Predictors of pathologic complete response in patients with residual flat mucosal lesions after neoadjuvant chemoradiotherapy for locally advanced rectal cancer

Objective:The accurate prediction of tumor response to neoadjuvant chemoradiotherapy(nCRT)remains challenging.Few studies have investigated pathologic complete response(ypCR)prediction in patients with residual flat mucosal lesions after treatment.This study aimed to identify variables for predicting ypCR in patients with residual flat mucosal lesions after nCRT for locally advanced rectal cancer(LARC).Methods:Data of patients with residual flat mucosal lesions after nCRT who underwent radical resection between 2009 and 2015 were retrospectively collected from the LARC database at Peking University Cancer Hospital.Univariate and multivariate analyses of the association between clinicopathological factors and ypCR were performed,and a nomogram was constructed by incorporating the significant predictors.Results:Of the 246 patients with residual flat mucosal lesions included in the final analysis,56(22.8%)had ypCR.Univariate and multivariate analyses showed that pretreatment cT stage(pre-cT)≤T2(P=0.016),magnetic resonance tumor regression grade(MR-TRG)1-3(P=0.001)and residual mucosal lesion depth=0 mm(Plibration curve showed that the nomogram model had good predictive consistency.The follow-up time ranged from 3.0 to 113.3 months,with a median follow-up time of 63.77 months.The multivariate Cox regression model showed that the four variables in the nomogram model were not risk factors for disease-free survival(DFS)or overall survival(OS).Conclusions:Completely flat mucosa,early cT stage and good MR-TRG were predictive factors for ypCR instead of DFS or OS in patients with LARC with residual flat mucosal lesions after nCRT.Endoscopic mucosal re-evaluation before surgery is important,as it may contribute to decision-making and facilitate nonoperative management or organ preservation.

Durable electromagnetic interference(EMI)shielding is highly desired,as electromagnetic pollution is a great concern for electronics’stable performance and human health.Although a superhydrophobic surface can extend the service lifespan of EMI shielding materials,degradation of its protection capability and insufficient self-healing are troublesome issues due to unavoidable physical/chemical damages under long-term application conditions.Here,we report,for the first time,an instantaneously self-healing approach via microwave heating to achieve durable shielding performance.First,a hydrophobic 1H,1H,2H,2H-perfluorooctyltriethoxysilane(POTS)layer was coated on a polypyrrole(PPy)-modified fabric(PPy@POTS),enabling protection against the invasion of water,salt solution,and corrosive acidic and basic solutions.Moreover,after being damaged,the POTS layer can,for the first time,be instantaneously self-healed via microwave heating for a very short time,i.e.,4 s,benefiting from the intense thermal energy generated by PPy under electromagnetic wave radiation.This self-healing ability is also repeatable even after intentionally severe plasma etching,which highlights the great potential to achieve robust and durable EMI shielding applications.Significantly,this approach can be extended to other EMI shielding materials where heat is a triggering stimulus for healing thin protection layers.We envision that this work could provide insights into fabricating EMI shielding materials with durable performance for portable and wearable devices,as well as for human health care.

Pain threshold, anxiety and other factors affect intensity of postoperative pain in gastric cancer patients: A prospective cohort study

Objective: This prospective cohort study explored factors related to postoperative pain in gastric cancer patients.Methods: A total of 236 patients who underwent gastrectomy were enrolled. All patients enrolled in the study completed the Hospital Anxiety and Depression Scale(HADS) questionnaire and life Orientation Test-Revised(LOT-R) questionnaire on the day before surgery. Heat pain threshold(HPT), cold pain threshold(CPT) and pressure pain threshold(PPT) were measured for all patients one day prior to surgery and demographic details were collected. All patients were connected to a patient-controlled intravenous analgesia(PCIA) pump at the end of the surgery. The occurrence of postoperative pain was used as a dependent variable, and multivariate logistic regression analyses were conducted to screen for factors affecting postoperative pain.Results: In total, 83 patients(35.2%) had postoperative pain. Body mass index(BMI) ≥28 kg/m^(2) [odds ratio(OR): 2.67;95% confidence interval(95% CI): 1.07-6.67], total gastrectomy(OR: 2.64;95% CI: 1.42-4.91),preoperative anxiety score ≥8(OR: 2.37;95% CI: 1.12-5.02), heat pain threshold ≤4.9 s(OR: 2.14;95% CI:1.06-4.32), pressure pain threshold ≤4 g(OR: 2.05;95% CI: 1.05-4.03), and female gender(OR: 1.99;95% CI:1.04-3.83) were risk factors for postoperative pain.Conclusions: Obesity, wide range of gastrectomy, high preoperative anxiety, low HPT and PPT, and female gender are associated with increased risk for postoperative pain.

The intelligent knee prosthesis is capable of human-like bionic lower limb control through advanced control systems and artificial intelligence algorithms that will potentially minimize gait limitations for above-knee amputees and facilitate their reintegration into society.In this paper,we sum up the control strategies corresponding to the prevailing control objectives(position and impedance)of the current intelligent knee prosthesis.Although these control strategies have been successfully implemented and validated in relevant experiments,the existing deficiencies still fail to achieve optimal performance of the controllers,which complicates the definition of a standard control method.Before a mature control system can be developed,it is more important to realize the full potential for the control strategy,which requires upgrading and refining the relevant key technologies based on the existing control methods.For this reason,we discuss potential areas for improvement of the prosthetic control system based on the summarized control strategies,including intent recognition,sensor system,prosthetic evaluation,and parameter optimization algorithms,providing future directions toward optimizing control strategies for the next generation of intelligent knee prostheses.

Levofloxacin(LVFX)as a representative drug of quinolone antibiotics is widely used in clinical,and its residues enriched in water bodies and sideline products seriously damage human health.It is imperative to develop a real-time/on-site sensing method for monitoring residual antibiotics.Here,we report a portable sensing platform by utilizing a composite fluorescent nanoprobe constructed by the cerium ions(Ce^(3+))coordination functionalized Cd Te quantum dots(QDs)for the visual and quantitative detection of LVFX residues.This fluorescent probe provides a distinct color variation from red to green,which shows a good linear relationship to LVFX residues concentrations in the range of 0-6.0μmol/L with a sensitive limit of detection(LOD)of 16.3 nmol/L.The smartphone platform with Color Analyzer App installed,which could accomplish quantified detection of LVFX in water,milk,and raw pork with a LOD of 27.9nmol/L.The facile sensing method we proposed realizes rapid visualization of antibiotics residual in the environment and provides a practical application pathway in food safety and human health.

The effects of La on the catalytic performance,SO_(2)and H2O resistance of Cu-Ce/TNU-9 catalyst were studied in the selective catalytic reduction of NO_(x)via ammonia(NH_(3)-SCR).The results show that the La doped Ce-Cu/TNU-9(CCL/T9)catalyst exhibits better SCR performance than Ce-Cu/TNU-9(CC/T9)and Cu/TNU-9(C/T9)in the wide temperature window(200-450℃)due to La benefiting from enhancing Cu^(+)+Ce^(4+)?Cu^(2+)-+Ce^(3+)to facilitate the electron transfer,which further accelerates the oxidation of NO into NO_(2)that leads to the"Fast SCR".The La can also enhance the surface acidic,redox property and is beneficial to the formation of oxygen vacancy.Meanwhile,the larger co ntent of Ce^(3+) can produce oxygen vacancy,resulting in rising SCR activity.The CCL/T9 has outstanding water and SO_(2) resistance,which may be due to the adso rbed S on the surface of these catalysts mainly existing in the form of a small amount of sulfate and sulfite,the decomposition of ammonium bisulphite under high temperature and the hydrophobicity of TNU-9 zeolite.

The family of voltage-gated potassium Kv2 channels consists of the Kv2.1 and Kv2.2 subtypes.Kv2.1 is constitutively highly phosphorylated in neurons and its function relies on its phosphorylation state.Whether the function of Kv2.2 is also dependent on its phosphorylation state remains unknown.Here,we investigated whether Kv2.2 channels can be phosphorylated by protein kinase C(PKC)and examined the effects of PKC-induced phosphorylation on their activity and function.Activation of PKC inhibited Kv2.2 currents and altered their steadystate activation in HEK293 cells.Point mutations and specific antibodies against phosphorylated S481 or S488 demonstrated the importance of these residues for the PKC-dependent modulation of Kv2.2.In layer Ⅱ pyramidal neurons in cortical slices,activation of PKC similarly regulated native Kv2.2 channels and simultaneously reduced the frequency of action potentials.In conclusion,this study provides the first evidence to our knowledge that PKC-induced phosphorylation of the Kv2.2 channel controls the excitability of cortical pyramidal neurons.

Repurposable drugs for SARS-CoV-2 and influenza sepsis with scRNA-seq data targeting post-transcription modifications

Coronavirus disease 2019(COVID-19)has impacted almost every part of human lifeworldwide,posing amassive threat to human health.The lack of time for new drug discovery and the urgent need for rapid disease control to reduce mortality have led to a search for quick and effective alternatives to novel therapeutics,for example drug repurposing.To identify potentially repurposable drugs,we employed a systematic approach to mine candidates from U.S.FDA-approved drugs and preclinical small-molecule compounds by integrating gene expression perturbation data for chemicals from the library of Integrated Network-Based Cellular Signatures project with a publicly available single-cell RNA sequencing dataset from patients withmild and severe COVID-19(GEO:GSE145926,public data available and accessed on 22 April 2020).We identified 281 FDA-approved drugs that have the potential to be effective against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,16 of which are currently undergoing clinical trials to evaluate their efficacy against COVID-19.We experimentally tested and demonstrated the inhibitory effects of tyrphostin-AG-1478 and brefeldin-a,two chemical inhibitors of glycosylation(a post-translational modification)on the replication of the single-stranded ribonucleic acid(ssRNA)virus influenza A virus as well as on the transcription and translation of host cell cytokines and their regulators(IFNs and ISGs).In conclusion,we have identified and experimentally validated repurposable anti-SARS-CoV-2 and IAV drugs using a systems biology approach,which may have the potential for treating these viral infections and their complications(sepsis).

Synthesis of a ternary amide lixKy（NH2）x+y and a novel li3K（NH2）4–xMgH2 combination system for hydrogen storage

The ternary amides liK2(NH2)3, liK(NH2)2, and li3 K(NH2)4 are successfully synthesized by ball milling mixtures of liNH2 and KNH2, and the hydrogen storage properties of li3K(NH2)4–xMgH2(x = 1, 2, 3,4) are systematically investigated. The li3K(NH2)4–2 Mg H2 sample displays optimized hydrogen storage properties, releasing 6.37 wt% of hydrogen in a two-stage reaction with an onset temperature of 60 °C.The first dehydrogenation stage exhibits good reaction kinetics and thermodynamic properties because of a lower activation energy and appropriate enthalpy change. After full dehydrogenation at 130 °C, the li3K(NH2)4–2 MgH2 sample absorbs 3.80 wt% of H2 below 160 °C in a variable temperature hydrogenation mode. Mechanistic investigations indicate that li3 K(NH2)4 reacts with Mg H2 to produce Mg(NH2)2, liH,and KH during ball milling. In the heating process, Mg(NH2)2 first reacts with li H to form li2 Mg2 N3 H3 and li NH2, while KH works as a catalyst, and then, KH reacts with li2Mg2N3H3 and li NH2 to generate a new K-containing compound.
