A lasting imbalance between fatty acid synthesis and consumption leads to non-alcoholic fatty liver disease(NAFLD),coupled with hepatitis and insulin *** the details of the underlying mechanisms are not fully ***,we unraveled that the expression of the transcription factor Zbtb18 is markedly decreased in the livers of both patients and murine models of *** Zbtb18 knockout promoted NAFLD features like impaired energy expenditure and fatty acid oxidation(FAO),and induced insulin ***,hepatic Zbtb18 overexpression alleviated hepato-steatosis,insulin resistance,and hyperglycemia in mice fed on a high-fat diet(HFD)or in diabetic ***,in vitro and in vivo mechanistic studies revealed that Zbtb18 transcriptional activation of Farnesoid X receptor(FXR)mediated FAO and Clathrin Heavy Chain(CLTC)protein hinders NLRP3 inflammasome *** key mechanism by which hepatocyte’s Zbtb18 expression alleviates NAFLD and consequent liver fibrosis was further verified by FXR’s deletion and forced expression in mice and cultured mouse primary hepatocytes(MPHs).Moreover,CLTC deletion significantly abrogated the hepatic Zbtb18 overexpression-driven inhibition of NLRP3 inflammasome activity in ***,Zbtb18 transcriptionally activates the FXR-mediated FAO and CLTC expression,which inhibits NLRP3 inflammasome’s activity alleviating inflammatory stress and insulin resistance,representing an attractive remedy for hepatic steatosis and fibrosis.

新时代以来当代文学总体经历了三次文化祛魅。自20世纪80年代后,科学技术的迅猛进步使媒介日新月异,而媒介的不断变迁又对文学表现出深刻的影响(杨博,2019)。80年代报纸与文学期刊作为清晰度低,参与度高的冷媒介推动着文化的“祛魅”与“赋魅”。90年代电子热媒介的发展迅速,直接促使大众文化的兴起,从而扩大了文学的生产域,并让精英文学逐渐被“祛魅”。到了21世纪,代表网络二代的新新媒介兴盛,作家身份被颠覆,文学丧失灵韵,这一次“祛魅”却让文学在媒介生态环境中面临挑战。所以媒介作为文学的载体的同时,也塑造着文学的特征。

桦褐孔菌黄酮对糖尿病肾病小鼠的治疗作用研究

目的:研究桦褐孔菌黄酮对糖尿病肾病小鼠的治疗作用。方法:取50只4周龄C57BL/6小鼠,随机抽取6只为正常对照组,其余小鼠以链脲佐菌素(STZ)联合高糖、高脂饲料喂养9周,建立糖尿病肾病模型,建模成功后取30只随机分为模型组、阳性对照组(缬沙坦20 mg/kg)和桦褐孔菌黄酮低(50 mg/kg)、中(100 mg/kg)、高(200 mg/kg)剂量组,每组6只。连续灌胃给药6周,6周后,观察小鼠免疫功能的变化。结果:与正常对照组小鼠相比,模型组小鼠中骨髓来源的树突状细胞及其上清液中分泌的外泌体表面MHC-Ⅱ、CD86、CD54与CD80等分子的表达均显著降低(P<0.01),脾中CD3^(+)、CD4^(+)、CD8^(+)T淋巴细胞及其分泌的IFN-γ因子数量均减少(P<0.01)。经治疗后,各组小鼠细胞及外泌体表面分子MHC-Ⅱ、CD86、CD54、CD80的表达均明显升高(P<0.05,P<0.01),CD3^(+)、CD4^(+)、CD8^(+)T淋巴细胞及IFN-γ的数量也不同程度增加(P<0.05,P<0.01)。结论:桦褐孔菌黄酮可以提高糖尿病肾病小鼠的免疫功能,对疾病有较好的治疗效果且安全性较高。

为解决由于电流密度分布不均导致的电铸镍片厚度不均匀性问题,研究了电铸工艺参数和阴极挂具结构对改善镍片厚度均匀性和电铸结瘤状况的影响。结果表明,当电流密度为11 A·dm^(-2)、阳极长度为3.5 cm、极间距为2.5 cm、温度为45℃时,镍片厚度均匀性最好。此时厚度平均值为0.484 mm,极差和均方差分别为0.157 mm和0.044 mm。在最佳工艺参数下,改善挂具和设计阴极遮蔽,解决了接触点厚度薄的问题,结瘤缓解,电铸时间由10 h缩至4 h,厚度极差和均方差分别降至0.116 mm和0.036 mm。实验确定的最佳工艺条件不仅提高了镍片厚度均匀性,电铸时间也缩短了60%,提升了阳极镍粒的利用率,节约了电铸成本。

HIF-1α信号通路对脂多糖诱导鸡巨噬细胞炎症的调节作用研究

【目的】探究鸡巨噬细胞(HD11)在脂多糖(LPS)诱导条件下,缺氧诱导因子-1α(HIF-1α)信号通路和线粒体功能对细胞炎症反应的影响。【方法】试验以HD11细胞为研究对象,分别用不同浓度LPS作用于细胞,培养24 h后检测细胞活力,筛选LPS最佳作用浓度;同时在最佳LPS作用浓度下,筛选LPS最佳作用时间。将HD11细胞分为对照组和模型组,模型组加入最佳作用浓度LPS培养,对照组加等量的完全培养液,按照LPS最佳作用时间培养后,提取细胞总RNA进行转录组测序,并对差异表达基因进行GO功能注释和KEGG通路富集分析。利用实时荧光定量PCR和Western blotting分别检测HIF-1α信号通路相关因子mRNA和蛋白表达量;通过流式细胞术检测线粒体膜电位和活性氧(ROS)水平变化。【结果】LPS诱导的HD11细胞炎症模型中最适条件为1μg/mL LPS培养12 h,以此条件成功建立了细胞炎症模型。与对照组相比,模型组共检测到2063个差异表达基因,其中1319个上调,744个下调。GO功能注释结果显示,差异表达基因显著富集到免疫系统应答、对外部刺激的反应和细胞因子受体结合等过程。KEGG通路富集分析结果显示,差异表达基因显著富集在50条信号通路,主要涉及免疫细胞介导的炎症反应和Toll样受体、核转录因子-κB(NF-κB)、HIF-1α等信号通路。其中有13个显著上调表达的基因集中在HIF-1α相关信号通路,包括Toll样受体4(TLR4)、NF-κB、HIF-1α、血管内皮生长因子(VEGF)基因等。实时荧光定量PCR和Western blotting结果显示,NF-κB p65、HIF-1α、VEGF等mRNA和蛋白表达水平均显著上调(P<0.05)。流式细胞术检测结果显示,与对照组相比,模型组线粒体膜电位极显著下降(P<0.01),ROS水平显著升高(P<0.05)。【结论】成功建立LPS诱导鸡巨噬细胞炎症模型,HIF-1α与NF-κB信号通路相互串扰共同参与LPS诱导的细胞炎症过程。同时,细胞线粒体功能下降,导致ROS生成增多,进而促进HIF-1α的表达,共同加重了炎性反应和代谢紊乱。

Selective extinction and habitat nestedness are the main drivers of lizard nestedness in the Zhoushan Archipelago

Nested subset pattern(nestedness)is an important part of the theoretical framework of island biogeography and community ***,most previous studies often used nestedness metrics or randomization algorithms that are vulnerable to type I *** this study,we inves-tigated the nestedness of lizard assemblages on 37 islands in the Zhoushan Archipelago,*** used the line-transect method to survey species occurrence,abundance,and habitat types of lizards on 37 islands during 2 breeding seasons in 2021 and *** applied the nested metric WNODF and the conservative rc null model to control for type I error and quantify the significance of *** rank cor relations were used to evaluate the role of 4 habitat variables(island area,2 isolation indices,and habitat diversity)and 4 ecological traits(body size,geographic range size,clutch size,and minimum area requirement)in generating *** results of WNODF analyses showed that lizard assemblages were significantly *** habitat-by-site matrix estimated by the program NODF was also significantly nested,support-ing the habitat nestedness *** nestedness of lizard assemblages were significantly correlated with island area,habitat *** size,and minimum area ***,our results suggest that selective extinction and habitat nestedness were the main drivers of lizard nestedness in our *** contrast,the nestedness of lizard assemblages was not due to passive sampling or selective *** maximize the number of species preserved,our results indicate that we should protect both large islands with diverse habitats and species with largearea requirement and clutch size.

Although DNA mutation drives stem cell aging,how mutation-accumulated stem cells obtain clonal advantage during aging remains poorly ***,using a mouse model of irradiation-induced premature aging and middle-aged mice,we show that DNA mutation accumulation in hematopoietic stem cells(HSCs)during aging upregulates their surface expression of major histocompatibility complex class II(MHCII).MHCII upregulation increases the chance for recognition by bone marrow(BM)-resident regulatory T cells(Tregs),resulting in their clonal expansion and accumulation in the HSC *** the basis of the establishment of connexin 43(Cx43)-mediated gap junctions,BM Tregs transfer cyclic adenosine monophosphate(cAMP)to aged HSCs to diminish apoptotic priming and promote their survival via activation of protein kinase A(PKA)***,targeting the HSC–Treg interaction or depleting Tregs effectively prevents the premature/physiological aging of *** findings show that aged HSCs use an active self-protective mechanism by entrapping local Tregs to construct a prosurvival niche and obtain a clonal advantage.

Interdecadal variation of drought patterns and associated influencing factors in the north-south transitional zone of China

In the context of global warming,escalating water cycles have led to a surge in drought frequency and ***,multidecadal fluctuations in drought and their multifaceted influencing factors remain inadequately *** study examined the multidecadal changes in drought characteristics(frequency,duration,and severity)and their geographical focal points within China's north-south transitional zone,the Qinling-Daba Mountains(QDM),from 1960 to 2017 using the Standardized Precipitation Evapotranspiration Index(SPEI).In addition,a suite of eight scenarios,correlation analysis,and wavelet coherence were used to identify the meteorological and circulation factors that influenced drought *** results indicate the following:(1)From 1960 to 2017,the QDM experienced significant interdecadal variations in drought frequency,duration,and severity,the climate was relatively humid before the 1990s,but drought intensified ***,the 1990s marked the period of the longest drought duration and greatest severity,while the years spanning 2010 to 2017 experienced the highest frequency of drought events.(2)Spatially,the Qinling Mountains,particularly the western Qinling Mountain,exhibited higher drought frequency,duration,and severity than the Daba *** disparity can be attributed to higher rates of temperature increase and precipitation decrease in the western Qinling Mountain.(3)Interdecadal variations in droughts in the QDM were directly influenced by the synergistic effects of interdecadal fluctuations in air temperature and *** factors modulate temperature and precipitation through phase transitions,thereby affecting drought dynamics in the *** Atlantic Multidecadal Oscillation emerges as the primary circulation factors influencing temperature changes,with a mid-1990s shift to a positive phase favoring *** East Asian Summer Monsoon and El Ni?o-Southern Oscillation are the main circulation factors a

禾草灵对铜绿微囊藻氧化应激及藻毒素释放的影响

以铜绿微囊藻(Microsystis aeruginosa)作为生物模型,通过测定藻细胞氧化损伤程度与藻细胞内外藻毒素LR(microcystin-LR,MC-LR)的含量,研究除草剂禾草灵的生态毒性。结果表明,禾草灵在实验室处理质量浓度1,2,5mg/L下,能诱导铜绿微囊藻细胞活性氧(reactive oxygen species,ROS)的产生,引起细胞脂质过氧化水平丙二醛(malondialdehyde,MDA)的提高和超氧化物歧化酶(superoxide dismutase,SOD)的活性增强。说明禾草灵对铜绿微囊藻产生了氧化胁迫。5mg/L禾草灵使铜绿微囊藻释放到细胞外的微囊藻毒素MC-LR显著增加,为空白对照的68.5,2.11,2.50,123.67倍(第2,3,4,5天);细胞内MC-LR含量与空白对照相比有一定的降低,为空白对照的0.34,0.82,2.76,0.11倍(第2,3,4,5天)。说明禾草灵在一定质量浓度下能对铜绿微囊藻产生氧化胁迫,引起藻细胞脂质过氧化,从而引起细胞膜损伤,导致微囊藻毒素MC-LR更多的释放到水体中,对水环境造成一定的影响。实验结果可对禾草灵的环境安全评价提供一定的参考。
