Dibutyl phthalate disrupts[Ca^(2+)]_(i),reactive oxygen species,[pH]_(i),protein kinases and mitochondrial activity,impairing sperm function

To explore the mechanism of sperm dysfunction caused by dibutyl phthalate(DBP),the effects of DBP on intracellular[Ca^(2+)]and[pH],reactive oxygen species(ROS),lipid peroxidation(LPO),mitochondrial permeability transition pore(mPTP)opening,mitochondrial membrane potential(MMP),adenosine triphosphate(ATP)levels,phosphorylation of protein kinase A(PKA)substrate proteins and phosphotyrosine(p-Tyr)proteins,sperm motility,spontaneous acrosome reaction,and tail bending were examined in mouse *** 100μg/mL,DBP significantly increased tail bending and[Ca^(2+)]***,DBP showed biphasic effects on[pH]*** at 10–100μg/mL significantly decreased sperm ***,Ca^(2+)ionophore A23187 decreased[pH]_(i)sperm motility,suggesting that DBP-induced excessive[Ca^(2+)]_(i)decreased sperm *** significantly increased ROS and *** at 100μg/mL significantly decreased mPTP closing,MMP,and ATP levels in spermatozoa,as did H2O2,indicative of ROS-mediatedmitochondrial dysfunction caused by *** as well as H2O2 increased p-Tyr sperm proteins and phosphorylated PKA substrate sperm *** at 1–10μg/mL significantly increased the spontaneous acrosome reaction,suggesting that DBP can activate sperm ***,DBP showed a biphasic effect on intracellular signaling in *** concentrations relevant to seminal ortho-phthalate levels,DBP activates[pH]i,protein tyrosine kinases and PKA via physiological levels of ROS generation,potentiating sperm *** at high doses excessively raises[Ca^(2+)]_(i)and ROS and disrupts[pH]i,impairing the mitochondrial function,tail structural integrity,and sperm motility.

Cardiac arrest attributable to dysfunction of the autonomic nervous system after traumatic cervical spinal cord injury

Bradycardia is the most common form of dysrhythmia developing after disruption of the sympathetic pathway by a spinal cord injury （SCI）, and it can have fatal consequences, including cardiac arrest. Here, we report a case of cardiac arrest developing after cervical SCI attributable to sympathetic hypoactivity. A 26-year-old male pedestrian was admitted after a traffic accident. Radiologically, fractures were apparent at the CG 7 bilateral articular facets, and cord contusion with hemorrhage was evident at （24 7. During his stay in ICU, intermittent bradycardia was noted, but the symptoms were not specific. On the 22nd postoperative day, the patient was taken to the computed tomography suite for further evaluation and experienced cardiac arrest during a positional change. After immediate cardiac massage, the patient was resuscitated. We scheduled Holter monitoring, which detected 26 pauses, the longest of which was 17.9 s. The patient underwent cardiac pacemaker insertion. No further cardiac events were noted.

Comprehensive molecular characterization to predict immunotherapy response in advanced biliary tract cancer:a phase II trial of pembrolizumab

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

Machine Learning-Based Detection and Selective Mitigation of Denial-of-Service Attacks in Wireless Sensor Networks

As the density of wireless networks increases globally, the vulnerability of overlapped dense wireless communications to interference by hidden nodes and denial-of-service (DoS) attacks is becoming more apparent. There exists a gap in research on the detection and response to attacks on Medium Access Control (MAC) mechanisms themselves, which would lead to service outages between nodes. Classifying exploitation and deceptive jamming attacks on control mechanisms is particularly challengingdue to their resemblance to normal heavy communication patterns. Accordingly, this paper proposes a machine learning-based selective attack mitigation model that detects DoS attacks on wireless networks by monitoring packet log data. Based on the type of detected attack, it implements effective corresponding mitigation techniques to restore performance to nodes whose availability has been compromised. Experimental results reveal that the accuracy of the proposed model is 14% higher than that of a baseline anomaly detection model. Further, the appropriate mitigation techniques selected by the proposed system based on the attack type improve the average throughput by more than 440% compared to the case without a response.

All-solid-state batteries(ASSBs)are pursued due to their potential for better safety and high energy ***,the energy density of the cathode for ASSBs does not seem to be satisfactory due to the low utilization of active materials(AMs)at high *** small amount of solid electrolyte(SE)powder in the cathode,poor electrochemical performance is often observed due to contact loss and non-homogeneous distribution of AMs and SEs,leading to high tortuosity and limitation of lithium and electron transport ***,we propose a novel cathode design that can achieve high volumetric energy density of 1258 Wh L^(-1)at high AM content of 85 wt%by synergizing the merits of AM@SE core–shell composite particles with conformally coated thin SE shell prepared from mechanofusion process and small SE *** core–shell structure with an intimate and thin SE shell guarantees high ionic conduction pathway while unharming the electronic *** addition,small SE particles play the role of a filler that reduces the packing porosity in the cathode composite electrode as well as between the cathode and the SE separator *** systematic demonstration of the optimization process may provide understanding and guidance on the design of electrodes for ASSBs with high electrode density,capacity,and ultimately energy density.

Evolution of microstructure and tensile properties of cold-drawn hyper-eutectoid steel wires during post-deformation annealing

Manufacturing temperatures of severely cold-drawn hyper-eutectoid steel wires are sufficiently high to influence the mobility of dislocations and alloy elements,thereby affecting the materials’mechanical ***,we describe the evolution of microstructure and tensile strength of the as-drawn 3.45 GPa steel wire during post-deformation annealing for 30 min at 150-450℃.Annealing at 150℃raised the strength to 3.77 GPa by age-hardening through activation of dislocations pinning by carbon,while further temperature rising up to 450℃caused a severe loss of *** was proved that annealing at 300 and 450℃destabilizes the lamellar microstructure,promoting the formation of carbon-deficient(Fe,Mn,Cr)3 C-type cementite particles with preferentially rounded and partially faceted *** at 450℃yielded the accumulation of Mn and Cr at the ferrite/particle interfaces,and their concentrations at the interfaces were dependent on the interface structure;i.e.,lower concentrations at rounded interfaces(formed through capillarity–driven coarsening of the spheroidized cementite),and higher concentrations at faceted interfaces(that are initially existing in the as-drawn state).Our proof-of-principle observations,supported by thermodynamic calculations and kinetic assessments,provide a pathway for understanding the changes in microstructural and tensile properties during manufacturing of the hyper-eutectoid steel wires.

Geometric Properties of Ribs and Fans of a Bézier Curve

Ribs and fans are interesting geometric entities that are derived from a given Bézier curve or surface based on the recent theory of rib and fan decomposition. In this paper, we present some of new geometric properties of ribs and fans for a Bézier curve including composite fans, rib-invariant deformation, and fan-continuity in subdivision. We also give some examples for the presented properties.

Owing to the tremendous demands for high-resolution pixel-scale thin lenses in displays,we developed a graphenebased ultrathin square subpixel lens(USSL)capable of electrically tuneable focusing(ETF)with a performance competitive with that of a typical mechanical refractive *** fringe field due to a voltage bias in the graphene proves that our ETF-USSL can focus light onto a single point regardless of the wavelength of the visible light—by controlling the carriers at the Dirac point using radially patterned graphene layers,the focal length of the planar structure can be adjusted without changing the curvature or position of the lens.A high focusing efficiency of over 60% at a visible wavelength of 405 nm was achieved with a lens thickness of change of 19.42% in the focal length with a 9% increase in transmission was exhibited under a driving *** design is first presented as an ETF-USSL that can be controlled in pixel units of flat panel displays for visible *** can be easily applied as an add-on to high resolution,slim displays and provides a new direction for the application of multifunctional autostereoscopic displays.
