Background Stroke is the leading cause of mortality in China,with limited evidence of in-hospital burden obtained from nationwide *** aimed to monitor and track the temporal trends and rural-urban disparities in cerebrovascular risk factors,management and outcomes from 2005 to *** We used a two-stage random sampling survey to create a nationally representative sample of patients admitted for ischaemic stroke in 2005,2010 and *** sampled participating hospitals with an economic-geographical region-stratified random-sampling approach first and then obtained patients with a systematic sampling *** weighed our survey data to estimate the national-level results and assess changes from 2005 to *** We analysed 28277 ischaemic stroke admissions from 189 participating *** 2005 to 2015,the estimated national hospital admission rate for ischaemic stroke per 100000 people increased(from 75.9 to 402.7,Ptrend<0.001),and the prevalence of risk factors,including hypertension,diabetes,dyslipidaemia and current smoking,*** composite score of diagnostic tests for stroke aetiology assessment(from 0.22 to 0.36,Ptrend<0.001)and secondary prevention treatments(from 0.46 to 0.70,Ptrend<0.001)were improved.A temporal decrease was found in discharge against medical advice(DAMA)(from 15.2%(95%CI 13.7%to 16.7%)to 8.6%(8.1%to 9.0%);adjusted Ptrend=0.046),and decreases in in-hospital mortality(0.7%in 2015 vs 1.8%in 2005;adjusted OR(aOR)0.52;95%CI 0.32 to 0.85)and the composite outcome of in-hospital mortality or DAMA(8.4%in 2015 vs 13.9%in 2005;aOR 0.65;95%CI 0.47 to 0.89)were *** between rural and urban hospitals narrowed;however,disparities persisted in in-hospital management(brain MRI:rural-urban difference from−14.4%to−11.2%;cerebrovascular assessment:from−20.3%to−16.7%;clopidogrel:from−2.1%to−10.3%;anticoagulant for atrial fibrillation:from−10.9%to−8.2%)and in-hospital outcomes(DAMA:from 2.7%to 5.0%;composite outcome

Targeted protein degradation(TPD)has rapidly emerged as a therapeutic modality to eliminate previously undruggable proteins by repurposing the cell’s endogenous protein degradation ***,the susceptibility of proteins for targeting by TPD approaches,termed“degradability”,is largely ***,we developed a machine learning model,model-free analysis of protein degradability(MAPD),to predict degradability from features intrinsic to protein *** shows accurate performance in predicting kinases that are degradable by TPD compounds[with an area under the precision–recall curve(AUPRC)of 0.759 and an area under the receiver operating characteristic curve(AUROC)of 0.775]and is likely generalizable to independent non-kinase *** found five features with statistical significance to achieve optimal prediction,with ubiquitination potential being the most *** structural modeling,we found that E2-accessible ubiquitination sites,but not lysine residues in general,are particularly associated with kinase ***,we extended MAPD predictions to the entire proteome to find964 disease-causing proteins(including proteins encoded by 278 cancer genes)that may be tractable to TPD drug development.

Organ fibrosis is a process in which cellular homeostasis is disrupted and extracellular matrix is excessively *** can lead to vital organ failure and there are no effective treatments *** epithelial-mesenchymal transition(EMT)may be one of the key cellular mechanisms,the underlying mechanisms of fibrosis remain largely *** is a cell phenotypic process in which epithelial cells lose their cell-to-cell adhesion and polarization,after which they acquire mesenchymal features such as infiltration and migration *** injurious stimulation in different organs,EMT can be triggered by multiple signaling pathways and is also regulated by epigenetic *** narrative review summarizes the current understanding of the underlying mechanisms of EMT in fibrogenesis and discusses potential strategies for attenuating EMT to prevent and/or inhibit *** better understanding the role of EMT in fibrosis development,targeting EMT and beyond in developing therapeutics to tackle fibrosis is challenging but likely feasible.

Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread rapidly around the world,posing a major threat to human health and the ***,long-term data on viral shedding and the serum antibody responses in COVID-19 patients are still ***,we report the clinical features,viral RNA loads,and serum antibody levels in a cohort of 112 COVID-19 patients admitted to the Honghu People’s Hospital,Hubei Province,***,5.36%(6/112)of patients showed persistent viral RNA shedding(>45 days).The peak viral load was higher in the severe disease group than in the mild group(median cycle threshold value,36.4 versus 31.5;P=0.002).For most patients the disappearance of IgM antibodies occurred approximately 4–6 weeks after symptoms onset,while IgG persisted for over 194 days after the onset of symptoms,although patients showed a 46%reduction in antibodies titres against SARS-CoV-2 nucleocapsid protein compared with the acute *** also studied18 asymptomatic individuals with RT-qPCR confirmed SARS-CoV-2 infection together with 17 symptomatic patients,and the asymptomatic individuals were the close contacts of these symptomatic *** IgG seroconversion and lower IgM seropositive rates were observed in asymptomatic *** data indicate that higher viral loads and stronger antibody responses are related to more severe disease status in patients with SARS-CoV-2 infection,and the antibodies persisted in the recovered patient for more than 6 months so that the vaccine may provide protection against SARS-CoV-2 infection.

在常规的宝石鉴定中,肉眼直接观察查尔斯滤色镜下祖母绿是否变红的判断依据存在一定的局限性。为了更加准确地描述查尔斯滤色镜下祖母绿的颜色变化,本文引入色度学方法,将透过查尔斯滤色镜后的光谱信息转换为色度坐标,将祖母绿的颜色变化和光谱变化以数据方式呈现。研究结果显示,光谱测试角度、祖母绿与查尔斯滤色镜的接触方式、查尔斯滤色镜型号等因素对色度结果有一定影响;分析了光线在透过查尔斯滤色镜过程中的光学损失,并基于光学损失建立了查尔斯滤色镜对祖母绿光谱的调制算法。经过查尔斯滤色镜调制后的天然与合成祖母绿样品的光谱所对应的CIE 1931色度图上的色度坐标分布具有可识别性,色度坐标的位置分布在一定程度上可以作为区分天然与合成祖母绿的依据。
