disease. Meanwhile, it is found that level of plasma homocysteic acid is positive correlation with injured degrees of cognitive function and brain damage. OBJECTIVE: To observe the changes of function of thyroid and level of homocysteic acid among patients with vascular dementia and compare with those patients without dementia cerebral infarction. DESIGN: Randomized grouping and contrast observation. SETTING: department of Neurology, People’s hospital affiliated to Yunyang medical college, South china hospital of wuhan University. PARTICIPANTS: A total of 38 patients with vascular dementia were hospitalized in the department of Neurology, People’s hospital affiliated to Yunyang medical college from February 2004 to December 2005. All patients met the diagnostic criteria of the Fourth Edition of Diagnostic and Statistical Manual of Mental Disorder (DSM-IV) established by American Psychiatric Association. Based on educational degrees, Mini-mental Status Examination (MMSE) was classified into illiteracy (≤ 17 points), education of primary school (educational duration ≤ 6 years, ≤ 24 points) and education of middle school or above (educational duration > 6 years, ≤ 24 points). Forty patients with non-dementia cerebral infarction were regarded as the control group and checked with CT examination. There were no significant differences of sex and age between the two groups. All patients and relatives were provided the consent. METHODS: Within 24 hours after hospitalization, patients with vascular dementia received MMSE scores, and the degrees were classified based on the scoring results: mild (20-24 points), moderate (10-19 points) and severe (below 10 points). Inhibitory effect of acetamide-45 on airway inflammation and phosphodiesterase 4 in allergic rats

Aim:To determine the effects of acetamide-45 on respiratory function,airwayinflammation,and the activity of phosphodiesterase 4（PDE4）in allergic ***:Rats were sensitized by a single intramuscular injection with ovalbumin（OVA）and were challenged with ovalbumin applied by using an aerosol repeat-edly for 7 d after 2 ***-45 at concentrations of 5,10,or 30 mg/kg wasthen administered by intraperitoneal *** in dynamic lung compli-ance and lung resistance,the accumulation of inflammatory cells in bronchoalveolarlavage,PDE4 activity,and the concentration of interleukin-4 in rat lung tissuewere ***:Seven days of treatment with acetamide-45 preventedeosinophil accumulation in allergic *** doses of 5,10,and 30 mg/kg,aceta-mide-45 decreased lung resistance to 0.20±0.04,0.25±0.07,and 0.22±0.05cmH2O·s-1·mL-1,respectively（P<0.05 vs OVA）,and it also increased dynamic lungcompliance to 0.41±0.07,0.39±0.06,and 0.42±0.09 mL/cmH20（P<0.05 vs OVA）.After being treated with different doses of acetamide-45,the PDE4 activities inlung tissue were 281±55,273±57,and 238±36 nmol·g-1·min-1（P<0.05 vs OVA）,andthe concentrations of interleukin-4 in lung tissue were 6.22±1.13,5.95±1.20,and 5.68±2.20 μtg/g protein（P<0.05 vs OVA）.Conclusions:Acetamide-45 wasfound to improve respiratory function and inhibit airway inflammation in thisanimal model,and the PDE4 activity of lung tissue was obviously ***-45 was an effective anti-inflammatory agent in respiratory inflammation,and the mechanism of its action might depend on inhibition of PDE4.

AIM: To investigate the role of the mitochondrial pathway in JTE-522-induced apoptosis and to investigate the relationship between cytochrome C release, caspase activity and loss of mitochondrial membrane potential (Deltapsim). METHODS: Cell culture, cell counting, ELISA assay, TUNEL, flow cytometry, Western blot and fluorometric assay were employed to investigate the effect of JTE-522 on cell proliferation and apoptosis in AGS cells and related molecular mechanism. RESULTS: JTE-522 inhibited the growth of AGS cells and induced the apoptosis. Caspases 8 and 9 were activated during apoptosis as judged by the appearance of cleavage products from procaspase and the caspase activities to cleave specific fluorogenic substrates. To elucidate whether the activation of caspases 8 and 9 was required for the apoptosis induction, we examined the effect of caspase-specific inhibitors on apoptosis. The results showed that caspase inhibitors significantly inhibited the apoptosis induced by JTE-522. In addition, the membrane translocation of Bax and cytosolic release of cytochrome C accompanying with the decrease of the uptake of Rhodamin 123, were detected at an early stage of apoptosis. Furthermore, Bax translocation, cytochrome C release, and caspase 9 activation were blocked by *** and Z-IETD-CHO. CONCLUSION: The present data indicate a crucial association between activation of caspases 8, 9, cytochrome C release, membrane translocation of Bax, loss of Deltapsim and JTE-522-induced apoptosis in AGS cells.

Epidermal growth factor receptor （EGFR） is fotmd to express at high levels in a variety of solid tumors including gliomas. This study was to examine the effect of an EGFR-tyrosine kinase inhibitor （AG1478） alone or in combination with cisplatin （CDDP） on the growth of glioma cells （U87）. U87 glioma cells were treated with AG1478 （10 μmol/L） or CDDP （25 μmol/L） as a single agent or in combination for 24 or 48 h. The expression of EGFR and the components in its downstream signaling pathway [extracellular signal-regulated kinase （ERK）, protein kinase B （AKT）] in U87 glioma cells was detected by Western blotting. Cell growth, cell cycle distribution and cell apoptosis were determined by MTT method and flow cytometry, respectively. The results showed that CDDP could induce the activation of EGFR and the components in its downstream signaling pathways in a concentration-dependent manner. The combined treatment of AG1478 with CDDP could inhibit the proliferation of U87 glioma cells, arrest the-cell cycle and promote cell apoptosis. In the EGFR signaling pathway, AG1478 decreased the phosphorylation of ERK, AKT and EGFR in U87 glioma cells. It was：concluded that the combined treatment of AG1478 and CDDP may exert synergistic inhibitory effects on the growth of glioma cells by suooressing the activities of EGFR, AKT and ERK.
