The use of aptamers in bioanalytical and biomedical applications exploits their ability to recognize cell surface protein receptors. Targeted therapeutics and theranostics come to mind in this regard. However,protein receptors occur on both cancer and normal cells; as such, aptamers are now taxed with identifying high vs. low levels of protein expression. Inspired by the flexible template mechanism and elegant control of natural nucleic acid-based structures, we report an allosteric regulation strategy for constructing a structure-switching aptamer for enhanced target cell recognition by engineering aptamers with DNA intercalated motifs(i-motifs) responsive to the microenvironment, such as p H. Structure-switching sensitivity can be readily tuned by manipulating i-motif sequences. However, structure-switching sensitivity is difficult to estimate, making it equally difficult to effectively screen modified aptamers with the desired sensitivity. To address this problem, we selected a fluorescent probe capable of detecting G-quadruplex in complicated biological media.

Traditional Chinese medicine(TCM)has been increasingly applied in both preventing and treating a variety of cancers in the last decades,attributing to its fewer side effects as compared with chemotherapy ***,a component of Chanpi from the skin of Bufo bufogargarizans Cantor or Duttaphrynus melanostictus has been reported to have an obvious anti-cancer activity on various ***,the effect and mechanism of hellebrigenin on colorectal cancers were still ***,the present study demonstrated hellebrigenin significantly reduced viability and triggered apoptosis via the intrinsic pathway in colorectal cancer cell lines HCT116 and HT29 in vitro and in ***,hellebrigenin led to a reduction of mitochondrial membrane *** addition,treatment with hellebrigenin could result in the induction of excessive reactive oxygen species,which led to cell *** results indicated that hellebrigenin had anti-cancer potential in the treatment of colorectal cancers.

A study of hybrid prediction method for ship parametric rolling

The parametric rolling（PR） in the head or following waves has been considered as one of the main stability failure modes in the development of the 2nd generation Intact Stability criterion by the International Maritime Organization（IMO）.According to previous studies,the estimation methods of the roll damping affect the prediction of the PR significantly,and most of them are based on experiment data or Ikeda＇s empirical *** accuracy of the estimation method for the roll damping could be a key aspect for the validity of its prediction for the full scale *** this research,a hybrid prediction method is developed for the numerical prediction of the parametric rolling when experiment data are not available for the roll *** study is also carried out between the hybrid method and a nonlinear dynamics method,where the roll damping is estimated by the simplified Ikeda＇s method and the direct CFD prediction method in a direct non-linear simulation based on the 3-D CFD approach in the model *** is shown that the results of the hybrid method are in satisfactory agreements with the model experiment results,and the method can be used for analysis especially at the early design stage where experiment data are often not available.

Background:Nutrient regulation has been proven to be an effective way to improve the flesh quality in *** a necessary nutrient for fish growth,protein accounts for the highest proportion in the fish diet and is *** our team found that the effect of protein on the muscle hardness of grass carp was probably related to an increased collagen content,the mechanism for this effect has not been deeply ***,few studies have explored the protein requirements of sub-adult grass crap(Ctenopharyngodon idella).Therefore,the effects of different dietary protein levels on the growth performance,nutritional value,muscle hardness,muscle fiber growth,collagen metabolism and related molecule expression in grass carp were ***:A total of 450 healthy grass carp(721.16±1.98 g)were selected and assigned randomly to six experimen-tal groups with three replicates each(n=25/replicate),and were fed six diets with 15.91%,19.39%,22.10%,25.59%,28.53%and 31.42%protein for 60 ***:Appropriate levels of dietary protein increased the feed intake,percentage weight gain,specific growth rate,body composition,unsaturated fatty acid content in muscle,partial free amino acid content in muscle,and muscle hardness of grass *** protein levels also increased the muscle fiber density,the frequency of new muscle fibers,the contents of collagen and IGF-1,and the enzyme activities of prolyl 4-hydroxylases and lysyloxidase,and decreased the activity of matrix *** the molecular level,the optimal dietary protein increased col-lagen type Iα1(Colα1),Colα2,PI3K,Akt,S6K1,La ribonucleoprotein domain family member 6a(LARP6a),TGF-β1,Smad2,Smad4,Smad3,tissue inhibitor of metalloproteinase-2,MyoD,Myf5,MyoG and MyHC relative mRNA *** levels of the myostatin-1 and myostatin-2 genes were downregulated,and the protein expression levels of p-Smad2,Smad2,Smad4,p-Akt,Akt,LARP6 and Smad3 were ***:The appropriate levels of dietary

紧扣新时代数字经济发展现状和基本原则,基于2011-2020年“一带一路”39个国家的面板数据构建并测算数字经济发展水平,运用基尼系数分析其差异,采用固定效应面板模型和两阶段最小二乘法识别其驱动因素。研究表明:“一带一路”沿线国家数字经济发展水平整体较低,异质性特征明显,互联互通程度不高,但总体呈上升态势,数字基础设施建设是其变化的主要原因;数字经济发展水平的差异总体呈扩大趋势,制度保障和创新环境差距始终不大,数字技术创新能力差距的扩大是其主要原因;制度保障与创新环境对数字经济发展的促进作用最大,数字基础设施建设次之,数字技术创新能力位居第三且对数字经济发展水平的影响存在滞后性,数字经济外向竞争力影响程度相对较小。

目的基于SERVQUAL(Service Quality)模型对重庆市某三甲医院的临床医学硕士专业学位研究生进行调查,深入分析教育服务的现状及问题并探讨改进措施。方法基于SERVQUAL模型设计5个维度、共30个题项的调查问卷,每个题项均有“期望值”与“感知值”2个分值,以2022年上半年重庆市某三甲医院在读临床医学硕士专业学位研究生为研究对象,测评研究生对教育服务的期望值(expectation service quality,E)与感知值(perceived service quality,P),计算服务质量(service quality,SQ)。用SPSS 26.0进行问卷的信度分析和效度分析、描述性统计分析。采用二维象限图分析测评结果。结果共回收有效问卷263份,有效率97.77%。期望量表和感知量表α系数均高于0.8,KMO值>0.7,问卷的信度和效度较好。教育服务质量的期望均值为4.28,感知均值为3.96,差值均值为-0.32;除“提供整洁的工作装”一项外,其余题项的SQ均为负值。二维象限分析结果显示,6个题项位于象限Ⅲ即“低期望低感知区”,7个题项位于象限Ⅳ即“高期望低感知区”。结论基于SERVQUAL模型的评价量表能较好地分析临床医学硕士专业学位研究生教育服务中存在的问题并提出解决策略,为提升教育服务质量提供重要参考。

Long scan depth optical coherence tomography on imaging accommodation: impact of enhanced axial resolution, signal-to-noise ratio and speed

Background:Spectral domain optical coherence tomography(SD-OCT)was a useful tool to study accommodation in human eye,but the maximum image depth is limited due to the decreased signal-to-noise ratio(SNR).In this study,improving optical resolutions,speeds and the SNR were achieved by custom built SD-OCT,and the evaluation of the impact of the improvement during accommodation was ***:Three systems with different spectrometer designs,including two Charge Coupled Device(CCD)cameras and one Complementary Metal-Oxide-Semiconductor Transistor(CMOS)camera,were *** measured the point spread functions of a mirror at different positions to obtain the axial resolution and the SNR of three OCT systems powered with a light source with a 50 nm bandwidth,centered at a wavelength of 840 *** normal subjects,aged 26 and 47,respectively,and one 75-year-old patient with an intraocular lens implanted were ***:The results indicated that spectrometers using cameras with 4096 camera pixels optimized the axial resolutions,due to the use of the full spectrum provided by the light *** CCD camera system with 4096 pixels had the highest SNR and the best image *** system with the CMOS camera with 4096 pixels had the highest speed but had a compromised SNR compared to the CCD camera with 4096 ***:Using these three OCT systems,we imaged the anterior segment of the human eye before and after accommodation,which showed similar results among the different *** system using the CMOS camera with an ultra-long scan depth,high resolution and high scan speed exhibited the best overall performance and therefore was recommended for imaging real-time accommodation.

As a versatile chiral ammonia equivalent, chiral tert-butylsulfinylamide (6) (TBSA) is a very useful auxiliary due to the characteristics of highly stereoselectivity in asymmetric induction and the sulfinyl group being easily removed compared with other amine auxiliaries since its introduction as a stable compound by Ellman in 1997.[1] Considering the importance of TBSA 6, search for efficient methods for the preparation of enantiopure TBSA (6) is the great interesting topic in synthetic chemistry. Visual measurement of the microscopic temperature of porous graphene based on cholesteric liquid crystal microcapsules

Measuring the microscopic temperature of graphene is *** used cholesteric liquid crystal microcapsules(CLCMs)as temperature sensors to detect the local temperature of three-dimensional porous graphene through quantitative *** on a CLCM(~20μm in size),we determined the temperature variation in a small area with an accuracy of 0.1℃.By analyzing the color changes between two CLCMs,we demonstrated the temperature changes dynamically in a region with a diameter of approximately 110μ***,by comparing the color evolution among the three CLCMs,we visualized the anisotropic thermal properties in the *** convenient and low-cost temperature measurement method is expected to further improve graphene-based devices.
