In matrix completion,additional covariates often provide valuable information for completing the unobserved entries of a high-dimensional low-rank matrix *** this paper,the authors consider the matrix recovery problem when there are multiple structural breaks in the coefficient matrix β under the column-space-decomposition model A=Xβ+B.A cumulative sum(CUSUM)statistic is constructed based on the penalized estimation of β.Then the CUSUM is incorporated into the Wild Binary Segmentation(WBS)algorithm to consistently estimate the location of ***,a nearly-optimal recovery of A is *** findings are further corroborated via numerical experiments and a real-data application.

A method for in-situ stress measurement via fiber optics was proposed. The method utilizes the relationship between rock mass elastic parameters and in-situ stress. The approach offers the advantage of long-term stress measurements with high spatial resolution and frequency, significantly enhancing the ability to measure in-situ stress. The sensing casing, spirally wrapped with fiber optic, is cemented into the formation to establish a formation sensing nerve. Injecting fluid into the casing generates strain disturbance, establishing the relationship between rock mass properties and treatment ***, an optimization algorithm is established to invert the elastic parameters of formation via fiber optic strains. In the first part of this paper series, we established the theoretical basis for the inverse differential strain analysis method for in-situ stress measurement, which was subsequently verified using an analytical model. This paper is the fundamental basis for the inverse differential strain analysis method.

Receptor-ligand interactions in blood flow are crucial to initiate such biological processes as inflammatory cascade,platelet thrombosis,as well as tumor *** mediate cell adhesion,the interacting receptors and ligands must be anchored onto two apposing surfaces of two cells or a cell and a substratum,i.e.,two-dimensional（2D）binding,which is different from the binding of a soluble ligand in fluid phase to a receptor,i.e.,three-dimensional（3D） *** numerous works have been focused on3 D kinetics of receptor-ligand interactions in the immune system,2D kinetics and its regulations have been less understood,since no theoretical framework or experimental assays were established until *** only does the molecular structure dominate 2D binding kinetics,but the shear force in blood flow also regulates cell adhesion mediated by interacting receptors and ***,we provide an overview of current progress in 2D binding and regulations,mainly from our *** issues of theoretical frameworks,experimental measurements,kinetic rates and binding affinities,and force regulations are discussed.

Neutrophil (PMN) spreading on endothelium, mediated by the interactions between surface-bound β2 integrin and intercellular adhesion molecule-1 (ICAM-1) in the inflammatory cascade, is crucial for PMN post-adhesion and trans-migration in blood flow. The underlying mechanisms by which shear flow regulates PMN spreading dynamics are not well understood. Here, a parallel-plate flow chamber assay was applied to quantify the time course of PMN adhesion and spreading on an ICAM-1-immobilized substrate. Two types of shear flow, steady flows at shear stresses of 0.2, 0.5, and 1 dyne/cm2 and stepwise flows at 0, 1, and 10 dyne/cm2, were used to elucidate the impact of shear flow on cell adhesion and spreading. The number of adhered PMNs, the fraction of spreading PMNs and the projected area of spread PMNs were determined and were found to correlate with the distribution of surface-bound β2 integrin subunit (CD11a, CD11b, or CD18). The results indicate that PMN spreading on an ICAM-1 substrate is bi-directionally regulated under shear flow. CD11a, CD11b and CD18 subunits of β2 integrin contribute distinctly to PMN spreading on ICAM-1 substrates. This work provides new insights into understanding PMN spreading on the endothelium, mediated by β2 integrin and ICAM-1 under shear flow.

The efficient development and exploitation of shale oil depends on long-distance horizontal wells. As the degreeof cleaning of the wellbore plays a key role in these processes, in this study, this problem is investigated experimentallyby focusing on the dimensionless cuttings bed height. A method is proposed to calculate the horizontalwellhydraulic extension taking into account the influence of the wellbore cleaning degree on the wellborepressure distribution and assess the effect of a variety of factors such as the bottom hole pressure, the circulatingpressure drop, the drilling pump performance and the formation properties. The analysis shows that the hydraulicextension of horizontal wells decreases with an increase in the cuttings bed height, and the higher the displacementof drilling fluid, the faster the hydraulic extension declines. The annular pressure drop of the horizontalsection increases with the increase of the cuttings bed height, resulting in a higher bottom-hole pressure. Severalarguments are provided to guide the safe drilling of shale oil horizontal wells and overcome the limits of currenttechnological approaches.

Neutrophil(PMN)accumulation on liver sinusoidal endothelial cells(lSECs)is crucial to pathogen clearance and tissue damage in the liver sinusoids and controlled by a series of adhesion molecules expressed on the surface of PMNs and *** role of lymphocyte function-associated antigen-1(lFA-1)and macrophage-1 antigen(Mac-1)in this process is still *** we compared the dynamic force spectra of the binding ofβ2 integrin to intercellular adhesion molecule-1(ICAM-1)on lSECs using atomic force microscopy(AFM)and performed free and steered molecular dynamics(MD)simulations to analyze their structural bases of lFA-1-or Mac-1-I-domain and ICAM-1-D1 or D3 pair in their force *** AFM data suggest that the mechanical strength of lFA-1-ICAM-1 bond is significantly stronger than that of Mac-1-ICAM-1 bond,implying a dominate role for lFA-1 to mediate PMN adhesion under shear *** simulations indicated that spontaneous dissociation of Mac-1-I-domain ***3-domain is slower with the stronger interaction energy than that for lFA-1 I-domain ***-D1-domain and that the rupture force for Mac-1 is lower than that for lFA-1,which are in qualitative agreement with the above experimental *** data indicate that the biomechanical features of lFA-1 and Mac-1 to mediate PMN adhesion on lSECs in vitro are similar with those in other tissues like cerebrovascular endothelium,while Mac-1-mediated PMN recruitment in liver sinusoids may stem from the slow blood flow in *** findings further the understandings of PMN recruitment under shear flow in liver sinusoids.

Receptor-ligand bond dissociation under applied force is crucial to elucidate its biological functionality when the molecular bond is usually connected to a mechanical probe. While the impact of probe stiffness, k, on bond rupture force has recently at- tracted more and more attention, the mechanism of how it affects the bond lifetime, however, remains unclear. Here we quanti- fied the dissociation lifetime of selectin-ligand bond using an optical trap assay with low stiffness ranging from 3.5×10^-3 to 4.7×10^-2 pN/nm. Our results indicated that bond lifetime yielded distinct distributions with different probe stiffness, implying the stochastic feature of bond dissociation. It was also found that the mean lifetime varied with probe stiffness and that the catch bond nature was visualized at k≥3.0×10^-2 pN/nm. This work furthered the understanding of the forced dissociation of se- lectin-ligand bond at varied probe stiffness, which is physiologically relevant to the tethered rolling of leukoeytes under blood flow.
