Background Renal angiomyolipoma(RAML)is the most common kidney lesion in patients with tuberous sclerosis complex(TSC),affecting about 80%of *** is a benign tumor that grows over time,usually bilaterally,and can easily lead to kidney complications such as acute ***,we investigated the efficacy and safety of sirolimus in children with TSC-associated RAML and explored the factors affecting tumor disappearance under sirolimus treatment through subgroup *** A prospective cohort study was *** was initiated at 1 mg/(m^(2)×day),and dose adjustments were made by a 2-week titration period to attain a trough blood concentration of 5-10 ng/*** disappearance of RAML in chil-dren after sirolimus treatment was observed,and Cox regression was used to screen the factors affecting tumor *** One hundred and twenty-six patients who met the criteria were *** 3 months,6 months,12 months,and 24 months of follow-up,tumors disappeared in 18(14.3%),30(23.8%),39(31.0%),and 42(33.3%)children,*** disappeared in 50(39.7%)children by the last visit of each individual,and 30(60%)of them occurred within 6 *** multivariate Cox regression analysis showed that patients with a smaller maximum tumor diameter at baseline had a higher tumor disappearance ***-six(29%)patients had stomatitis during the entire treatment period,and no serious *** Sirolimus could promote the disappearance of TSC-related *** disappearance rate was correlated with the maximum diameter at baseline,and the smaller the tumor was,the higher the disappearance *** is well tolerated in the treatment of RAML associated with TSC.

变循环发动机起动建模及控制规律研究

为研究双外涵变循环发动机起动控制规律,基于部件级建模方法,考虑可调部件变几何特性处理和旋转部件低转速特性外推,采用流量平衡方法构建共同工作方程组,建立变循环发动机起动数学模型。通过改变单个几何调节量,包括风扇、核心机驱动风扇(CDFS)和压气机导叶角度,低压涡轮导向器面积,前涵道引射器(FVABI)面积、后涵道引射器(RVABI)面积和尾喷管喉道面积,采用单因素分析方法,研究其对起动性能的影响,获得几何变量影响矩阵,并基于此开展起动控制规律研究。研究表明:控制规律的风扇导叶、核心机驱动风扇导叶、前涵道引射器和喷管均置于全关闭位置,低压涡轮导向器处于最大位置,后涵道引射器面积和压气机导叶角度随转速下降。经试验验证,起动控制规律正确可行,变循环发动机起动成功。研究结论对变循环发动机起动控制规律和起动性能设计提供了参考。

Dear Editor,Electric vehicle(EV)sales have significantly grown over the years to fulfill growing demands for economic travel and greenhouse gas mitigation.1 However,the surge in the number of EVs has led to charging anxiety as users struggle to find an available charging station before running out of electricity,resulting in longer reserve and waiting times.2 Moreover,severe mobility restrictions caused by infectious diseases,such as coronavirus disease 2019(COVID-19),have greatly affected people’s travel behavior3,4 and hindered their willingness to use EVs,given that charging in public spaces consumes time and increases the risk of contracting the virus.5 This implies that in the postpandemic era,in which individuals coexist with the virus,the interplay between the two important trends,namely vehicle electrification and mobility restrictions,can extensively affect people’s daily commuting by using EVs.

传统研究往往把马克思技术思想凝结为“决定论”“异化”等命题,本文通过对《马克思恩恩格斯全集》(MEGA^(2))等文献分析,发现其技术思想大致经过三个阶段、两次转向:第一阶段(1845年之前)以“巴黎手稿”为标志,表现出技术本质主义的特征;第二阶段(1845-1850年)以“布鲁塞尔笔记”为标志,转向对技术应用问题的研究;第三阶段(1850年之后)以“伦敦笔记”等为代表,转向对工艺本身的经验性研究。其思想的独到之处在于,以技术的经验性考察为基础展开了对资本主义技术应用的批判性反思,在方法论上实现了经验性与规范性的统一,体现了哲学与科学的贯通与融合。由此可见,马克思的技术思想对马克思主义、技术哲学以及当代的人工智能研究都具有借鉴价值。

Rational design of a highly active N-glycosyltransferase mutant using fragment replacement approach

The modularity of carbohydrate-active enzymes facilitates that enzymes with different functions have similar ***,because of the complex structure of the enzyme active sites and the epistatic effects of various mutations on enzyme activity,it is difficult to design enzymes with multiple mutation sites using conventional *** this study,we designed multi-point mutants by fragment replacement in the donor-acceptor binding pocket of Actinobacillus pleuropneumoniae N-glycosyltransferase(ApNGT)to obtain novel *** fragments were selected from a customized glycosyltransferase *** stability and substrate-binding energy of the three fragment replacement mutants were calculated in comparison with wild-type ApNGT,and mutants with top-ranking stability and middle-ranking substrate-binding energy were chosen for priority experimental *** found that a mutant called F13,which increased the glycosylation efficiency of the natural substrate by 1.44 times,the relative conversion of UDP-galactose by 14.2 times,and the relative conversion of UDP-xylose from almost 0 to 78.6%.Most importantly,F13 mutant acquired an entirely new property,the ability to utilize UDP-glucuronic *** one hand,this work shows that replacing similar fragments in the donoracceptor binding pocket of the enzyme might provide new ideas for designing mutants with new properties;on the other hand,F13 mutant is expected to play an important role in targeted drug delivery.

Background Respiratory syncytial virus(RSV)is the leading global cause of respiratory infections and is responsible for about 3 million hospitalizations and more than 100,000 deaths annually in children younger than 5 years,representing a major global healthcare *** is a great unmet need for new agents and universal strategies to prevent RSV infections in early life.A multidisciplinary consensus development group comprising experts in epidemiology,infectious diseases,respiratory medicine,and methodology aims to develop the current consensus to address clinical issues of RSV infections in *** sources The evidence searches and reviews were conducted using electronic databases,including PubMed,Embase,Web of Science,and the Cochrane Library,using variations in terms for"respiratory syncytial virus","RSV","lower respiratory tract infection","bronchiolitis","acute","viral pneumonia","neonatal","infant""children",and"pediatric".Results Evidence-based recommendations regarding diagnosis,treatment,and prevention were proposed with a high degree of *** supportive care remains the cornerstone for the management of RSV infections,new monoclonal antibodies,vaccines,drug therapies,and viral surveillance techniques are being rolled *** This consensus,based on international and national scientific evidence,reinforces the current recommendations and integrates the recent advances for optimal care and prevention of RSV *** improvements in the management of RSV infections will require generating the highest quality of evidence through rigorously designed studies that possess little bias and sufficient capacity to identify clinically meaningful end points.

In the domain of point cloud registration,the coarse-to-fine feature matching paradigm has received significant attention due to its impressive *** paradigm involves a two-step process:first,the extraction of multilevel features,and subsequently,the propagation of correspondences from coarse to fine ***,this approach faces two notable ***,the use of the Dual Softmax operation may promote one-to-one correspondences between superpoints,inadvertently excluding valuable ***,it is crucial to closely examine the overlapping areas between point clouds,as only correspondences within these regions decisively determine the actual *** these issues,we propose OAAFormer to enhance correspondence *** the one hand,we introduce a soft matching mechanism to facilitate the propagation of potentially valuable correspondences from coarse to fine *** the other hand,we integrate an overlapping region detection module to minimize mismatches to the greatest extent ***,we introduce a region-wise attention module with linear complexity during the fine-level matching phase,designed to enhance the discriminative capabilities of the extracted *** on the challenging 3DLoMatch benchmark demonstrate that our approach leads to a substantial increase of about 7%in the inlier ratio,as well as an enhancement of 2%-4%in registration ***,to accelerate the prediction process,we replace the Conventional Random Sample Consensus(RANSAC)algorithm with the selection of a limited yet representative set of high-confidence correspondences,resulting in a 100 times speedup while still maintaining comparable registration performance.

Background and Aims:SARS-CoV-2 vaccines-associated autoimmune liver diseases have been reported in several case ***,the safety and immunogenicity after primary and booster inactivated SARS-CoV-2 vaccination in patients with autoimmune liver diseases(AILD)is still ***:Eighty-four patients with AILD were prospectively followed up after the second dose(primary)of inactivated SARS-CoV-2 *** of them received the third dose(booster)of inactivated *** events(AEs),autoimmune activation,and liver inflammation exacerbation after primary and booster vaccination were ***,dynamics of antireceptor-binding-domain IgG(anti-RBD-IgG),neutralizing antibodies(NAbs)and RBD-specific B cells responses were ***:The overall AEs in AILD patients after primary and booster vaccination were 26.2%and 13.3%,*** decrease of C3 level and increase of immunoglobulin light chain K andλlevels were observed in AILD patients after primary vaccination,however,liver inflammation was not exacerbated,even after booster *** the seroprevalence and titers of anti-RBD-IgG and NAbs were decreased over time in AILD patients after primary ***,the antibody titers were significantly elevated after booster vaccination(10-fold in anti-RBD-IgG and 7.4-fold in NAbs,respectively),which was as high as in healthy ***,the inferior antibody response was not enhanced after booster vaccination in patients with *** of atypical memory B cells were inversely related to antibody levels,which indicate that the impaired immune memory was partially restored partly by the booster ***:The well tolerability and enhanced humoral immune response of inactivated vaccine supports an additional booster vaccination in AILD patients without immunosuppressants.

Rheumatoid arthritis(RA)is a worldwide public health *** to delay or prevent the onset of RA have attracted much attention in recent years,and researchers are now exploring various prevention *** present,there is still no unified consensus for RA prevention,but targeting therapeutic windows and implementing interventions for at-risk individuals are extremely *** to the limited number of clinical trials on pharmacologic interventions,further studies are needed to explore and establish optimal intervention regimens and effective measures to prevent progression to *** this review,we introduce the RA disease process and risk factors,and present research on the use of both Western and Chinese medicine from clinical perspectives regarding RA ***,we describe several complete and ongoing clinical studies on the use of Chinese herbal formulae for the prevention of RA.
