Multi-modal radiomics model to predict treatment response to neoadjuvant chemotherapy for locally advanced rectal cancer

BACKGROUND Neoadjuvant chemotherapy is currently recommended as preoperative treatment for locally advanced rectal cancer(LARC);however,evaluation of treatment response to neoadjuvant chemotherapy is still challenging.AIM To create a multi-modal radiomics model to assess therapeutic response after neoadjuvant chemotherapy for LARC.METHODS This retrospective study consecutively included 118 patients with LARC who underwent both computed tomography(CT)and magnetic resonance imaging(MRI)before neoadjuvant chemotherapy between October 2016 and June 2019.Histopathological findings were used as the reference standard for pathological response.Patients were randomly divided into a training set(n=70)and a validation set(n=48).The performance of different models based on CT and MRI,including apparent diffusion coefficient(ADC),dynamic contrast enhanced T1 images(DCE-T1),high resolution T2-weighted imaging(HR-T2WI),and imaging features,was assessed by using the receiver operating characteristic curve analysis.This was demonstrated as area under the curve(AUC)and accuracy(ACC).Calibration plots with Hosmer-Lemeshow tests were used to investigate the agreement and performance characteristics of the nomogram.RESULTS Eighty out of 118 patients(68%)achieved a pathological response.For an individual radiomics model,HR-T2WI performed better(AUC=0.859,ACC=0.896)than CT(AUC=0.766,ACC=0.792),DCE-T1(AUC=0.812,ACC=0.854),and ADC(AUC=0.828,ACC=0.833)in the validation set.The imaging performance for extramural venous invasion detection was relatively low in both the training(AUC=0.73,ACC=0.714)and validation(AUC=0.578,ACC=0.583)sets.The multi-modal radiomics model reached an AUC of 0.925 and ACC of 0.886 in the training set,and an AUC of 0.93 and ACC of 0.875 in the validation set.For the clinical radiomics nomogram,good agreement was found between the nomogram prediction and actual observation.CONCLUSION A multi-modal nomogram using traditional imaging features and radiomics of preoperative CT and MRI adds accuracy to the prediction of

Restrictive Cardiomyopathy Resulting from a Troponin Ⅰ Type 3 Mutation in a Chinese Family

Objective To identify the pathogenic variant responsible for restrictive cardiomyopathy(RCM) in a Chinese family.Methods Next generation sequencing was used for detecting the mutation and results verified by sequencing.We used restriction enzyme digestion to test the mutation in the family members and 200 unrelated normal subjects without any cardiac inherited diseases when the mutation was identified.Results Five individuals died from cardiac diseases,two of whom suffered from sudden cardiac death.Two individuals have suffered from chronic cardiac disorders.Mutation analysis revealed a novel missense mutation in exon 7 of troponin Ⅰ type 3(TNNI3),resulting in substitution of serine(S) with proline(P) at amino acid position 150,which cosegregated with the disease in the family,which is predicted to be probably damaging using PolyPhen-2.The mutation was not detected in the 200 unrelated subjects we tested.Conclusion Using next generation sequencing,which has very recently been shown to be successful in identifying novel causative mutations of rare Mendelian disorders,we found a novel mutation of TNNI3 in a Chinese family with RCM.

Analysis of the key networks,metabolic pathways,and regulation substances of hypoxia based on the omics and zebrafish model

OBJECTIVE Hypoxia is associated with many complicated pathophysiological and biochemical processes that integrated and regulated via the key gene,protein and endogenous metabolite levels.Up to date,the exact molecular mechanism of hypoxia still remains unclear.In this work,we further explore the molecular mechanism of hypoxia and adaption to attenuate the damage in zebrafish model that have potential to resist hypoxic environment.METHODS The hypoxic zebrafish model was established in different concentration of oxygen with 3%,5%,10%,21%in water.The brain tissue was separated and the RNA-seq was used to identify the differentially expressed genes.The related endogenous metabolites profiles were obtained by LC-HDMS,and the multivariate statistics was applied to discover the important metabolites candidates in hypoxic zebrafish.The candidates were searched in HMDB,KEGG and Lipid Maps databases.RESULTS The zebrafish hypoxic model was successfully constructed via the different concentration of oxygen,temperature and hypoxic time.The activities of the related hypoxic metabolic enzymes and factors including HIF-1a,actate dehydrogenase(LDH)and citrate synthase(CS)were evaluated.Significant differences(P2)in the expression of 422 genes were observed between the normal and 3% hypoxic model.Among them,201 genes increased depended on the lower concentration of oxygen.53 metabolites were identified that had significant difference between the hypoxia and control groups(P1.5 and VIP>1.5).The ten key metabolites were increased gradually while six compounds were decreased.The endogenous hypoxic metabolites of phenylalanine,D-glucosamine-6P and several important lipids with the relevant hub genes had similar change in hypoxic model.In addition,the metabolic pathways of phenylalanine,glutamine and glycolipid were influenced in both the levels of genes and metabolites.CONCLUSION The up-regulation of phenylalanine,D-glucosamine-6P and lipid may have further understanding of protective effect in

随着高速列车速度的不断提升,列车尾部非定常尾迹引起的列车风安全问题越来越凸显。为探究转向架构型对列车风和非定常尾迹的影响,以CRH3两节编组的高速列车缩比模型为研究对象,采用增强型延迟分离涡模拟对三种不同转向架构型,即不对称转向架、对称转向架和无转向架,进行列车非定常流动的数值模拟研究。通过对比分析列车风时均和脉动速度分布以及尾迹流场结构,同时采用SPOD (Spectral Proper Orthogonal Decomposition)方法对非定常尾迹进行模态分解和流场重构。研究结果表明:三种转向架构型的列车尾迹均是由一对反向旋转的半环形流向交替涡脱落主导;相对于对称转向架构型,不对称转向架底部带来的扰动与尾迹流向涡对的相互作用更强,增加流向交替涡脱落的强度,继而诱导出更高的列车风脉动速度;无转向架的转向架舱及其后缘流动分离产生的大尺度涡脱落导致尾迹流向涡对展向范围更宽,列车风时均速度最大;SPOD方法通过增强相关矩阵的对角相似性,使得提取的主导模态配对性增强,模态峰值频率更加凸显,模态空间分布更加清晰。以上研究结果对高速列车转向架的设计优化有一定参考意义,显示了SPOD方法在高速列车非定常尾迹分析中的优势。

The earth-abundant and high-performance catalysts are crucial for commercial implementation of hydrogen evolution reaction(HER).Herein,a multifunctional site strategy to construct excellent HER catalysts by incorporating iridium(Ir)ions on the atomic scale into orthorhombic-CoSe2(Ir-CoSe_(2))was reported.Outstanding hydrogen evolution activity in alkaline media such as a low overpotential of 48.7 mV at a current density of 10 mA cm^(-2)and better performance than commercial Pt/C catalysts at high current densities were found in the Ir-CoSe_(2) samples.In the experiments and theoretical calculations,it was revealed that Ir enabled CoSe_(2)to form multifunctional sites to synergistically catalyze alkaline HER by promoting the adsorption and dissociation of H_(2)O(Ir sites)and optimizing the binding energy for H^(*)on Co sites.It was noticeable that the electrolytic system comprising the Ir-CoSe_(2)electrode not only produced hydrogen efficiently via HER,but also degraded organic pollutants(Methylene blue).The cell voltage of the dual-function electrolytic system was 1.58 V at the benchmark current density of 50 mA cm^(-2),which was significantly lower than the conventional water splitting voltage.It was indicated that this method was a novel strategy for designing advanced HER electrocatalysts by constructing multifunctional catalytic sites for hydrogen production and organic degradation.

Molecular targets for modulating the protein translation vital to proteostasis and neuron degeneration in Parkinson’s disease

Parkinson’s disease(PD)is the most common neurodegenerative movement disorder,which is characterized by the progressive loss of dopaminergic neurons in the Substantia Nigra pars compacta concomitant with Lewy body formation in affected brain areas.The detailed pathogenic mechanisms underlying the selective loss of dopaminergic neurons in PD are unclear,and no drugs or treatments have been developed to alleviate progressive dopaminergic neuron degeneration in PD.However,the formation ofα-synuclein-positive protein aggregates in Lewy body has been identified as a common pathological feature of PD,possibly stemming from the consequence of protein misfolding and dysfunctional proteostasis.Proteostasis is the mechanism for maintaining protein homeostasis via modulation of protein translation,enhancement of chaperone capacity and the prompt clearance of misfolded protein by the ubiquitin proteasome system and autophagy.Deregulated protein translation and impaired capacities of chaperone or protein degradation can disturb proteostasis processes,leading to pathological protein aggregation and neurodegeneration in PD.In recent years,multiple molecular targets in the modulation of protein translation vital to proteostasis and dopaminergic neuron degeneration have been identified.The potential pathophysiological and therapeutic significance of these molecular targets to neurodegeneration in PD is highlighted.

永磁同步电机的结构参数选择往往是多模态、非线性的优化问题,同时其结构优化设计又存在计算分析时间过长、计算效率过低、收敛慢等问题。文中针对此类问题提出了一种基于改进萤火虫算法的新型多模态优化算法。改进萤火虫算法主要引用了等值线概念并通过克里金(Kriging)代理模型自适应确定子计算区域,从而获得多个全局最优解和局部解,并且引入单纯形法和改进停止条件以提高算法效率。此外,研究了分层余一法用于检验代理模型精度。最后,将其应用到永磁同步电机的结构优化中,使齿槽转矩成功降低了38.33%。

在飞行器气动外形优化设计中,复杂约束条件导致设计空间可行域呈现不连续的特征,且理想解大多靠近约束边界,传统高效代理模型方法难以适用。研究了参考点对优化设计的影响,提出了一种考虑约束的参考点选择机制;对于最优解靠近边界的问题,罚函数法更加有效,但惩罚因子的设置对于罚函数方法影响很大,不合适的惩罚因子反而会损害优化效率,分析了优化过程中罚函数方法对惩罚因子的要求,提出了一种无参数自适应罚函数的代理模优化设计方法,引入基于样本分析的惩罚项,结合归一化目标值和约束值,在优化过程中动态调整惩罚因子,使优化能够尽可能地聚焦于可行域内,迅速收敛到最优解,实现样本的高效配置。通过带约束的函数算例和翼型优化算例证实,所提方法可以大幅提高飞行器气动外形优化设计效率。

Hepatocellular carcinoma: Can LI-RADS v2017 with gadoxetic-acid enhancement magnetic resonance and diffusion-weighted imaging improve diagnostic accuracy?

BACKGROUND The Liver Imaging Reporting and Data System(LI-RADS), supported by the American College of Radiology(ACR), has been developed for standardizing the acquisition, interpretation, reporting, and data collection of liver imaging examinations in patients at risk for hepatocellular carcinoma(HCC). Diffusionweighted imaging(DWI), which is described as an ancillary imaging feature of LI-RADS, can improve the diagnostic efficiency of LI-RADS v2017 with gadoxetic acid-enhanced magnetic resonance imaging(MRI) for HCC.AIM To determine whether the use of DWI can improve the diagnostic efficiency of LIRADS v2017 with gadoxetic acid-enhanced magnetic resonance MRI for HCC.METHODS In this institutional review board-approved study, 245 observations of high risk of HCC were retrospectively acquired from 203 patients who underwent gadoxetic acid-enhanced MRI from October 2013 to April 2018. Two readers independently measured the maximum diameter and recorded the presence of each lesion and assigned scores according to LI-RADS v2017. The test was used to determine the agreement between the two readers with or without DWI. In addition, the sensitivity(SE), specificity(SP), accuracy(AC), positive predictive value(PPV), and negative predictive value(NPV) of LI-RADS were calculated.Youden index values were used to compare the diagnostic performance of LIRADS with or without DWI.RESULTS Almost perfect interobserver agreement was obtained for the categorization of observations with LI-RADS(kappa value: 0.813 without DWI and 0.882 with DWI). For LR-5, the diagnostic SE, SP, and AC values were 61.2%, 92.5%, and71.4%, respectively, with or without DWI; for LR-4/5, they were 73.9%, 80%, and75.9% without DWI and 87.9%, 80%, and 85.3% with DWI; for LR-4/5/M, they were 75.8%, 58.8%, and 70.2% without DWI and 87.9%, 58.8%, and 78.4% with DWI; for LR-4/5/TIV, they were 75.8%, 75%, and 75.5% without DWI and 89.7%,75%, and 84.9% with DWI. The Youden index values of the LI-RADS classification without or with DWI were as follows: LR-4/5: 0.

基于注意力机制的高容量通用图像隐写模型

随着深度学习与隐写技术的发展,深度神经网络在图像隐写领域的应用越发广泛,尤其是图像嵌入图像这一新兴的研究方向.主流的基于深度神经网络的图像嵌入图像隐写方法需要将载体图像和秘密图像一起输入隐写模型生成含密图像,而最近的研究表明,隐写模型仅需要秘密图像作为输入,然后将模型输出的含密扰动添加到载体图像上,即可完成秘密图像的嵌入过程.这种不依赖载体图像的嵌入方式极大地扩展了隐写的应用场景,实现了隐写的通用性.但这种嵌入方式目前仅验证了秘密图像嵌入和恢复的可行性,而对隐写更重要的评价标准,即隐蔽性,未进行考虑和验证.提出一种基于注意力机制的高容量通用图像隐写模型USGAN,利用注意力模块,USGAN的编码器可以在通道维度上对秘密图像中像素位置的扰动强度分布进行调整,从而减小含密扰动对载体图像的影响.此外,利用基于CNN的隐写分析模型作为USGAN的目标模型,通过与目标模型进行对抗训练促使编码器学习生成含密对抗扰动,从而使含密图像同时成为攻击隐写分析模型的对抗样本.实验结果表明,所提模型不仅可以实现不依赖载体图像的通用嵌入方式,还进一步提高了隐写的隐蔽性.
