Towards predicting biochar impacts on plant-available soil nitrogen content

Biochars can improve soil health but have been widely shown to reduce plant-available nitrogen(N)owing to their high carbon(C)content,which stimulates microbial N-immobilization.However,because biochars contain large amounts of C that are not microbially available,their total elemental C:N ratio does not correspond well with impacts on soil N.We hypothesized that impacts on soil plant-available N would relate to biochar mineralizable-C(C_(min))content,and that C:N ratios of the mineralizable biochar component could provide a means for predicting conditions of net soil N-mineralization or-immobilization.We conducted two laboratory experiments,the first measuring biochar C_(min)from respiration of isotopically labeled barley biochars manufactured at 300,500,and 750℃,and the second characterizing C_(min)by proxy measurements for ten biochars from six feedstocks at several temperatures.For both experiments,soils were incubated with 2%biochar by mass to determine impacts to soil N-mineralization.Contrary to expectation,all the biochars increased soil N-mineralization relative to unamended soils.Also unexpected,higher temperature(500 and 700℃)barley biochars with less C_(min)stimulated more soil decomposition and more soil N-mineralization than a 350℃barley biochar.However,across diverse biochar feedstocks and production methods,none of the biochar characteristics correlated with soil N-mineralization.The finding of improved soil N-mineralization adds complexity to the range of soil N responses that can be expected in response to biochar amendment.Because of the limited ability to predict soil N responses from biochar properties,users should monitor soil N to manage soil fertility.

Applicability of five models to simulate water infiltration into soil with added biochar

As a soil amendment, biochar can reduce soil bulk density, increase soil porosity, and alter soil aggregates and thus affect the infiltration. Researchers have proposed and revised several theoretical models to describe the process of soil infiltration. Although these models have been successfully used to evaluate the soil infiltration in different scenarios in agricultural fields, little effort has been devoted to assess their performances in arid and semi-arid soils after the addition of biochar. A laboratory experiment was performed to study the infiltration characteristics of two typical Loess Plateau soils at three particle sizes（2–1, 1–0.25, and 〈0.25 mm） and five biochar application amounts（0, 10, 50, 100, and 150 g/kg）. The performance of five models（i.e., the Philip model, Kostiakov model, Mezencev model, USDA-NRCS model, and Horton model） in simulating the infiltration process was then evaluated based on the adjusted coefficient of determination and a reduced Chi-Square test. Results indicated that the Horton model best simulated the water-infiltration process in an aeolian sandy soil with added biochar. However, the Mezencev model best simulated the infiltration process in a loamy clay soil（eum-Orthic Anthrosol）. The three-parameter model, i.e., Mezencev and Horton models can better describe the relationship between cumulative infiltration and infiltration time. In conclusion, biochar reduced the soil infiltration capacity of the aeolian sandy soil and increased that of the eum-Orthic Anthrosol.

Muscle energy technique for chronic obstructive pulmonary disease:A feasibility study

Objective:This study aimed to investigate the feasibility of implementing a manual therapy technique(muscle energy technique,MeT)protocol in a hospital pulmonary rehabilitation(PR)program for patients with moderate to severe chronic obstructive pulmonary disease(COPD).Methods:Participants aged 40 years and over,with moderate to severe COPD,were recruited into this 12-week study.The primary outcome measures were feasibility(acceptability of the intervention and attendance/adherence to the trial)and safety(adverse events,Aes).All participants received the MeT and PR therapies.Participants and assessors were unblinded.Semi-standardized MeT was delivered on 6 occasions(a maximum of once per week)at the hospital directly before a PR session.Participants undertook PR sessions as per the hospital program at a frequency of two days per week for 8 weeks.Participants were contacted 4 weeks after their final MeT treatment via a telephone call to assess acceptability of the intervention.Results:Thirty-three participants were enrolled,with a median age of 74 years(range 45-89 years).The median number of MeT sessions that participants attended was 5(range O-6)out of a possible 6 sessions(83%attendance).At follow-up,participants overwhelmingly enjoyed the MeT treatment with some subjectively reporting improved breathing.There were no major Aes related to the intervention,with the majority of Aes classified as expected events related to COPD exacerbations.Conclusion:It is feasible to implement a manual therapy protocol using MeT as an adjunct to PR in a hospital setting.Recruitment rates were satisfactory and there were no Aes related to the MeT component of the intervention.

CRISPR/Cas-mediated genome editing in human pluripotent stem cells(hPSCs)offers unprecedented opportunities for developing in vitro disease modeling,drug screening and cell-based therapies.To efficiently deliver the CRISPR components,here we developed two all-in-one vectors containing Cas9/gRNA and inducible Cas13d/gRNA cassettes for robust genome editing and RNA interference respectively.These vectors utilized the PiggyBac transposon system,which allows stable expression of CRISPR components in hPSCs.The Cas9 vector PB-CRISPR exhibited high efficiency(up to 99%)of inducing gene knockout in both protein-coding genes and long non-coding RNAs.The other inducible Cas13d vector achieved extremely high efficiency in RNA knockdown(98%knockdown for CD90)with optimized gRNA designs.Taken together,our PiggyBac CRISPR vectors can serve as powerful toolkits for studying gene functions in hPSCs.

The majority of patients with congenitally corrected transposition of the great arteries,also known as transposition of the great arteries{S,L,L}have ventricular septal defects(VSD),most commonly perimembranous VSD(pmVSD).Transcatheter device closure of pmVSD in these patients has not been widely described.We present a case of device closure of pmVSD in L-TGA with an Amplatzer Duct Occluder II(ADOII)device using a deployment starting in the subpulmonary left ventricle.The case demonstrates some of the technical advantages of the ADOII device for VSD closure,specifically its low profile,symmetric shape,and soft material.These characteristics are advantageous in closing conventional pmVSD but are especially useful in patients with challenging anatomic substrates.

BACKGROUND Diabetes rates among pregnant women in the United States have been increasing and are associated with adverse pregnancy outcomes.AIM To investigate differences in birth outcomes(preterm birth,macrosomia,and neonatal death)by diabetes status.MeTHODS Cross-sectional design,using linked Missouri birth and death certificates(singleton births only),2010 to 2012(n=204057).exposure was diabetes non-diabetic,pre-pregnancy diabetes-insulin dependent(PD-I),pre-pregnancy diabetes-non-insulin dependent(PD-NI),gestational diabetes-insulin dependent(GD-I),and gestational diabetes-non-insulin dependent(GD-NI).Outcomes included preterm birth,macrosomia,and infant mortality.Confounders included demographic characteristics,adequacy of prenatal care,body mass index,smoking,hypertension,and previous preterm birth.Bivariate and multivariate logistic regression assessed differences in outcomes by diabetes status.ReSULTS Women with PD-I,PD-NI,and GD-I remained at a significantly increased odds for preterm birth(aOR 2.87,aOR 1.77,and aOR 1.73,respectively)and having a very large baby[macrosomia](aOR 3.01,aOR 2.12,and aOR 1.96,respectively);in reference to non-diabetic women.Women with GDNI were at a significantly increased risk for macrosomia(aOR1.53),decreased risk for their baby to die before their first birthday(aOR 0.41)and no difference in risk for preterm birth in reference to non-diabetic women.CONCLUSION Diabetes is associated with the poor birth outcomes.Clinical management of diabetes during pregnancy and healthy lifestyle behaviors before pregnancy can reduce the risk for diabetes and poor birth outcomes.

This study aims to investigate the ability of an imidazolium biobased Zwitterionic Ionic Liquids(ZILs)in enhancing the phytoavailability of copper from garden(G)and vineyard(V)soils using the model plant ryegrass.Uncontaminated and artificially contaminated CuSO_(4) soils,unamended and ZIL-amended soil modalities were designed.The copper/ZIL molar ratio(1/4)introduced was rationally established based on molecular modeling and on the maximal copper concentration in artificially contaminated soil.Higher accumulation of copper in the shoots was detected for the uncontaminated and copper contaminated ZIL amended V soils(18.9 and 23.3 mg/kg,respectively)contrary to G soils together with a ZIL concentration of around 3%(W/W)detected by LC-MS analyses.These data evidenced a Cu-accumulation improvement of 38%and 66%compared to non-amended V soils(13.6 and 13.9 mg/kg respectively).ZIL would be mainly present under Cu(II)-ZIL_(4) complexes in the shoots.The impact on the chemical composition of shoot was also studied.The results show that depending on the soils modalitity,the presence of free copper and/or ZIL led to different chemical compositions in lignin and monomeric sugar contents.In the biorefinery context,performances of enzymatic hydrolysis of shoots were also related to the presence of both ZIL and copper under free or complex forms.ecotoxicity assessment of the vineyard soil samples indicated that the quantity of copper and ZIL remaining in the soils had no significant toxicity.ZIL amendment in a copper-contaminated soil was demonstrated as being a promising way to promote the valorization of phytoremediation plants.

Global Transcriptional Profile of Tranosema rostrale Ichnovirus Genes in Infected Lepidopteran Hosts and Wasp Ovaries

The ichnovirus TrIV, transmitted by the endoparasitic wasp Tranosema rostrale to its lepidopteran host during oviposition, replicates asymptomatically in wasp ovaries and causes physiological dysfunctions in parasitized caterpillars. The need to identify ichnoviral genes responsible for disturbances induced in lepidopteran hosts has provided the impetus for the sequencing and annotation of ichnovirus genomes, including that of TrIV. In the latter, 86 putative genes were identified, including 35 that could be assigned to recognized ichnoviral gene families. With the aim of assessing the relative importance of each TrIV gene, as inferred from its level of expression, and evaluating the accuracy of the gene predictions made during genome annotation, the present study builds on an earlier qPCR quantification of transcript abundance of TrIV rep ORFs, in both lepidopteran and wasp hosts, extending it to other gene families as well as to a sample of unassigned ORFs. We show that the majority (91%) of putative ORFs assigned to known gene families are expressed in infected larvae, while this proportion is lower (67%) for a sample taken among the remaining ORFs. Selected members of the TrV and rep gene families are shown to be transcribed in infected larvae at much higher levels than genes from any other TrIV gene family, pointing to their likely involvement in host subjugation. In wasp ovaries, the transcriptional profile is dominated by a rep gene and a member of a newly described gene family encoding secreted proteins displaying a novel cysteine motif, which we identified among previously unassigned ORFs.

Challenges for cysteamine stabilization,quantification,and biological effects improvement

The aminothiol cysteamine,derived from coenzyme A degradation in mammalian cells,presents several biological applications.However,the bitter taste and sickening odor,chemical instability,hygroscopicity,and poor pharmacokinetic profile of cysteamine limit its efficacy.The use of encapsulation systems is a good methodology to overcome these undesirable properties and improve the pharmacokinetic behavior of cysteamine.Besides,the conjugation of cysteamine to the surface of nanoparticles is generally proposed to improve the intra-oral delivery of cyclodextrin-drug inclusion complexes,as well as to enhance the colorimetric detection of compounds by a gold nanoparticle aggregation method.On the other hand,the detection and quantification of cysteamine is a challenging mission due to the lack of a chromophore in its structure and its susceptibility to oxidation before or during the analysis.Derivatization agents are therefore applied for the quantification of this molecule.To our knowledge,the derivatization techniques and the encapsulation systems used for cysteamine delivery were not reviewed previously.Thus,this review aims to compile all the data on these methods as well as to provide an overview of the various biological applications of cysteamine focusing on its skin application.

Long-term efficacy of a computer-tailored physical activity intervention for prostate and colorectal cancer patients and survivors:A randomized controlled trial

Background Physical activity(PA)can improve the physical and psychological health of prostate and colorectal cancer survivors,but PA behavior change maintenance is necessary for long-term health benefits.OncoActive is a print-and web-based intervention in which prostate and colorectal cancer patients and survivors receive automatically generated,personalized feedback aimed at integrating PA into daily life to increase and maintain PA.We evaluated the long-term outcomes of OncoActive by examining the 12-month follow-up differences between OncoActive and a control group,and we explored whether PA was maintained during a 6-month non-intervention follow-up period.Methods Prostate or colorectal cancer patients were randomly assigned to an OncoActive(n=249)or a usual care waitlist control group(n=229).OncoActive participants received PA advice and a pedometer.PA outcomes(i.e.,ActiGraph and self-report moderate-to-vigorous intensity PA(MVPA)min/week and days with≥30 min PA)and health-related outcomes(i.e.,fatigue,depression,physical functioning)were assessed at baseline,6 months,and 12 months.Differences between groups and changes over time were assessed with multilevel linear regressions for the primary outcome(ActiGraph MVPA min/week)and all additional outcomes.Results At 12 months,OncoActive participants did not perform better than control group participants at ActiGraph MVPA min/week,self-report MVPA min/week,or ActiGraph days with PA.Only self-report days with PA were significantly higher in OncoActive compared to the control group.For health-related outcomes only long-term fatigue was significantly lower in OncoActive.When exploratively examining PA within OncoActive,the previously found PA effects at the end of the intervention(6 months follow-up)were maintained at 12 months.Furthermore,all PA outcomes improved significantly from baseline to 12 months.The control group showed small but non-significant improvements from 6 months to 12 months(and from baseline to 12 months),resulting in a decline of differences bet
