Phosphorylated tau as a toxic agent in synaptic mitochondria: implications in aging and Alzheimer’s disease

During normal aging,there is a decline in all physiological functions in the organism.One of the most affected organs is the brain,where neurons lose their proper synaptic function leading to cognitive impairment.Aging is one of the main risk factors for the development of neurodegenerative diseases,such as Alzheimer’s disease.One of the main responsible factors for synaptic dysfunction in aging and neurodegenerative diseases is the accumulation of abnormal proteins forming aggregates.The most studied brain aggregates are the senile plaques,formed by Aβpeptide;however,the aggregates formed by phosphorylated tau protein have gained relevance in the last years by their toxicity.It is reported that neurons undergo severe mitochondrial dysfunction with age,with a decrease in adenosine 5′-triphosphate production,loss of the mitochondrial membrane potential,redox imbalance,impaired mitophagy,and loss of calcium buffer capacity.Interestingly,abnormal tau protein interacts with several mitochondrial proteins,suggesting that it could induce mitochondrial dysfunction.Nevertheless,whether tau-mediated mitochondrial dysfunction occurs indirectly or directly is still unknown.A recent study of our laboratory shows that phosphorylated tau at Ser396/404(known as PHF-1),an epitope commonly related to pathology,accumulates inside mitochondria during normal aging.This accumulation occurs preferentially in synaptic mitochondria,which suggests that it may contribute to the synaptic failure and cognitive impairment seen in aged individuals.Here,we review the main tau modifications promoting mitochondrial dysfunction,and the possible mechanism involved.Also,we discuss the evidence that supports the possibility that phosphorylated tau accumulation in synaptic mitochondria promotes synaptic and cognitive impairment in aging.Finally,we show evidence and argue about the presence of phosphorylated tau PHF-1 inside mitochondria in Alzheimer’s disease,which could be considered as an early event in the neurodegenerative process.Thus,phosphoryl

Development of an analytical method based on solid-phase extraction and LC-mS/mS for the monitoring of current-use pesticides and their metabolites in human urine

Pyrethroids,organophosphorus pesticides and fipronil have been listed as priority chemicals in human biomonitoring studies because of their wide use and potential health effects in humans.The determination of 13 pesticides,including pyrethroids(deltamethrin,cypermethrin,permethrin,cyfluthrin,bifenthrin),organophosphorus(chlorpyrifos,chlorpyrifosmethyl,and malathion),fipronil,neonicotinoids(imidacloprid,acetamiprid and thiacloprid)and triazole(prothioconazole),together with 13 corresponding metabolites in human urine samples was achieved by solid-phase extraction and analysis by liquid chromatography coupled to tandem mass spectrometry.All targeted compounds,except malathion dicarboxylic acid,were measured with a mean within-accuracy(n=5)of 71%–114%(RSD:1%–14%)and between-run(n=15)accuracy of 80%–118%(RSD:2%–14%).Limits of quantitation of the targeted analytes ranged from 0.1 to 16 pg/m L.The detection result of urine samples from 25 volunteers indicated that the detection frequencies of 3,5,6-trichloro-2-pyridinol(median:448 pg/m L),6-chloropyridine-3-carboxylic acid(median:193 pg/m L),2-methyl-3-phenylbenzoic acid(median:181 pg/m L),3-phenoxybenzoic acid(median:99 pg/m L),2-isopropyl-6-methyl-4-pyrimidinol(median:77 pg/m L),cyfluthrin(median:59 pg/m L),cis-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylic acid(cis-DCCA,median:53 pg/m L),trans-DCCA(median:25 pg/m L),prothioconazole(median:21 pg/m L),imidacloprid(median:7 pg/m L),and prothioconazole-desthio(median:1 pg/m L)were>50%.The obtained results show that the validated method is suitable for the human biomonitoring of these currentuse pesticides and their metabolites.

The genus Thelonectria(Nectriaceae,Hypocreales,Ascomycota)and closely related species with cylindrocarpon-like asexual states

The genus Thelonectria and closely related species with cylindrocarpon-like asexual states are a group of perithecial ascomycetes in the family Nectriaceae that occur as saprobes and in few cases as pathogens of hardwood trees,shrubs or other plants.Although a key component of forest ecosystems around the world,species relationships and distributions of these fungi are largely unknown.The objectives of this study were to:1)infer species rank phylogenetic relationships of the genus Thelonectria and closely related species with cylindrocarpon-like asexual states and test the monophyly of each of the groups studied;2)delimit taxa establishing taxon circumscriptions;3)resolve nomenclatural issues by identifying redundantly used names and synonyms;and 4)provide an updated outline to the genus,geographical distributions data and identification tools,specifically diagnostic keys and molecular data that can be used as molecular barcodes.The recovered consensus phylogeny resulted in a narrow circumscription of the genus Thelonectria,based on the type T.discophora,excluding one of the common species T.jungneri.According to the phylogenetic analyses,T.jungneri belongs in a segregate clade that should be recognized as a different genus.In the genus Thelonectria,a total of four new species and three new combinations are recognized.Additionally,three new genera,closely related to Thelonectria,are described to accommodate species displaying a morphological resemblance to those of Thelonectria:Cinnamomeonectria gen.***.with C.cinnamomea as type species,macronectria gen.***.with m.jungneri as type species and including four additional newly described species,and Tumenectria gen.***.with T.laetidisca as type species.

Diversity patterns, environmental drivers and changes in vegetation composition in dry inter-Andean valleys

Aims We studied diversity,patterns of endemism and turnover of veg-etation composition in dry inter-Andean valleys(DIAVs)where lit-tle is known about the influence of the abiotic drivers controlling plant species composition and occurrences,and the life forms that contribute most toα-andβ-diversity,respectively.This study was focused on DIAVs located in the highlands(800-2800 m)around the Equator(1°N-5°S).The following questions were addressed:(i)what differences exist between endemic and non-endemic species in terms of species number,frequency and abundance?(ii)are pat-terns ofα-andβ-diversity correlated with latitude?(iii)what are the major environmental drivers controlling spatial patterns in species composition and occurrence?methods We established 63 transects of 5×100 m in areas with DIAV vegetation,impacted as little as possible by human activities.In each transect,all mature trees and shrubs were identified and counted to determine their density.The coverage of terrestrial herbs was estimated.Generalized additive models were used to quantify the relationship betweenα-andβ-diversity with latitude.To recordα-diversity,we used the exponential Shannon index.The Sørensen index was used to measureβ-diversity or species turnover.We used canonical correspondence analysis to deter-mine species composition and generalized linear mixed models to quantify simultaneously the determinants of species occur-rence across species and sites.The models were evaluated using the Akaike information criterion.All analyses were run separately for trees,herbs and shrubs.Important Findings Overall,we found that shrubs was the most diverse life form,constituting 43%(12%endemic)of the 313 species,followed by herbs(36%;11.5%endemic)and trees(20%;5%endemic).Trees comprise mostly species with low abundance(e.g.mber of abundant species(>30 individuals ha^(−1)),includ-ing four endemic species.most of the endemic species were locally rare,and most of the

A wide spectrum of thoracic tumors have known or suspected viral etiologies.Oncogenic viruses can be classified by the type of genomic material they contain.Neoplastic conditions found to have viral etiologies include post-transplant lymphoproliferative disease,lymphoid granulomatosis,Kaposi's sarcoma,Castleman's disease, recurrent respiratory papillomatosis,lung cancer,malignant mesothelioma,leukemia and lymphomas.Viruses involved in these conditions include Epstein-Barr virus, human herpes virus 8,human papillomavirus,Simian virus 40,human immunodeficiency virus,and Human T-lymphotropic virus.Imaging findings,epidemiology and mechanism of transmission for these diseases are reviewed in detail to gain a more thorough appreciation of disease pathophysiology for the chest radiologist.

Red pigment production by Penicillium purpurogenum GH2 is influenced by pH and temperature

The combined effects of pH and temperature on red pigment production and fungal morphology were evaluated in a submerged culture of Penicillium purpurogenum GH2, using Czapek-Dox media with D-xylose as a carbon source. An experimental design with a factorial fix was used: three pH values (5, 7, and 9) and two temperature levels (24 and 34 °C) were evaluated. The highest production of red pigment (2.46 g/L) was reached with a pH value of 5 and a temperature of 24 °C. Biomass and red pigment production were not directly associated. This study demon- strates that P. purpurogenum GH2 produces a pigment of potential interest to the food industry. It also shows the feasibility of producing and obtaining natural water-soluble pigments for potential use in food industries. A strong combined effect (pmperature was associated with maximal red pigment production (2.46 g/L).

Post-transcriptional gene silencing, transcriptional gene silencing and human immunodeficiency virus

While human immunodeficiency virus 1(HIV-1) infectionis controlled through continuous, life-long use of a combination of drugs targeting different steps of the virus cycle, HIV-1 is never completely eradicated from the body. Despite decades of research there is still no effective vaccine to prevent HIV-1 infection. Therefore, the possibility of an RNA interference(RNAi)-based cure has become an increasingly explored approach. Endogenous gene expression is controlled at both, transcriptional and post-transcriptional levels by noncoding RNAs, which act through diverse molecular mechanisms including RNAi. RNAi has the potential to control the turning on/off of specific genes through transcriptional gene silencing(TGS), as well as finetuning their expression through post-transcriptional gene silencing(PTGS). In this review we will describe in detail the canonical RNAi pathways for PTGS and TGS, the relationship of TGS with other silencing mechanisms and will discuss a variety of approaches developed to suppress HIV-1 via manipulation of RNAi. We will briefly compare RNAi strategies against other approaches developed to target the virus, highlighting their potential to overcome the major obstacle to finding a cure, which is the specific targeting of the HIV-1 reservoir within latently infected cells.

Specimens regarded as Thelonectria discophora(Thelonectria,Nectriaceae,Hypocreales)constitute a conspicuous group of saprobic fungi on decaying plant material,characterized by red perithecia each with a broad mammiform(nipple-like)apex.The asexual state is characterized by a cylindrocarpon-like morphology,with 3–5 septate macroconidia,unicellular microconidia and chlamydospores that are rarely produced in culture.In the past,T.discophora was regarded as one species with a wide geographic distribution.However,a recent study rejected the monophyly and cosmopolitan distribution of this species,and showed the existence of at least 16 cryptic species distributed in three main groups.By combining the results of phylogenetic analyses of six nuclear loci and morphological studies,we revised the taxonomy of the T.discophora species complex,resulting in the description of 12 new species and four new combinations based on historic names.Even though molecular phylogenetic analyses strongly support the segregation of these species,and are in agreement with previous studies,individual diagnostic morphological characters for each species could not be identified.However,discrete morphological traits corresponding to each of the three main groups of species were discovered.Lineages could be differentiated based on the average values of morphological traits as well as the presence/absence of characteristic asexual propagules and colony growth at 30C.Descriptions,illustrations are provided for the recognized species.

At present, Argentina does not count with a production of indigenous yeast strains with suitable technological and oenological features to be used in the regional winery industry. Isolation and molecular characterization of these microorganisms and its fermentation attributes would be relevant to the sustainable development of the activity in the country and to recognize and preserve the biodiversity of the region. Eight strains isolated from grapes and musts from the North Patagonian region and genetically identified as Saccharomyces cerevisiae, were studied for their fermentation behavior, emphasizing in hexose transport through the plasma membrane, which is the limiting step of the process. Thus, sugar consumption profiles were analyzed in different media at laboratory scale, to be subsequently applied to the fermentation of natural musts. Three of the eight initial strains were selected, named NNm10, NIF8 and NmN16 according to their fermentation profiles. The expression of hexose transporters during fermentations revealed interesting differences in the response of each strain to sugar consumption, where transporters HXT2 and HXT5 showed significant changes in expression in Patagonian strains, which are normally associated to endurance to culture stress conditions. The results obtained by combining the characteristics studied, at molecular and physiological level, are extremely encouraging. Native strain NmN16, showed a high potential for application in local winemaking. Assays carried out on a pilot scale will determine the feasibility of applying this strain with promising technological features at industrial scale.
