Interactions of thymic stromal lymphopoietin with interleukin-4 in adaptive immunity during Aspergillus fumigatus keratitis

AIM:To investigate the potential interactions of thymic stromal lymphopoietin(TSLP)with interleukin-4(IL-4)in adaptive immunity during fungal keratitis(FK).METHODS:An FK mouse model was induced with Aspergillus fumigatus(AF)hyphal *** were divided into several groups:untreated,phosphate buffer saline(PBS),infected with AF,and pretreated with a scrambled siRNA,a TSLP-specific siRNA(TSLP siRNA),murine recombinant TSLP(rTSLP),immunoglobulin G(IgG),murine recombinant IFN(rIFN-γ),murine recombinant IL-4(rI L-4),rIL-13,murine recombinant IL-17A(rIL-17A),and murine recombinant IL-17F(rIL-17F)*** realtime reverse transcription-polymerase chain reaction(qRTPCR)and enzyme-linked immunosorbent assay(ELISA)or Western blot were performed to determine mRNA and protein levels in the inflamed *** locations were observed by immunofluoresence staining after AF hyphal ***:Compared to those in the untreated group,TSLP and T helper type 1(Th1)cytokine levels in the AF group were upregulated at 24 h post infection(hpi),and those of T helper type 2(Th2)and T helper type 17(Th17)cytokines were increased at 5 d post infection(dpi).Th2 cytokine levels were decreased in the TSLP siRNA-pretreated group and increased in the rTSLP-pretreated group compared with the AF *** TSLP level was increased in the rIL-4-pretreated group,but there were no significant changes among the other *** staining showed cytokine locations after AF hyphal ***:TSLP induces a Th2 immune response and promots Th2 T cell differentiation in ***-4 promotes TSLP ***,TSLP with IL-4 regulates adaptive immunity in FK.

What are the practical limits for the specific surface area and capacitance of bulk sp^2 carbon materials?

The possible practical limits for the specific surface area and capacitance performance of bulk sp^2 carbon materials were investigated experimentally and theoretically using a variety of carbon materials. We find the limit for the specific surface area to be 3500–3700 m^2 g^(-1), and based on this, the corresponding best capacitance was predicted for various electrolyte systems. A model using an effective ionic diameter for the electrolyte ions was proposed and used to calculate the theoretical capacitance. A linear dependence of experimental capacitance versus effective specific surface area of various sp^2 carbon materials was obtained for all studied ionic liquid, organic and aqueous electrolyte systems. Furthermore, excellent agreement between the theoretical and experimental capacitance was observed for all the tested sp^2 carbon materials in these electrolyte systems, indicating that this model can be applied widely in the evaluation of various carbon materials for supercapacitors.

Response of winter cohort abundance of Japanese common squid Todarodes pacificus to the ENSO events

The Japanese common squid Todarodes pacificus is an economically important species with one year lifespan,which is significantly influenced by climatic and environmental variability. According to the fishery data of the winter cohort of T. pacificus from 2003 to 2012, as well as environmental data and the Oceanic Ni？o index（ONI,which was defined by the sea surface temperature（SST） anomaly in the Ni？o 3.4 region）, variations in the SST,chlorophyll a（Chl a） concentration, suitable spawning area（SSA） and sea surface height anomaly（SSHA） on the spawning ground of T. pacificus were examined under the El Ni？o and La Nina conditions. Their influences on squid abundance（defined by catch per unit effort, CPUE） were further assessed. The results showed that seasonal changes were found in SST, Chl a and SSA on the spawning ground of T. pacificus. Correlation analysis suggested that annual CPUE was significantly positively correlated with Chl a and SSA（p〈0.05）, but had insignificant relationship with SST（p〉0.05）. Moreover, the El Ni？o and La Ni？a events tended to dominate the changes of SSA and Chl a concentration in the key area between 25°–29°N and 122.5°–130.5°E, driving the variability of squid abundance. However, this influence varied with the intensity of each anomalous climatic event： the weak El Ni？o event occurred, the spawning ground was occupied by waters with enlarged SSA but with extremely low Chl a concentration, leading to low squid recruitment, the CPUE then decreased; the moderate intensity of El Ni？o event resulted in shrunk SSA but with high Chl a concentration on the spawning ground, the squid recruitment and CPUE increased; the moderate intensity of La Ni？a events yielded elevated SSA and high Chl a concentration on the spawning ground, the squid recruitment and CPUE dramatically increased. Multi-Layer Superposed Coalbed Methane System in Southern Qinshui Basin, Shanxi Province, China

Multi-coalbed developed in Carboniferous–Permian coal-bearing strata of southern Qinshui Basin, and different coal-bearing segments have different coalbed methane（CBM） reservoiring characteristics. Analysis of previous studies suggests that the essence of an unattached CBM system is to possess a unified fluid pressure system, which includes four key elements, namely, gas-bearing coal-rock mass, formation fluid, independent hydrodynamic system and capping layer condition. Based on the exploration and exploitation data of CBM, it is discovered that the gas content of coal seams in southern Qinshui Basin presents a change rule of non-monotonic function with the seam dipping, and a turning point of the change appears nearby coal seam No. 9, and coal seams of the upper and the lower belong to different CBM systems respectively; well test reservoir pressure shows that the gradient of coal seam No. 15 of the Taiyuan Formation is significantly higher than that of coal seam No. 3 of the Shanxi Formation; the equivalent reservoir pressure gradient of coal seam No. 15 ＂jumps＂ obviously compared with the reservoir pressure gradient of coal seam No. 3 in the same vertical well, that is, the relation between reservoir pressure and burial depth takes on a characteristic of nonlinearity; meanwhile, the vertical hydraulic connection among the aquifers of Shanxi Formation and Taiyuan Formation is weak, constituting several relatively independent fluid pressure systems. The characteristics discussed above reveal that the main coal seams of southern Qinshui Basin respectively belong to relatively independent CBM systems, the formation of which are jointly controlled by sedimentary, hydrogeological and structural conditions.

Grazing exclusion-induced shifts,the relative importance of environmental filtering,biotic interactions and dispersal limitation in shaping desert steppe communities,northern China

Grazing exclusion is one of the most efficient approaches to restore degraded grassland but may negatively affects the recovery of species diversity. Changes in plant species diversity should be a consequence of the ecological assembly process. Local community assembly is influenced by environmental filtering, biotic interactions, and dispersal. However, how these factors potentially contribute to changes to species diversity is poorly understood, especially in harsh environments. In this study, two management sites within a Stipa breviflora desert steppe community（typical natural steppe） were selected in northern China. In one of the two management sites, grazing has been excluded since 2010 and in the other with open grazing by sheep. In August 2016, three plots were established and 100 sampling units were created within each plot in a 5 m×5 m area at the two management sites. To assess the effects of grazing exclusion on S. breviflora steppe, we analyzed the vegetation biomass, species diversity,soil organic carbon, and soil particle size distribution using paired T-tests. In addition, variation partitioning was applied to determine the relative importance of environmental filtering and dispersal limitation. Null mode analysis was used to quantify the influence of biotic interactions in conjunction with Eco Sim niche overlap and co-occurrence values. Our results demonstrated that（1） species diversity significantly decreased and the main improvements in soil quality occurred in the topsoil 0–10 cm after the grazing exclusion;（2） environmental filtering was important for community assembly between grazed and fenced grassland and this appears particularly true for soil particle size distribution, which may be well correlated with soil hydrological processes; and（3） however, competitive exclusion may play a significant role within the exclusion. Sedimentary environment and major controlling factors of organic matter-rich shale from the Wufeng-Longmaxi formation in eastern Sichuan Basin,China

The major controlling factors of organic matter and its enrichment model of the black shale from the Wufeng-Longmaxi Formation were explored by investigating the vertical variation characteristics,as well as major element and trace element abundances in the Wuxi Bailu *** results show that the sedimentary tectonic setting of the Wufeng-Longmaxi Formation in the north-east margin of the upper yangtze platform is located on the active continental margin,which is a passive continental margin and continental island *** parent rock in the source area is mainly felsic volcanic rocks mixed with small amounts of sedimentary recycling *** to increased plate activity and a drop in sea levels,terrigenous pyroclastic input *** palaeoclimate was semi-humid,and a robust dysoxic-reduction environment and a high level of palaeoproductivity,causing the formation of the organic-rich shale in the Wufeng *** the base of the Longmaxi Formation,the sedimentary water body was affected by global transgression,showing a strong anoxic-reductive environment,and the paleoclimate was a warm and humid *** palaeoproductivity level was high,resulting in the formation of organic *** to the sea level drop at the top of the Longmaxi Formation,the sedimentary water was in an oxic-reduced environment,but the input of terrigenous pyroclastic matter *** the paleoclimate was warm and humid and the palaeoproductivity level was high,organic-rich shale was *** findings demonstrate that terrigenous clastic input circumstances,palaeoproductivity conditions,and paleo-redox conditions had the greatest influence on the enrichment of organic matter in the Wufeng-Longmaxi ***,organic matter enrichment was controlled by multiple paleoenvironmental factors.

Optical properties of estuarine dissolved organic matter isolated using cross-flow ultrafiltration

Dissolved organic matter （DOM） from freshwater, mid-salinity, and seawater endmember samples in the Jiulong River Estuary, China were fractionated using cross-flow ultrafiltration with a 10-kDa membrane. The colloidal organic matter （COM; 10 kDa-0.22 pm） retentate, low molecular weight （LMW） DOM （〈10 kDa） permeate, and bulk samples were analyzed using absorption spectroscopy and three-dimensional fluorescence excitation-emission-matrix spectroscopy. The UV-visible spectra of COM were very similar to those obtained for permeate and bulk samples, decreasing monotonically with increasing wavelength. Most of the chromophoric DOM （CDOM, expressed as the absorption coefficient a355） occurred in the LMW fraction, while the percentage of CDOM in the colloidal fraction was substantially higher in the freshwater endmembet （13.4% of the total） than in the seawater endmember （6.8%）. The bulk CDOM showed a conservative mixing behavior in the estuary, while there was removal of the COM fraction and a concurrent addition of the permeate fraction in the mid-salinity sample, implying that part of the colloidal CDOM was transformed into LMW CDOM. Two humic-like components （CI：〈250, 325/402 nm; and C2： 265, 360/458 nm） and one protein-like component （C3：275/334 nm） were identified using parallel factor analysis. The contributions of the C1, C2, and C3 components of the COM fraction to the bulk sample were 2.5%-8.7%, 4.8%-12.6%, and 7.4%-14.7%, respectively, revealing that fluorescent DOM occurred mainly in the LMW fraction in the Jiulong River Estuary. The C1 and C2 components in the retentate and permeate samples showed conservative mixing behavior, but the intensity ratio of C2/C1 was higher in the retentate than in the permeate fractions for all salinity samples, showing that the humic component was more enriched in the COM than the fulvic component.
