Responses to oral glucose challenge differ by physical activity volume and intensity: A pilot study

Background:One-hour postprandial hyperglycemia is associated with increased risk of type 2 diabetes and cardiovascular *** activity(PA)has short-term beneficial effects on post-meal glucose *** study compared the oral glucose tolerance test results of 3 groups of people with habitually different levels of ***:Thirty-one adults without diabetes(age 25.9±6.6 years;body mass index 23.8±3.8 kg/m^2;mean±SD)were recruited and divided into 3 groups based on self-reported PA volume and intensity:low activityn/day of moderate-intensity activity(n=11),moderately active≥30 min/day of moderate-intensity PA(n=10),and very active≥60 min/day of PA at high intensity(n=10).Participants completed an oral glucose tolerance test(50 g glucose)with capillary blood samples obtained at baseline,15 min,30 min,45 min,60 min,90 min,and 120 min ***:There were no significant differences between groups for age or body fat percentage or glycated hemoglobin(p>0.05).The groups were significantly different in terms of baseline glucose level(p=0.003)and,marginally,for gender(p=0.053)and BMI(p=0.050).There was a statistically significant effect of PA on the 1-h postprandial glucose results(p=0.029),with differences between very active and low activity groups(p=0.008)but not between the moderately active and low activity groups(p=0.360),even when baseline glucose level and gender differences were accounted *** incremental area under the curve there was no significant effect of activity group once gender and body fat percentage had been accounted for(p=0.401).Those in the low activity group took 15 min longer to reach peak glucose level than those in the very active group(p=0.012).Conclusion:The results suggest that high levels of PA have a beneficial effect on postprandial blood glucose profiles when compared to low and moderate levels of activity.

Sensitivity, repeatability and reproducibility study with a leg prototype of arecently developed knee arthrometer: The DYnEELAX®

Sagittal knee laxity is often quantified using arthrometers. Adding rotational laxity and compliance measurementsis relatively new and could bring new information for anterior cruciate ligament (ACL) injury diagnosis. TheDYnEELAX® is a new knee arthrometer able to evaluate simultaneously tibial translation and rotation. Thepurpose is to assess the sensitivity, reproducibility and repeatability of the DYnEELAX® with a prototype leg toprovide accurate instructions before using it in clinical practice. Sensitivity is studied by varying 7 parameters(positioning and sensors), and reproducibility by repeating measurement series with a group of two experiencedoperators and a group of two non-experienced operators. Repeatability is assessed throughout the study. Theresults showed that DYnEELAX® is poorly sensitive to the angle and the position of the displacement sensor, andthe angle of the rotation sensor. It is a bit sensitive to positioning of patella and ankle supports. It is very sensitiveto the tightening of the patella and ankle supports. It exists a significant difference only between groups (p no significant difference between the two experienced operators (p > 0.215), or between thetwo non-experienced operators (p > 0.229). Variation coefficients for intra-series are on average inferior to 5% fortranslation and rotation tests. Then, the DYnEELAX® presented encouraging results with a good accuracy and agood reliability but operators must be careful about positioning.

Plant Cell Wall Proteomics： Mass Spectrometry Data, a Trove for Research on Protein Structure/Function Relationships

Proteomics allows the large-scale study of protein expression either in whole organisms or in purified organelles. In particular, mass spectrometry （MS） analysis of gel-separated proteins produces data not only for protein identification, but for protein structure, location, and processing as well. An in-depth analysis was performed on MS data from etiolated hypocotyl cell wall proteomics ofArabidopsis thaliana. These analyses show that highly homologous members of multigene families can be differentiated. Two lectins presenting 93% amino acid identity were identified using peptide mass fingerprinting. Although the identification of structural proteins such as extensins or hydroxyproline/proline-rich proteins （H/PRPs） is arduous, different types of MS spectra were exploited to identify and characterize an H/PRP. Maturation events in a couple of cell wall proteins （CWPs） were analyzed using site mapping. n-glycosylation of CWPs as well as the hydroxylation or oxidation of amino acids were also explored, adding information to improve our understanding of CWP structure/function relationships. A bioinformatic tool was developed to locate by means of MS the n-terminus of mature secreted proteins and n-glycosylation.

Determination of 3-MeO-PCP in human blood and urine in a fatal intoxication case,with a specific focus on metabolites identification

3-Methoxyphencyclidine(3-MeO-PCP)is a new psychoactive substance that belongs to the phencyclidines family,first identified in Europe in *** drug presents a stronger binding to n-methyl-D-aspartate(nMDA)receptors when compared to phencyclidine,which results in more potent effects,even at low *** few articles have been published regarding 3-MeO-PCP in forensic *** this paper,the authors present a fatal 3-MeO-PCP intoxication *** addition to the detection of the parent drug,metabolites were investigated in urine and,for the first time in the scientific literature,in blood.3-MeO-PCP and its metabolites were quantitated by liquid chromatography-tandem mass spectrometry system(LC-MS/MS).Identification was confirmed by liquid chromatography-high resolution mass spectrometry(LC-HRMS).3-MeO-PCP tested positive in femoral blood(3525 ng/mL)and urine(7384 ng/mL).The femoral blood concentration was higher than the fatal concentrations range already reported in the literature(from 50 to 3200 ng/mL).3-MeO-PCP metabolites,including O-demethyl-3-MeO-PCP,piperidine-OH-3-MeO-PCP,O-demethyl-piperidine-di-OH-3-MeO-PCP and piperidine-di-OH-3-MeO-PCP,were detected in *** addition,two new metabolites,O-demethyl-piperidine-OH-3-MeO-PCP and O-demethyl-cyclohexyl-OH,were identified in both blood and ***,due to the lack of reference material on the market,it was not possible to measure the concentration of these ***,the ratios between the metabolites and the parent drug were useful to estimate their analytical response and *** this time,considering the low ratios(n metabolites and parent drug,metabolites testing does not seem useful to increase the detection window of the drug.

The SlSHn2 transcription factor contributes to cuticle formation and epidermal patterning in tomato fruit

The use of non-noble nickel-based catalysts for low temperature CO methanation has been a challenge in recent ***,MgAl layered double oxides sample with high dispersion synthesized by a facile n-(2-Hydroxyethyl)ethylenediaminetriacetic acid assisted wetness impregnation approach,demonstrates much superior catalytic activity and exceptional stability for CO methanation in comparison with the classical ni/MgAl-LDO catalyst prepared by the ordinary wetness impregnation *** results showed that n-(2-Hydroxyethyl)ethylenediaminetriacetic acid played a positive role in the dispersion of ni,as well as ni-support ***-dispersed ni particles with a size of about 5 nm were formed in the presence of n-(2-Hydroxyethyl)ethylenediaminetriacetic *** to the ni/MgAl-LDO prepared by conventional impregnation method,the nH-ni/MgAl-LDO exhibited superior catalytic performance,especially excellent thermal *** nH-ni30/MgAl-LDO catalyst was found to keep a 70%CO conversion even at 160◦C which demonstrates its good low temperature *** the in situ FTIR observations,this good performance at low temperatures may be linked to the delocalization of electrons around CO caused by surface hydroxyl groups.

Outcomes in randomized controlled trials of exercise interventions in solid organ transplant

AIM To identify the outcome measures that have been used in randomized controlled trials(RCTs) of exercise training in solid organ transplant(SOT) recipients and to link these outcomes to the International Classification of Functioning, Disability and Health(ICF) *** Electronic literature searches of MEDLInE, EMBASE, CInAHL, Cochrane, Scopus, and Web of Science were performed. We sought RCTs that investigated the effect of exercise training in SOT recipients. Reference lists of all eligible publications were searched for other appropriate studies not identified by the electronic search. A complete list of outcome measures used in the RCTs was generated and each of these was linked to an ICF *** Four hundred and thirteen articles were retrieved, of which 35 met our inclusion criteria. The studies included were designed to compare the effects of exercise training programs to usual care or to another exercise training program and reported on recipients of heart(n = 21), kidney(n = 9), lung(n = 3) or liver(n = 2) transplant. Of the 126 outcome measures identified, 62 were used as primary outcome measures. The most commonly occurring primary outcomes were aerobic capacity using the peak VO2(n = 13), quality of life using the shortform-36(n = 8), and muscle strength(n = 7). Theseoutcome measures were linked to 113 ICF categories and the majority of outcomes fall into the body function domain(n = 93). COnCLUSIOn There is little standardization in outcome measures used in RCTs of exercise interventions in SOT recipients. The ICF framework can be used to select a core set of outcomes that cross all domains of ICF and that would be appropriate to all SOT recipients.

A novel Process Using Ion Exchange Resins for the Coproduction of Ethyl and Butyl Acetates

Before proposing an innovative process for the coproduction of ethyl and butyl acetates, the individual syntheses of ethyl acetate and butyl acetate by two different routes were first studied. These syntheses involved the reaction of ethanol or n-butanol with acetic acid or acetic anhydride in the presence of ion exchange resins: Amberlyst 15, Amberlyst 16, Amberlyst 36 and Dowex 50WX8. Kinetic and thermodynamic studies were performed with all resins. The lowest activation energy (Ea) value was obtained with Dowex 50WX8, which was identified as the best-performing resin, able to be reused at least in four runs without regeneration. The presence of water-azeotropes during the synthesis of ethyl acetate makes its purification difficult. A new strategy was adopted here, involving the use of ethanol and acetic anhydride as the starting material. In order to minimize acetic acid as co-product of this reaction, a novel two-step process for the coproduction of ethyl and butyl acetates was developed. The first step involves the production of ethyl acetate and its purification. Butyl acetate was produced in the second step: n-butanol was added to the mixture of acetic acid and the resin remaining after the first-step distillation. This process yields ethyl acetate and butyl acetate at high purity and shows an environmental benefit over the independent syntheses by green metrics calculation and life cycle assessment.
