Identification and Molecular Mapping of the Rs Dm R Locus Conferring Resistance to Downy Mildew at Seedling Stage in Radish(Raphanus sativus L.)

Downy mildew （DM）, caused by the fungus Peronospora parasitica, is a destructive disease of radish （Raphanus sativus L.） worldwide. Host resistance has been considered as an attractive and environmentally friendly approach to control the disease. However, the genetic mechanisms of resistance in radish to the pathogen remain unknown. To determine the inheritance of resistance to DM, F1, F2 and BC1F1 populations derived from reciprocal crosses between a resistant line NAU-dhp08 and a susceptible line NAU-qtbjq-06 were evaluated for their responses to DM at seedling stage. All F1 hybrid plants showed high resistance to DM and maternal effect was not detected. The segregation for resistant to susceptible individuals statistically iftted a 3：1 ratio in two F2 populations （F2（SR） and F2（RS））, and 1：1 ratio in two BC1F1 populations, indicating that resistance to DM at seedling stage in radish was controlled by a single dominant locus designated as RsDmR. A total of 1 972 primer pairs （1 036 SRAP, 628 RAPD, 126 RGA, 110 EST-SSR and 72 ISSR） were screened, and 36 were polymorphic between the resistant and susceptible bulks, and consequently used for genotyping individuals in the F2 population. Three markers （Em9/ga24370, NAUISSR826700 and Me7/em10400） linked to the RsDmR locus within a 10.0 cM distance were identiifed using bulked segregant analysis （BSA）. The SRAP marker Em9/ga24370 was the most tightly linked one with a distance of 2.3 cM to RsDmR. These markers tightly linked to the RsDmR locus would facilitate marker-assisted selection and resistance gene pyramiding in radish breeding programs.

Climate change and rapid urbanization are pressing environmental and social concerns, with approximately 56% of the global population living in urban areas. This number is expected to rise to 68% by 2050, leading to the expansion of cities and encroachment upon natural areas, including wetlands, causing their degradation and fragmentation. To mitigate these challenges, green and blue infrastructures (GBIs), such as constructed wetlands, have been proposed to emulate and replace the functions of natural wetlands. This study evaluates the potential of eight constructed wetlands near Beijing, China, focusing on their ecosystem services (ESs), cost savings related to human health, growing/maintenance expenses, and disservices using an emergy-based assessment procedure. The results indicate that all constructed wetlands effectively purify wastewater, reducing nutrient concentrations (e.g., total nitrogen, total phosphorus, and total suspended solids). Among the studied wetlands, the integrated vertical subsurface flow constructed wetland (CW-4) demonstrates the highest wastewater purification capability (1.63E+14 sej/m^(2)/yr) compared to other types (6.78E+13 and 2.08E+13 sej/m^(2)/yr). Additionally, constructed wetlands contribute to flood mitigation, groundwater recharge, wildlife habitat protection, and carbon sequestration, resembling the functions of natural wetlands. However, the implementation of constructed wetlands in cities is not without challenges, including greenhouse gas emissions, green waste management, mosquito issues, and disturbances in the surrounding urban areas, negatively impacting residents. The ternary phase diagram reveals that all constructed wetlands provide more benefits than costs and impacts. Sequential carbohydrate synthesis is important for plant survival because it guarantees energy supplies for growth and development during plant ontogeny and reproduction. Starch and fructan are two important carbohydrates in many flowering plants and in human diets. Understanding this coordinated starch and fructan synthesis and unraveling how plants allocate photosynthates and prioritize different carbohydrate synthesis for survival could lead to improvements to cereals in agriculture for the purposes of greater food security and production quality. Here, we report a system from a single gene in barley employing two alternative promoters, one intronic/exonic, to generate two sequence-overlapping but functionally opposing transcription factors, in sensing sucrose, potentially via sucrose/gluco lose 6-phosphate signaling. The system employs an autoregulatory mechanism in perceiving a sucrose- controlled trans activity on one promoter and orchestrating the coordinated starch and fructan synthesis by competitive transcription factor binding on the other promoter. As a case in point for the physiological roles of the system, we have demonstrated that this multitasking system can be exploited in breeding barley with tailored amounts of fructan to produce healthy food ingredients. The identification of an intron/exon-spanning promoter in a hosting gene, resulting in proteins with distinct functions, adds to the complexity of plant genomes.
