Osteoarthritis(OA)is characterized by cartilage degradation,pain,and synovitis.1 Joint inflammation driven by cytokines has been demonstrated to cause cartilage degradation and pain.2 However,approaches to neutralize cytokines,such as IL-1 and TNF-α,known to be involved in OA have shown poor clinical efficacy.3 There is an unmet clinical need to find better anti-inflammatory and pain targets for OA therapy and to elucidate the role of other cytokines in OA pathogenesis.Previous studies have shown that IL-22 and its receptor IL-22R play central roles in inflammation and diseases such as psoriasis,ulcerative colitis,graft-versus-host disease,certain infections and tumors,as well as in liver and pancreas damage.4,5 The role of IL-22/IL-22R and the potential for therapeutic targeting of both proteins in OA remain largely unknown,which we sought to investigate.

The ongoing coronavirus disease 2019(COVID-19)pandemic is a global public health crisis,causing social and economic disasters in many countries.In China,two-consecutive negative results of nucleic acid tests for SARS-CoV-2 from the respiratory samples are required to end the quarantine of COVID-19 patients.However,clinicians face a dilemma in case of patients with long-term viral shedding.This report described an unusual COVID-19 case who had persistent viral RNA positivity for more than 4 months after initial illness in the presence of low neutralizing antibodies,but without prolonged clinical symptoms.Multiple anti-viral drug treatments had no impact and there was no evidence of re-infection.When the patient was self-quarantined at home,no infection occurred to the three family members living with her for 15 to 19 days.Sputum viral culture in BSL-3 laboratory on the 102nd day after symptom onset was negative.From the 129th day on,8 continuous nucleic acid tests of sputum samples showed negative results.The patient was discharged on 137th days since symptom onset.In conclusion,viral RNA shedding in the sputum of the COVID-19 patient may last over 4 months.As no evidence shows the existence of infectious virus,two-consecutive negative nucleic acid tests may not be the prerequisite for ending quarantine of COVID-19 patients with prolonged viral shedding.

Dear Editor,Influenza affects humans of all ages;however,elderly people have increased susceptibility to infections and are especially predisposed to complications.1 A number of global efforts have increased influenza vaccination administration in elderly people to reduce complications,particularly when seasonal influenza infections peak.2 To date,much is known about how influenza infection and host immunity interact during pathogenesis,which has led to a number of vaccines.3,4 However,the impact of influenza vaccines in older people is modest.2 Hence,the underlying pathophysiological mechanisms responsible for the worsening of influenza infection in old age are still elusive.

Recently, giant intrinsic anomalous Hall effect(AHE) has been observed in the materials with kagome lattice.Here, we systematically investigate the influence of high pressure on the AHE in the ferromagnet LiMn_(6)Sn_(6) with clean Mn kagome lattice. Our in situ high-pressure Raman spectroscopy indicates that the crystal structure of LiMn_(6)Sn_(6) maintains a hexagonal phase under high pressures up to 8.51 GPa. The anomalous Hall conductivity(AHC) σ_(xy)^(A) remains around 150 Ω^(-1)·cm^(-1), dominated by the intrinsic mechanism. Combined with theoretical calculations, our results indicate that the stable AHE under pressure in Li Mn_(6)Sn_(6) originates from the robust electronic and magnetic structure.

为了构建出表达新布尼亚病毒(SFTSV)Gn基因的重组5型腺病毒,试验根据GenBank上发表的SFTSV Gn基因序列(登录号为ADZ04482.1)设计合成引物,采用特异性PCR方法在Gn基因前添加了KOZAK序列和tPA信号肽序列;然后通过酶切、连接等方法构建重组穿梭质粒PGA-KOZAK-tPA-Gn,与腺病毒骨架质粒pAd5-ΔE1ΔE3-5E4在BJ5183感受态细胞中进行同源重组,得到重组腺病毒质粒rAd5-KOZAK-tPA-Gn,用PacⅠ酶酶切重组腺病毒质粒,并将线性化的质粒转染至HEK 293细胞中进行病毒包装、扩繁和纯化;采用PCR扩增、Western-blot等技术检测病毒基因的表达情况,并测定重组腺病毒效价。结果表明:通过PCR扩增获得了1 546 bp的KOZAK-tPA-Gn基因,并成功构建了重组穿梭质粒;SFTSV Gn基因在重组腺病毒传代过程中稳定存在;重组腺病毒在HEK 293细胞中表达出分子量约为61 ku的SFTSV Gn蛋白;测得重组腺病毒效价为1×10^(-7.63)/0.1 mL TCID_(50)。说明试验成功构建出表达SFTSV Gn基因的重组5型腺病毒。

Clathrate-like or caged compounds have attracted great interest owing to their structural flexibility,as well as their fertile physical properties.Here,we report the pressure-induced reemergence of superconductivity in BaIr2Ge7 and Ba3Ir4Ge16,two new caged superconductors with two-dimensional building blocks of cage structures.After suppression of the ambient-pressure superconducting(SC-I)states,new superconducting(SC-II)states emerge unexpectedly,with Tc increased to a maximum of 4.4 and 4.0 K for BaIr2Ge7 and Ba3Ir4Ge16,respectively.Combined with high-pressure synchrotron x-ray diffraction and Raman measurements,we propose that the reemergence of superconductivity in these caged superconductors can be ascribed to a pressure-induced phonon softening linked to cage shrinkage.

The recent discovery of superconductivity up to 32 K in the pressurized MoBreignites the interest in exploring high-Tc superconductors in transition-metal diborides. Inspired by that work, we turn our attention to the 5 d transition-metal diborides.Here we systematically investigate the responses of both structural and physical properties of WBand ReBto external pressure,which possess different types of boron layers. Similar to MoB, the pressure-induced superconductivity was also observed in WBabove 60 GPa with a maximum Tcof 15 K at 100 GPa, while no superconductivity was detected in ReBin this pressure range. Interestingly, the structures at ambient pressure for both WBand ReBpersist to high pressure without structural phase transitions. Theoretical calculations suggest that the ratio of flat boron layers in this class of transition-metal diborides may be crucial for the appearance of high Tc. The combined theoretical and experimental results highlight the effect of the geometry of boron layers on superconductivity and shed light on the exploration of novel high-Tcsuperconductors in borides.
