EKLF is an erythroid-specific,zinc finger-containing transcription factor essential for the activation of the mamma-lian beta globin gene in erythroid cells of definitive *** have prepared a polyclonal anti-mouse EKLF antibodysuitable for Western blotting and immunoprecipitation(IP)qualities,and used it to define the expression patterns of theEKLF protein during mouse erythroid *** have also used this antibody for the chromatin-immunoprecipi-tation(ChIP)*** was found to bind in vivo at both the mouse beta-major-globin promoter and the HS2 site ofbeta-LCR in the mouse erythroleukemia cells(MEL)in a DMSO-inducible *** DMSO-induced bindings ofEKLF as well as three other proteins,namely,RNA polymerase Ⅱ,acetylated histone H3,and methylated histone H3,were not abolished but significantly lowered in CB3,a MEL-derived cell line with null-expression of p45/NF-E2,anerythroid-enriched factor needed for activation of the mammalian globin ***,binding of EKLF in vivowas also detected in the mouse alpha-like globin locus,at the adult alpha globin promoter and its far upstream regula-tory element alpha-MRE(HS26).This study provides direct evidence for EKLF-binding in vivo at the major regulatoryelements of the mouse beta-like globin gene clusters the data also have interesting implications with respect to the roleof EKLF-chromatin interaction in mammalian globin gene regulation.

Template-assisted growth of Co-BaTiO_(3) vertically aligned nanocomposite thin films with strong magneto-optical coupling effect

Oxide-metal based nanocomposite thin films have attracted great interests owing to their unique anisotropic structure and physical properties.A wide range of Au-based oxide-metal nanocomposites have been demonstrated,while other metal systems are scarce due to the challenges in the initial nucleation and growth as well as possible interdiffusions of the metallic *** this work,a unique anodic aluminum oxide(AAO)template was used to grow a thin Co seed layer and the following self-assembled metal-oxide(Co-BaTiO_(3))vertically aligned nanocomposite thin film *** AAO template allows the uniform growth of Co-seeds and successfully deposition of highly ordered Co pillars(with diameter<5 nm and interval between pillars<10 nm)inside the oxide *** magnetic anisotropy and strong magneto-optical coupling properties have been observed.A thin Au-BaTiO_(3) template was also later introduced for further enhanced nucleation and ordered growth of the *** the advantage of such a unique nanostructure,a large out-of-plane(OP)coercive field(Hc)of~5000 Oe has been achieved,making the nanocomposite an ideal candidate for high density perpendicular magnetic tunneling junction(p-MTJ).A strong polar magneto-optical Kerr effect(MOKE)has also been observed which inspires a novel optical-based reading method of the MTJ states.

The improvement in the efficiency of inverted perovskite solar cells(PSCs)is significantly limited by undesirable contact at the NiO_(x)/perovskite *** this study,a novel microstructure-control technology is proposed for fabrication of porous NiO_(x)films using Pluronic P123 as the structure-directing agent and acetylacetone(AcAc)as the coordination *** synthesized porous NiO_(x)films enhanced the hole extraction efficiency and reduced recombination defects at the NiO_(x)/perovskite ***,without any modification,the power conversion efficiency(PCE)of the PSC with MAPbl_(3)as the absorber layer improved from 16.50%to 19.08%.Moreover,the PCE of the device composed of perovskite Cs0.05(MA_(0.15)FA_(0.85))_(0.95)Pb(I_(0.85)Br_(0.15))_(3)improved from 17.49%to 21.42%.Furthermore,the application of the fabricated porous NiO_(x)on fluorine-doped tin oxide(FTO)substrates enabled the fabrication of large-area PSCs(1.2 cm^(2))with a PCE of 19.63%.This study provides a novel strategy for improving the contact at the NiO_(x)/perovskite interface for the fabrication of high-performance large-area perovskite solar cells.

A group of massive galaxies at redshifts of z■7 have been recently detected by the james Webb Space Telescope(JWST),which were unexpected to form at such an early time within the standard Big Bang *** this work,we propose that this puzzle can be explained by the presence of some primordial black holes(PBHs)with a mass of~1000M_(⊙).These PBHs act as seeds for early galaxy formation with masses of~10^(8)-10^(10)M_(⊙)at high redshift,hence accounting for the JWST *** use a hierarchical Bayesian inference framework to constrain the PBH mass distribution models,and find that the Lognormal model with the M_(c)~750M_(⊙)is preferred over other *** rapidly growing BHs are expected to have strong radiation and may appear as high-redshift compact objects,similar to those recently discovered by *** we focused on PBHs in this work,the bound on the initial mass of the seed black holes remains robust even if they were formed through astrophysical channels.

Studies have proposed that there is evidence for cosmological coupling of black holes(BHs)with an index of k≈3;hence,BHs serve as the astrophysical source of dark ***,the data sample is limited for the redshifts of≤*** recent years,the james Webb Space Telescope(JWST)has detected many high-redshift active galactic nuclei(AGNs)and *** the JWST NIRSpec-/NIRCam-resolved AGNs,three are determined to be in early-type host galaxies with a redshift of z~***,their M_(*)and MBH are in tension with the predicted cosmological coupling of black holes with k=3 at a confidence level of~2σ,which challenges the hypothesis that BHs serve as the origin of dark *** work on high-redshift AGNs using the JWST will further assess such a hypothesis by identifying more early-type host galaxies in the higher mass range.

Hereditary spastic paraplegia(HSP)is a group of genetic motor neuron diseases resulting from length-dependent axonal degeneration of the corticospinal upper motor *** to the advancement of next-generation sequencing,more than 70 novel HSP disease-causing genes have been identified in the past *** this,our understanding of HSP physiopathology and the development of efficient management and treatment strategies remain *** major challenge in studying HSP pathogenicity is selective neuronal vulnerability,characterized by the manifestation of clinical symptoms that are restricted to specific neuronal populations,despite the presence of germline disease-causing variants in every cell of the ***,disease genes may exhibit ubiquitous expression patterns and involve a myriad of different pathways to cause motor neuron *** the current review,we explore the correlation between transcriptomic data and clinical manifestations,as well as the importance of interspecies models by comparing tissue-specific transcriptomic profiles of humans and mice,expression patterns of different genes in the brain during development,and single-cell transcriptomic data from related ***,we discuss the potential of emerging single-cell RNA sequencing technologies to resolve unanswered questions related to HSP pathogenicity.

NADPH oxidase-1 deficiency offers little protection in Salmonella typhimurium-induced typhlitis in mice

AIM To test whether Nox1 plays a role in typhlitis induced by Salmonella enterica serovar Typhimurium(S. Tm) in a mouse *** Eight-week-old male wild-type(WT) and Nox1 knockout(KO) C57BL6/J(B6) mice were administered metronidazole water for 4 d to make them susceptible to S. Tm infection by the oral route. The mice were given plain water and administered with 4 different doses of S. Tm by oral gavage. The mice were followed for another 4 d. From the time of the metronidazole application, the mice were observed twice daily and weighed daily. The ileum, cecum and colon were removed for sampling at the fourth day post-inoculation. Portions of all three tissues were fixed for histology and placed in RNAlater for m RNA/c DNA preparation and quantitative real-time PCR. The contents of the cecum were recovered for estimation of S. Tm *** We found Nox1-knockout(Nox1-KO) mice were not more sensitive to S. Tm colonization and infection than WT B6 mice. This conclusion is based on the following observations:(1) S. Tm-infection induced similar weight loss in Nox1-KO mice compared to WT mice;(2) the same S. Tm CFU was recovered from the cecal content of Nox1-KO and WT mice regardless of the inoculation dose, except the lowest inoculation dose(2 × 106 CFU) for which the Nox1-KO had one-log lower CFU than WT mice;(3) there is no difference in cecal pathology between WT and Nox1-KO groups; and(4) there are no S. Tm infection-induced changes in gene expression levels(IL-1b, TNF-α, and Duox2) between WT and Nox1-KO groups. The Alpi gene expression was more suppressed by S. Tm treatment in WT than the Nox1-KO cecum. CONCLUSION Nox1 does not protect mice from S. Tm colonization. Nox1-KO provides a very minor protective effect against S. Tm infection. Using NOX1-specific inhibitors for colitis therapy should not increase risks in bacterial infection.

Tonstein layers are found worldwide in the Permo-Carboniferous coal-bearing *** study investigates the geochronology,mineralogy,and geochemistry of four tonstein samples from the Permo-Carboniferous Benxi Formation,Ordos Basin,North China Craton(NCC).The typical features of the studied tonsteins include thin beds,lateral continuity,angular quartz grains,and euhedral zircons with similar U-Pb ages,indicating a significant pyroclastic *** addition,the tonstein samples have low TiO_(2)/Al_(2)O_(3)ratios(<0.02)and rare earth elements and yttrium(REY)concentrations with obvious negative Eu anomalies,indicating that the tonsteins have a felsic magma ***,compared with the mean composition of clay shale,the studied tonsteins are characterized by high concentrations of the elements Nb and Ta,which may affect the concentration of the corresponding elements in surrounding coal *** zircon U-Pb ages of the tonsteins(293.9-298.8 Ma)provide a precise chronological framework on the Benxi Formation in the Ordos Basin,constraining the Gzhelian-Aselian *** tonsteins were probably sourced from arc volcanism along the western margin of the NCC during the early Permian,implying that the Alxa Terrane had not amalgamated with the NCC at that time.
