Role of osteopontin in diet-induced brown gallstone formation in rats

Background:Although osteopontin(OPN)is expressed in the liver and pigment gallstones of patients with hepatolithiasis,its role in pigment gallstone formation remains unclear.This study aimed to explore the function of OPN in pigment gallstone formation.Methods:Rats were fed a chow diet(CD)or lithogenic diet(LD)for 10 consecutive weeks;blocking tests were then performed using an OPN antibody(OPN-Ab).Incidence of gallstones and levels of several bile components,OPN,tumor necrosis factor alpha(TNF-α),and cholesterol 7 alpha-hydroxylase(CYP7A1)were analyzed.To determine TNF-αexpression in hepatic macrophages and both CYP7A1 and bile acid(BA)expression in liver cells,recombinant rat OPN and recombinant rat TNF-αwere used to treat rat hepatic macrophages and rat liver cells,respectively.Chi-square or Fisher exact tests were used to analyze qualitative data,Student t-test or one-way analysis of variance were used to analyze qualitative data.Results:Incidence of gallstones was higher in LD-fed rats than in CD-fed rats(80%vs.10%,P<0.05).BA content significantly decreased in bile(t=-36.08,P<0.01)and liver tissue(t=-16.16,P<0.01)of LD-fed rats.Both hepatic OPN protein expression(t=9.78,P<0.01)and TNF-αlevel(t=8.83,P<0.01)distinctly increased in the LD group;what’s more,CYP7A1 mRNA and protein levels(t=-12.35,P<0.01)were markedly down-regulated in the LD group.Following OPN-Ab pretreatment,gallstone formation decreased(85%vs.25%,χ^(2)=14.55,P<0.01),liver TNF-αexpression(F=20.36,P<0.01)was down-regulated in the LD group,and CYP7A1 expression(F=17.51,P<0.01)was up-regulated.Through CD44 and integrin receptors,OPN promoted TNF-αproduction in macrophage(F=1041,P<0.01),which suppressed CYP7A1 expression(F=48.08,P<0.01)and reduced liver BA synthesis(F=119.4,P<0.01).Conclusions:We provide novel evidence of OPN involvement in pigmented gallstone pathogenesis in rats.

Coherent beam combining of two all‑PM thulium‑doped fiber chirped pulse amplifiers

In this paper,we report a coherent beam combining(CBC)system that involves two thulium-doped all-polarization maintaining(PM)fiber chirped pulse amplifiers.Through phase-locking the two channels via a fiber stretcher by using the stochastic parallel gradient descent(SPGD)algorithm,a maximum average power of 265 W is obtained,with a CBC efficiency of 81%and a residual phase error ofλ/17.After de-chirping by a pair of diffraction gratings,the duration of the combined laser pulse is compressed to 690 fs.Taking into account the compression efficiency of 90%and the main peak energy proportion of 91%,the corresponding peak power is calculated to be 4 MW.The laser noise characteristics before and after CBC are examined,and the results indicate that the CBC would degrade the low frequency relative intensity noise(RIN),of which the integration is 1.74%in[100 Hz,2 MHz]at the maximum combined output power.In addition,the effects of the nonlinear spectrum broadening during chirped pulse amplification on the CBC efficiency are also investigated,showing that a higher extent of pulse stretching is effective in alleviating the spectrum broadening and realizing a higher output power with decent combining efficiency.

Gastric peroral endoscopic myotomy(G-POME)is an emerging minimally invasive endoscopic technique involving the establishment of a submucosal tun-nel around the pyloric sphincter.In 2013,Khashab et al used G-POME for the first time in the treatment of gastroparesis with enhanced therapeutic efficacy,prov-iding a new direction for the treatment of gastroparesis.With the recent and rapid development of G-POME therapy technology,progress has been made in the treatment of gastroparesis and other upper digestive tract diseases,such as congenital hypertrophic pyloric stenosis and gastric sleeve stricture,with G-POME.This article reviews the research progress and future prospects of G-POME for the treatment of upper digestive tract gastrointestinal diseases.

Dynamic Analysis of A Deepwater Drilling Riser with A New Hang-off System

The safety of risers in hang-off states is a vital challenge in offshore oil and gas engineering.A new hang-off system installed on top of risers is proposed for improving the security of risers.This approach leads to a challenging problem:coupling the dynamics of risers with a new hang-off system combined with multiple structures and complex constraints.To accurately analyze the dynamic responses of the coupled system,a coupled dynamic model is established based on the Euler-Bernoulli beam-column theory and penalty function method.A comprehensive analysis method is proposed for coupled dynamic analysis by combining the finite element method and the Newmarkβmethod.An analysis program is also developed in MATLAB for dynamic simulation.The simulation results show that the dynamic performances of the risers at the top part are significantly improved by the new hang-off system,especially the novel design,which includes the centralizer and articulation joint.The bending moment and lateral deformation of the risers at the top part decrease,while the hang-off joint experiences a great bending moment at the bottom of the lateral restraint area which requires particular attention in design and application.The platform navigation speed range under the safety limits of risers expands with the new hang-off system in use.

Cumulative evidence suggests that O-linkedβ-N-acetylglucosaminylation(OGlcNAcylation)plays an important regulatory role in pathophysiological processes.Although the regulatory mechanisms of O-GlcNAcylation in tumors have been gradually elucidated,the potential mechanisms of O-GlcNAcylation in bone metabolism,particularly,in the osteogenic differentiation of bone marrow mesenchymal stromal cells(BMSCs)remains unexplored.In this study,the literature related to O-GlcNAcylation and BMSC osteogenic differentiation was reviewed,assuming that it could trigger more scholars to focus on research related to OGlcNAcylation and bone metabolism and provide insights into the development of novel therapeutic targets for bone metabolism disorders such as osteoporosis.

Benefit of aerosol reduction to winter wheat during China's clean air action:A case study of Henan Province

A strongly declining aerosol radiative effect has been observed in China since 2013 after implementing the clean air action,yet its impact on wheat(Triticum aestivum L.)production remains unclear.We use satellite measures and a biophysical crop model to assess the impact of aerosol-induced radiative perturbations on winter wheat production in the agricultural belt of Henan province from 2013 to 2018.After calibrating parameters with the extended Fourier Amplitude Sensitivity Test(EFAST)and the generalized likelihood uncertainty estimation(GLUE)method,the DSSAT CERES-Wheat model was able to simulate crop biomass and yield more accurately.We found that the aerosol negatively impacted wheat biomass by 21.87%and yield by 22.48%from 2006 to 2018,and the biomass effects from planting to anthesis were more significant compared to anthesis to maturity.Due to the strict clean air action,under all-sky conditions,the surface solar shortwave radiation(SSR)in 2018 increased by about 7.08%over 2006-2013 during the wheat growing seasons.As a result of the improvement of crop photosynthesis,winter wheat biomass and yield increased by an average of 5.46%and 2.9%,respectively.Our findings show that crop carbon uptake and yield will benefit from the clean air action in China,helping to ensure national food and health security.

In winter,wind turbines are susceptible to blade icing,which results in a series of energy losses and safe operation problems.Therefore,blade icing detection has become a top priority.Conventional methods primarily rely on sensor monitoring,which is expensive and has limited applications.Data-driven blade icing detection methods have become feasible with the development of artificial intelligence.However,the data-driven method is plagued by limited training samples and icing samples;therefore,this paper proposes an icing warning strategy based on the combination of feature selection(FS),eXtreme Gradient Boosting(XGBoost)algorithm,and exponentially weighted moving average(EWMA)analysis.In the training phase,FS is performed using correlation analysis to eliminate redundant features,and the XGBoost algorithm is applied to learn the hidden effective information in supervisory control and data acquisition analysis(SCADA)data to build a normal behavior model.In the online monitoring phase,an EWMA analysis is introduced to monitor the abnormal changes in features.A blade icing warning is issued when themonitored features continuously exceed the control limit,and the ambient temperature is below 0℃.This study uses data fromthree icing-affected wind turbines and one normally operating wind turbine for validation.The experimental results reveal that the strategy can promptly predict the icing trend among wind turbines and stably monitor the normally operating wind turbines.

Hierarchical Task Planning for Power Line Flow Regulation

The complexity and uncertainty in power systems cause great challenges to controlling power grids.As a popular data-driven technique,deep reinforcement learning(DRL)attracts attention in the control of power grids.However,DRL has some inherent drawbacks in terms of data efficiency and explainability.This paper presents a novel hierarchical task planning(HTP)approach,bridging planning and DRL,to the task of power line flow regulation.First,we introduce a threelevel task hierarchy to model the task and model the sequence of task units on each level as a task planning-Markov decision processes(TP-MDPs).Second,we model the task as a sequential decision-making problem and introduce a higher planner and a lower planner in HTP to handle different levels of task units.In addition,we introduce a two-layer knowledge graph that can update dynamically during the planning procedure to assist HTP.Experimental results conducted on the IEEE 118-bus and IEEE 300-bus systems demonstrate our HTP approach outperforms proximal policy optimization,a state-of-the-art deep reinforcement learning(DRL)approach,improving efficiency by 26.16%and 6.86%on both systems.

Prognostic significance of grade of malignancy based on histopathological differentiation and Ki-67 in pancreatic ductal adenocarcinoma

Objective:Tumor cell malignancy is indicated by histopathological differentiation and cell proliferation.Ki-67,an indicator of cellular proliferation,has been used for tumor grading and classification in breast cancer and neuroendocrine tumors.However,its prognostic significance in pancreatic ductal adenocarcinoma(PDAC)remains uncertain.Methods:Patients who underwent radical pancreatectomy for PDAC were retrospectively enrolled,and relevant prognostic factors were examined.Grade of malignancy(GOM),a novel index based on histopathological differentiation and Ki-67,is proposed,and its clinical significance was evaluated.Results:The optimal threshold for Ki-67 was determined to be 30%.Patients with a Ki-67 expression level>30%rather than≤30%had significantly shorter 5-year overall survival(OS)and recurrence-free survival(RFS).In multivariate analysis,both histopathological differentiation and Ki-67 were identified as independent prognostic factors for OS and RFS.The GOM was used to independently stratify OS and RFS into 3 tiers,regardless of TNM stage and other established prognostic factors.The tumor-nodemetastasis-GOM stage was used to stratify survival into 5 distinct tiers,and surpassed the predictive performance of TNM stage for OS and RFS.Conclusions:Ki-67 is a valuable prognostic indicator for PDAC.Inclusion of the GOM in the TNM staging system may potentially enhance prognostic accuracy for PDAC.
