Hot water-extracted Lycium barbarum and Rehmannia glutinosa inhibit proliferation and induce apoptosis of hepatocellular carcinoma cells

AIM： To investigate the effect of hot water-extracted Lydurn barbarum （LBE） and Rehrnannia glutinosa （RGE） on cell proliferation and apoptosis in rat and/or human hepatocellular carcinoma （HCC） cells. METHODS： Rat （H-4-Ⅱ-E） and human HCC （HA22T/ VGH） cell lines were incubated with various concentrations （0-10 g/L） of hot water-extracted LBE and RGE. After 6-24 h incubation, cell proliferation （n = 6） was measured by a colorimetric method. The apoptotic cells （n = 6） were detected by flow cytometry. The expression of p53 protein （n = 3） was determined by SDS-PAGE and Western blotting. RESULTS： Crude LBE （2-5 g/L） and RGE （2-10 g/L） dose-dependently inhibited proliferation of H-4-Ⅱ-E cells by 11% （P 〈 0.05） to 85% （P 〈 0.01） after 6-24 h treatment. Crude LBE at a dose of 5 g/L suppressed cell proliferation of H-4-Ⅱ-E cells more effectively than crude RGE after 6-24 h incubation （P 〈 0.01）. Crude LBE （2-10 g/L） and RGE （2-5 g/L） also dose-dependently inhibited proliferation of HA22T/VGH cells by 14%-43% （P 〈 0.01） after 24 h. Crude LBE at a dose of 10 g/L inhibited the proliferation of HA22T/VGH cells more effectively than crude RGE （56.8% ＋ 1.6% vs 70.3% ＋ 3.1% of control, P = 0.0003 〈 0.01）. The apoptotic cells significantly increased in H-4-Ⅱ-E cells after 24 h treatment with higher doses of crude LBE （2-5 g/L） and RGE （5-10 g/L）（P 〈 0.01）. The expression of p53 protein in H-4-Ⅱ-E cells was 119% and 143% of the control group compared with the LBE-treated （2, 5 g/L） groups, and 110% and 132% of the control group compared with the RGE -treated （5, 10 g/L） groups after 24 h. CONCLUSION： Hot water-extracted crude LBE （2-5 g/L） and RGE （5-10 g/L） inhibit proliferation and stimulate p53-mediated apoptosis in HCC cells.

Three well-preserved fossil Pelecinids from the Late Mesozoic Yixian Formation,Liaoning Province,China are described and assigned to two new species,Eopelecinus huangi *** Eopelecinus tumidus *** Eopelecinus Zhang,Rasnitsyn and Zhang,*** of now,17 species have been included in this genus,which is the most diverse in the Pelecinidae *** new information and characters obtained from these new specimens,the diagnosis of Eopelecinus Zhang, Rasnitsyn and Zhang,2002 is emended.

New Primitive Fossil Earwig from Daohugou,Inner Mongolia,China(Insecta:Dermaptera:Archidermaptera)

A new genus and species Palaeodermapteron dicranum gen. et sp. nov. of the subfamily Dermapterinae and family Dermapteridae is described from the Middle Jurassic Jiulongshan Formation of Daohugou, Inner Mongolia in China. This article documents the earliest record of fossil earwig, extending the earwig existence to the Middle Jurassic. Detailed description and illustration of the specimen along with a brief review of the subfamily Dermapterinae are given. Evolutional trend of earwig morphology and its associated living environment are also discussed.

New discovery of Palaeontinid fossils from the Middle Jurassic in Daohugou, Inner Mongolia (Homoptera, Palaeontinidae)

Three new species of fossil Palaeontinidae are described from Daohugou Village, Inner Mongolia, China: Daohugoucossus shii sp. nov., D. parallelivenius sp. nov., D. lii sp. nov. The diagnosis of Daohugoucossus Wang, Zhang and Fang, 2006 is revised. These materials are the first complete fossil palaeontinids in the Middle Jurassic of the world. Based on observation of these new specimens, Sc of this new genus has the following combined characters: joined with R at base, diverged near M separation, coalesced with R before the nodal line, extended beyond the nodal line and fused with R1 before distal end. Due to the Sc characteristics mentioned above, the genus Liaocossus Ren, Yin and Dou, 1998 can be well distinguished from “Ilerdocosus-complex”.

Lattice Boltzmann method for simulating particle-fluid interactions

The lattice Boltzmann method （LBM） has gained increasing popularity in the last two decades as an alternative numerical approach for solving fluid flow problems. One of the most active research areas in the LBM is its application in particle-fluid systems, where the advantage of the LBM in efficiency and parallel scalability has made it superior to many other direct numerical simulation （DNS） techniques. This article intends to provide a brief review of the application of the LBM in particle-fluid systems. The numerical techniques in the LBM pertaining to simulations of particles are discussed, with emphasis on the advanced treatment for boundary conditions on the particle-fluid interface. Other numerical issues, such as the effect of the internal fluid, are also briefly described. Additionally, recent efforts in using the LBM to obtain closures for particle-fluid drag force are also reviewed.

EFFECT OF NOZZLE FAN ANGLE ON SPRAYS IN GAS-SOLID RISER FLOW

A three-dimensional simulation study is performed for investigating the hydrodynamic behaviors of a cross-flow liquid nitrogen spray injected into an air-fluidized catalytic cracking （FCC） riser of rectangular cross-section. Rectangular nozzles with a fixed aspect ratio but different fan angles are used for the spray feeding. While our numerical simulation reveals a generic three-phase flow structure with strong three-phase interactions under rapid vaporization of sprays, this paper tends to focus on the study of the effect of nozzle fan angle on the spray coverage as well as vapor flux distribution by spray vaporization inside the riser flow. The gas-solid （air-FCC） flow is simulated using the multi-fluid method while the evaporating sprays （liquid nitrogen） are calculated using the Lagrangian trajectory method, with a strong two-way coupling between the Eulerian gas-solid flow and the Lagrangian trajectories of spray. Our simulation shows that the spray coverage is basically dominated by the spray fan angle. The spray fan angle has a very minor effect on spray penetration. The spray vaporization flux per unit area of spray coverage is highly non-linearly distributed along the spray penetration. The convection of gas-solid flow in a riser leads to a significant downward deviation of vapor generated by droplet vaporization, causing a strong recirculating wake region in the immediate downstream area of the spray.

A new fossil weevil (Coleoptera, Curculionoidea, Belidae) from the Yixian Formation of western Liaoning, China

This paper reports a new genus and a new species Microprobelus liuae gen. et sp. nov. referred to the family Belidae. This fossil was collected from the Late Jurassic Yixian Formation of western Liaoning, China. Detailed description and illustration of the specimen along with a brief review of fossil belids are given. And the age of the Yixian Formation and the early diversification of the weevils are discussed.

Human mesenchymal stem cells,such as human adipose-derived stem cells(hASCs),are typically cultured on a two-dimensional(2 D)monolayer material surface,on which 2 D culturing methods are easily performed and ***,hASCs usually suffer from decreased pluripotency and differentiation ability when cultured with a 2 D monolayer culturing method compared to hASCs cultured with a three-dimensional(3 D)culturing method,such as suspension *** this study,we evaluated whether the pluripotency and differentiation ability of hASCs can be reversibly changed during sequential cultivation with 2 D and 3 D culturing *** hASCs cultivated with a 3 D culturing process after 2 D culture showed at least 2-fold enhanced pluripotency(Sox2,Nanog,and OCT4)compared with that of hASCs cultured with the 2 D culture process ***,hASCs obtained from the 3 D culture process expressed increased levels of differentiation markers of chondrocytes and osteoblasts compared with hASCs obtained from the 2 D culture process when hASCs were induced to ***,their pluripotency and differentiation ability were extensively reduced when hASCs were shifted from 3 D culture to 2 D culture and vice versa,which indicates that hASCs show reversibility in terms of their pluripotency and differentiation ability depending on their environment in 2 D and 3 D *** reversibility of pluripotency and differentiation ability were found to last for at least 5 passages in culture during the alternative and sequential culture of cells with 2 D and 3 D culturing *** study revealed the importance of the culture microenvironment in maintaining the pluripotency and differentiation ability of hASCs,which may reduce the effects of the aging process in *** discuss whether the environment of stem cell culture(i.e.,2 D or 3 D cultivation)can affect stem cell fate in terms of pluripotency and differentiation reversibility.
