OSW-1 triggers necroptosis in colorectal cancer cells through the RIPK1/RIPK3/MLKL signaling pathway facilitated by the RIPK1- p62/SQSTM1 complex

BACKGROUND Necroptosis has emerged as a novel molecular pathway that can be targeted by chemotherapy agents in the treatment of ***-1,which is derived from the bulbs of Ornithogalum saundersiae Baker,exerts a wide range of pharmaco-logical *** To explore whether OSW-1 can induce necroptosis in colorectal cancer(CRC)cells,thereby expanding its range of clinical *** We performed a sequence of functional experiments,including Cell Counting Kit-8 assays and flow cytometry analysis,to assess the inhibitory effect of OSW-1 on CRC *** utilized quantitative proteomics,employing tandem mass tag label-ing combined with liquid chromatography-tandem mass spectrometry,to analyze changes in protein *** bioinformatic analysis was conducted to elucidate the biological processes associated with the identified *** electron microscopy(TEM)and immunofluorescence studies were also performed to examine the effects of OSW-1 on ***,western blotting,siRNA experiments,and immunoprecipitation were employed to evaluate protein interactions within CRC *** The results revealed that OSW-1 exerted a strong inhibitory effect on CRC cells,and this effect was accompanied by a necroptosis-like morphology that was observable via ***-1 was shown to trigger necroptosis via activation of the RIPK1/RIPK3/MLKL ***,the accumulation of p62/SQSTM1 was shown to mediate OSW-1-induced necroptosis through its interaction with *** We propose that OSW-1 can induce necroptosis through the RIPK1/RIPK3/MLKL signaling pathway,and that this effect is mediated by the RIPK1-p62/SQSTM1 complex,in CRC *** results provide a theoretical foundation for the use of OSW-1 in the clinical treatment of CRC.

Computer Simulation Study on Adsorption and Conformation of Polymer Chains Driven by External Force

In this work,Monte Carlo simulations are used to study the critical adsorption behaviors of flexible polymer chains under the action of an external driving force F parallel to an attractive flat *** critical adsorption temperature Tc decreases linearly with increasing F,indicating that the driving force suppresses the adsorption of *** conformation of polymer is also affected by the driving ***,the effect of F is dependent on the competition between the driving force and *** strong force or at low temperature,the polymer is stretched along the direction of the force,while under weak force or at high temperature,the polymer is not *** the force is comparable to the temperature,the polymer may be stretched perpendicular to the driving force,and below Tc,we observe conformational transitions from parallel to perpendicular and again to parallel by decreasing the *** found that the perpendicular stretched conformation leads the polymer chain to synchronously move along the direction of the driving ***,the conformational transitions are attributed to the competition and cooperation between the driving force and the temperature.

OsPDR20 is an ABCG metal transporter regulating cadmium accumulation in rice

Cadmium(Cd)is a non-essential toxic heavy metal,seriously posing high environmental risks to human *** genetic resources relevant to functional genes is important for understanding the metal absorption and accumulation in crops and bioremediation of Cd-polluted *** study investigated a functionally uncharacterized ATP binding cassette transporter G family(ABCG)gene encoding a Pleiotropic Drug Resistance 20(PDR20)type metal transporter which is localized to the plasma membrane of ***20 was transcriptionally expressed in almost all tissues and organs in lifespan and was strongly induced in roots and shoots of young rice under Cd *** expression of OsPDR20 in a yeast mutant ycf1 sensitive to Cd conferred cellular tolerance with less Cd *** of OsPDR20 by RNA interference(RNAi)moderately attenuated root/shoot elongation and biomass,with reduced chlorophylls in rice grown under hydroponic medium with 2 and 10μmol/L Cd,but led to more Cd accumulation.A field trial of rice grown in a realistic Cd-contaminated soil(0.40 mg/kg)showed that RNAi plants growth and development were also compromised compared to wild-type(WT),with smaller panicles and lower spikelet fertility but little effect on yield of ***,OsPDR20 suppression resulted in unexpectedly higher levels of Cd accumulation in rice straw including lower leaves and culm and *** results suggest that OsPDR20 is actively involved in Cd accumulation and homeostasis in rice *** increased Cd accumulation in the RNAi plants has the potential application in phytoremediation of Cd-polluted wetland soils.

Application of multi-algorithm ensemble methods in high-dimensional and small-sample data of geotechnical engineering:A case study of swelling pressure of expansive soils

Geotechnical engineering data are usually small-sample and high-dimensional,which brings a lot of challenges in predictive *** paper uses a typical high-dimensional and small-sample swell pressure(P_(s))dataset to explore the possibility of using multi-algorithm hybrid ensemble and dimensionality reduction methods to mitigate the uncertainty of soil parameter *** on six machine learning(ML)algorithms,the base learner pool is constructed,and four ensemble methods,Stacking(SG),Blending(BG),Voting regression(VR),and Feature weight linear stacking(FWL),are used for the multi-algorithm ***,the importance of permutation is used for feature dimensionality reduction to mitigate the impact of weakly correlated variables on predictive *** results show that the proposed methods are superior to traditional prediction models and base ML models,where FWL is more suitable for modeling with small-sample datasets,and dimensionality reduction can simplify the data structure and reduce the adverse impact of the small-sample effect,which points the way to feature selection for predictive *** on the ensemble methods,the feature importance of the five primary factors affecting P_(s) is the maximum dry density(31.145%),clay fraction(15.876%),swell percent(15.289%),plasticity index(14%),and optimum moisture content(13.69%),the influence of input parameters on P_(s) is also investigated,in line with the findings of the existing literature.

Recycling spent lithium-ion batteries is integral to today's low-carbon environmental protection *** concept of direct regeneration,acknowledged for its environmental sustainability,economic viability,and consistent performance of recycled materials,is gaining *** study presents an efficient and nondestructive approach by utilizing an ultrafast microwave technology to directly regenerate spent lithium cobaltate(LCO)cathode *** contrast to conventional furnacebased processes,this method significantly reduces the regeneration *** subjecting the spent LCO mixed with lithium sources to three microwave heating cycles(at approximately 1,350 K),LCO regeneration is achieved,yielding a specific capacity of 140.8 mAh g^(-1)(0.2 C)with a robust cycle *** further environmental and economic benefits,the ultrafast microwave technology holds scientific promise for directly regenerating cathode materials,while establishing competitiveness for industrial applications.

Insights into Formation and li-Storage Mechanisms of Hierarchical Accordion-Shape Orthorhombic CuNb2O6 toward lithium-Ion Capacitors as an Anode-Active Material

The orthorhombic CuNb2O6（O-CNO） is established as a competitive anode for lithium-ion capacitors（liCs） owing to its attractive compositional/structural ***,the high-temperature synthesis（>900 ℃） and controversial charge-storage mechanism always limit its ***,we develop a low-temperature strategy to fabricate a nano-blocksconstructed hierarchical accordional O-CNO framework by employing multilayered Nb2CTxas the niobium *** intrinsic stress-induced formation/transformation mechanism of the monoclinic CuNb2O6to O-CNO is tentatively put ***,the integrated phase conversion and solid solution lithium-storage mechanism is reasonably unveiled with comprehensive in（ex） situ *** to its unique structural merits and lithium-storage process,the resulted O-CNO anode is endowed with a large capacity of 150.3 mAh g-1at 2.0 A g-1,along with longduration cycling ***,the constructed O-CNO-based liCs exhibit a high energy(138.9 Wh kg-1) and power(4.0 kW kg-1) densities with a modest cycling stability（15.8% capacity degradation after 3000consecutive cycles）.More meaningfully,the in-depth insights into the formation and charge-storage process here can promote the extensive development of binary metal Nb-based oxides for advanced liCs.

A bifunctional POM-based Cu-viologen complex with mixed octamolybdate clusters for rapid oxidation desulfurization and effective photogeneration of hydrogen

The design and synthesis of catalysts for the oxidation desulfurization and production of hydrogen are extremely important for solving environmental pollution and energy ***,a novel bifunctional [α-Mo_(8)O_(26)]^(4-)/[β-Mo_(8)O_(26)]^(4-)-based Cu-viologen complex H_(4)[Cu_(2)~ⅠCl_(2)(Hbcbpy)_(4)] [α-Mo_(8)O_(26)] [β-Mo8O_(26)]·H_(2)O(BHU-2,Hbcbpy=1-(4-carboxybenzyl)-4,4'-bipyridinium) was synthesized and characterized by single-crystal X-ray diffraction(XRD),infrared radiation spectra,powder X-ray diffraction(PXRD) and X-ray photoelectron spectroscopy(XPS) *** structural characteristic of BHU-2 is the presence of two types of octamolybdate clusters [α-Mo_(8)O_(26)]^(4-)/[β-Mo_(8)O_(26)]^(4-)and a new binuclear Cu~Ⅰ-Hbcbpy complex linked by Cl-bridges[Cu_(2)ⅠCl_(2)(Hbcbpy)_(4)]^(4+).BHU-2 as a heterogeneous catalyst exhibits excellent activities to the oxidation desulfurization and photocatalytic hydrogen *** room temperature,BHU-2 can catalyze 96% conversion of methyl phenyl sulfide with 98% selectivity,and the process obeys the pseudo-first-order reaction kinetic with the half-life of9.6 *** notorious 2-chloroethyl ethyl sulfide can achieve 99% conversion with 98% selectivity within only1 min at the presence of BHU-2,and the turnover frequency(TOF) is up to 7400 h^(-1).BHU-2 also exhibits high catalytic activity for the oxidation of other aromatic and aliphatic thioethers within short time at room ***,BHU-2 shows a high catalytic activity for visible-light-driven hydrogen evolution with an H_(2) evolution rate of 1677.85 μmol·g^(-1)·h^(-1) within 10 ***,the catalytic activities do not decrease evidently after three cycles,revealing the prominent structural stability and recyclability.

The miRNA let-7al inhibits the expression of insulin-like growth factor 1 receptor （IGFIR） in prostate cancer PC-3 cells

Reduced microRNA （miRNA） let-7a expression and the activation of insulin-like growth factor-1 receptor （IGFIR） signalling are both involved in prostate cancer and progression. In the present study, we demonstrated that the growth inhibitory effect of let-7al is directly related to targeting IGFIRgene expression in PC-3 cells. TargetScan predicted three potential target sites （T1, T2 and T3） of let-7a in the 3＇ untranslational region （3＇ UTR） of IGFIR mRNA. Real-time PCR, Western blot and luciferase reporter assays were used to detect the effects of let-7al overexpression or let-7al inhibitor on the IGFIRgene expression in PC-3 cells. The results indicated that let-7al could inhibit IGFIR expression by directly targeting the T1 and T2 sites in the 3＇ UTR of the IGFIR mRNA. We then used RT-PCR, luciferase reporter assays, 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyl-2H-tetrazolium bromide （MTT） assay, flow cytometry and Hoechst 33342 staining to examine whether let-7al-mediated inhibition of IGFIR expression also affects the IGFIR-mediated signalling events, including Elk1 activity and c-fos gene expression, proliferation, apoptosis and cell cycle. We demonstrated that let-7al-mediated IGFIR downregulation was accompanied by attenuation of Elk1 activity and c-fos expression, inhibition of cell proliferation, enhanced apoptosis and cell cycle arrest, and that loss function of let-7al via inhibition can upregulate IGF1R accompanied by an increase of Elk1 activity and c-fos expression, thereby enhancing cell proliferation. Altogether, these findings sueeest that let-7a mav be novel therapeutic candidate for Drostate cancer.
