Orally administered peptides or proteins are garnering increasing preference owing to their superiority in terms of patient compliance and ***,the development of oral protein formulations has stalled due to the low bioavailability of macromolecules that encounter the aggressive gastrointestinal environment and harsh mucus villi ***,we propose an ideal reverse micelle/self-emulsifying drug delivery system(RM/SEDDS)nanoplatform that is capable of improving the oral bioavailability of hydrophilic peptides by preventing enzymatic degradation and enhancing mucosal *** the passage through the mucus,the self-emulsifying drug delivery system with optimal surface properties effectively penetrates the viscoelastic mucosal barrier,followed by the exposure of the inner reverse micelle amphipathic vectors,which autonomously form continua with the lipidic cell membrane and facilitate the internalization of *** membrane-fusion mechanism inaugurates a new way for hydrophilic peptide delivery in the free form,circumventing the traditional impediments of the cellular internalization of nanocarriers and subsequent poor release of *** more importantly,reverse micelles are not spatially specific to the laden drugs,which enables their delivery for a myriad of peptide clinical *** conclusion,as an exquisitely designed nanoplatform,RM/SEDDS overcomes multiple physiological barriers and opens a new path for drug cellular entry,providing new prospects for the development of oral drug delivery systems.

Although iron oxide(Fe_(3)O_(4)) nanoparticles have broad application prospects as magnetic resonance imaging(MRI) contrast agent, their biocompatibility and biotoxicity still need to be improved. In this study, we prepared Fe_(3)O_(4)@Angelica sinensis polysaccharide nanoparticles(Fe_(3)O_(4)@ASP NPs) with a 9 nm Fe_(3)O_(4) core and ASP as the coating material. The Fe_(3)O_(4)@ASP NPs are superparamagnetic, can be taken up by liver and spleen macrophages in the circulatory system in vivo, and are a good-biocompatibility and low-toxicity transverse relaxation time(T_(2)) and T_(2)-star(T_(2)^(*)) magnetic resonance imaging(MRI) contrast agent for the liver. The cytotoxicity assessment using HeLa cells and the pathological tests in mice validate that Fe_(3)O_(4)@ASP NPs have low toxicity and good biocompatibility in vivo, which can be attributed to the ASP as a natural polysaccharide with good biocompatibility and its function of protecting the liver. Fe_(3)O_(4)@ASP NPs are a potential new MRI contrast agent with high signal intensity in vivo.

目的:建立黄蜀葵花药材、饮片、标准汤剂与配方颗粒的高效液相色谱(UPLC)特征图谱,并同时测定黄酮类成分的含量,考察四者之间化学成分的传递与差异。方法:采用Shim-packGISTC_(18)-AQ色谱柱(2.1 mm×150 mm,1.9μm),以乙腈(A)和0.1%磷酸溶液(B)为流动相梯度洗脱,检测波长为360 nm,流速为0.30 mL·min^(-1),柱温为30℃。以含量、特征图谱共有峰传递数及相似度为主要评价指标,分析其量值传递规律。结果:18批黄蜀葵花药材、饮片、标准汤剂及3批配方颗粒特征图谱均呈现8个共有特征峰,与各自对照特征图谱相似度均>0.90,辨别出杨梅素-3-O-β-D-葡萄糖苷(峰2)、金丝桃苷(峰3)、异槲皮苷(峰4)、棉皮素-8-O-β-D-葡萄糖醛酸苷(峰5)、杨梅素(峰6)、槲皮素-3′-O-葡萄糖苷(峰7)、槲皮素(峰8)7个成分,并同时测定其含量,未有离散数据出现。结论:黄蜀葵花药材、饮片、标准汤剂与配方颗粒的主要化学成分基本相同,表明特征图谱具有较高的相关性。7个成分在测定范围内有良好的线性关系,所建立的整体质量评价方法系统能够客观、全面地反映黄蜀葵花4种形态多个成分的综合情况,为黄蜀葵花配方颗粒后续质量标准的控制提供一定的参考依据。

Bio-assembled smart nanocapsules for targeted delivery of KRAS shRNA and cancer cell bioimage

The five-year survival rate for pancreatic cancer is less than 5%. However, the current clinical multimodal therapy combined with first-line chemotherapy drugs only increases the patient’s median survival from 5.0 months to 7.2 months. Consequently, a new strategy of cancer treatments is urgently needed to overcome this high-fatality disease. Through a series of biometric analyses, we found that KRAS is highly expressed in the tumor of pancreatic cancer patients, and this high expression is closely related to the poor prognosis of patients. It shows that inhibiting the expression of KRAS has great potential in gene therapy for pancreatic cancer. Given those above, we have exploited the possibility of targeted delivery of KRAS shRNA with the intelligent and bio-responsive nanomedicine to detect the special oxidative stress microenvironment of cancer cells and realize efficient cancer theranostics. Our observations demonstrate that by designing the smart self-assembled nanocapsules of melanin with fluorescent nanoclusters we can readily achieve the bio-recognition and bioimaging of cancer cells in biological solution or *** self-assembled nanocapsules can make a significant bio-response to the oxidative stress microenvironment of cancer cells and generate fluorescent zinc oxide Nanoclusters in situ for targeted cell bioimaging. Moreover, it can also readily facilitate cancer cell suppression through the targeted delivery of KRAS shRNA and low-temperature hyperthermia. This raises the possibility to provide a promising theranostics platform and self-assembled nanomedicine for targeted cancer diagnostics and treatments through special oxidative stress-responsive effects of cancer cells.

Replacement of Dietary Fish Oil with Vegetable Oils Improves the Growth and Flesh Quality of Large Yellow Croaker(Larmichthys crocea)

We investigated the effect of the replacement of dietary fish oil with vegetable oils on the growth and flesh quality of large yellow croaker(Larmichthys crocea). The basal diet(FO) was formulated to contain 66.5% fish meal and 6.4% menhaden fish oil; whereas the other 3 experimental diets were formulated by replacing the fish oil with 50% soybean oil(SO50), 100% soybean oil(SO100) and 100% palm oil(PO100), respectively. The 4 diets were randomly assigned to 4 floating sea cages(3.0 m × 3.0 m × 3.0 m), and each was stocked with 250 fish individuals with an initial average weight of 245.29 g ± 7.45 g. The fish were fed to apparent satiation twice a day at 5:00 and 17:00, respectively, for 12 weeks. Experimental analysis showed that the specific growth rate of fish fed SO50 or PO100 were significantly higher than that of fish fed FO or SO100(Plues of fish among the dietary treatments were observed(P>0.05). Compared to FO diet, SO50, SO100 and PO100 diets led to substantial decreases in the liquid loss and water loss from fresh fillets(1 d, 4℃)(Plues of fillets under different storage conditions(1 d, 4℃; 7 d, 4℃; 4 weeks,-20℃; 8 weeks,-20℃) decreased significantly after partial or complete replacement of fish oil with vegetable oils. These findings indicated that the growth performance and selected flesh quality properties(liquid holding capacity and TBARS value) of large yellow croaker were substantially improved by replacing dietary fish oil with vegetable oils.

针对现有的工业钢丝绳损伤监测系统的局限性,设计并实现了一款基于LabVIEW、Flask和***的钢丝绳远程智慧监测系统。该系统兼具传感器数据采集、信号处理、可视化展示、远程检测、数据分析等多项功能,以满足钢丝绳检测的全面需求。系统使用LabVIEW进行传感器数据采集和实时处理,Flask用于构建后端服务器以及数据处理,***负责前端的可视化展示和用户界面,使工程师和操作人员能够远程监测钢丝绳状态并及时作出反应。试验结果表明,采用的综合性系统设计不仅提升了工业生产中钢丝绳的质量控制水平,也大幅提高了远程监测能力,为工程人员提供了全方位的实时监测和管理工具。
