Sonocatalytic hydrogen/hole-combined therapy for anti-biofilm and infected diabetic wound healing

It is a great challenge to effectively eradicate biofilm and cure biofilm-infected diseases because dense extracellular polymeric substance matrix prevents routine antibacterial agents from penetrating into biofilm.H2is an emerging energy-regulating molecule possessing both high biosafety and high tissue permeability.In this work,we propose a concept of sonocatalytic hydrogen/hole-combined ’inside/outside-cooperation’anti-biofilm for promoting bacteria-infected diabetic wound healing based on two-dimensional piezoelectric nanomaterials.Proof-of-concept experiments using C3N4nanosheets as a representative piezoelectric catalyst with wide band gap and high biosafety have verified that sonocatalytically generated H2and holes rapidly penetrate into biofilm to inhibit bacterial energy metabolism and oxidatively deprive polysaccharides/NADH in biofilm to destroy the bacterial membrane/electron transport chain,respectively,inside/outside-cooperatively eradicating biofilm.A bacteria-infected diabetic wound model is used to confirm the excellent in vivo antibacterial performance of sonocatalytic hydrogen/hole-combined therapy,remarkably improving bacteria-infe cted diabetic wound healing.The proposed strategy of sonocatalytic hole/hydrogen-combined ’inside/outside-cooperation’ will make a highway for treatment of deep-seated biofilm infection.

Detecting building changes with off-nadir aerial images

The tilted viewing nature of the off-nadir aerial images brings severe challenges to the building change detection(BCD) problem: the mismatch of the nearby buildings and the semantic ambiguity of the building facades. To tackle these challenges, we present a multi-task guided change detection network model,named as MTGCD-Net. The proposed model approaches the specific BCD problem by designing three auxiliary tasks, including:(1) a pixel-wise classification task to predict the roofs and facades of buildings;(2) an auxiliary task for learning the roof-to-footprint offsets of each building to account for the misalignment between building roof instances; and(3) an auxiliary task for learning the identical roof matching flow between bi-temporal aerial images to tackle the building roof mismatch problem. These auxiliary tasks provide indispensable and complementary building parsing and matching information. The predictions of the auxiliary tasks are finally fused to the main BCD branch with a multi-modal distillation module. To train and test models for the BCD problem with off-nadir aerial images, we create a new benchmark dataset,named BANDON. Extensive experiments demonstrate that our model achieves superior performance over the previous state-of-the-art competitors.

Tissue distribution of Lycium barbarum polysaccharides in rat tissue by fluorescein isothiocyanate labeling

To date, in vivo investigations of polysaccharide’s pharmacokinetics are significantly restricted by the difficulty in their detection. This study was conducted to establish the quantitative determination of Lycium barbarum polysaccharides(LBPs) based on fluorescein isothiocyanate(FITC) pre-labeling and to investigate their tissue distribution in rat. We obtained the calibration curves linear over the range of 0.0–25 μg/m L in rat tissue samples with correlation coefficients greater than 0.99. The inter-day and intra-day precisions(RSD, %) were within 15%, and the relative recovery ranged 95.2%–102.4%, with RSD range 1.48%–9.58%, indicating that this experiment was suitable for the determination of LBPs. The fluorescence intensity was measured after 24 h storage at room temperature, 3 times of freeze-cycle and cryopreservation at –20 ℃ for 15 day, these results indicated that the stability of the samples was good. LBP-FITC was mainly absorbed by the small intestine and stomach, and mainly excreted in the urine through the kidney;this distinct difference in the tissue distribution of LBPs could be attributed to the size of these LBPs in relation to the pore sizes of the vascular beds in the kidney and liver. Results showed in this study enable us to comprehensively understand the biological effects of LBPs following its oral ingestion.

Effects ofiron and temperature on solubility of light rare earth sulfates in multicomponent system of Fe2(SO4)3-H3PO4-H2SO4 synthetic solution

Numerous light rare earth elements(LREE)minerals containing Fe and P were processed by sulfuric acid roasting method,and the leaching solution mainly comprises LREE sulfate,Fe2(SO4)3,H3PO4,and H2SO4,however,the solubility data of LREE sulfates in this system is few.This work studies the solubility of LREE sulfates in independent LREE sulfate system RE2(SO4)3-Fe2(SO4)3-H3 PO4-H2SO4(RE=La,Ce,Pr or Nd)and mixed LREE sulfates system(La,Ce,Pr,Nd)2(SO4)3-Fe2(SO4)3-H3 PO4-H2SO4 at different temperature(25-65℃)and concentrations of Fe2(SO4)3(Fe2 O3,0-50.13 g/L),H2SO4(0.5 mol/L),and H3 PO4(P2 O5,20.34 g/L)based on the industrial operating condition at low liquid and solid ratio 2:1.The solubility of each LREE sulfate in the independent system(La2 O3,12.25-20.88 g/L;CeO2,41.93-62.35 g/L;Pr6 O11,37.34-56.69 g/L;Nd2 O3,26.60-37.63 g/L)is much higher than that of the mixed system(La2 O3,6.95-11.03 g/L;CeO2,10.63-21.51 g/L;Pr6 O11,11.56-20.36 g/L;Nd2 O3,12.36-19.79 g/L)under the same other conditions.The results also indicate that,in the two systems,both Fe and the temperature have negative effects on the solubility of LREE sulfates.That may occur due to the complication reactions between the complexes of RESO4+and Fe(SO4)2-.However,the influence degree of temperature and iron concentration on the LREE sulfates solubility varies in the two systems and among different LREE species.This research is of theoretical significance for optimizing the conditions of the sulfuric acid process for recovering the LREE from the mixed LREE bearing minerals as well as the single LREE containing secondary rare earth scraps.

Case analysis of water exchange between the Bohai and Yellow Seas in response to high winds in winter

Based on the data from a special project titled China's Offshore Marine Integrated Investigation and Evaluation as well as Regional Ocean Modeling Systems(ROMS)diagnostic numerical model,we studied the influence of high wind processes on the circulation and water exchange between the Bohai and Yellow Seas(BYS)in winter.The results show that the vertical structure of the Yellow Sea Warm Current(YSWC)is relatively uniform under condition of high winds,showing obvious barotropic features.However,this flow is not a stable mean flow,showing strong paroxysmal and reciprocating characteristics.A comparison of the changes in sea level suggests that the intensity of the northwards upwind flow is consistent with the abnormal fluctuations in the sea level.It indicates that the upwind flow is closely related to the water exchange between the BYS.The impact of high wind processes on the water exchange between the BYS is enormous.It can make the flux through the Bohai Strait,as well as that through the mouth of each constituent bay(i.e.,Liaodong Bay,Bohai Bay,and Laizhou Bay)far greater than usual,resulting in a significant increase in the water exchange rate.The exchange capacity,which is about 8%of the total volume of the Bohai Sea,can be completed in a few days.Therefore,the water exchange of the Bohai Sea may be completed by only a few occasional high wind processes in winter.

Human skin can function steadily regardless of surrounding circumstarices(dry or wet),while it is still a challenge for artificial ionic skins,which tend to release solvents in dry air and leach electrolytes in wetted state.Herein,a series of hierarchically crosslinked ionogels containing hydrophobic ionic liquids(ILs)is fabricated by combining a crystalline fluorinated copolymer with hydrophobic ILs.With a reasonable combination of nonvolatility,transparency,stretchablility,and sensitivity,such ionogels can work as reliable sensors for real-time monitoring huma n motions and operate steadily in complex environments as human skin does,which can contribute to the developme nt of durable sen sing devices with a simple design.

Effect of mechanical stresses on degradation behavior of high-purity magnesium in bone environments

High-purity(HP)magnesium(Mg)has emerged as a promising biomaterial for supporting functional bone tissue.Our previous study found that mechanical stresses and the surrounding fibrotic tissue(subcuta-neous)both play crucial roles in the degradation of HP Mg.However,due to challenges in the degradation and regeneration process in vivo,it remains unclear how stress affects HP Mg degradation in bone en-vironments,limiting its further application.In this study,novel loading devices were designed and the effects of tensile and compressive stresses on HP Mg degradation in vivo and in vitro bone environments were quantitatively analyzed.In addition,bone osteointegration around HP Mg was explored preliminar-ily.Tensile stress increases the degradation rate of HP Mg in vivo and in vitro.HP Mg degradation in vivo is more sensitive to stress factors than in vitro,but the sensitivity decreases with corrosion time.The volume loss rate of HP Mg is multilinear with the applied stress and degradation time.The volume of bone tissue surrounding HP Mg is larger in the no-stress group compared to the stressed groups,which is more pronounced with increasing implantation time.These results provide valuable insights for optimiz-ing the design of HP Mg-based implants considering load conditions.This will help to achieve a balance between the degradation rate of the implant and the regeneration rate of the surrounding bone.

An unprecedented visible-light-induced transitionmetal-free alkynylation of alkyl pinacol boronates has been demonstrated with alkynyl phenylsulfones as the alkynylation reagents and 4CzIPN as the organic photocatalyst.Common sodium methoxide(NaOMe)or sodium hydroxide(NaOH)was used as the Lewis base togeneratephotoactivetetra-coordinatedboron species.Various functional groupswerewell tolerated.The selective monoalkynylation of diboronates was achieved to keep one boryl group in the product for potential further transformations.Radical trapping experiments and electron paramagnetic resonance(EPR)analysis confirmed the generation of alkyl radical intermediate.

An XBi-CFAO Method for the Optimization of Multi-Layered Variable Stiffness Composites Using Isogeometric Analysis

This paper presents an effective fiber angle optimization method for two and multi-layered variable stiffness composites.A gradient-based fiber angle optimization method is developed based on isogeometric analysis(IGA).Firstly,the element densities and fiber angles for two and multi-layered composites are synchronously optimized using an extended Bi-layered continuous fiber angle optimization method(XBi-CFAO).The densities and fiber angles in the base layer are attached to the control points.The structure response and sensitivity analysis are accomplished using the non-uniform rational B-spline(NURBS)based IGA.By the benefit of the B-spline space,this method is free from checkerboards,and no additional filtering is needed to smooth the sensitivity numbers.Then the curved fiber paths are generated using the streamline method and the discontinuous fiber paths are smoothed using a partitioned selection process.The proposed method in the paper can alleviate the phenomenon of fiber discontinuity,enhance information retention for the optimized fiber angles of the singular points and save calculating resources effectively.

The mechanistic target of rapamycin complex 1 pathway involved in hepatic gluconeogenesis through peroxisome-proliferator-activated receptorγcoactivator-1a

Cattle can efficiently perform de novo generation of glucose through hepatic gluconeogenesis to meet post-weaning glucose demand.Substantial evidence points to cattle and non-ruminant animals being characterized by phylogenetic features in terms of their differing capacity for hepatic gluconeogenesis,a process that is highly efficient in cattle yet the underlying mechanism remains unclear.Here we used a variety of transcriptome data,as well as tissue and cell-based methods to uncover the mechanisms of high-efficiency hepatic gluconeogenesis in cattle.We showed that cattle can efficiently convert propionate into pyruvate,at least partly,via high expression of acyl-CoA synthetase short-chain family member 1(ACSS1),propionyl-CoA carboxylase alpha chain(PCCA),methylmalonyl-CoA epimerase(MCEE),methylmalonyl-CoA mutase(MMUT),and succinate-CoA ligase(SUCLG2)genes in the liver(P<0.01).Moreover,higher expression of the rate-limiting enzymes of gluconeogenesis,such as phosphoenolpyruvate carboxykinase(PCK)and fructose 1,6-bisphosphatase(FBP),ensures the efficient operation of hepatic gluconeogenesis in cattle(P<0.01).Mechanistically,we found that cattle liver exhibits highly active mechanistic target of rapamycin complex 1(mTORC1),and the expressions of PCCA,MMUT,SUCLG2,PCK,and FBP genes are regulated by the activation of mTORC1(P<0.001).Finally,our results showed that mTORC1 promotes hepatic gluconeogenesis in a peroxisome proliferator-activated receptor γ coactivator 1a(PGC-1a)dependent manner.Collectively,our results not only revealed an important mechanism responsible for the quantitative differences in the efficiency of hepatic gluconeogenesis in cattle versus non-ruminant animals,but also established that mTORC1 is indeed involved in the regulation of hepatic gluconeogenesis through PGC-1a.These results provide a novel potential insight into promoting hepatic gluconeogenesis through activated mTORC1 in both ruminants and mammals.
