Differential protein expression in low-grade astrocytomas and peritumoral human brain tissues

Differential protein expression between various pathological grades of glioma has been shown in studies of glioma proteomics. However, very little data is available regarding normal brain tissues and glioma differential protein expression, because normal human brain tissues are difficult to harvest. The present study selected samples from low-grade astrocytomas and peritumoral brain tissues to analyze differential protein expression by two-dimensional （2D） electrophoresis and mass spectrometry techniques. Results revealing 36 protein spots by 2D electrophoresis, including 23 spots revealing increased expression and 13 spots revealing decreased expression. However, 25 differential proteins were identified by mass spectrometry, including 16 proteins with increased expression and 9 with decreased expression. Western blot analysis confirmed the mass spectrometry results, i.e., heat shock protein 70 （HSP70） and human transthyretin （TTR） expressions were increased, but glial fibrillary acidic protein （GFAP） was decreased, in astrocytomas. The present study constructed a 2D electrophoresis pattern between low-grade astrocytomas in the human brain and peritumoral tissues. Results demonstrated that a majority of differential proteins, such as HSP70, TTR, and GFAP, participate in malignant progression of gliomas.

In this study, the impact of atmospherewave coupling on typhoon intensity was investigated using numerical simulations of an idealized typhoon in a coupled atmospherewaveocean modeling system. The coupling between atmosphere and sea surface waves considered the effects of wave state and sea sprays on airsea momentum flux, the atmospheric lowlevel dissipative heating, and the wavestateaffected sea spray heat flux. Several experiments were conducted to examine the impacts of wave state, sea sprays, and dissipative heating on an idealized typhoon system. Results show that considering the wave state and seasprayaffected seasurface roughness reduces typhoon intensity, while including dissipative heating intensifies the typhoon system. Taking into account sea spray heat flux also strengthens the typhoon system with increasing maximum wind speed and significant wave height. The overall impact of atmospherewave coupling makes a positive contribution to the intensification of the idealized typhoon system. The minimum central pressure simulated by the coupled atmospherewave experiment was 16.4 hPa deeper than that of the control run, and the maximum wind speed and significant wave height increased by 31% and 4%, respectively. Meanwhile, within the area beneath the typhoon center, the average total upward airsea heat flux increased by 22%, and the averaged latent heat flux increased more significantly by 31% compared to the uncoupled run.

建立考虑车体和构架横移与侧滚自由度的车辆系统滚摆振动模型,利用复模态分解法推导系统响应公式;借助于相位同步模态分析揭示车体与构架的横移和侧滚响应与模态响应的物理关系。研究结果表明:车辆系统滚摆振动由4阶模态组成,分别为车体和构架的同相下心滚摆、同相上心滚摆、反相下心滚摆以及反相上心滚摆;轴箱外置车辆各阶模态阻尼比均比轴箱内置车辆的阻尼比大;在频率为10 Hz以内时,轴箱外置车辆的车体和构架响应明显比轴箱内置车辆的响应低,当频率为10 Hz以上时,这2种车型相差较小;当频率低于2 Hz时,这2种车型的车体响应比构架的响应大;当频率高于2 Hz时,构架响应比车体的响应大;采用合理柔性的二系横向刚度以及优化二系横向减振器阻尼,对降低车体低频振动最为有效。

Study and application of a new blockage remover in polymer injection wells

With the extensive application of polymer flooding technology in offshore oilfields,the plugging in polymer injection wells has become more and more severe,which seriously affects the oil displacement effect and regular production of oilfields.In this paper,a new kind of blockage remover has been developed and evaluated by rheological behavior experiments,dissolution experiments and core flooding experiments.The results reveal that this new blockage remover can effectively reduce the viscosity of polymer and completely degrade the reservoir blockage with low corrosion rate.It is beneficial to long-term production of oil wells in offshore oilfield.Results of core flooding experiments show that this new blockage remover can relieve polymer damage and improve permeability.The agent has been applied in LD10-1 oilfield in 2016,the daily injection rate increased significantly after stimulation.

地铁钢轨双光带形成机理分析

针对国内某地铁线路上直线段钢轨出现双光带的现象,选取标准钢轨廓形与不同镟后运行里程的车轮廓形匹配,利用建立的地铁车辆动力学模型与Archard磨耗模型,对钢轨双光带的形成机理进行分析;同时利用仿真得到的钢轨磨耗廓形与车轮标准踏面外形LM匹配,研究钢轨双光带对车辆动力学性能的影响。研究结果表明:当车轮标准踏面外形LM与标准钢轨廓形匹配时,在钢轨轨距角侧会产生明显的单峰值磨耗,在钢轨上表现为单光带;随车轮镟后运行里程增加,除了轨距角侧的磨耗峰值外,在轨顶外侧也开始出现磨耗峰值;随车轮镟后运行里程进一步增大,钢轨在轨距角侧和轨顶外侧将分别形成2个明显磨耗峰值,对应钢轨上双光带出现的位置,并且双光带的位置趋于稳定;钢轨产生双光带后,车辆的蛇行临界速度明显下降,横向平稳性变差,垂向平稳性几乎不受影响。

纳米CrN_(x)涂层相组成、结构及力学性能研究

采用高功率调制脉冲磁控溅射(modulated pulsed power magnetron sputtering,MPPMS)和脉冲直流磁控溅射(pulsed direct current magnetron sputtering,PDCMS)复合沉积CrN_(x)涂层,通过调节氮气流量比及溅射功率,研究了氮气/氩气流量比、PDCMS溅射功率及MPPMS溅射功率等工艺参数对CrN_(x)涂层成分、相组成、微结构和力学性能的影响。通过电子探针(EPMA)、X射线衍射仪(XRD)、扫描电子显微镜(SEM)、纳米压痕仪及维氏硬度计等,分别对CrN_(x)涂层的成分、相组成、微结构、形貌、硬度及断裂韧性等进行表征。结果表明,当PDCMS溅射功率从700 W增加到1000 W,MPPMS峰值功率增加43.5%,涂层中Cr含量(原子分数)由61.0%增加到65.4%,N含量由39.0%减少到34.6%,而CrN_(x)涂层主要由Cr_(2)N相组成。随着溅射功率的增大,CrN_(x)涂层硬度变化不大,均在20 GPa左右,而涂层的致密性和断裂韧性均得到明显提升。流量比由15%增加至50%,MPPMS的峰值电流和峰值功率均先减小后增大,涂层的物相逐渐由Cr_(2)N向CrN转变。当氮气流量比为35%时,CrN_(x)涂层由Cr_(2)N和CrN两相组成,受两相结构的影响,CrN_(x)涂层的硬度值、残余压应力值和断裂韧性值均达到最大值,分别为20.1 GPa、-901.8 MPa和6.5 MPa·m1/2。电子温度应为CrN_(x)涂层相结构变化的主要驱动力,致密性和两相结构是影响CrN_(x)涂层断裂韧性的主要原因。

Cucumber,Cucumis sativus L.(2n=2x=14),is an important vegetable crop worldwide.It was the first specialty crop with a publicly available draft genome.Its relatively small,diploid genome,short life cycle,and selfcompatible mating system offers advantages for genetic studies.In recent years,significant progress has been made in molecular mapping,and identification of genes and QTL responsible for key phenotypic traits,but a systematic review of the work is lacking.Here,we conducted an extensive literature review on mutants,genes and QTL that have been molecularly mapped or characterized in cucumber.We documented 81 simply inherited trait genes or major-effect QTL that have been cloned or fine mapped.For each gene,detailed information was compiled including chromosome locations,allelic variants and associated polymorphisms,predicted functions,and diagnostic markers that could be used for marker-assisted selection in cucumber breeding.We also documented 322 QTL for 42 quantitative traits,including 109 for disease resistances against seven pathogens.By alignment of these QTL on the latest version of cucumber draft genomes,consensus QTL across multiple studies were inferred,which provided insights into heritable correlations among different traits.Through collaborative efforts among public and private cucumber researchers,we identified 130 quantitative traits and developed a set of recommendations for QTL nomenclature in cucumber.This is the first attempt to systematically summarize,analyze and inventory cucumber mutants,cloned or mapped genes and QTL,which should be a useful resource for the cucurbit research community.

Chinese cabbage is the most consumed leafy crop in East Asian countries.However,premature bolting induced by continuous low temperatures severely decreases the yield and quality of the Chinese cabbage, and therefore restricts its planting season and geographic distribution.In the past 40years,spring Chinese cabbage with strong winterness has been selected to meet the market demand.Here,we report a genome variation map of Chinese cabbage generated from the resequencing data of 194 geographically diverse accessions of three ecotypes.In-depth analyses of the selection sweeps and genome-wide patterns revealed that spring Chinese cabbage was selected from a specific population of autumn Chinese cabbage around the area of Shandong peninsula in northern China.We identified 23 genomic loci that underwent intensive selection,and further demonstrated by gene expression and haplotype analyses that the incorporation of elite alleles of VERNAliSATION INSENTIVE 3.1(BrVIN3.1)and FLOWER LOCUS C 1(BrFLC1)is a determinant genetic source of variation during selection.Moreover,we showed that the quantitative response of BrVIN3.1 to cold due to the sequence variations in the cis elements of the BrVlN3.1 promoter significantly contributes to bolting-time variation in Chinese cabbage.Collectively, our study provides valuable insights into the genetic basis of spring Chinese cabbage selection and will facilitate the breeding of bolting-resistant Varieties by molecular-marker-assisted selection,transgenic or gene editingapproaches.

Bacillus thuringiensis（B.thuringiensis） is a soil-dwelling Gram-positive bacterium and its plasmid-encoded toxins（Cry） are commonly used as biological alternatives to pesticides.In a pangenomic study,we sequenced seven B.thuringiensis isolates in both high coverage and base-quality using the next-generation sequencing platform.The B.thuringiensis pangenome was extrapolated to have 4196 core genes and an asymptotic value of 558 unique genes when a new genome is added.Compared to the pangenomes of its closely related species of the same genus,B.thuringiensis pangenome shows an open characteristic,similar to B.cereus but not to B.anthracis;the latter has a closed pangenome. We also found extensive divergence among the seven B.thuringiensis genome assemblies,which harbor ample repeats and single nucleotide polymorphisms（SNPs）.The identities among orthologous genes are greater than 84.5%and the hotspots for the genome variations were discovered in genomic regions of 2.3-2.8 Mb and 5.0-5.6 Mb.We concluded that high-coverage sequence assemblies from multiple strains, before all the gaps are closed,are very useful for pangenomic studies.
