ABA Signaling in Guard Cells Entails a Dynamic Protein-Protein Interaction Relay from the PYL-RCAR Family Receptors to Ion Channels

Plant hormone abscisic acid （ABA） serves as an integrator of environmental stresses such as drought to trig-ger stomatal closure by regulating specific ion channels in guard cells. We previously reported that SLAC1, an outward anion channel required for stomatal closure, was regulated via reversible protein phosphorylation events involving ABA signaling components, including protein phosphatase 2C members and a SnRK2-type kinase （OST1）. In this study, we reconstituted the ABA signaling pathway as a protein-protein interaction relay from the PYL/RCAR-type receptors, to the PP2C-SnRK2 phosphatase-kinase pairs, to the ion channel SLAC1. The ABA receptors interacted with and inhibited PP2C phosphatase activity against the SnRK2-type kinase, releasing active SnRK2 kinase to phosphorylate, and activate the SLAC1 channel, leading to reduced guard cell turgor and stomatal closure. Both yeast two-hybrid and bimolecular fluorescence complementation assays were used to verify the interactions among the components in the pathway. These biochemical assays demonstrated activity modifications of phosphatases and kinases by their interaction partners. The SLAC1 channel activity was used as an endpoint readout for the strength of the signaling pathway, depending on the presence of different combinations of signaling components. Further study using transgenic plants overexpressing one of the ABA receptors demonstrated that changing the relative level of interacting partners would change ABA sensitivity.

Optimization of post-warranty sequential inspection for second-hand products

This paper considers an optimal sequential inspection schedule for a second-hand product after that the free nonrenewable warranty is expired. The length of warranty is prespecified and during the warranty period, the product is minimally repaired by the dealer when it fails. Following the expiration of the non-renewing warranty, the product is inspected and upgraded sequentially a fixed number of times at the expenses of the *** each inspection, the failure rate of the product is reduced proportionally so that the product is upgraded. The product is assumed to deteriorate as it ages and the replacement of the product occurs when a fixed number of inspections are rendered. In addition,the intervals between two successive inspections are assumed to decrease monotonically. The main objective of this paper is to determine the optimal improvement level to upgrade the product at each inspection so that the expected maintenance cost during the life cycle of the product is minimized from the perspective of the customer. Under the given cost structures, we derive an explicit formula to obtain the expected maintenance cost incurred during the life cycle of the product and discuss the method to find the optimal level of the improvement analytically in case the failure times follow the Weibull distribution. Numerical results are analyzed to observe the impact of relevant parameters on the optimal solution.

Newcastle Disease in Chickens of Smallholder Farmers at Krang Yov Commune S’ang District,Kandal Province

The study was conducted in Krang Yov commune,S’ang district,Kandal province and was conducted in three villages,including Kampong Po,Andong and Svay Damnak *** the total,88 families were selected for interview by using snowball technic of nonprobability *** to the results of the interviews,it showed that the number of farmers who raised chickens and had participated in training was very low number,ranging from 10.71%to 34.38%.For chicken raising system,the free ranging was the most popular by allowing the chicken to scavenge around the village,however,the total confinement was also *** local breed was more popular and the purpose of raising was for selling meat,however there were few households that raised the fighting cock for *** number of chickens owned per household,was mostly less than 20 birds,while,the average number of birds per household ranged from 16.82 birds to 37.66 ***,the average number of birds per household was significant different between the households who have participated in chicken raising training and those who never participated(phose three villages used the concentration feed to offer the chicken,especially for supplementation,but,the whole rice was also used to supplement *** vaccination for chicken was significant different among the villages(phousehold in Kampong Po village had highest percentage of vaccinating their chicken,40.63%,than those in Svay Damnak and Andong *** Newcastle vaccination was the more popular in those ***,all of the households have experienced with the disease,especially ND(Newcastle Disease)accounting for 67.05%,followed by influenza and fowl *** chicken got sick,most of farmers had treated their chicken with different methods,especially buying the medicine from the store and treatment by themselves were mostly applicabl

Inhibitory effects of Cnidium monnieri fruit extract on pulmonary inflammation in mice induced by cigarette smoke condensate and lipopolysaccharide

The aim of this study was to investigate the inhibitory effect of Cnidium monnieri fruit(CM) extracts on pulmonary inflammation induced in mice by cigarette smoke condensate(CSC) and lipopolysaccharide(LPS). Pulmonary inflammation was induced by intratracheal instillation of LPS and CSC five times within 12 days. CM extract was administered orally at a dose of 50 or 200 mg·kg-1. The number of inflammatory cells in the bronchoalveolar lavage fluid was counted using a fluorescence activated cell sorter. Inflammatory mediator levels were determined by enzyme-linked immunosorbent assay. The administration of LPS and CSC exacerbated airway hyper-responsiveness(AHR) and induced an accumulation of inflammatory cells and mediators, and led to histological changes. however, these responses are modulated by treatment with CM, and the treatment with CM extract produces similar or more extensive results than the treatment with cyclosporin A(CSA). CM extract may have an inhibitory effect on pulmonary inflammation related with chronic obstructive pulmonary disease.

Validation of four Helicobacter pylori rapid blood tests in a multi-ethnic Asian population

AIM: To validate the accuracy of four rapid blood tests in the diagnosis of Helicobacter ***: Consecutive dyspeptic patients scheduled for endoscopy at the National University hospital,Singapore, were interviewed and had blood drawn for serology. The first 109 patients were tested with BM-test (BM), Pyloriset Screen (PS) and QuickVue (QV), and the next 99 subjects were tested with PS and Unigold (UG).Endoscopies were performed blinded to rapid blood test results and biopsies were taken for culture and rapid urease test. Urea breath tests were performed after endoscopies. The rapid blood test results were compared with four reference tests (rapid urease test, culture,serology, and breath test).RESULTS: The study population composed of 208patients (mean age 43.1 years; range 18-73 years; 119males; 174 Chinese). The number of evaluable patientsfor BM, QV, UG and PS were 102, 102, 95, and 197,respectively. The sensitivity and specificity, respectively were: PS 80.2%, 95.8%; UG 55.9%, 100%; QV 43.3%,100%; BM 67.2%, 97.1%.CONCLUSION: The rapid blood test kits showed high specificity and positive predictive value (97-100%), while sensitivity and negative predictive value ranged widely (43%-80% and 47%-73%, respectively). Among test kits, PS showed the best sensitivity (80%), best negative predictive value (73%) and best negative likelihood ratio (0.207). PS had a specificity of 96%, positive predictive value of 97% and positive likelihood ratio of 19.1.

Bidirectional quantum teleportation of unknown photons using path-polarization intra-particle hybrid entanglement and controlled-unitary gates via cross-Kerr nonlinearity

We propose an arbitrary controlled-unitary （CU） gate and a bidirectional quantum teleportation （BQTP） scheme. The proposed CU gate utilizes photonic qubits （photons） with cross-Kerr nonlinearities （XKNLs）, X-homodyne detectors, and linear optical elements, and consists of the consecutive operation of a controlled-path （C-path） gate and a gathering-path （G- path） gate. It is almost deterministic and feasible with current technology when a strong coherent state and weak XKNLs are employed. Based on the CU gate, we present a BQTP scheme that simultaneously teleports two unknown photons between distant users by transmitting only one photon in a path-polarization intra-particle hybrid entangled state. Consequently, it is possible to experimentally implement BQTP with a certain success probability using the proposed CU gate.

Appendicitis-mimicking presentation in fishbone induced microperforation of the distal duodenum:A case report

BACKGROUND Upper gastrointestinal fishbone microperforations are rare and not commonly reported in medical *** the increasing use of computer tomography(CT)imaging and the employment of the Alvardo criteria,misdiagnosis of acute appendicitis can still *** report the rare case of an elderly Chinese gentleman who had a fish-bone induced microperforation of the duodenum that closely mimicked the symptoms of acute *** SUMMARY This 79-year-old man presented with migratory lower abdominal pain that localized at his periumbilical region and right lower *** had associated pyrexia,general malaise and was noted to have an elevated white cell *** investigations initially revealed a distended appendix which was resected laparoscopically but showed no obvious signs of gross *** patient then deteriorated clinically and had increased oxygen requirements immediately after the *** prompted further investigations.A further review of his CT scan revealed a fine fishbone microperforation in the distal duodenum associated with retroperitoneal abscess formation and seepage extending into the right lower *** was then started on broad spectrum intravenous antibiotics and subsequently underwent a laparotomy 12 h later to manage the obscure aetiology and to drain the *** post-operative course was uneventful and he was discharged 11 d later including a 2-d stay in the intensive care *** This case offers an insight into a potential mimic of acute appendicitis and the diagnostic difficulties experienced in such presentations.
